Abstract
【解決手段】βアミロイド前駆体タンパク質(APP)の全ての3つの型、APP695、APP751、およびAPP770の発現、そして、ロンドンおよびインディアナ家系性アルツハイマー病(FAD)のようなアミノ酸717位での変異およびAPP遺伝子中で予測された変異のような天然に生じる変異に基づく特徴を有するトランスジェニック動物を提供し、該トランスジェニク動物はアルツハイマー病の病理学的経過を変える化合物についてのスクリーニングに使用できる。

アルツハイマー病に対する可能な治療をテストするためのトランスジェニック動物モデルの構築が記載されている。このモデルは、アルツハイマー病を引き起こす天然に存在する条件に非常に類似することで特徴付けられる。すなわち、βアミロイド前駆体タンパク質(APP)の3つの型、APP695、APP751、およびAPP770、あるいはそのサブフラグメントのうちの1つ以上の発現を制御する能力に加えて、アミノ酸717位でのFAD変異およびAPP遺伝子中で予測された変異のような天然に生じる変異に基づいた種々の位置での変異に基づく。APP遺伝子構築物は、ヒト、マウス、またはラット起源の天然に生じるAPPプロモーターに加えて、マウスメタロチオネイン(metallothionine)プロモーターのような誘導プロモーターを使用して調製され、動物の水あるいは食餌に亜鉛のような重金属を添加することにより調節され得る。構築物のニューロン特異的発現は、ラットニューロン特異的エノラーゼプロモーターを使用して調製される。
構築物ならびにトランスジェニック動物および動物細胞が、下記の方法および材料を用いて調製される。
制限エンドヌクレアーゼは、New England Biolabs(Beverly, MA.)、Promega Biological Research Products(Madison, WI.)およびStratagene(LaJolla CA.)などの、従来の市販のソースから得られる。放射性材料は、Dupont/NENまたはAmershamのような従来の市販のソースから得られる。部位特異的変異誘起用のために注文設計されたオリゴヌクレオチドは、BioSynthesis Inc., Lewisville, TX.のような、数社の材料販売業者から入手可能である。部位特異変異誘起を行うためのキットは、Promega Biological Research Products、Stratageneなどの販売業者から入手可能である。APP mRNAのAPP695、APP751、およびAPP770型を含むcDNAクローンは、Dr. Dmitry Goldgaber、NIHから直接得た。DNAライブラリーは、Stratagene, La Jolla, CA.またはClontech, Palo Alto, CA.ような販売業者から入手可能である。PC12および3T3細胞は、ATCCから得た(それぞれ、#CRL1721および#CCL92)。さらにPC12細胞系は、Harvard Medical SchoolのDr. Charles Marotta、Massachusetts General Hospital、およびMcLean Hospitalから得た。細胞系に適した標準細胞培養培地は、Gibco/BRLのような従来の市販のソースから得られる。マウス幹細胞、株D3は、Dr. Rolf Kemlerから得られた(Doetschmanら、J. pWE15コスミドベクターにヒト胎盤DNAから構築されたコスミドライブラリーは、APP770 cDNAクローンのニックトランスレーション(Maniatisら、Molecular Cloning:a laboratory manual(Cold Spring Harbor Laboratory, Cold Spring Harbor, NY 1989))により調製された32P標識プローブとのハイブリダイゼーションによってスクリーニングした。プローブとハイブリダイズされたクローンを、取り出して精製し、制限マッピング、ハイブリダイゼーション、およびDNA塩基配列決定により特徴付けた。長い5'フランキング領域を含むこのようなクローンの1つから、約25 kbのNotIからNruIの制限DNAフラグメントを単離した。このフラグメントは、アルツハイマータンパク質コード領域の開始メチオニンコドンの2つ前のヌクレオチドで終わる。このフラグメントまたはそのサブフラグメントは、本明細書に記載の構築物のためのヒトAPPプロモーターのソースである。マウスまたはラットゲノムライブラリーから同じ方法を用いて単離された類似のDNAフラグメントは、マウスまたはラットプロモーターのソースである。
cDNAクローンAPP-695は、Kangら、Nature325:733-735(1987)に記載のcDNAの型であり、脳中のアルツハイマータンパク質の最も優勢な型を示す。cDNAクローンAPP-751は、Ponte, P, Nature 331, 525-527(1988)に記載された型である。cDNAクローンAPP-770は、Kitaguchiら、Nature331:530-532(1988)に記載された型である。この型には、695型に関連する225ヌクレチド挿入断片が含まれる。225ヌクレオチド挿入断片は、KIドメインに加えて、OX-2ドメインもコードする。
以下の表に示すように、ヒトゲノムDNAクローンのファージおよびコスミドクローンにより、アルツハイマー遺伝子の最小の大きさが本来少なくとも100 kbであることが特徴付けられた。APP遺伝子中には、合計18個のエキソンが存在する(Lemaireら、Nucl. Acid Res, 17;517-522, 1989;Yoshikaiら、1990)。これらの結果を併せて、アルツハイマー遺伝子の最小の大きさは175 kbであることが示される。
図1〜図5に示すヒトAPP遺伝子配列の種々の部分を有するクローンを、同様の方法で構築する。まず、253塩基対のBclIからBamHIのフラグメントとして、SV40ウイルス由来のポリA付加シグナル(Reddyら、Science200;494-502(1978))を、一連のpUCからの改変されたベクター内にクローン化する。次に、cDNAコード配列(770、751、または695)を挿入する。挿入されたフラグメントの正確な方向および構成を、制限エンドヌクレアーゼマッピングおよび限定的配列決定により、決定する。
種々のプロモーター配列は、APPコード配列の発現を制御するために用いられる。トランスジェニック動物におけるAPP遺伝子の発現を調節する能力は、ADにおいて種々のAPP遺伝子産物のはたらきを評価するのに有用であると考えられている。培養された細胞におけるAPP遺伝子の発現を調節する能力は、種々のAPP遺伝子産物の発現およびプロセシングを評価するのに有用であると考えられており、そして細胞培養された薬剤スクリーンの基準を提供し得る。
図7に示される構築物は、以下のように構築されている。ヒトゲノムDNAの大きなセグメントは、ある種のベクター内にクローン化される場合、酵母細胞内で自律複製単位として増殖され得る。このようなベクター産生(vector-borne)セグメントは、酵母人工染色体と呼ばれている(YAC;Burkeら、Science236, 806(1987))。ヒトYACライブラリーは、市販され入手可能で(Clontech, Palo Alto, CA)、平均挿入断片サイズ250,000塩基対(180,000塩基対〜500,000塩基対の範囲)を有している。アルツハイマーの遺伝子のYACクローンは、ヒトAPP770 cDNAを有するライブラリーをスクリーニングすることによって直接単離され得る。クローンにおける本質的な遺伝子領域のすべての含有物は、PCR解析によって確認され得る。
ヒトのアルツハイマータンパク質遺伝子とネズミのアルツハイマータンパク質遺伝子との間のヌクレオチド配列の相同性は、およそ85%である。βペプチドのコード領域内には、この2つの配列の間に3つのアミノ酸の相違がある。717位のアミノ酸で変異しているval残基は、マウス、ラット、およびヒトの間で保存される。野生型のげっ歯動物は、アルツハイマー病を起こさないか、またはヒトアルツハイマー患者において存在するものに類似のそれらのCNSにおける堆積物またはプラークを生じない。従って、βペプチドのげっ歯動物形態ではなく、ヒトの形態は、疾患を引き起こし得る。相同組換え(Capecchi, MR Science 244. 1288-1292(1989))は、インサイチューでマウスのアルツハイマー遺伝子をヒトβペプチドをコードする遺伝子に変換するのに用いられ得る。この組換えは、KIおよびOX-2ドメイン由来の下流部位、例えば、エキソン9内でなされる。その結果、トランスジェニック動物内の全ての細胞に適切な自然選択的なスプライシング機構は、最終遺伝子産物を発現するのに用いられ得る。
FAD変異部位である、タンパク質全長のアミノ酸717位での変異を有すること以外はこれらと同一の発現クローンもまた、構築される。アミノ酸717位での変異は、部位特異的変異誘発によってつくられ(Vincentら、1989)、野生型valコドンの以下のコドンの1種への変異を包含する;ile、phe、gly、tyr、leu、ala、pro、trp、met、ser、thr、asn、gln。val-717からile、phe、およびglyへの変異は、記載されている(Goateら、1991;Murrellら、1991;Chartier-harlinら、1991)。これらの天然に生じる変異は、荷電されたアミノ酸でもバルキーアミノ酸でもない。従って、val-717と列挙された他のアミノ酸との置換はまた、FAD症候群を促進し得、そして動物のADモデルに有用な特性を有し得ると考えられている。
マイクロインジェクションのためのDNAクローンを、適切な酵素(例えば、SalI、NotIなど)で切断し、そしてこのDNAフラグメントをTBE緩衝液中の1%アガロースゲルで電気泳動する(Maniatisら、1989)。DNAバンドを、エチジウムブロマイドで染色することにより可視化し、切り出し、そして0.3 Mの酢酸ナトリウム、pH7.0を入れた透析バッグ中に入れる。DNAを透析バッグ中で電気溶離し、フェノール-クロロホルム(1:1)で抽出し、そして2容量のエタノールによって沈澱させる。このDNAを低塩緩衝液(0.2 M NaCl、20 mMトリスTM、pH7.4、および1 mM EDTA)1 mlに再溶解し、そしてElutip-DTMカラムで精製する。このカラムを、まず、高塩緩衝液(1 M NaCl、20 mMトリスTM、pH7.4、および1 mM EDTA)3 mlで満たし、次いで、低塩緩衝液5 mlで洗浄する。DNA溶液をこのカラムに3回通し、DNAをカラムマトリックスに結合させる。低塩緩衝液3 mlで1回洗浄した後、このDNAを高塩緩衝液0.4 mlで溶出し、そして2容量のエタノールによって沈澱させる。DNAの濃度をUV分光光度計における260 nmでの吸光度によって測定する。マイクロインジェクションのために、DNAの濃度を、5 mMトリスTM、pH7.4および0.1 mM EDTAで3μg/mlに調整する。マイクロインジェクションのためのDNAの精製の他の方法はまた、Hoganら、Manipulating the mouse embryo(Cold Spring Harbor Laboratory, Cold Spring Harbor, NY(1986)、Palmiterら、Nature 300, 611(1982)、“The Qiagenologist. 動物のソース
トランスジェニック試験に適切な動物は、標準的な市販のソース、例えば、Charles River(Wilmington, MA)、Taconic(Germantown, NY)、Harlan Sprague Dawley(Indianapolis, IN)などから得られ得る。Swiss Webster雌マウスは、胚の回収(embryo retrieval)および胚移植に好ましい。B6D2F1雄は、交配に用いられ得、そして精管切除されたSwiss Webster種ウマは、偽妊娠を刺激するのに用いられ得る。精管切除されたマウスおよびラットは、供給業者から得られ得る。

6週齢の雌マウスに、過剰***を誘発するため、妊娠ウマ血清ゴナドトロピン(PMSG;Sigma)5 IU(0.1 cc、ip)を注入し、48時間後にヒト絨毛性ゴナドトロピン(hCG;Sigma)5 IU(0.1 cc、ip)を注入する。雌は、hCG注入後直ちに雄と共に置かれる。hCG注入から21時間後、交配した雌をCO2で窒息させるか、または頸部を脱臼させて屠殺し、そして切り出した輸卵管から胚を回収し、さらにこれを0.5%ウシ血清アルブミン(BSA;Sigma)が入ったダルベッコリン酸緩衝生理食塩水中に入れる。周辺の丘細胞をヒアルロニダーゼ(1 mg/ml)で取り除く。次いで、前核胚を洗浄し、そしてこれをCO2 5%、空気95%の加湿雰囲気下の37.5℃のインキュイベーター中で、0.5%BSA(EBSS)を含有するアール調節塩溶液(Earle's balanced salt solution)に注入時間まで入れておく。
トランスジェニックラットを生産するための手順は、マウスに対する手順と同様である(Hammerら、Cell 63;1099-112(1990))。30日齢の雌ラットにPMSG 20 IU(0.1 cc)を皮下注入し、そして48時間後に、個々の雌を試験に供した雄と共にする。同時に、40〜80日齢の雌を精管切除した雄と共にケージの中に入れる。これらは胚移植の仮親(foster mothers)を提供する。翌朝、雌の膣栓をチェックする。精管切除した雄と交配した雌は、移植のときまで別に置いておく。交配した雌のドナーを屠殺し(CO2による窒息)、そしてそれらの輸卵管を取り出し、これを0.5%BSAが入ったDPBS(ダルベッコリン酸緩衝生理食塩水)に入れ、そして胚を集める。胚周辺の丘細胞をヒアルロニダーゼ(1 mg/ml)で取り除く。次いで、胚を洗浄し、そしてこれを37.5℃のインキュベーター中で、0.5%BSAを含有するEBSS(アール調節塩溶液)にマイクロインジェクションの時間まで入れておく。
cDNAのES細胞への導入
ES細胞の培養方法およびそれに続くトランスジェニック動物の生産、種々の方法によるDNAのES細胞への導入(例えば、エレクトロポレーション、リン酸カルシウム/DNA沈降、および直接注入(direct injection))が、Teratocarcinomas and embryonic stem cells, a practical approach, E.J. Robertson編(IRL Press 1987)に詳細に記載されており、その教示は本明細書中に援用される。トランスジーン(transgene)含有ES細胞の望ましいクローンの選択は、いくつかの手法の1つにより達成される。ランダムな遺伝子組込みを含む場合には、APPクローンは、ネオマイシン耐性をコードする遺伝子と共沈する。トランスフェクションは、Lovell-Badge、Teratocarcinomas and embryonic stem cells, a practical approach, E.J. Robertson編(IRL Press 1987)またはPotterら、Proc. Natl. Acad. Sci. USA 81, 7161(1984)で詳細に記載されるいくつかの方法のいずれかにより達成される。リン酸カルシウム/DNA沈降、直接注入、およびエレクトロポレーションは好ましい方法である。これらの手順においては、0.5×106個のES細胞を組織培養皿にプレートし、そして、最終的に100μlの体積になるようにして50 mgのリポフェクチン(lipofectin)の存在下で沈澱させた、線状化したAPPクローンと1 mgのpSV2neo DNAとの混合物によりトランスフェクトした(SouthernおよびBerg、J. Mol. Appl. Gen. 1:327-341(1982))。この細胞を、10%胎仔ウシ血清のG418で補足した(200と500μg/mlとの間)DMEM溶液を含有する選択培地で培養する。G418耐性の細胞のコロニーをクローニングリングを利用して単離し、そして増大させた。薬剤耐性のクローンからDNAを抽出し、そしてAPP配列を有するこれらのクローンを同定するために用いられるAPP770 cDNAプローブを用いて、サザーンブロッティング実験を行う。いくつかの実験では、重要なクローンを同定するのにPCR法を用いる。
雄と交配した自然周期または過剰***の雌マウスが、ES細胞移植のための胚を採取するのに用いられる。この目的にマウスを用いるときは、C57株を用いることが望ましい。交配が成功して約3.5日後に適齢の胚を回収する。交配した雌をCO2で窒息させるか、または頸部を脱臼させて屠殺し、そして切開した子宮角から胚を取り出して洗浄し、そしてES細胞注入のため、ダルベッコの10%ウシ血清を加えた修正必須培地にこれを入れる。約20μmの内径を有するガラス製顕微針を用いて、約10〜20のES細胞を胚盤胞へ注入する。
不定周期の成熟雌マウスを精管切除した雄と対にする。Swiss Webster、ICRまたはその他のマウス系統がこの目的のために用いられ得る。ES細胞を含有する胚盤胞の移植に要求される、交配してから2.5〜3.5日となるように受容雌を交配させる。胚移植の際に、体重1グラムあたり2.5%アベルチンの0.015 mlを腹腔内注入して、受容雌を麻酔する。体壁から直接輸卵管への切開により卵巣を露出させ、そして卵巣および子宮を外にさらす。25ゲージ針を用いて子宮角に穴をあけ、そこから胚盤胞を移植する。移植後、卵巣および子宮を体内へ押し戻し、そして2箇所縫合して切り口を閉じる。さらに移植をするときは、反対側でこの手順を繰り返す。
3週齢の動物から尾のサンプル(1〜2 cm)を取り出す。DNAを調製し、サザーンブロットおよびPCRの両方法により分析し、トランスジェニック親(F0)動物およびそれらの子孫(F1およびF2)を検出する。
マイクロインジェクションを行ったトランスジーンを有する種々のF0、F1およびF2動物をCO2で窒息させて屠殺し、そして脳内の神経炎によるプラーク(neuritic plaques)および神経細線維のもつれ(NFT)を免疫組織学的に分析する。各トランスジェニックラインからのマウスおよびラットの脳を4%パラフォルムアルデヒド中で固定し、そしてクリオスタット上で分割する。APPおよび/またはβペプチドと反応性の抗体で切片を染色する。フルオレセイン、ローダミン、西洋ワサビペルオキシダーゼまたはアルカリ性ホスファターゼと結合した二次抗体が、一次抗体を検出するために用いられる。これらの実験は、アミロイドプラークの同定、および脳の特定の領域へそれらのプラークを局所化させることを可能にする。9〜50μmの範囲の大きさを有するプラークが、大脳皮質中においてAD患者の脳内に特徴的に生じるだけでなく、へん桃核、線条体および間脳を含むさらに奥の灰白質にも観察され得る。切片は、アルツハイマープラーク診断用の他の抗体でもまた染色され、抗原(例えば、Alz-50、tau、A2B5、神経フィラメント、ニューロン特異的エノラーゼ、およびアルツハイマープラークに特有の他の抗原)を認識する(Wolozinら、Science232, 648(1986);HardyおよびAllsop、Trends in Pharm. Acad. Sci. 82, 4245-4249;Majochaら、Can J Biochem Cell Biol 63;577-584(1985))。また、チオフラビン類およびコンゴーレッドによる染色も、神経炎によるプラークおよびNFT内のβペプチド沈着物の位置を共に確認するのに行われる。
mRNA: 酸グアニジニウムチオシアネート-フェノール/クロロホルム抽出法によりトランスジェニック動物の細胞系および細胞組織から、mRNAを単離し(ChomczynskiおよびSacchi, Anal Biochem 162, 156-159(1987))、ノーザンブロット法により発現レベルを決定する。
病理学的研究
免疫組織的手法(Immunohistology)およびチオフラビンS染色は、本明細書の別項に記載のように行われる。
学習および記憶の欠損を評価するように考案された行動試験を用いる。このような試験には、例えばMorris Water迷路(Morris、Learn Motivat.12;239-260(1981))がある。この手順によれば、水で満たした円形のプールの中に動物が入れられ、水面のすぐ下にはエスケーププラットフォームが沈められている。その動物が近位の視覚的手がかりに向かって泳いでいくことによって見つけられるように、視覚的マーカーがプラットフォームの上に置かれている。あるいは、プラットフォームの位置を示す形式的手がかりがないより複雑な形式の試験をその動物に課すこともできる。その形式の場合、動物は遠位の視覚的手がかりに関連してプラットフォームの位置を学習しなければならない。
標準的な方法論によってアルツハイマー病の治療をする時に潜在的な効果をもたらす化合物をスクリーニングするために、トランスジェニック動物と動物細胞を用いる。この化合物を一定期間の間に投与量を変化させて動物に投与するか、培地に導入する。次いで、その動物あるいは動物細胞は、上記の手順を用いてAPP発現、組織病理学、および/または行動における変化を調べた。
クローンpMTAPP-1は、図1aに示される発現ベクターの一例である。ここで使用されているプロモーターは、メタロチオネインプロモーターである。NIH3T3およびPC12細胞系(ATCC#CCL92およびCRL1721)をトランスフェクトすることによって安定な細胞系を得た。0.5×106個のNIH3T3またはPC12細胞を100 mm皿にプレートし、そして、50 mgのリポフェクチン(Gibco, BRL)の存在下で沈澱させた5 mgのSalIフラグメントおよび1 mgのpSV2neo DNA(46)の混合物を最終容積が100μlになるようにしてトランスフェクトした。PC12細胞には、ポリリジンでコートしたプレートを用いた。この細胞は通常、組織培養皿にあまり付着しないためである。DMEMまたはRPMI中10%のウシ胎児血清を含有し、G418を補給した選択培地を細胞に与えた。500 mg/ml(生物学的重量)および250 mg/mlのG418を用いて、NIH3T3およびPC12細胞由来のコロニーをそれぞれ選択した。トランスフェクションの15日後、G418に対して耐性を有する細胞のコロニーをクローニングリングで単離し、Tフラスコ内で増した。細胞内のAPP cDNAの存在は、MullisおよびFaloona, Methods Enzymol. 155;335-350(1987)の手法を用いたPCRで検出した。本明細書中に上記教示するところを援用する。
pEAPP-1は、図1Aの構築物と同様なヒトAPP770 cDNAに結合し、その発現を制御する25 kbヒトAPP遺伝子プロモーターの一例である。この構築物からのDNAを上記のようにしてPC12細胞にトランスフェクトした。pEAPP-1トランスフェクト細胞の特定のクローンは、神経成長因子(NGF)で処理されたPC12細胞によって示されるのと形態的に同様な分化表現型を示した。PC12細胞は通常、ほぼ円形および平坦な細胞である。pEAPP-1でトランスフェクトされた細胞は、神経突起に類似した細胞質伸長部を有する。PC12細胞をNGFで処理すると、神経突起伸長が非常に長く伸長する。pEAPP-1でトランスフェクトされた13個のPC12細胞クローンを選択し、増殖させた。これらの細胞クローンのうち8個が自発分化表現型を示し、クローン1-8、1-1および1-4が最も強力な表現型を示した。上記のようにβペプチドに対する抗体でpEAPP-1トランスフェクトPC12細胞を染色したところ、分化を示す細胞もAPPを発現していることが示された。APP770 cDNAは発現されるが、pMTAPP1クローンでトランスフェクトされたPC12細胞はこの表現型を示さなかったので、これらの結果により、ヒトプロモーター由来のAPP770の発現が、細胞生理学に関する新規な特徴を有することが示唆される。
pMTA4は、図4Aに示される構築物のタイプの一例である。ここで使用されているプロモーターは、メタロチオネインプロモーターである。この構築物でコードされるタンパク質は、図4に示されるタンパク質とはわずかに異なる。APP770 cDNAクローンを、56位(Kangらの番号付けシステムによる、1987年)より後を切断するAsp718で消化した。その結果得られた5'エクステンションをクレノウ酵素(Maniatis)を用いてうめた。同一のDNA調製物もまた、1795位より後を切断するEcoRIで切断し、その結果得られた5'エクステンションをクレノウ酵素(Maniatis)を用いてうめた。この分子が自己連結することにより発現クローンとなる。この発現クローンでは、このようにしてコードされた欠失タンパク質は、リーダー配列、βペプチドのphe-arg-val-gly-ser配列で開始する短縮βペプチド、そしてAPPの56末端アミノ酸をこの順で含んでいる。この構築部からのDNAを上記のようにPC12細胞にトランスフェクトした。
Claims (11)
- 組み合わせcDNA/ゲノム発現クローンに対して作動可能に連結したプロモーターを含む遺伝子構築物を含有するトランスジェニックげっ歯動物であって、
前記発現クローンはAPP770をコードするcDNA配列または天然に存在する変異を有するAPP770をコードするcDNA配列を含有し、
エキソン6およびスプライシングのために十分な領域を有する隣接する下流イントロン、KIコード領域およびOX-2コード領域およびスプライシングのために十分な領域を有するそれぞれのそれらの上流および下流イントロン、そしてエキソン9およびスプライシングのために十分な領域を有する隣接する上流イントロンからなるゲノムAPP DNA配列を、APP770をコードするcDNA配列または天然に存在する変異を有するAPP770をコードするcDNAの対応する領域に置換し、そして
前記構築物は、トランスジェニックげっ歯動物中で、転写され、そして選択的にスプライシングされて、APP695、APP751およびAPP770をコードしそしてAPP695、APP751およびAPP770に翻訳されるmRNA分子を形成する、

- 請求項3に記載のトランスジェニックげっ歯動物から培養された細胞。
- 野生型APP770タンパク質の717位でバリン残基をコードするコドンが、Ile、Phe、Gly、Tyr、Leu、Ala、Pro、Trp、Met、Ser、Thr、Asn、およびGlnからなる群から選択されるアミノ酸をコードするように変異している、請求項1に記載のトランスジェニックげっ歯動物。
- 請求項1に記載のトランスジェニックげっ歯動物であって、前記プロモーターが、そのトランスジェニックげっ歯動物の起源であるげっ歯動物と同一種のAPPプロモーターである、請求項1に記載のトランスジェニックげっ歯動物。
- 組み合わせcDNA/ゲノム発現クローンに対して作動可能に連結したプロモーターを含む遺伝子構築物を、げっ歯動物細胞またはげっ歯動物胚に導入することを含む、アルツハイマー病のトランスジェニックモデルを形成するための方法であって;
前記発現クローンはAPP770をコードするcDNA配列または天然に存在する変異を有するAPP770をコードするcDNA配列を含有し、
エキソン6およびスプライシングのために十分な領域を有する隣接する下流イントロン、KIコード領域およびOX-2コード領域およびスプライシングのために十分な領域を有するそれぞれのそれらの上流および下流イントロン、そしてエキソン9およびスプライシングのために十分な領域を有する隣接する上流イントロンからなるゲノムAPP DNA配列を、APP770をコードするcDNA配列または天然に存在する変異を有するAPP770をコードするcDNAの対応する領域に置換し、
前記構築物は、哺乳類細胞中で、転写され、そして選択的にスプライシングされて、APP695、APP751およびAPP770をコードしそしてAPP695、APP751およびAPP770に翻訳されるmRNA分子を形成する、
