Description
本出願は、2004年9月9日出願の米国特許仮出願第60/608,151号の特典および優先権を主張し、前記出願の内容を参照により本出願に組み込む。
本発明は、一般的に新規なペプチド、および特に薬剤設計のためのペプチドまたはタンパク質の骨格として有用な新規ペプチドに関する。
本明細書中における先行して発行された文献の掲載および考察は、必ずしも、その文献が現況技術の一部または一般的共通の知識であるとの認識と受け取るべきではない。これにより、掲載した全ての文献は参照により本明細書に参照により組み込む。
本研究の発見は、α−コノトキシンImIの折り畳みを起こす鍵となる構造決定基の同定に関する。
1つの態様において、本発明は、1つまたは複数の配座誘導アミノ酸を、例えば欠失または置換により除去することによってタンパク質の配座を変更する方法を提供する。

ペプチド合成:
ペプチド変異体はABI Pioneer Model 433A Peptide Synthesizerで、Fmoc法を用いて固相ペプチド合成により合成した。アミノ酸残基はN−[(ジメチルアミノ)−1H−1,2,3−トリアゾロ[4,5−b]ピリジン−1−イルメチレン]−N−メチルメタンアミニウムヘキサフルオロホスフェートN−オキシド/N,N−ジイソプロピルエチルアミンin situ中和法を使用して結合した。アミド化C末端を有する合成ペプチドは、Fmoc−PAL−PS支持体を使用して合成し、一方遊離カルボキシル末端を有する変異体はプレロードしたFmoc−L−Cys(Trt)−PEG−PS(ポリエチレングリコール−ポリスチレン)支持体樹脂上で構築した。配列中の4つのシステインは全て選択的脱保護のないトリフルオロ酢酸(TFA)に反応し易いトリチル基により保護した。次に合成したペプチドは、トリフルオロ酢酸:エタンジチオール:チオアニソール:水(92.5:2.5:2.5:2.5)を使用して、樹脂から開裂させて外し、同時に側鎖保護基を除去した。続いて粗ペプチドを逆相HPLCによって精製した(図1)。精製した還元ImIコノトキシンはSynpep社(カリフォルニア州Dublin所在)から特注で得た。次に精製ペプチドを分子量により確認した(図2)。精製したペプチドの空気酸化を2mM EDTAを添加したpH8.5の100mMとトリス・HCl中で実施し、空気中で48時間撹拌を継続した。
選択した所望の強制折り畳みジスルフィド結合を有するペプチド変異体を、選択的脱保護により生成させた。これには、2つのジスルフィド架橋形成において、選択した特定のシステイン対形成を生じさせるような、4つのシステイン残基の直交する側鎖保護が必要になる。第1および第2ジスルフィド架橋の生成に関連するシステイン対は、それぞれS−トリチルおよびS−アセトアミドメチル保護基を使用して保護した。開裂ステップ中に除去されるS−トリチル基は、空気中0.1M重炭酸アンモニウム(pH8.5)中において0.1mg/mlの濃度で48時間撹拌することにより、第1ジスルフィド結合を生成させる。
空気酸化により得られた主要なイソ型の同定
各変異体において優勢なイソ型の配座を同定するために、種々のペプチド変異体に対する強制折り畳み配座異性体の保持時間を、対応する空気酸化クロマトグラフプロファイルと比較した(図3)。使用した折り畳み条件で天然のα−コノトキシンの折り畳みのバイアスが維持されたことを確認するために、ImIコノトキシンの空気酸化物を対照として使用した。合成ImIコノトキシン、ImI酸変異体、P6Kアミド変異体、およびP6K酸変異体のクロマトグラフプロファイリングから、ImIコノトキシンのみが優勢なイソ型として球状配座を有する折り畳みバイアスを維持していて、一方他の3つの変異体はリボン状配座に変化したことが見て取れた(図2B、表3)。
次いで各変異体に対する空気酸化の検討で主として生成したイソ型を、Bruker 300MHz分光器で分析して、1次元NMRスペクトルを得た。次いで1−D NMRスペクトルを、選択的脱保護により得た種々の可能な配座のスペクトルと比較した。1−D NMR分析から得た結論は、HPLCを使用して得た配座のデータと一致する。
α−コノトキシンImIおよびχ/λ−コノトキシンの配列の分析から、合成変異体の設計の基礎となる構造上の相違が明らかになった。
実施例1 宿主配列としての使用
前記配列は剛直な構造フレームワークとして使用することができ、そのフレームワークに生物活性ペプチド配列の短いセグメントを挿入することができる。その結果、この挿入されたセグメントは、所望の活性を獲得するように構造骨格により決定される配座を使用することができる。本発明者らはよく研究されたトリペプチド配列(Arg−Gly−Asp)をコノトキシンのフレームワークに挿入することにより、配列を試験した。その結果、RGD−コノトキシンのキメラペプチドは、本発明者らがトリペプチド配列に期待する抗血小板活性を発揮する。

Claims (11)
- 式Iの配列を含むペプチドを修飾してCys3に対して2位置N末端側にプロリン残基を導入するか、またはCys3に対して2位置N末端側であるプロリン残基を除去することを含む、ペプチドの配座を変更する方法であって、
式Iは−Cys1−Cys2−Xm−Cys3−Xn−Cys4−であり;
Cys1、Cys2、Cys3およびCys4は、Cys1とCys3との間およびCys2とCys4との間、Cys1とCys2との間およびCys3とCys4との間あるいはCys1とCys4との間およびCys2とCys3との間で、一緒になって2つのジスルフィド結合を形成するシステイン残基であり;
Xは任意のアミノ酸であり;および
mとnとは同じかまたは異なり、各々1に等しいかまたは1を超える
方法。 - ペプチドがカルボキシ基またはアミド基のいずれかであるC末端基を有する請求項1に記載の方法であって、C末端基をカルボキシ基またはアミド基の他方に変換することをさらに含む方法。
- 式Iの配列とカルボキシ基またはアミド基のいずれかであるC末端基とを含むペプチドを修飾してC末端基をカルボキシ基またはアミド基の他方に変換することを含む、ペプチドの配座を変更する方法であって、
式Iは−Cys1−Cys2−Xm−Cys3−Xn−Cys4−であり;
Cys1、Cys2、Cys3およびCys4は、Cys1とCys3との間およびCys2とCys4との間、Cys1とCys2との間およびCys3とCys4との間あるいはCys1とCys4との間およびCys2とCys3との間で、一緒になって2つのジスルフィド結合を形成するシステイン残基であり;
Xは任意のアミノ酸であり;および
mとnとは同じかまたは異なり、各々1に等しいかまたは1を超える
方法。 - プロリン残基をCys3に対して2位置N末端側に導入するか、またはCys3に対して2位置N末端側にあるプロリン残基を除去することをさらに含む、請求項3に記載の方法。
- 式Iにより定義されるコノトキシンのコンセンサス配列を含み、Xmにより規定される領域が如何なる野生型のコノトキシン配列中の対応領域とも異なるように、Cys2とCys3の間に挿入もしくは置換された1つもしくは複数のアミノ酸残基を有するか、またはXnにより規定される領域が如何なる野生型のコノトキシン配列中の対応領域とも異なるように、Cys3とCys4の間に挿入もしくは置換された1つもしくは複数のアミノ酸残基を有するペプチドであって、
式Iは−Cys1−Cys2−Xm−Cys3−Xn−Cys4−であり;
Cys1、Cys2、Cys3およびCys4は、Cys1とCys3との間およびCys2とCys4との間、Cys1とCys2との間およびCys3とCys4との間あるいはCys1とCys4との間およびCys2とCys3との間で、一緒になって2つのジスルフィド結合を形成するシステイン残基であり;
Xは任意のアミノ酸であり;および
mとnとは同じかまたは異なり、各々1に等しいかまたは1を超える

- Cys3に対して2位置N末端側にプロリン残基を有さず、C末端カルボキシ基を有し、リボン状配座をとる傾向を有する請求項5に記載のペプチド。
- アミノ酸配列RGDがCys2とCys3との間またはCys3とCys4との間に挿入された請求項5に記載のペプチド。
- アミノ酸配列RGDWがCys2とCys3との間またはCys3とCys4との間に挿入された請求項5に記載のペプチド。
- 配列番号2、3、4、6、7または8のいずれか1つで表される配列を含むペプチド。
- 配列番号2、3、4、6、7または8のいずれか1つで表される配列からなるペプチド。
