ポリ−L−オルニチンコーティングした35mm皿に細胞を 1 dishあたり10000個撒き、1〜3日培養した。培養後、培地を細胞外溶液 (97mM N−メチル−D−グルカミン(NMDG)、10mM BaCl2、10mM HEPES、40mMテトラエチルアンモニウム(TEA)−Cl、5.6mMグルコース(pH7.4))に交換し、下記の条件でランプパルスあるいはステップパルス刺激を行い、カルシウムチャネル電位を測定してTタイプのカルシウムチャネルおよびHVA(L、N、P/Q、R−タイプのカルシウムチャネル)の発現について検討した。なお、ランプおよびステップパルスの条件は、下記のとおり。
・ランプパルス:保持電位を−90mVとし、1000msの三角波で−120から70mVで刺激した。
・ステップパルス:保持電位を−90mVとし、200msの長さで電気刺激を与えた。結果を図3〜7に示す。それらは、電機生理学的指標からNaHSが、神経突起伸長を促すことを示す。
