発明の分野
本発明は細胞性免疫に関する。
樹状細胞(「DC」)は、免疫系における効果的な抗原提示細胞(「APC」)である。DCはT細胞活性化および増殖に必要な信号すべてを与えることが判っている。これらの信号は2つのタイプに分類することができる。免疫反応に対する特異性を与える第1のタイプは、T細胞受容体/CD3(「TCR/CD3」)複合体と、APC表面にある主要組織適合性遺伝子複合体(「MHC」)のクラスIまたはクラスIIタンパク質により提示される抗原ペプチドとの相互作用によって伝達される。この相互作用は、T細胞活性化を起こさせるために必要ではあるがこれだけでは十分でない。実際、第2のタイプの信号がなくても、第1のタイプの信号はT細胞アネルギーを起こすことができる。補助的刺激(costimulatory signal)信号と呼ばれる第2タイプの信号は、抗原特異性でもなくまたMHC拘束性でもないが、T細胞の完全増殖反応をもたらし、第1タイプの信号の存在下T細胞エフェクター機能を誘導することができる。

本発明は、免疫系を刺激するための組成物を特徴とする。これらの組成物はそれぞれ複数の細胞を含有しており、少なくともその半数(例えば、70〜80%より多い)は融合細胞であり、その融合細胞はそれぞれ少なくとも1つの哺乳類樹状細胞(例えば、骨髄培養液または末梢血細胞培養液由来のDC)と、細胞表面抗原(例えば、癌抗原)を発現する少なくとも1つの哺乳類非樹状細胞(例えば、癌細胞やトランスフェクトされた細胞)との融合によって生成される。「癌抗原」とは、その癌に罹患している個体の正常細胞とは対照的に、主としてまたは完全に癌細胞により発現される抗原分子を意味する。組成物中の融合細胞のうち少なくとも半数(例えば、少なくとも70%、好ましくは少なくとも80%)は、免疫系を刺激する(例えば、T細胞を活性化する)のに有効な量において、MHCクラスII分子、B7および細胞表面抗原を発現する。「B7」とは、補助的刺激分子のB7族の任意のメンバーを意味する(例えば、B7-1またはB7-2)。
融合細胞の生成に使用される親細胞は、単一の個体(例えば、ヒト、マウスまたはラット)から得ることができる。この親細胞はまた適合もしくは非適合MHC分子を用いて、同一の種(例えば、ホモ・サピエンス)の異なる個体から得ることもできる。
本発明のその他の特徴および利点は、以下の図面、詳細な説明および請求の範囲から明らかとなると思われる。
本発明は、(1)DCと非樹状細胞との融合によって得られる融合細胞を含有する免疫系を刺激する組成物、(2)その組成物で免疫系を刺激する方法、および(3)融合細胞を生成する方法を特徴とする。
MHCクラスII分子、B7、またはその他所望のT細胞刺激分子を発現する融合細胞は、これらの分子に対する抗体でパンニングするかまたは蛍光標示式細胞ソーティングによっても選択することができる。
以下の実施例により、本発明の組成物および方法を例示的に示す、これらに限定されるものではない。
材料および方法
細胞培養および融合
マウス(C57BL/6)MC38腺癌細胞をDF3/MUC1 cDNAにより安定的にトランスフェクトしてMC38/MUC1細胞株を作った(Siddiqui et al., Proc. Natl. Acad. Sci. USA 85: 2320-2323, 1988; Akagi et al., J. Immunother. 20: 38-47, 1997)。10%熱不活化胎児ウシ血清(「FCS」)、2 mMグルタミン、100 U/mlペニシリン、および100μg/mlストレプトマイシンを添加したDMEM培地に、MC38、MC38/MUC1および同系MB49膀胱癌細胞を維持した。
Inabaらに記載の方法(J. Exp. Med. 176: 1693-1702, 1992)の変法を用いて、骨髄培養液からDCを得た。要約すれば、骨髄を長骨からフラッシュし、赤血球を塩化アンモニウムで溶解した。以下のモノクローナル抗体(「mAb」)とインキュベートし、続いて補体と共にインキュベートすることによって、骨髄細胞から、リンパ球、顆粒球およびIa+細胞を放出させた:
(1) 2.43、抗CD8 [TIB 210; アメリカンタイプカルチャーコレクション(ATCC), Rockville, MD];
(2) GK1.5、抗CD4 (TIB 207, ATCC);
(3) RA3-3A1/6.1、抗B220/CD45R (TIB 146, ATCC);
(4) B21-2、抗Ia (TIB 229, ATCC);および
(5) RB6-8C5、抗Gr-1 (Pharmingen, San Diego, CA)。
5%熱不活化FCS、50μM 2-メルカプトエタノール、1 mM HEPES (pH 7.4)、2 mMグルタミン、10 U/mlペニシリン、10μg/mlストレプトマイシンおよび500 U/mlリコンビナントマウスGM-CSF (Boehringer Mannheim, Indiana)を添加したRPMI 1640培地中、非溶解細胞を6ウェル培養プレートに播種した。培養7日目、付着していない細胞とゆるく付着している細胞を回収し、再び100 mmのペトリ皿(106細胞/ml; 8 ml/皿)に再び播種した。30分間培養した後、非付着性細胞を洗い流し、付着細胞にGM-CSF含有RPMI培地を加えた。MC38/MUC1細胞もしくはMC38と融合させるため、培養18時間後に非付着性細胞集団を除いた。
細胞をPBSで洗浄し、氷上で30分間、mAb DF3 (抗MUC1)、mAb M1/42/3.9.8(抗MHCクラスI)、mAb M5/114(抗MHCクラスII)、mAb 16-10A1(抗B7-1)、mAb GL1(抗B7-2)およびMAb 3E2(抗ICAM-1)とインキュベートした。PBSで洗浄後、フルオレセインイソチオシアネート(FITC)結合抗ハムスター、抗ラットおよび抗マウスIgGを加えて氷上でさらに30分間放置した。その後試料を洗浄・固定してFACSCAN (Becton Dickinson, Mount View, CA)で分析した。
細胞傷害性T細胞(「CTL」)活性は、乳酸デヒドロゲナーゼ(「LDH」)(CytoTox, Promega, Madison, WI)の放出により測定した。
DC、MC38/MUC1およびFC/MUC1細胞をイオン照射(30 Gy)し、96ウェル平底培養プレートで5日間1 x 105の同系または同種T細胞に加えた。T細胞は、脾細胞懸濁液をナイロンウールに通して残留APCを除いて作り、100ミリの組織培養皿で90分間平板培養した。付着していない細胞における[3H]チミジンの取り込みはウェルあたり1μCiパルス後6時間目に測定した(GBq/mmol; Du Pont-New England Nuclear, Wilmington, DE)。反応はそれぞれ3回行った。
mAb GK1.5(抗CD4)、mAb 2.43(抗CD8)またはラットIgGを1日おきにマウスに静脈内、腹腔内の両方による投与を行った。投与は、FC/MUC1による2回にわたる免疫のうち1回目の4日前からMC38/MUC1細胞によるチャレンジの4日前まで行った。フローサイトメトリーおよびCTL活性分析を行うため、脾細胞を回収した。
マウスMC38腺癌細胞を骨髄由来のDCと融合させた。融合が成功したことを示すため、まずDF3/MUC1腫瘍関連抗原を安定的に発現するMC38細胞を使用した(Siddiqui et al., Proc. Natl. Acad. Sci. USA 75: 5132-5136, 1978)。融合細胞(FC/MUC1)は、MHCクラスIおよびII、B7-1、B7-2およびICAM-1と同様、DF3/MUC1を発現した(図1A)。

白血球泳動(leukophoresis)により得られたバフィーコート(またはロイコパック)の白血球をフィコール遠心分離によって分画した。(末梢血)単核細胞を含むフラクションを、10%胎児ウシ血清(「FCS」)添加RPMI 1640培地を入れたフラスコ中37℃で30分間インキュベーションした。付着していない細胞を静かに洗い流した(これら非付着細胞の一部はDCでもあった)。これらのDCを回収するため、該細胞を、20% FCS添加RPMI 1640培地で1時間30分インキュベーションした。浮遊細胞は除去した。残った付着細胞を20% FCS添加RPMI 1640培地で2〜3日間インキュベーションした。ゆるやかに付着している細胞はDCであった。残留付着細胞を10% 胎児ウシ血清添加RPMI 1640培地で一晩インキュベーションした。次に、ゆるく付着している細胞を回収し、GM-CSF(1000 U/ml)およびIL-4 (100 U/ml)含有培地中106細胞/mlの濃度で5〜6日間培養した。得られた細胞は融合実験に用いたDCであった。
フローサイトメトリーに示すように、DC/MY5細胞は親細胞の表現型特徴を持っていた。DC/MY5は、HLA-DR、CD38(骨髄腫細胞表面マーカー)、DF3(腫瘍細胞表面マーカー)、CD83(DC細胞表面マーカー)、B7-1およびB7-2のmAbによる染色に対して陽性であった。MLRアッセイから、これらの融合細胞はT細胞の強力な刺激剤でもあることが示された。
材料および方法
MUC1トランスジェニックマウス
Rowseら(Cancer Res. 58: 315-321, 1998)に記載の通り、ヒトMUC1に対してトランスジェニックなC57B1/6マウス株を樹立した。ヌクレオチド745〜765およびヌクレオチド1086〜1065に相当するMUC1プライマーを用いて、テイルDNA 500 ngをそれぞれPCRにより増幅し、MUC1配列の存在を確認した。PCR生成物は1%アガロースゲル中の電気泳動により検出した(前記Rowse et al.)。
マウス(C57B1/6)のMC38、MB49癌細胞をMUC1 cDNAにより安定にトランスフェクトした(Siddiqui et al., Proc. Natl. Acad. Sci. USA 85: 2320-2323, 1988; Akagi et al., J. Immunotherapy 20: 38-47, 1997; Chen et al., J. Immunol. 159: 351-359, 1997)。細胞は、10%熱不活化FCS、2 mM L-グルタミン、100 U/mlペニシリン、および100μg/mlストレプトマイシンを添加したDMEM培地中に維持した。骨髄培養液からDCを得、前記実施例Iに記載の通り、癌細胞と融合させた。
10%熱不活化FCS、50μMβ-メルカプトエタノール、2 mM L-グルタミン、100 U/mlペニシリン、および100μg/mlストレプトマイシンを添加したRPMI培地に、脾臓およびリンパ節の単一細胞調製物を懸濁した。細胞は、5 U/mlの精製MUC1抗原で刺激した(Sekine et al., J. Immunol. 135: 3610-3616, 1985)。培養1日後、3日後および5日後に、ウェルあたり1μCiの[3H]チミジンで12時間細胞のパルスを行い、半自動細胞ハーベスターによりフィルター上に細胞を回収した。液体シンチレーションによって放射活性を定量した。
5 U/ml MUC1抗原を含有する完全RPMI培地にリンパ節細胞(「LNC」)を懸濁した。培養5日後、10 U/mlのマウスIL-2を添加した。10日目と15日目に、細胞は5 U/ml MUC1抗原およびAPCとして1:5照射(30 Gy)同系脾細胞による再刺激を行った。フィコール遠心分離によって死細胞を除去し、ナイロンウールに通して残留APCを除去した後、T細胞培養液を分析した。このT細胞を、CD3e (145-2C11)、CD4 (H129, 19)、CD8 (53-6.7)、αβTcR (H57-597)およびγδTcR (UC7-13D5) (PharMingen)に対するFITC結合抗体で染色した。氷上で1時間インキュベーション後、細胞を洗浄・固定してFACSCAN (Becton-Dickinson)により分析した。
標準51Cr放出アッセイにより、インビトロにおける細胞傷害性を測定した。簡単に述べると、37℃で60分間、細胞を51Crで標識した後、取り込まれなかった放射性同位元素を洗い流した。96ウェルV型プレートのウェルに標的細胞(1 x 104)を加え、エフェクター細胞と共に37℃で5時間インキュベーションした。ガンマカウンターで上清の51Crを測定した。51Crの自然放出は、エフェクターの非存在下、標的細胞をインキュベーションすることによって測定した。一方、51Crの最大または総放出量は、標的細胞を0.1% Triton-X-100でインキュベーションすることにより測定した。特異的51Cr放出のパーセンテージは、以下の式によって求めた:
%特異的放出 = [(実験値−自然放出値)/(最大値−自然放出値)]x 100
1ウェルあたり5 Uの精製MUC1抗原により4℃で一晩、マイクロタイタープレートを被覆した。ウェルを5%ウマ血清アルブミン含有PBSで洗浄した後、マウス血清の4倍希釈液で1時間インキュベーションした。洗浄してホースラディッシュペルオキシダーゼ結合ヤギ抗マウスIgG(Amersham Life Sciences)とインキュベーション後、o-フェニレンジアミン(Sigma)で展開し、OD 490 nmにおいてELISAマイクロプレートオートリーダーEL310で測定することにより、抗体複合体を検出した。
新しく取り出した組織を液体窒素中で凍結した。クリオスタットで幅5μmの組織切片を調製し、アセトン中で10分間固定した。次いで、該切片を、室温に30分間、モノクローナル抗体DF3 (抗MUC1)、抗CD4 (H129, 19)または抗CD8 (53-6.7)とインキュベーションした。その後、VECTASTAIN ABCキット(Vector Laboratories)を用いて、間接イムノペルオキシダーゼ染色を行った。
実施例1に示した通り、DCとMC38/MUC1癌細胞(FC/MUC1)との融合により得られたワクチンは、強力な抗腫瘍免疫を誘導する。MUC1トランスジェニックマウスに対するFC/MUC1ワクチン接種効果を調べるため、マウスを2回5 x 105 FC/MUC1で免疫した。対照として、106照射MC38/MUC1細胞またはPBSによる免疫も行った。106 MC38またはMC38/MUC1細胞によるチャレンジ後に、照射MC38/MUC1細胞またはPBSで免疫したマウスはすべて腫瘍を発生した。これに対し、FC/MUC1で免疫したマウスでは腫瘍成長は観察されなかった。MUC1トランスジェニックマウスのFC/MUC1免疫は、関連のないMB49膀胱癌の成長には何ら影響を与えなかった(Chen et al., J. Immunol. 159: 351-359, 1997)。しかしながら、MUC1 (MB49/MUC1)を発現するMB49細胞はFC/MUC1免疫マウスでは成長しなかった。
詳細な説明と共に本発明を説明してきたが、上記説明は例示にとどまるものであって、添付の請求の範囲に定義されている本発明の範囲を限定するものではないことを理解すべきである。その他の局面、利点および変更も以下の請求の範囲内である。
Claims (32)
- 免疫系を刺激する組成物であって、該組成物が複数の細胞からなり、少なくともその半数が融合細胞であり、該融合細胞がそれぞれ少なくとも1つの哺乳類樹状細胞と、細胞表面抗原を発現する少なくとも1つの哺乳類非樹状細胞との融合によって生成され、融合細胞のうち少なくとも半数は免疫系を刺激するのに有効な量の(a)MHCクラスII分子、(b)B7、および(c)細胞表面抗原を発現する組成物。
- 哺乳類非樹状細胞が癌細胞である、請求項1記載の組成物。
- 哺乳類樹状細胞および哺乳類非樹状細胞が同一の個体から得られる、請求項1記載の組成物。
- 前記個体がヒトである、請求項3記載の組成物。
- 細胞表面抗原が癌抗原である、請求項4記載の組成物。
- 哺乳類樹状細胞および哺乳類非樹状細胞が同一種の異なる個体から得られる、請求項1記載の組成物。
- 前記の種がホモサピエンス(Homo sapiens)である、請求項6記載の組成物。
- 細胞表面抗原が癌細胞抗原である、請求項7記載の組成物。
- 下記の段階を含む、免疫系を刺激するのに有用な融合細胞を製造する方法:
少なくとも1つの哺乳類樹状細胞と、細胞表面抗原を発現する少なくとも1つの哺乳類非樹状細胞との融合によって生成される第一の融合細胞を提供する段階、および
第一の融合細胞を少なくとも1つの哺乳類樹状細胞と融合させて免疫系を刺激するのに有用な第二の融合細胞を生成させる段階。 - 第二の融合細胞が(i)MHCクラスII分子、(ii)B7、および(iii)細胞表面抗原を発現する、請求項9記載の方法。
- 哺乳類樹状細胞および哺乳類非樹状細胞がすべてヒト細胞である、請求項9記載の方法。
- 細胞表面抗原が癌抗原である、請求項11記載の方法。
- 下記の段階を含む、融合細胞を製造する方法:
(i)第一の複数の哺乳類樹状細胞と、(ii)細胞表面抗原を発現する複数の哺乳類非樹状細胞とを含む細胞試料を提供する段階、
該細胞試料を融合剤と接触させて少なくとも一つの樹状細胞と少なくとも一つの非樹状細胞との融合生成物である融合細胞を含む融合後細胞集団を生成させる段階、
培地中で該融合後細胞集団を培養して培養融合細胞を含む培養細胞集団を生成させる段階、ならびに
培養融合細胞と融合していない細胞との付着性の差に基づき、培養細胞集団中の培養融合細胞を非融合細胞から分離して単離融合細胞を製造する段階。 - 培地がヒポキサンチン、アミノプテリン、およびチミジンを含有する、請求項13記載の方法。
- 単離融合細胞が免疫系を刺激するのに有用である、請求項13記載の方法。
- 単離融合細胞が(a)MHCクラスII分子、(b)B7、および(c)細胞表面抗原を発現する、請求項15記載の方法。
- 哺乳類樹状細胞が(i)骨髄細胞または(ii)末梢血細胞から培養される、請求項13記載の方法。
- 接触段階と分離段階との間の時間が10日間より少ない、請求項17記載の方法。
- 単離融合細胞を少なくとも1つの哺乳類樹状細胞と融合させて第二の融合細胞を製造する段階をさらに含む、請求項13記載の方法。
- 第二の融合細胞が(i)MHCクラスII分子、(ii)B7、および(iii)細胞表面抗原を発現する、請求項19記載の方法。
- 哺乳類樹状細胞および哺乳類非樹状細胞がすべてヒト細胞である、請求項20記載の方法。
- 細胞表面抗原が癌抗原である、請求項21記載の方法。
- 個体の免疫系を刺激する方法であって、該個体に請求項1記載の組成物を投与することを含む方法。
- 個体が以下からなる群より選択される状態を有する、請求項23記載の方法:
細胞内病原体による感染に対する感受性;
細胞内病原体による感染;
癌;および
癌発症前の疾病素質。 - ヒトの免疫系を刺激する方法であって、ヒトに請求項4記載の組成物を投与することを含む方法。
- 哺乳類樹状細胞がヒトまたはヒトの一卵性双生児(identical twin)から得られる、請求項25記載の方法。
- 非樹状細胞がヒトから得られる癌細胞である、請求項26記載の方法。
- 細胞表面抗原が癌抗原である、請求項26記載の方法。
- 細胞表面抗原が病原体由来の抗原である、請求項26記載の方法。
- 病原体がウイルスである、請求項29記載の方法。
- 癌抗原がMUC1である、請求項28記載の方法。
- 個体が以下の状態または以下の状態の一つを生じる疾病素質を有する、請求項31記載の方法:乳癌、子宮癌、膵臓癌、前立腺癌、肺癌、および骨髄腫。
