Abstract
Description
The present invention relates to an inspection apparatus for inspecting the amount and interaction of a target substance in a liquid specimen.
In recent years, with increasing awareness of health issues, environmental issues, and safety issues, detection methods for biological or chemical substances involved in these issues have been desired. These substances are mainly contained in living body-derived liquid specimens, and understanding the content of these substances and their interactions with other biomolecules is important for measures against health problems and environmental problems. In order to solve this problem, a detection method using surface plasmon resonance has been put into practical use. In the detection method using surface plasmon resonance, since it is not necessary to add a label for detection, the reaction process can be monitored sequentially, not only the content of the substance but also the interaction between the substance and the biomolecule. There is an advantage that it is possible to simultaneously measure the strength of.
In order to apply a detection method using surface plasmon resonance to a clinical test for medical use, it is required to efficiently process a large amount of specimens generated in a medical field. However, with the proposed method disclosed in Patent Document 1, although simplification of the apparatus is achieved, accurate alignment is required for each detection, and continuous processing of multiple specimen tests or multiple target substance tests is not possible. It was impossible.
On the other hand, in the method disclosed in Patent Document 2, although the alignment for each detection is improved, in addition to the parallel movement position control for angle control, the vertical accuracy, that is, the height accuracy when the element is fixed The point that is necessary was left as a problem.
One of the objects of the present invention is to reduce the required accuracy of alignment of the detection element while maintaining the simplicity of the optical configuration. And the irradiation angle of the said detection light to the said sensor site | part is changed by moving the said detection element by the said moving means, and changing the incident position of the said detection light to the said deflection | deviation means, It is characterized by the above-mentioned. .

It is characterized by.
According to the inspection apparatus of the present invention, it is possible to reduce the required accuracy of alignment of the detection element while maintaining the simplicity of the optical configuration. In addition, it is possible to inspect a plurality of detection elements continuously. Furthermore, with regard to the monitoring of the reaction process, which is a feature of the plasmon resonance inspection, it is possible to capture the change in the reaction process over time only by passing through the inspection region a plurality of times.
The inspection apparatus of the present invention is an inspection apparatus that inspects a liquid specimen using plasmon resonance.

(Detection element)
Although the detection element is not a component of the inspection apparatus of the present invention, it plays an important role when the inspection apparatus of the present invention is used, and will be described here.
The
In the
In FIG. 2,
Next, the
Therefore, the detection light (parallel light) 204 orthogonal to the
Next, the

EXAMPLES Hereinafter, although an Example is given and this invention is demonstrated further more concretely, these Examples do not limit the scope of the present invention at all.
<Device configuration>
A schematic configuration of the inspection apparatus of this example is shown in FIG.

<Detection element>
In the inspection apparatus of this example, the

In this example, the substrate is manufactured using K-PSFn2 optical glass made by Sumita Optical Glass, which has a refractive index of 1.97 with respect to light having a wavelength of 780 nm. The
Furthermore, as shown in FIGS. 6A and 6B, the
A sputtering apparatus was used to form a gold thin film on the
Here, a method for immobilizing a capture molecule will be described with reference to FIG. FIG. 5 shows the detection region when the reaction at the time of detection is completed. It is necessary to immobilize a solid phase antibody (capturer) 503 in the detection region. An ethanol solution of 11-mercaptodecanoic acid having a thiol group having a high affinity for gold is dropped on the gold thin film to modify the surface of the fine particles. In that state, an aqueous solution of N-hydroxysulfuccinimide and an aqueous solution of 1-Ethyl-3- [3-dimethylamino] propyl] carbohydrate hydrate are added and incubated at room temperature for 15 minutes. In this example, N-Hydroxysulfuccinimid and 1-Ethyl-3- [3-dimethylamino] propyyl] carbohydrate hydride manufactured by Dojindo Laboratories were used. Thus, the succinimide group is exposed on the gold pattern surface. In this state, an anti-human CRP-mouse monoclonal antibody (manufactured by Biogenesis) solution that binds to human C-reactive protein is dropped onto the gold thin film pattern region and incubated. Then, an anti-human CRP-mouse monoclonal antibody is immobilized on the gold thin film. If necessary after the fixation, treatment for suppressing nonspecific adsorption reaction may be performed using a reagent having an inhibitory effect on nonspecific adsorption such as bovine serum albumin. The detection element can be manufactured by the above operation.
<光学系>
<Optical system>
The configuration of the optical system provided in the inspection apparatus of this example will be further described with reference to FIGS. Light emitted from the

<Operation during measurement>
Next, the measurement operation of the inspection apparatus of this example will be described with reference to FIG.
Next, the measurement procedure will be described. When the user inputs a sample setting instruction using the

The configuration of the optical system provided in the inspection apparatus of this example will be described with reference to FIGS. However, the description of the same configuration as that shown in FIG.
