Abstract
Description
本発明は、低酸素誘導性因子(HIF)オリゴヌクレオチドデコイ分子、およびHIF関連の疾患もしくは病的状態の処置におけるHIFオリゴヌクレオチドデコイ分子の使用に関する。
低酸素誘導性因子(HIF)は、組織内の酸素添加の変化に対する適応応答を媒介する、ヘテロダイマーの転写因子である。3種のHIFのサブタイプが現在公知であり(HIF−1、HIF−2、HIF−3);HIF−1およびHIF−2は、保存されるHREを介して遺伝子調節に影響することが示されている。HIF−1は、構成的に発現されるHIF−1βサブユニットおよび高度に調節されるHIF−1αサブユニットからなるヘテロダイマーである。HIF−1αの合成は酸素非依存性であり;しかし、分解は主に酸素依存性の機構を通して調節される。活性化されたHIF−1αサブユニットは、核内へ移動し、ARNT(アリールレセプター核内トランスロケーター)サブユニットと二量体化して、活性な転写因子HIF−1を形成する。HIF−1は、多くの遺伝子のエンハンサーもしくはプロモーター中に存在する低酸素応答性エレメント(hypoxia−response element)(HRE、もしくは5’−ACGTG−3’(配列番号126))を認識し、HREの発現をもたらす。

一実施形態において、本発明のHIF dsODN分子は、ワトソン−クリック型塩基対形成によって互いに結合する二本のオリゴヌクレオチド鎖からなる。代表的にはこの二本の鎖の全てのヌクレオチドが塩基対形成に参加するが、このことは必要条件ではない。一つ以上(例えば1〜3個または1個もしくは2個)のヌクレオチドが塩基対形成に関係していないオリゴヌクレオチドデコイ分子もまた、包含される。加えて、二本鎖デコイは3’および/または5’の一本鎖突出部を含み得る。
本発明のHIF dsODNデコイ分子は、市販の自動合成機を用いて、標準的なホスホジエステル化学またはホスホルアミデート化学によって合成され得る。実施例において記載される特定のdsODN分子は、自動DNA合成機(Model 380B;Applied Biosystems,Inc.,Foster City,CA)を用いて合成されている。これらのデコイは、カラムクロマトグラフィーによって精製され、凍結乾燥され、そして培養培地中に溶解された。各々のデコイの濃度は、分光測光的に決定された。
本発明のHIFデコイ分子は、ゲルシフトアッセイまたは電気泳動移動度シフト(EMSA)アッセイにおいて、最初に便宜よく試験されて特徴付けられ得る。このアッセイは転写因子のDNAへの結合を検出するための迅速かつ高感度な方法を提供する。このアッセイは、タンパク質とDNAとの複合体が、非変性ポリアクリルアミドゲルの中を、遊離の二本鎖オリゴヌクレオチドよりゆっくりと移動するという観察に基づいている。このゲルシフトアッセイは、転写因子結合部位を含む32P末端標識化DNAフラグメントと共に、精製されたタンパク質またはタンパク質の複合混合物(例えば、核抽出物)をインキュベートすることによって行われる。次いで、この反応生成物は、それから非変性ポリアクリルアミドゲルで解析される。結合部位についての転写因子の特異性は、目的のタンパク質に対する結合部位を含むオリゴヌクレオチドまたはスクランブルされたDNA配列のオリゴヌクレオチドのいずれかの過剰量を用いる競合実験によって確立される。この複合体に含まれるタンパク質の同一性は、このタンパク質を認識する抗体を用い、次いでDNA−タンパク質−抗体複合体の低下した移動度、または放射性標識オリゴヌクレオチドプローブへのこの複合体の結合の分断のいずれかを探すことによって確立される。
先に議論されるように、HIF−1は、新脈管形成および解糖ならびに転移を含む腫瘍の成長において重要な役割を果たすことが示されており、抗癌治療戦略の潜在的な標的として同定されている(Semenza,Nature Rev.3:721−732(2003);Williamsら、Oncogene 21:282−90(2002);Griffithsら、Cancer Res.62:688−95(2002);Welshら、Mol.Cancer Ther.2:235−43(2003))。HIF−1は、乳癌において過剰発現されること、および、さらに浸潤性の腫瘍と潜在的に関連することが示されている(Bosら、J.Natl.Cancer Inst.93:309−314(2001))。加えて、HIF−1は、炎症と腫瘍形成との間の重要な結合として、最近同定されている(Jungら、The FASEB Journal Express Article 10.1096/fj.03−0329fjc、2003年9月4日オンライン公開)。脳腫瘍、乳癌、子宮頚部癌、食道癌、口腔咽頭癌、および卵巣癌の生検におけるHIF−1αの過剰発現は、処置の不全および死亡率と相関する。増加したHIF−1活性は、腫瘍の進行を促進し、HIF(例えば、HIF−1および/またはHIF−2)の阻害は、癌治療への新規のアプローチを表し得る。従って、HIF−1および/またはHIF−2を本発明のデコイ分子によって遮断することは、癌の予防および処置における有用性を見出し、単独でまたは他の処置オプションとの併用で、新しい抗癌戦略を提供する。本発明のdsODN分子を投与することによるHIF−1および/またはHIF−2の阻害はまた、他の癌治療(例えば、放射線治療および/または化学療法薬を用いる処置)の効力を増強し得る。特定の癌標的としては、固形腫瘍の悪性疾患および非ホジキンリンパ腫が挙げられるが、これらに限定されない。
本発明のHIFデコイの送達の一つの可能な様式は、例えば米国特許第5,922,687号および同第6,395,550号(これらの開示の全体が本明細書によって特に参考として援用される)において記載されるような、圧力媒介トランスフェクションである。簡単に述べると、HIFデコイ分子は、このデコイ核酸を細胞の細胞外環境に置き、そして細胞の周囲および細胞外環境でインキュベーション圧を確立することによって、組織内の細胞へ送達される。このインキュベーション圧の確立は、細胞による核酸の取り込みを促進し、細胞核への局在化を高める。
(HIFデコイ分子の設計および試験)
(設計)
初めに、HIF−1結合DNAの共通配列を、HIF−1に関係するDNA−タンパク質相互作用についての刊行物より選択し、BioBase TRANSFAC(バージョン7.2およびバージョン8.2)データベースにおいて要約される、公開された配列のセットから選んだ。これらの共通配列が対応する調節領域の局在を確認し、伸長された隣接ゲノムDNA配列を、最新のゲノムデータベースより検索した(表1を参照)(ヒトに関して、2003年7月バージョン;マウスに関して、2003年2月バージョン;ラットに関して、2003年6月バージョン)。
HIFのコア結合部位付近の塩基組成を定義するために、上記の利用可能なHIF−1コア結合配列に基づくコア結合部位を、計算的にアラインメントした。そのアラインメントに基づいて、このコアおよび直接隣接する領域の両方についての塩基組成を計算的に記載する、表もしくは「マトリックス」を作成した(図1を参照)。この解析を、TRANSFACデータベースの最新のバージョン(8.2、2004年6月)を用いて行った(例えば、Heinemeyerら、Nucleic Acids Res.27:318−22(1999);Knuppelら、J.Comput.Biol.1:191−8(1994);Matysら、Nucleic Acids Res.31:374−8(2003);Schachererら、Bioinformatics 17:1053−7(2001)を参照)。TRANSFACは、DNA−TF結合についての位置−重量−マトリックスを集積する。Matchツール(Kelら、Nucleic Acids Res.31:3576−3579(2003))は、結合親和力を計算的に予測するために、これらのマトリックスを用いる。図1は、所定の塩基が所定の位置に見出される確率を統計学的に示唆する。
HIF−1は、基本的なヘリックス−ループ−ヘリックス(bHLH)DNA結合タンパク質のファミリーに属する。(HIF−1aサブユニットについての合計826中の)位置30から位置70までに位置するアミノ酸は、DNA認識およびDNA結合親和力に関与する。HIF−1に関するDNA結合モチーフの結晶構造は存在しないが、同様のDNA結合モチーフを共有する他のbHLHメンバーの利用可能な構造の情報が、有用な構造の情報を提供する(Michelら、Theor Chem Acc.101:51−56(1999);Michelら、J.Biomolecular Structure & Dynamics 18:169−179(2000);Michelら、Biochemica et Biophysica Acta 1578:73−83(2002))。これらの研究は、HIF−1の結合モチーフ内に位置する数個の残基の重要性を示唆した。公知のコア結合配列は、CGTG(配列番号134)であるが、中心のコアACGTG(配列番号126)がHIF−1複合体(HIF−1αおよびARNT)の最大限の結合のために必須であること、ならびにこのコアからすぐ5’側の上流の塩基組成もまた、HIF−1結合の特異性および親和性のために非常に重要であることが見出されている。DNAフットプリント研究もまた、5’側の上流領域がHIF−1誘導性遺伝子発現のために重要であり得ることを示唆した。従って、5’側の隣接部分の配列および長さが変化する候補のデコイを設計し、試験した。
公開されたHIF−1結合の研究からの知識、利用可能なHIF−1コア結合配列からの知識、計算的なコア結合マトリックスからの知識、bHLHファミリーについての結晶構造のモデルからの知識、および(他の転写因子に結合し得るデコイを排除するための)特定の生命情報科学のアプローチからの知識に基づいて、1セットのデコイを最初のスクリーニングのために生成した(表2Aを参照)。これらのデコイは、「変異デコイ」、「スクランブル(scramble)デコイ」、5’末端もしくは3’末端で異なる長さを有するデコイ、および、コア領域でもしくはコア領域に隣接して代替的な塩基組成を有するデコイを含む。

HIF−1αを含む複合体のためのオリゴヌクレオチドの相対的な親和力を評価するために、HIF−1 TransAMアッセイ(Active Motif、カタログ番号47096)を利用した。製造者の指示書に従ってアッセイを行った。簡単に述べると、低酸素応答性エレメント(hypoxia response element)(HRE)を含む二本鎖オリゴヌクレオチドを、96ウェルのプレートに固定した。HIF−1α複合体を含む核抽出物をインキュベートし、固定されたオリゴヌクレオチドに結合させた。結合していない物質を洗い流し、結合したHIF−1αをHIF−1αを特異的に認識する抗体を用いて検出した。この抗HIF−1α抗体を、西洋ワサビペルオキシダーゼ(HRP)で標識された二次抗体によって検出し、各ウェル内のHRPの量を、比色定量法の基質反応を用いて測定し、マイクロプレート分光光度計を用いて読み取った。
異なるデコイ濃度を用いてスクリーニングを行った。各UV吸光度読み取り値について、サンプル 対 野生型コントロールの吸光度読み取り値の比を計算することによって、正規化をおこなった。その結果を表3において要約する。より大きな比は、野生型コントロールと比較した場合の、HIF−1αとの結合のより低い競合を表す。より小さな(1.0より小さいか、もしくは1.0に近い)比は、より良い結合もしくはより良い競合を表す。
(1)このデータは、最大限の結合親和性のために、コア配列(位置1〜5)は、ACGTG(配列番号126)でなければならないことを示す。位置+1に「A」を有するデコイの優れた結合親和性は、著しく新しく、予想外の知見である。
(2)別の重要な知見は、位置+1に「G」を有することは、結合親和性を著しく低下させ、従って避けられるべきであることである。
(3)位置−1に「A」を有するデコイもまた、弱められた結合を示した。
(4)位置−2にTもしくはAを有するデコイは、弱められた結合を示した。
(5)位置−3に「T」を有するデコイは、優れた結合親和性を有する。
(6)位置+5にAもしくはGを有するデコイは、増大された親和性を有する。
(7)このデータはまた、位置−4もしくは+2での塩基組成について特別な要求が存在せず、従って本明細書中のデコイ分子はこれらの位置で任意の塩基を含み得ることを、明確に示す。
(8)隣接する5’配列の比較は、GCAGまたはGGAGまたはGCATまたはCCCTまたはCCGTの塩基組成が、低い競合(例えば、野生型デコイと比べてより大きい比)をもたらし得ることを示唆する。
(9)本発明者らが比を分類するならば、より良い競合(例えば、より小さい比)を有するデコイは、大部分が位置「−4」および「−1」に各々塩基「G」および塩基「T」を共有する(図3)。より良い競合デコイのために、コア(ACGTG;配列番号126)のすぐ前の4塩基は、「GCGT」(配列番号127)のようである(図2)。図2はまた、各々、位置「−4」での「G」と位置「−1」での「G」との組合せ、同じく、位置「−3」での「A」と位置「−2」での「A」との組合せが、結合親和性に有利にはならないことを、示唆する。
HIFゲルシフトアッセイ(EMSA)を以下のように行った。共通HIF結合部位を含む二本鎖オリゴヌクレオチドを、T4ポリヌクレオチドキナーゼ(Promega)を用いてγ32P−ATPで末端標識した。LPSで刺激されたTHP−1細胞(ヒト単球細胞株)から調製された1マイクログラムの核抽出物を、競合する、標識されていないHIF二本鎖オリゴヌクレオチド(dsODN)もしくはスクランブルされたdsODNの存在下または非存在下で、35fmolの放射性標識プローブと共にインキュベートした。10mMのTris−HCl pH8、100mMのKCL、5mMのMgCl2、2mMのDTT、10%のグリセロール、0.1%のNP−40、0.025%のBSAおよび1μgのポリ−dIdCからなる20μlの反応容積中で、室温にて30分間インキュベーションを行った。この反応物を、6%のポリアクリルアミドゲルにロードし、電気泳動に供し、乾燥させた。乾燥させたゲルを、Typhoon 8600 PhosphorImager(Amersham)およびImageQuantソフトウェアを用いて、画像化し定量した。この放射性標識オリゴヌクレオチドプローブに結合された複合体中に含まれるHIFタンパク質の同一性を、HIFファミリーの各々のメンバーに特異的な個々の抗体と共に、この反応物を5分間プレインキュベートし、放射性標識プローブを添加することによって、同定した。
(結合特性への5’隣接配列の長さの効果)
以下の表4は、全てが最適のコアおよび公知の望ましい3’隣接配列を含む、一連のデコイ分子の比較を示す。これらの配列の間の主要な相違は、5’隣接配列の長さである。7個以上の塩基の5’隣接部分を有する多数の付加的なデコイもまた解析し、最適のコアおよび望ましい3’隣接部分を有する付加的なデコイの全てが、1.25またはより好ましい範囲にスコア(競合比)を有することを見出した。従って、5個以下の塩基の5’隣接部分は、一般的に、望ましいHIF結合を支持するために十分ではない。6塩基を有する3’隣接部分は望ましい結合を示し得るが、3’隣接領域において7個以上の塩基を有する配列は、一般的に、はるかに好ましい結合特性を有する。この研究の結果はまた、3’隣接配列中の位置+1ではAについての選択性が存在するが、Gはこの位置では好まれないことを、示唆する。加えて、3’隣接配列中では、より高いGC含有量についての選択性が存在する。
(結合親和性への骨格の置換の効果)
骨格のホスホジエステル結合(PO)に硫黄置換を有さない単一のデコイ配列(895/896)の結合親和性を、3’末端を始点としてホスホジエステル結合に6個までの硫黄置換を有するデコイ配列と比較する、一連の実験を行った。3’末端を始点としてホスホジエステル結合に6個までの硫黄置換を有するデコイ配列を、ハイブリッド骨格としてのH、および3’末端を始点とする置換の数で標識した。例えば、H3は、3’末端を始点として第1、第2、第3の結合位置に置換を有するハイブリッド骨格を示す。全てのホスホジエステル結合が置換されている場合、その分子をPSと称する。
(HIFデコイ分子はHIF−1α/HIF−1β複合体に結合する)
(方法)
HIF−1αゲルシフトアッセイを、以下のように行った。HIF−1αDecoy(5’CACCAGCGTACGTGCCTCAGG 3’(配列番号130))に対するHIF−1α結合部位を含む二本鎖オリゴヌクレオチド(Sigma Genosys)を、T4ポリヌクレオチドキナーゼ(Promega)を用いてγ32P−ATPで末端標識した。酸素正常状態または低酸素状態のいずれかのMiaPaCa(膵臓腫瘍細胞株)から調製された5μgの核抽出物を、HIF−1αまたはHIF−1βのいずれかに特異的な抗体の存在下または非存在下で、35fmolの放射性標識プローブと共にインキュベートした。このインキュベーションを、25mMのTris pH7.6、100mMのKCL、0.5mMのEDTA、1mMのDTT、10%のグリセロール、0.2MのPMSF、0.2Mのオルトバナジン酸ナトリウムおよび1μgのポリ−dIdC(Roche)からなる20μlの反応容積中で、室温にて30分間行った。この反応物を、5%のポリアクリルアミドゲルにロードし、電気泳動に供し、乾燥させた。乾燥させたゲルを、Typhoon 8600 PhosphorImager(Amersham)およびImageQuantソフトウェアを用いて、画像化し定量した。この放射性標識オリゴヌクレオチドプローブに結合された複合体中に含まれるHIF−1αタンパク質の同一性を、HIF−1αファミリーの各々のメンバーに特異的な個々の抗体と共に、この反応物を5分間プレインキュベートすることによって同定した後、放射性標識プローブを添加した。
低酸素状態に暴露される場合、HIFα放射性標識プローブに結合するタンパク質複合体が誘導される。図3に示すように、HIF−1αおよびHIF−1βの両方に対する抗体は、バンドをスーパーシフト(supershift)し得、これらの抗体がその標的に特異的に結合し、従ってこの複合体の運動性を低下させることを示す。このことは、このバンドがHIF−1α/HIF−1βヘテロダイマーからなることを示す。
(HIFデコイはHIFに結合してHIFを遮断するが他のTFを阻害しない)
(方法)
HIFデコイ895:896H3が、その標的であるHIF−1ならびに他の非標的のTFに結合し、従ってそれらの活性を遮断する能力を、実施例4に記載される低酸素状態で誘導された細胞由来の核抽出物を用いて、TransAMTM法プレートアッセイ(Active Motif,Carlesbad,CA 92008)によって決定した。
HIFデコイ895:896H3は、試験された4種の細胞株の全てについて、核抽出物中でのHIF結合について、固定されたEPOプロモーター結合部位と競合することが可能であった。図5に示すように、標的のTFに対するデコイは、固定された標的結合部位への結合について競合することが可能であったが、HIFデコイはこれらの非標的の転写因子のいずれの結合をも遮断することは出来なかった。
(HIFデコイはHIF−1α/HIF−1βを二つの天然のプロモーターに結合させることについて競合する)
この研究の目的は、HIF−1αデコイが、二つの天然のプロモーターであるエリスロポエチン(EPO)およびトランスフェリンレセプター由来のHIF−1α/HIF−1β複合体の結合について、競合し得ることを、ゲルシフトアッセイを用いて示すことであった。
HIF−1αゲルシフトアッセイを、以下のように行った。トランスフェリンレセプター由来のHIFα結合部位(5’CGCGAGCGTACGTGCCTCAGG 3’;配列番号131)もしくはエリスロポエチン(EPO)プロモーター(5’GCCCTACGTGCTGTCTCA 3’;配列番号132)内に含まれるHIFα結合部位を含む二本鎖オリゴヌクレオチド(Sigma Genosys)を、T4ポリヌクレオチドキナーゼ(Promega)を用いてγ32P−ATPで末端標識した。低酸素状態のSHP−77細胞(小細胞肺癌腫瘍細胞株)から調製された5μgの核抽出物を、競合する未標識HIFα二本鎖オリゴヌクレオチドデコイ(ODN)の漸増するモル量の存在下または非存在下で、35fmolの放射性標識プローブと共にインキュベートした。このインキュベーションを、25mMのTris pH7.6、100mMのKCL、0.5mMのEDTA、1mMのDTT、10%のグリセロール、0.2MのPMSF、0.2Mのオルトバナジン酸ナトリウム、および1μgのポリ−dIdC(Roche)からなる20μlの反応容積中で、室温で30分間行った。この反応物を、5%のポリアクリルアミドゲルにロードし、電気泳動に供し、乾燥させた。乾燥したゲルを、Typhoon 8600 PhosphorImager(Amersham)およびImageQuantソフトウェアを用いて、画像化し定量した。この放射性標識オリゴヌクレオチドプローブに結合された複合体中に含まれるHIFαタンパク質の同一性を、HIFαファミリーの各々のメンバーに特異的な個々の抗体と共に、この反応物を5分間プレインキュベートすることによって前もって同定した後、放射性標識プローブを添加した(データは示さず)。
図6に示すように、HIF−1αデコイは、HIFαの結合について、試験された二つの天然のプロモーターから効果的に競合することが可能であった。EPOプロモーターの場合において、HIFαデコイは、20倍のモル過剰で、HIF−1α/HIF−1β複合体の結合について効果的に競合することが可能であった(より低い濃度はこの時点においては試験されない)。トランスフェリンレセプターのプロモーターについては、HIF−1αデコイは、20倍のモル過剰で、ほとんどのHIF−1α/HIF−1β複合体の結合について、効果的に競合することが可能であった。
低酸素状態に暴露された場合に高レベルのHIF−1α転写因子を発現するように腫瘍細胞を誘導すること、およびその複合体をゲルシフトアッセイを用いて同定することが可能であった。HIF−1αデコイは、二つの天然のプロモーターであるエリスロポエチンおよびトランスフェリンレセプター由来のHIF−1α結合部位から離れてHIF−1α/HIF−1β複合体の結合について、競合することが可能であった。
(HIFデコイは転写因子Oct−1に結合しない)
HIF-1αデコイが偏在性の転写因子であるOct−1に結合しないことを示すために、電気泳動移動度シフトアッセイ(EMSA)を用いて研究を行った。
Oct−1ゲルシフトアッセイを、以下のように行った。Oct−1結合部位(5’TGTCGAATG CAAATCACTAGAA 3’;配列番号133)を含む二本鎖オリゴヌクレオチド(Promega)を、T4ポリヌクレオチドキナーゼ(Promega)を用いてγ32P−ATPで末端標識した。MiaPaCa細胞から調製された5μgの核抽出物を、競合する未標識HIF−1α二本鎖オリゴヌクレオチドデコイ(ODN)の漸増するモル量の存在下または非存在下で、35fmolの放射性標識プローブと共にインキュベートした。このインキュベーションを、10mMのTris pH8.0、100mMのKCL、5mMのMgCl2、2mMのDTT、6%のグリセロール、0.1%のNP−40、0.02%のBSAおよび1μgのポリ−dIdC(Roche)からなる20μlの反応容積中で、室温で30分間行った。この反応物を、6%のポリアクリルアミドゲルにロードし、電気泳動に供し、乾燥させた。乾燥させたゲルを、Typhoon 8600 PhosphorImager(Amersham)およびImageQuantソフトウェアを用いて、画像化し定量した。この放射性標識オリゴヌクレオチドプローブに結合された複合体中に含まれるOct−1タンパク質の同一性を、Oct−1オリゴヌクレオチドが取り除かれた結合された複合体をスクランブルされた配列に対して競合させることによって、同定した。
図7に示すように、無関係な転写因子であるOct−1のその特異的な結合部位への結合は、HIF−1αデコイによって阻害されなかった。
(HIFデコイを用いる癌細胞の処理は低酸素誘導性のHIF活性を誘導する)
(方法)
HT−29(ヒト結腸癌)、MiaPaCa2およびBxPc3(ヒト膵臓癌)、ならびにSHP−77(NSCLC)腫瘍細胞株を、ATCCから得、そして適切な培地中で5%のCo2中に維持した。HIF活性を、これらの細胞を1%のO2条件において24時間までインキュベートすることによって、または、BehroozおよびIsmail−Beigi(J.Biol.Chem.133:151−60(1997))によって報告されるように、260μMのCoCl2の培地への添加によって、誘導した。HIFデコイがこれらの細胞においてHIF活性を遮断する能力を測定するために、これらの細胞を、10分間の6psiでの加圧処理を用いて、様々な量のHIF−1デコイ895:896H3でトランスフェクトした。核抽出物を、このデコイの添加の24時間後にこれらの細胞から調製した。
ゲルシフトによって測定されるHIF−1αの活性、およびELISAによって測定される分泌されるVEGFは、全ての細胞株において低酸素状態によって増加した(図8Aおよび図8B)。
(異種移植研究におけるHIFデコイの効力)
(異種移植腫瘍モデル)
6〜8週齢のnu/nuマウスに、ヒトの腫瘍細胞株を皮下移植した。腫瘍が150mm3〜250mm3の容積に達した場合、これらの腫瘍を各々の群が同等の平均容積を有するように無作為に6〜15個の群に分け、そして、動物を、背側に挿入されるAlzet浸透ミニポンプを介した連続的な皮下送達によって、または、ボーラスな腹腔内注射もしくは静脈内注射によって、処置する。全てのデコイを、生理食塩水中に再懸濁し、研究ごとに適切なビヒクルのコントロールを含めた。
各々の実験の最後(処置開始の1〜6週間後)に、動物を麻酔下の全採血によって安楽死させ、各々の群からの腫瘍(および他の組織)を採取し、重量を計測し、そして10%の中性緩衝化ホルマリン中で固定するかもしくは液体窒素中で急速(snap)凍結した。固定された組織を、低酸素症、アポトーシス、血管(CD−31検出)、HIF−1、VEGFなどのような種々のマーカーの組織学的解析のために処理した。血清サンプルを、先に記載されるように、R&D Systems製のQuantikineキットを用いるELISAによって、mVEGFレベルおよびmEPOレベルについて解析した。
結腸癌の処置のための標準的な治療法の一つは、5−FUである。HT−29腫瘍を有するマウスに送達されるHIFデコイ895:896H3を、5−FU単独、および5−FUとの組合せと比較する研究を行った。
生理食塩水処置した4個のコントロール腫瘍、およびHIFデコイ895:896H3処置した群由来の4個の腫瘍を、固定し、切片を作成し、断片化された染色体DNAを蛍光FITC標識を用いて検出するためにTumorTACSTM(Trevigen,Inc.Gaithersburg,MD 20877)を用いてアポトーシス小体を染色した。核の対比染色を、Hoescht染色を用いて行った。SPOTデジタルカメラ(Diagnostic Inst.Inc.)を備えたZeiss Axioskop 2 Plus顕微鏡を用いて、腫瘍の中心断面から10×倍率で無作為に5枚の画像を取得することによって、この染色の画像化を行った。ImagePROソフトウェアを用いてアポトーシスの定量を行った。存在する核の数を、Hoeschtの蛍光を捕捉することにより決定し、TumorTACSTMによってもまた染色された核の数を、アポトーシス細胞の比率として見なした。HIFデコイ処置は、アポトーシス細胞の数の2.5倍の増加をもたらした(図11)。
(AvastinTMを用いる併用処置)
腫瘍の新脈管形成の阻害が、その腫瘍をより低酸素状態にし、HIFの量を増加させ、それによってHIFデコイについての治療の窓(therapeutic window)を増加させることを、仮定した。このことは、HIFデコイ895:896H3と新脈管形成因子との併用治療が、より治療的であることを含意する。この仮説を検証するために、HIFデコイ895:896H3を、2群の動物に、2種の用量(30mg/kg/日および45mg/kg/日)で、連続的な注入によって投与した。抗新脈管形成因子AvastinTM(抗VEGF抗体、Genentech,South San Francisco,CA)を、2群のマウスに0.4mg/kg/投与(低用量)および2mg/kg/投与(最大用量)の用量で毎週2回静脈内注射によって送達した。第六群においては、低用量のAvastinTMおよび低用量のHIFデコイ895:896H3の両方を用いた。この研究の結果を、図13に示す。8日間の処置の後、30mg/kg/日のデコイ処置群は、24%のTGIを示し、45mg/kg/日のデコイ処置群は、52%のTGIを示した。この用量は、低用量のAvastinTMと30mg/kg/日のデコイとを合わせた場合と同じであった。低用量のAvastinTM単独は、39%のTGIをもたらし、高用量のAvastinTM単独は、63%のTGIをもたらした。
(さらなる腫瘍モデルにおける効力の研究)
(SHP−77における効力の研究)
先の実施例に記載されるように、細胞を増殖させ、移植した。HIF1デコイ895:896H3を、2群の動物に、2種の用量(30mg/kg/日および45mg/kg/日)で、連続的な注入によって投与した。抗新脈管形成因子であるAvastinTM(抗VEGF抗体、Genentech,South San Francisco,CA)を、2群のマウスに、0.4mg/kg用量(低用量)および2mg/kg用量(最大用量)の用量で毎週2回静脈内注射によって送達した。第六群においては、低用量のAvastinTMおよび低用量のHIFデコイ895:896H3の両方を用いた。この研究の結果を、図14に示す。わずか7日間の処置の後に、45mg/kg/日のHIFデコイ895:896H3の効果は、低用量のAvastinTMを用いた場合に見られる効果と同等もしくはそれより高く、併用群は高用量のAvastinTMを用いた場合に見られる効果と同様であった。これらのマウスからの血漿を分析し、VEGFのレベルを計測した。全ての処置について、VEGFの有意な用量依存性減少が存在し、この効果は併用処置において最も高かった。
Matrigelを用いるMiaPaCa2マウスの異種移植モデルを、記載されるように確立した。動物群を28日間3種の用量(1.7mg/kg/日、5mg/kg/日、および15mg/kg/日)のHIFデコイ895:896H3で処置した一つの研究において、TGIの用量依存性の増加が存在し、このTGIの増加は、図15の左のパネルに示すように、15mg/kg/日の用量で有意(p=0.0139 Mann−Whitney)であった。
Claims (53)
- 一本のセンス鎖および一本のアンチセンス鎖を含むHIF二本鎖オリゴデオキシヌクレオチド(dsODN)分子であって、該センス鎖は、5’から3’方向に式FLANK1−CORE−FLANK2の配列を含み、
COREは配列ACGTG(配列番号126)であり、
ヌクレオチドの位置が負(−)の番号によって表されるFLANK1は、少なくとも6ヌクレオチド長であり、そして
ヌクレオチドの位置が正(+)の番号によって表されるFLANK2は、少なくとも約50%のGC含量を有し、
そしてここで、該dsODN分子は、HIFに特異的に結合し得る、
dsODN分子。 - 前記FLANK2は、位置+1にG以外のヌクレオチドを有する、請求項1に記載のdsODN分子。
- 前記FLANK2は、位置+1にヌクレオチドAを有する、請求項1に記載のdsODN分子。
