Abstract
Description
The present invention relates to a gene detection method for detecting a specific gene sequence present in a sample, and particularly to a gene detection method for electrochemically detecting a gene with a labeling agent.
Conventionally, as a gene detection method for electrochemically detecting a specific gene sequence, a DNA chip in which a single-stranded nucleic acid probe having a base sequence complementary to a target gene to be detected is immobilized on an electrode surface is used. There is something. In this method, the nucleic acid probe is hybridized with a target gene sample that has been denatured into a single strand, and then specifically binds to a double-stranded nucleic acid formed with the nucleic acid probe and the target gene sample. A chemically active labeling agent is added to the reaction system of the nucleic acid probe and the gene sample, and the labeling agent bound to the double-stranded nucleic acid is detected by electrochemical measurement through the electrode of the DNA chip. Thus, the nucleic acid probe hybridized with the target gene sample is detected, and the presence of the target gene is confirmed (see, for example, Patent Document 1 and Patent Document 2).
The labeling agent used in this technique refers to a substance that recognizes the double-stranded nucleic acid and specifically binds to the double-stranded nucleic acid. Each of the labeling agents has a planar insertion group such as a phenyl group in the molecule, and the insertion group is inserted between the base pair of the double-stranded nucleic acid, thereby Join. The binding between the labeling agent and the double-stranded nucleic acid is binding by electrostatic interaction or hydrophobic interaction, and the insertion of the labeling agent between the base pairs of the double-stranded nucleic acid and between the base pairs. This is a binding by an equilibrium reaction in which the release from is repeated at a constant rate.
Furthermore, the above-mentioned labeling agent has a substance that causes an electrically reversible redox reaction. If an intercalating agent that causes such an electrochemically reversible redox reaction is used, the electrochemical change can be measured. Thus, the presence of the labeling agent bound to the double-stranded nucleic acid can be detected. Examples of the output signal of the electrochemical change include current generated during oxidation / reduction and light emission.
Therefore, in such a conventional gene detection method, it is important to specifically bind the labeling agent only to the double-stranded nucleic acid and accurately detect the amount of the labeling agent bound to the double-stranded nucleic acid. Become.
However, the labeling agent is non-specifically adsorbed on a single-stranded nucleic acid probe or the electrode surface on which the nucleic acid probe is immobilized. The non-specifically adsorbed labeling agent becomes background noise when detecting the amount of the labeling agent bound to the double-stranded nucleic acid, and causes a decrease in detection sensitivity.
One technique for solving this is a technique using magnetic beads. In this method, a capture probe immobilized on a magnetic bead, a labeled probe bound with an electrochemically active substance as a labeling agent, and a gene sample are hybridized, and the labeling agent is attached to the magnetic bead via the gene sample. A bound complex is formed. Then, magnetic beads are collected by magnetic force and washed to remove unreacted gene samples and labeled probes that have not been hybridized (B / F separation). Then, the presence of the target gene is detected by developing magnetic beads and electrolyte on the electrode, applying voltage to the electrode, and detecting the electrochemical signal from the labeled probe bound to the gene sample. (See Patent Document 3 and Patent Document 4).
This technique is a technique using so-called sandwich hybridization, and the capture probe, the gene sample, and the labeled probe are complexed through specific interaction of each component. Therefore, it is possible to improve detection sensitivity.
In the methods of Patent Document 3 and Patent Document 4 described above, the labeling agent bound to the magnetic bead surface via a gene sample exchanges electrons with the electrode, and the current generated by being oxidized or reduced It detects light emission. The above-described electron transfer between the labeling agent and the electrode is performed in a very narrow range such as a region of several nm from the electrode surface called an electric double layer.
However, the magnetic beads used in the above-mentioned method are obtained by dispersing a magnetizable substance such as iron oxide in polystyrene, which is an electrically insulating material, as represented by DYNABEADS (trade name) sold by Dynal of Norway. Since the main component is polystyrene, the magnetic beads are an insulator as a whole.
Therefore, among the labeling agents bound to the magnetic bead surface via the gene sample, only the labeling agent that contacts the electrode surface causes electron transfer with the electrode, that is, only a part of the magnetic bead surface undergoes an electrochemical reaction. There was a problem that the utilization efficiency of the labeling agent was poor without involvement.
The present invention has been made to solve the above-mentioned problems, and provides a gene detection method that improves the utilization efficiency of a labeling agent immobilized on magnetic beads and can detect a gene sample to be detected with high sensitivity. The purpose is to provide.
In order to solve the above problems, the gene detection method of the present invention is a gene detection method for detecting a gene having a specific sequence, wherein the gene to be detected is denatured into a single strand to produce a gene sample. Sample preparation step, and labeled probe preparation step of preparing a single-stranded labeled probe having a base sequence complementary to a part of the gene sequence of the gene sample, labeled with an electrochemically active substance And a single-stranded capture probe having a base sequence complementary to a part of the gene sequence of the gene sample and having a base sequence different from the labeled probe, and immobilizing the capture probe on a magnetic bead A magnetic bead preparation step, a single-stranded capture probe fixed to the magnetic bead, the labeled probe, and a gene sample to be detected are hybridized. A complex forming step for forming the complex, a washing step for collecting the complex by magnetic force, removing a non-hybridized gene sample and a labeled probe, and adding the collected magnetic beads to the conductive fine particles. And a detection step of developing the electrochemically active substance in the complex with an electrochemical solution and detecting the electrochemically active substance in the complex by electrochemical measurement.
According to the gene detection method of the present invention, during the electrochemical measurement, the electric double layer region extends to a part away from the electrode surface via the conductive particles dispersed in the electrolytic solution, and is in contact with the electrode surface. Since the electrochemical reaction occurs even in a portion of the labeling agent that is not present, the utilization efficiency of the labeling agent can be increased, and highly sensitive detection is possible.
Hereinafter, the gene detection method of the present invention will be described in detail. In addition, the gene sample in the following embodiments refers to, for example, blood, leukocytes, serum, urine, feces, semen, saliva, cultured cells, tissue cells such as various organ cells, and any sample containing other genes. The cells in the sample are destroyed to release double-stranded nucleic acids, which are dissociated into single-stranded nucleic acids by heat treatment or alkali treatment. In addition, the gene sample in this embodiment may be a nucleic acid fragment that has been cut with a restriction enzyme and purified by electrophoresis separation or the like.
(Embodiment 1)
Hereinafter, the gene detection method in Embodiment 1 is demonstrated. First, a gene sample to be tested is prepared. As described above, this gene sample destroys cells in an arbitrary sample to release double-stranded nucleic acid, and is denatured to single-stranded by heat treatment or alkali treatment.
At this time, the destruction of the cells in the sample can be performed by a conventional method, for example, by applying a physical action such as shaking or ultrasonic waves from the outside. In addition, nucleic acid can be released from cells using a nucleic acid extraction solution (for example, a solution containing a surfactant such as SDS, Triton-X, or Tween-20, or a saponin, EDTA, or protease). .
Next, a solution containing a labeled probe described later is brought into contact with the single-stranded gene sample. Thereby, the labeled binding probe of the labeled probe and the gene sample having a sequence complementary to the labeled binding probe are hybridized to form a labeled gene sample. Since the method of hybridizing the labeled probe and the gene sample is well known, the description thereof is omitted here.
Hereinafter, the labeled probe used in the present invention will be described. The labeled probe is a single-stranded nucleic acid labeled with a substance that has a base sequence complementary to the gene sequence to be detected and is electrochemically active.
Any electrochemically active substance may be used as long as it is an electrochemically detectable substance. For example, by measuring an oxidation-reduction current generated during a reversible oxidation-reduction reaction. Examples thereof include compounds having redox properties capable of detecting substances. Examples of such redox compounds include ferrocene, catecholamine, metal complexes having a heterocyclic compound as a ligand, rubrene, anthracene, coronene, pyrene, fluoranthene, chrysene, phenanthrene, perylene, binaphthyl, There are octatetraene and viologen.
In addition, the aforementioned metal complexes having a heterocyclic compound as a ligand, rubrene, anthracene, coronene, pyrene, fluoranthene, chrysene, phenanthrene, perylene, binaphthyl, and octatetraene emit electrochemiluminescence during the oxidation-reduction reaction. Some occur, and the gene sample can be detected by measuring the luminescence in the detection step described later.
Examples of the metal complex having a heterocyclic compound in the above-mentioned ligand include a heterocyclic compound containing oxygen, nitrogen, etc., for example, a metal complex having a pyridine moiety, a pyran moiety, etc. in the ligand. In particular, a metal complex having a pyridine moiety as a ligand is preferable, and examples of the metal complex having the pyridine moiety as a ligand include metal bipyridine complexes and metal phenanthroline complexes.
Further, as the central metal of the metal complex having a heterocyclic compound as the ligand, for example, ruthenium, osnium, zinc, cobalt, platinum, chromium, molybdenum, tungsten, technetium, rhenium, rhodium, iridium, palladium, copper , Indium, lanthanum, praseodymium, neodymium, samarium and the like. In particular, a complex in which the central metal is ruthenium or osmium has good electrochemiluminescence properties, and examples of the material having such good electrochemiluminescence properties include ruthenium bipyridine complexes, ruthenium phenanthroline complexes, males. Examples thereof include a nium bipyridine complex and an osnium phenanthroline complex.
After that, the solution containing the labeled gene sample formed as described above is brought into contact with the capture probe immobilized on the magnetic beads. Thereby, the capture probe and a gene sample having a sequence complementary to the capture probe are hybridized, and the labeled gene sample is immobilized on the magnetic beads. Since the method of hybridizing the capture probe and the labeled gene sample is well known, description thereof is omitted here.
As the capture probe, a single-stranded nucleic acid obtained by chemical synthesis, or a nucleic acid obtained by cleaving a nucleic acid extracted from a biological sample with a restriction enzyme and separating it by electrophoresis or the like can be used. In the case of nucleic acid extracted from a biological sample, it is preferably dissociated into single-stranded nucleic acid by heat treatment or alkali treatment.
The magnetic beads for immobilizing the capture probe are not particularly limited, and magnetic beads generally used for medical use can be used. The magnetic beads are obtained by dispersing a magnetizable substance such as iron oxide in a polymer such as polystyrene or dextran, which is an electrically insulating material. The particle size of the magnetic beads can be selected from a wide range of 10 nm to 10 μm and is not particularly limited, but is preferably 300 nm to 5 μm from the viewpoint of dispersibility in the solution, separation and recovery.
As a method for immobilizing the capture probe on the magnetic bead, for example, a known method such as applying avidin coating to the magnetic bead and binding the end to a biotin-modified nucleic acid probe can be used.
In the above description, first, after the gene sample labeling step in which the gene sample and the labeled probe are hybridized to form a labeled gene sample, the labeled gene sample and the capture probe immobilized on the magnetic beads The gene sample capturing step for immobilizing the gene sample and immobilizing the gene sample on the magnetic beads is performed, but the gene sample capturing step and the gene sample labeling step may be performed simultaneously. In this case, the gene sample, the labeled probe, and the capture probe immobilized on the magnetic beads are simultaneously contacted to immobilize the labeled gene sample on the magnetic beads.
After immobilizing the labeled gene sample to the magnetic beads as described above, the magnetic beads are collected by magnetic force, and the unreacted gene sample and the labeled probe are removed to perform B / F separation.
After performing the B / F separation, the magnetic beads are spread on the electrode together with the electrolyte, and the magnetic beads are captured on the electrode surface by magnetic force. Then, by applying a voltage to the electrode, an electrochemical signal derived from an electrochemically active substance of the labeled gene sample immobilized on the magnetic beads is measured.
The measurement of the electrochemical signal differs depending on the labeled electrochemically active substance, and an electrochemically active substance that generates a redox current is used as the electrochemically active substance. In some cases, an electrochemical signal derived from the electrochemically active substance can be measured by a measurement system including the potentiostat, function generator, and the like. On the other hand, when a substance that generates electrochemiluminescence is used as the electrochemically active substance, measurement can be performed using a photomultiplier or the like.
Hereinafter, the electrolytic solution used in the present invention will be described. The electrolytic solution of the present invention has conductive fine particles added thereto, and the electric double layer region extends to a part away from the electrode surface through the conductive particles dispersed in the electrolytic solution, and contacts the electrode surface. Since an electrochemical reaction occurs even in an electrochemically active portion of the portion that is not present, the utilization efficiency of the labeling agent can be increased.
The conductive particle may be a substance that can transfer electrons between a metal electrode and the electrochemically active substance, and can oxidize and reduce the electrochemically active substance. Anything can be used without particular limitation.
Suitable materials include metal materials that are chemically stable and have a high dissolution potential, such as gold, platinum, silver, palladium, rhodium, iridium, tungsten, and carbon having a wide potential window in the oxidation and reduction directions. Is mentioned.
The particle size of the conductive particles is not particularly limited, but is preferably equal to or smaller than the particle size of the magnetic beads. The magnetic beads trapped on the electrode surface by the magnetic force exist on the electrode surface in an aggregated and laminated state. Therefore, by using the conductive particles having the above-mentioned particle size, the conductive particles are placed in the gap between the magnetic beads. The labeling agent bonded to the aggregated and laminated magnetic beads can be reacted via the entering and entering conductive particles, and the utilization efficiency of the labeling agent can be increased.
Furthermore, the particle diameter of the conductive particles is more preferably 1 nm to 5 μm, and particularly preferably 1 nm to 300 nm. By using such conductive particles having a small particle diameter, it becomes easy to enter the gaps between the aggregated and laminated magnetic beads.
Examples of such conductive fine particles include colloidal particles used in immunoassay colorants, fuel cell catalysts, and the like.
The amount of the conductive fine particles added to the electrolytic solution is determined by measuring the optical density of the electrolytic solution to which the conductive fine particles are added. In general, the concentration of the fine particles in the solution is indicated by the optical density of the solution containing the fine particles, and the optical density is measured with an absorptiometer using the light having the maximum absorption wavelength of the solution containing the fine particles. For example, when gold colloid is used for the conductive fine particles, the measurement is performed using light having a wavelength of 520 nm.
The optical density of the electrolytic solution to which the conductive fine particles are added is preferably 0.0001 to 1, and particularly preferably 0.01 to 0.3. By using such an optical density, a good dispersion state of the conductive fine particles can be obtained, and the utilization efficiency of the labeling agent can be increased. Furthermore, when a substance that generates electrochemiluminescence is used for the labeling agent, if the optical density of the electrolytic solution increases, the conductive fine particles also have an effect of inhibiting light emission from the labeling agent. By setting such an optical density, the influence of the conductive fine particles added to the electrolysis solution inhibiting the light emission from the labeling agent can be suppressed.

(4) Hybridization 14 μL of 2XSSC was added to the magnetic beads on which the capture probe was immobilized, and 4 μL of the gene sample and labeled probe prepared to 5 μM were added thereto, and gently shaken at 70 ° C. After shaking for 1 hour, the magnetic beads were collected with a magnet, and then the solution was decanted and washed with 2XSSC heated to 40 ° C. to obtain magnetic beads α on which double-stranded nucleic acids were formed.
The same processing as described above was performed on the magnetic beads on which the non-complementary capture probe was immobilized, thereby obtaining magnetic beads β on which no double-stranded nucleic acids were formed.
(5) Electrochemical measurement After the above steps, 10 μL of 0.2 M PBS and 0.2 M triethylamine are added to magnetic beads α in which double-stranded nucleic acids are formed and magnetic beads β in which double-stranded nucleic acids are not formed. 10 μL of a mixed solution was added and suspended, and 5 μL each was added dropwise to the electrode.
As the electrode, a glass substrate was used in which a gold film of 200 nm was formed on a glass substrate with a sputtering apparatus (SH-350, ULVAC, Inc.), and an electrode pattern was formed by a photolithography process. In addition, a permanent magnet sheet is attached under the electrode so that the magnetic beads are concentrated only on the working electrode.
After standing for 5 minutes, 75 μL of an electrolytic solution was dropped on each of the electrodes xα and yβ where the magnetic beads α and β were aggregated.
Here, in the electrolytic solution, a solution prepared by mixing 500 μL of 0.2 M PBS and 500 μL of 0.2 M triethylamine, and further gold colloid (EMGC20 manufactured by Funakoshi Co., Ltd., optical diameter of 20 nm, 520 nm) as conductive fine particles. An electrolytic solution A prepared by adding 100 μL of a concentration 1.0) was used.
In addition, as a comparative object for clarifying the effect of adding conductive fine particles, an electrolytic solution B in which 500 μL of 0.2 M PBS was mixed and 500 μL of 0.2 M triethylamine was added and no gold colloid was added was also used. .
Thereafter, voltages were applied to the respective electrodes xα and yβ, and the electrochemiluminescence generated at this time was measured. The voltage was applied by scanning from 0 V to 1.3 V and performing electrochemical measurement for 3 seconds. The electrochemiluminescence amount was measured using a photomultiplier tube (H7360-01 manufactured by Hamamatsu Photonics), and the maximum luminescence amount during voltage scanning was measured.
FIG. 1 shows the maximum amount of electrochemiluminescence detected at an electrode xα of a magnetic bead on which a double-stranded nucleic acid is formed and an electrode yβ of a magnetic bead on which no double-stranded nucleic acid is formed.
FIG. 1 shows that the amount of light emitted from the magnetic bead electrode xα on which the double-stranded nucleic acid is formed is higher in the electrolytic solution A, and the amount of light emitted is increased by the addition of gold colloid. In addition, the amount of light emitted from the magnetic bead electrode yβ in which no double-stranded nucleic acid is formed is not different between the electrolytic solution A and the electrolytic solution B, and it can be seen that there is no change in background noise due to the addition of gold colloid. .
Therefore, it can be seen that the addition of colloidal gold can increase only the amount of luminescence without increasing background noise, and can detect a double-stranded nucleic acid, that is, a target gene sample with high sensitivity.
