Abstract
Description
The present invention has a microreactor that immobilizes a biochemical substance and measures the weight of a specific substance such as an enzyme, an antibody, a protein, a hormone, a sugar chain, or a compound that specifically adsorbs or binds thereto, and the microreactor. It relates to a microreactor system.
In recent years, the analysis of the human genome (human genetic information) has been completed, and the elucidation of the relationship between abnormal proteins that produce abnormal gene structures and diseases is being promoted. Along with this, the new drug development method has changed from an existing method that relies on the developer's experience and intuition for drugs and compounds to a method that searches for compounds that act directly on abnormal proteins and makes them new drugs. . By adopting this method, it is expected that the period of new drug development that has been necessary for nearly 20 years will be shortened to about 5 years in the future.
In the search for a new drug candidate compound, the physical binding amount between the abnormal protein and the new drug candidate compound is generally used as an index. As a method for measuring this amount of binding, previously, a compound in which a labeling substance such as an enzyme, a luminescent substance, or a radioisotope was bound was used. After further binding this compound and an abnormal protein, the amount of the labeling substance was determined. The bound compound was quantified by measuring. However, at present, a method of performing measurement without using a labeling substance has attracted attention.
As an example of this measurement method, a measurement method using a reactor will be briefly described. First, a reactor is a device in which an introduction path and a waste liquid path are formed in a chip formed of a semiconductor, glass, resin or the like, and a reaction tank is provided therebetween. In the reaction tank, a sensor having an abnormal protein immobilized thereon is installed in advance.
In the reactor configured as described above, when the sample liquid to be measured containing the compound is poured from the introduction path by a pump or the like, the compound in the sample liquid to be measured reacts with the abnormal protein of the sensor previously installed in the reaction tank, Waste liquid after the reaction is discharged from the waste liquid path. A substance fixed in advance to the sensor is called a ligand, and a substance supplied as a solution is called an analyte.
That is, in the reaction tank, some of the analytes in the sample liquid to be measured are fixed to the sensor by binding to the ligand. Here, when the reaction between the analyte and the ligand reaches an equilibrium state (that is, the amount of the analyte immobilized on the sensor becomes equal to the amount of the analyte separated from the sensor), the analyte immobilized on the ligand The amount is constant. This amount is the data required for new drug development.
By the way, the following are known as techniques relating to such a reactor. That is, this is a technique for measuring the viscosity of a sample in contact with the surface of the piezoelectric vibrator and the minute weight attached to the surface of the piezoelectric vibrator by using the vibration of a piezoelectric vibrator (particularly a quartz crystal vibrator) as a sensor. More specifically, when an AC voltage is applied between the electrodes formed on both surfaces of the piezoelectric vibrator, that is, the detection electrode and the counter electrode, resonance occurs at a specific frequency determined from the material and shape of the piezoelectric vibrator. And if a substance adheres to a detection electrode, the resonant frequency of the whole piezoelectric vibrator will change according to the attached weight. This technique is to measure the weight of the substance attached to the detection electrode by measuring this change.
However, since this method cannot detect a specific substance, a technique for detecting only a specific substance by fixing a means for adsorbing or capturing only the specific substance at a predetermined position is used. As an example, a technique using an antigen-antibody reaction for protein detection is known (see Patent Document 1). By applying such technology to sensors, it becomes possible to measure the minute weight of a specific substance to be measured, and as described above, the amount of the analyte immobilized on the ligand can be measured with high accuracy. Is possible.
When measurement is performed using a microreactor, the measurement is started by flowing a liquid into the reaction vessel. By the way, this reaction tank is created by packing bonded between a chip substrate on which an inlet and an outlet are formed and a sensor. This packing is made of silicon resin or the like, and is formed in a ring shape so as to surround the periphery of the detection electrode of the sensor. However, if the area where the packing contacts the sensor is large, vibration is suppressed and the impedance becomes high. As a result, the oscillation characteristics are deteriorated, and the reliability of the measurement results is lowered. In some cases, it is difficult to perform the measurement itself.
Further, when the packing width is narrowed in order to reduce the contact area of the packing, the liquid flowing into the reaction tank may leak out, and the original function as packing may not be performed.
Further, if the contact area of the packing is made as close as possible to the detection electrode, the inclination of the phase angle at the time of oscillation becomes small, and the inclination becomes gentle rather than steep. As a result, the phase rise when the phase angle is 0 (deg) is deteriorated, and the oscillation characteristics are also deteriorated.
The present invention has been made in consideration of such circumstances, and the object thereof is to reduce the impedance and steepen the inclination of the phase angle to improve the oscillation characteristics, so that measurement can be performed reliably and with high accuracy. It is to provide a microreactor that can be performed and a microreactor system having the microreactor.
In order to achieve the above object, the present invention provides the following means.
A microreactor according to the present invention is a microreactor for measuring the weight of a specific substance contained in a liquid, and includes a substrate having a liquid inlet for introducing the liquid and a liquid outlet for discharging the liquid; A sensor that is bonded to the surface of the substrate with a packing interposed therebetween and resonates the piezoelectric substrate at a predetermined frequency by a pair of electrodes provided on both surfaces of the piezoelectric substrate, wherein the packing is one of the pair of electrodes. A ring-shaped packing that forms a reaction tank between the liquid inlet and the liquid outlet for storing the liquid surrounding the electrode and adsorbing or binding the specific substance to the one electrode. The inner diameter D1 is sized to be 0.25 mm or more away from the one electrode, and the packing width W between the inner diameter D1 and the outer diameter D2 is 0.3 mm to 0.7 mm. The one in which the features.
In the microreactor according to the present invention, the sensor is activated in advance before starting the measurement. That is, a predetermined voltage is applied between the pair of electrodes to resonate the piezoelectric substrate. Next, the liquid is supplied to the liquid introduction path through the liquid introduction port. Then, the supplied liquid flows into the reaction tank from the liquid introduction path, fills the reaction tank, and then is discharged to the outside through the liquid discharge path. In particular, when the reaction vessel is filled with a liquid, one electrode is immersed in the liquid. Therefore, a specific substance (for example, an antigen or the like) contained in the liquid is captured by adsorbing or binding to one electrode.
Therefore, the weight of one electrode increases by the weight of the specific substance attached to the adsorption film. Then, along with this weight change, the resonance frequency of the sensor changes. Therefore, the weight of the specific substance can be measured by detecting this frequency change. As a result, various biochemical reactions such as an antigen-antibody reaction, a DNA hybridization reaction, a protein binding reaction, and an enzyme reaction can be measured.
In particular, the ring-shaped packing that creates a reaction tank between the substrate and the sensor is sized such that the inner diameter D1 is 0.25 mm or more away from one electrode. Therefore, the packing can be separated from one electrode, and the inclination of the phase angle during oscillation can be made steep. Therefore, the phase rising characteristic when the phase angle is 0 (deg) can be improved, and the oscillation characteristic can be improved.
Moreover, since the packing width W is suppressed within the range of 0.3 mm to 0.7 mm while improving the oscillation characteristics, the contact area with the sensor can be suppressed as much as possible. Therefore, it is possible to suppress the vibration of the sensor from being suppressed as much as possible, and to reduce the impedance. In this respect as well, the oscillation characteristics can be improved.
Furthermore, since the packing width W is secured at least 0.3 mm, there is no possibility that the liquid introduced into the reaction tank leaks. Therefore, it can surely fulfill the role as packing. Thus, according to the microreactor according to the present invention, the impedance can be lowered and the oscillation characteristics can be improved, so that reliable and highly accurate measurement can be performed. In addition, liquid leakage can be reliably prevented.
The microreactor according to the present invention is characterized in that, in the microreactor of the present invention, the one electrode is formed with a diameter of 3 mm.
In the microreactor according to the present invention, since one of the electrodes is an electrode having a diameter of 3 mm, the impedance can be suppressed to 1 KΩ or less and oscillation can be stably performed in the liquid. Therefore, the measurement can be performed more accurately.
The microreactor according to the present invention is characterized in that, in the microreactor according to the present invention, the packing has the inner diameter D1 of 3.9 mm and the packing width W of 0.4 mm. It is.
In the microreactor according to the present invention, the packing size is such that the inner diameter D1 is 3.9 mm, the packing width W is 0.4 mm, and the outer diameter D2 is 4.7 mm. In general, it is known that the larger the inner diameter D1 of the packing, the better the oscillation characteristics, and the larger the outer diameter D2 of the packing, the worse the oscillation characteristics. That is, in order to improve the oscillation characteristics, it is necessary to make the outer diameter D2 as small as possible while making the inner diameter D1 as large as possible.
Here, since the packing is formed with the above-described size, the balance between the inner diameter D1 and the outer diameter D2 is very excellent, and the oscillation characteristics can be further improved while minimizing the impedance. Therefore, measurement can be performed with higher accuracy. In addition, since the inner diameter D1 is 3.9 mm, the volume in the reaction layer can be made as small as possible (1 μl or less). Therefore, efficient measurement can be performed with a small amount of liquid without wastefully consuming the liquid.
The microreactor of the present invention is the microreactor of the present invention described above, wherein the packing has a thickness T of 10 μm to 80 μm.
In the microreactor according to the present invention, since the packing thickness T is as thin as 10 μm to 80 μm, the liquid in the reaction tank is easily affected by the surface tension of the packing. Therefore, even if air remains or is mixed in the reaction layer at the start of measurement or during measurement, it is easy to eliminate the air by flowing it out of the reaction tank together with the liquid by capillary action due to surface tension. Therefore, measurement can be performed without being affected by residual air, and reliable and highly accurate measurement can be performed.
In addition, the effect mentioned above becomes so remarkable that the thickness T of packing approaches 10 micrometers which is the minimum value. Further, when the thickness T of the packing is made thinner than 10 μm, the flow path resistance becomes too large and it becomes difficult to feed the liquid, and therefore the thickness T is preferably 10 μm or more.
Further, the microreactor system of the present invention is connected to the microreactor of any of the present invention, a measuring means for measuring a frequency change of the sensor, the liquid inlet and the liquid outlet, and And a liquid feeding means for feeding liquid.
In the microreactor system according to the present invention, various liquids can be fed at an arbitrary timing and at an arbitrary flow rate by the liquid feeding means while measuring the frequency change of the sensor by the measuring means. Measurement can be performed reliably and with high accuracy.
According to the microreactor and the microreactor system according to the present invention, measurement can be performed using a sensor having low impedance and improved oscillation characteristics, so that reliable and highly accurate measurement can be performed.

As shown in FIG. 5, the packing 23 configured in this manner has the center of the
This bonding method is not particularly limited to one method. For example, the bonding is performed by siloxane bonds in which silicon and oxygen are alternately bonded. In the case of performing this joining, first, the packing 23 is superimposed on a predetermined position of the

As shown in FIG. 1, the measuring means 3 includes a frequency variable

Similarly, the
Next, the case where the antigen-antibody reaction is measured by measuring the weight of the antigen attached to the antibody by the
Before starting measurement, the
Next, the
By the way, when the
Particularly, in the
Moreover, since the packing width W is suppressed to 0.4 mm, which is in the range of 0.3 mm to 0.7 mm, while improving the oscillation characteristics, the contact area with the
Further, since the packing width W is 0.4 mm, there is no possibility that the sample liquid F introduced into the
Furthermore, since the thickness T of the packing 23 is 80 μm, the sample solution F stored in the
As described above, according to the
In addition, in this embodiment, since the
In particular, the packing 23 of the present embodiment has an inner diameter D1 of 3.9 mm, a packing width W of 0.4 mm, and an outer diameter D2 of 4.7 mm. In general, it is known that as the inner diameter D1 of the packing 23 is increased, the oscillation characteristics are improved, and as the outer diameter D2 of the packing 23 is increased, the oscillation characteristics are deteriorated. That is, in order to improve the oscillation characteristics, it is necessary to make the outer diameter D2 as small as possible while making the inner diameter D1 as large as possible. In this regard, since the packing 23 of the present embodiment is formed in the above-described size, the balance between the inner diameter D1 and the outer diameter D2 is very excellent, the impedance is low, and the oscillation characteristics are extremely excellent.
Further, since the inner diameter D1 is 3.9 mm, the volume in the
In the above embodiment, after introducing the sample liquid F into the
The technical scope of the present invention is not limited to the above embodiment, and various modifications can be made without departing from the spirit of the present invention.
For example, in the above-described embodiment, the antigen-antibody reaction has been described as an example. However, the present invention is not limited to this, and by appropriately selecting an analyte and a ligand, a DNA hybridization reaction, a protein binding reaction, It can also be applied to various biochemical reactions such as enzyme binding reactions.
In the above embodiment, the inner diameter D1 of the packing 23 is set to 3.9 mm, the outer diameter D2 is set to 4, 7 mm, and the packing width W is set to 0.4 mm. You can design freely.
That is, as long as the inner diameter D1 is 0.25 mm or more away from the
Moreover, in the said embodiment, although the thickness of the packing 23 was 80 micrometers, it is not limited to this thickness, You may change freely. In particular, the thickness T of the packing 23 is preferably in the range of 10 μm to 80 μm. By doing so, even if air remains or mixes in the
In addition, it is more preferable that the thickness T of the packing 23 is closer to 10 μm because the above-described effect can be remarkably exhibited. In addition, an effect that the volume in the

(Example)
Next, based on the above embodiment, the following description is based on the data actually measured how the impedance changes when the inner diameter D1, outer diameter D2 and packing width W of the packing 23 are appropriately changed. To do. The measurement was performed under the following conditions.
First, the thickness T of the packing 23 was set to 80 μm. The
FIG. 13 and FIG. 14 show the results of impedance measured when the inner diameter D1 is changed in three stages while maintaining the packing width W at 0.3 mm under this condition. FIG. 13 summarizes the numerical values of the inner diameter D1, the outer diameter D2, and the packing width W and the measured impedance values, and FIG. 14 is a graph plotting the numerical values of the inner diameter D1 and the impedance values. It has become.
As shown in FIGS. 13 and 14, it was confirmed that the larger the inner diameter D1 of the packing 23, the smaller the impedance and the better the oscillation characteristics.
Next, FIG. 15 and FIG. 16 show the impedance results measured when the outer diameter D2 is changed in two stages while keeping the inner diameter D1 constant. The inner diameter D1 was set to 3.5 mm, 3.8 mm, and 4.0 mm, and the outer diameter D2 was changed in two steps in each case. FIG. 15 summarizes the numerical values of the inner diameter D1, the outer diameter D2, and the packing width W and the measured impedance values, and FIG. 16 plots the outer diameter D2 values and the impedance values. It is a graph.
As shown in FIGS. 15 and 16, it was confirmed that the larger the outer diameter D2 of the packing 23, the higher the impedance and the worse the oscillation characteristics.
Finally, FIG. 17 and FIG. 18 show the impedance results measured when the packing width W is changed in four stages while the outer diameter D2 is maintained at 5.2 mm. FIG. 17 summarizes the numerical values of the inner diameter D1, the outer diameter D2, and the packing width W and the measured impedance values, and FIG. 18 plots the numerical values of the packing width W and the impedance values. It is a graph.
As shown in FIGS. 17 and 18, it was confirmed that the larger the packing width W, the higher the impedance, and the worse the oscillation characteristics. This is because the larger the packing width W is, the larger the area in contact with the
From the results described above, the larger the inner diameter D1 of the packing 23 is, the better the oscillation characteristics are. On the other hand, the tendency that the larger the outer diameter D2 of the packing 23 is, the worse the oscillation characteristics can be confirmed. . In addition, when the outer diameter D2 was constant, it was confirmed that the oscillation characteristics deteriorated as the packing width W increased. However, in this case, as the packing width W is increased, the inner diameter D1 is gradually decreased. Therefore, in addition to the influence of the packing width W, the influence of impedance due to the reduction of the
