つぎに、本発明の実施の形態2にかかる自動合焦装置および顕微鏡について説明する。上述した実施の形態1では、本発明にかかる自動合焦装置および顕微鏡は、標本Sの表面部に透明膜が形成された場合に、その透明膜に起因して生じる膜合焦誤差を補正するものとして説明したが、本実施の形態2では、標本Sの表面部に反射防止膜等、計測光の反射を抑制する膜が形成された場合について説明する。

設定条件記憶テーブル56aは、例えば図12に示すように、対物レンズごとに標本Sの各種類に対して計測光の波長と色収差補正レンズ9の配置位置とを対応付けて記憶している。特にここでは、標本Sの種類として、標本Sの表面部に形成される反射防止膜の各種類に対し、その反射防止膜が反射防止対象とする波長以外の計測光の波長を対応付けて記憶している。また、一般に反射防止膜は薄膜であるため、色収差補正レンズ9の配置位置として、主に色合焦誤差を補正するための配置位置が対応付けられて記憶されている。

これによって、自動合焦装置103では、対物レンズによらず、標本Sの種類、つまり標本Sの表面部に形成された反射防止膜の種類に応じて、合焦誤差を確実に補正することができる。また、自動合焦装置103では、反射防止膜によって計測光が減衰されることがなく、標本検出光学系に残存する色収差によって標本Sの検出感度が低下することもないため、常に高感度に標本Sの位置を検出し、正確に合焦を行うことができる。

なお、自動合焦装置103では、自動合焦装置100と同様に、計測光の光源および波長を2つに限定する必要はなく、3つ以上とすることができる。また、自動合焦装置101と同様に、色収差補正レンズ9を固定配置し、計測光の波長切換のみで合焦誤差を補正することもできる。さらに、自動合焦装置103と同様に、光源4A,4Bに替えて広帯域光を発する光源44を備えるとともに波長選択フィルタDF等を備え、単色光もしくは狭帯域光を計測光として択一的に抽出することもできる。また、計測光の光源として、複数の異なる波長の光を選択的に射出可能なチューナブルレーザ等を用いてもよい。

波長が異なる複数の計測光のうち1つを択一的に射出させる光源手段を有し、前記対物レンズを介して前記計測光を前記標本に投射するとともに、該標本が反射した前記計測光を受光して前記標本の位置を検出する標本検出手段と、
あらかじめ前記標本の種類ごとに前記計測光の波長を対応付けて記憶する記憶テーブルと、
前記標本の種類を示す標本情報を取得する情報取得手段と、
前記光源手段によって、前記標本情報が示す前記標本の種類に対応付けられた波長の前記計測光を射出させるとともに、前記標本検出手段の検出結果をもとに、前記焦準手段によって前記相対距離を変化させ、前記標本を前記対物レンズの焦点面に合焦させる制御を行う制御手段と、
