In general, for new chemical substances that are active ingredients of drugs such as agricultural chemicals and pharmaceuticals, inhalation toxicity tests of the chemical substance are conducted at the same time as the safety evaluation, and the chemical substance is applied to the test mammal. It is required to evaluate the inhalation toxic effect to be given (for example, see Non-Patent
When conducting such an evaluation, the concentration of exposure is freely adjusted depending on the test conditions (for example, depending on the type of chemical substance), while centrally monitoring and managing the status of the inhalation toxicity test.
However, there is no inhalation toxicity test apparatus that meets the above requirements, and the conventional inhalation exposure apparatuses described in
That is, the inhalation toxicity test apparatus needs to be able to always grasp in real time the test implementation status during the inhalation toxicity test in the case where the inhalation toxicity test under a plurality of test conditions is performed simultaneously. The inhalation toxicity test apparatus needs to be able to record the test implementation status during the inhalation toxicity test in order to analyze the test results after such a test.
Therefore, the present invention provides inhalation toxicity under a plurality of test conditions (for example, freely adjusting the exposure concentration depending on the type of chemical substance) while intensively monitoring and managing the test implementation status during the inhalation toxicity test. In the invention according to
According to the seventh aspect of the present invention, a small amount of external air is drawn from the leak hole of the leak valve to the downstream side of the discharge system, and dust or mist is discharged from the inhalation exposure chamber. Since a small amount of air is sucked in from the outside and the small amount of air is exhausted together with dust or mist, for example, even if a change occurs in the exhaust flow rate at the feeding means, the amount of air sucked from the leak hole It only changes, and the pressure in the inhalation exposure chamber is always maintained at a slight negative pressure. Further, since the flow rate of pushing into the hollow chamber by the feeding means is constant and the leak hole is provided downstream of the discharge port, dust or mist can always maintain a constant concentration at the discharge port. . In addition, since the concentration of dust or mist is not diluted, a high concentration of the test substance can be inhaled by the animal under test.
The invention according to claim 8 is an inhalation toxicity test under a plurality of test conditions while intensively monitoring and managing the test implementation status during the inhalation toxicity test using the inhalation toxicity test apparatus according to
According to the invention described in claim 8, the inhalation toxicity test under a plurality of test conditions can be simultaneously performed while intensively monitoring and managing the test implementation status during the inhalation toxicity test. The inhalation toxicity test can be easily performed with high accuracy and reliability.
The present invention makes it possible to provide an inhalation toxicity test apparatus that allows anyone to easily perform an inhalation toxicity test under a plurality of test conditions at the same time. Thus, the analysis of the test result after the test can be easily performed with high accuracy and reliability.
The present invention relates to an apparatus suitable for conducting inhalation toxicity tests such as agricultural chemicals and medicines on small animals such as mice or rats. Here, the “inhalation toxicity test” refers to a mist in which liquid test substances such as agricultural chemicals and pharmaceuticals are dispersed in a mist in a gas such as air, or a solid test substance powder dispersed in a gas such as air. The dust or the like is taken into a small animal such as a mouse or a rat (inhalation exposure), and the toxicity of the test substance is evaluated based on the biological damage caused thereby. Representative examples of the inhalation toxicity test include, for example, acute inhalation toxicity test (a test in which a large amount of a chemical substance is taken into a mammal at one time (exposure to inhalation)) And sub-acute inhalation toxicity test (in the case where a mammal is taken into a mammal by continuous administration over several months (inhalation exposure)). The behavior of mammals, blood, urine, effects on the whole body of various organs and tissues, etc. are investigated.) Etc., and the present invention may be applied to any test. In addition, the present invention is a mist in which a liquid test substance is dispersed in a mist form in a gas such as air, compared to a test in which powder of a solid test substance is dispersed in a gas such as air. More suitable for some tests.
In the acute inhalation toxicity test, the toxicity of the test substance is usually evaluated by the concentration at which half the test animal dies when exposed to a certain period of time (ie, the half-lethal concentration LC50).
Inhalation toxicity test methods include the whole body exposure method in which the whole body of a small animal such as a mouse or rat is exposed to the atmosphere containing the test substance, and the head / nose exposure method in which only the head or nose is exposed. In the former systemic exposure method, the test substance is administered dermally in addition to inhalation by the respiratory tract. Therefore, in order to correctly evaluate toxicity by inhalation alone, it is desirable to use the latter head / nose exposure method.
Hereinafter, specific examples of the present invention will be described.
The inhalation
Specifically, for example, in inhalation toxicity tests under test conditions in multiple groups of 1 to 5, in addition to grasping the air flow rate and test substance flow rate, the air flow rate and test in the necessary group as necessary The substance flow rate setting can be changed from the control unit.
In addition, after the test is completed, the test execution status during the test is output / displayed, and the output / display after the test facilitates analysis of the test results after the test.
In addition, you may manufacture this invention apparatus by adding an improvement so that each structural requirement of this invention may be satisfy | filled to a commercially available inhalation exposure apparatus.
FIG. 1 is a schematic diagram for explaining an inhalation
The inhalation
As shown in FIG. 1, the
The
そして吸入曝露チャンバー本体31aの頂部31d(尚、本発明装置における「頂部」とは、吸入曝露チャンバー本体31aが全体として円筒状をなした構成の場合には上部の円状平面の中心と意味し、また吸入曝露チャンバー本体31aが全体として円筒状をなすとともに上部を円錐状に形成した構成の場合には上部の円錐面の頂部を意味する。)
In FIG. 1, an inhalation exposure chamber main body 31a which is a main component of the
The upper end portion of the inhalation exposure chamber main body 31a may be a cylindrical surface or a conical surface. The top 31d of the inhalation exposure chamber main body 31a (the "top" in the apparatus of the present invention means the center of the upper circular plane in the case where the inhalation exposure chamber main body 31a has a cylindrical shape as a whole. In the case where the inhalation exposure chamber main body 31a has a cylindrical shape as a whole and has an upper portion formed in a conical shape, it means the top of the upper conical surface. The supply nozzle 21 is arranged in the chamber 31e so that the small hole-shaped jet port 21b is located.
Furthermore, an inhalation exposure opening 37 to which a
A

In the inhalation
As an apparatus of another form other than the inhalation
(If a leak valve is provided, a small amount of external air is drawn from the leak hole 73 of the leak valve 72 to the downstream side of the discharge system, and dust or mist is discharged from the inhalation exposure chamber at the same time. Also in 73, a small amount of air is sucked from the outside, and the small amount of air is exhausted together with dust or mist. For example, even if a change occurs in the exhaust flow rate at the feeding means, the air is sucked from the leak hole 73. The pressure of the inhalation exposure chamber is always maintained at a slight negative pressure, the pushing flow rate into the hollow chamber by the feeding means is constant, and the leak hole 73 is a discharge port. The dust or mist can always be maintained at a constant concentration at the outlet, and the dust or mist concentration is diluted. The test substance can be inhaled into the test animal.)
Dust or mist used for inhalation toxicity tests is generated as follows. First, a desired amount of the test substance (for example, the chemical solution 8) held in advance in the test substance reservoir 25 is released into the test

With such a centralized monitoring control system including a measurement unit and a control unit, a control terminal of the system can be installed (for example, in a centralized monitoring room), and operation control can be performed by centralized monitoring. The apparatus of the present invention is an example of a system for acute inhalation toxicity tests of agricultural chemicals, pharmaceuticals, etc. Specifically, for example, a centralized monitoring control apparatus capable of simultaneously operating five systems of four concentrations and one control. Can be mentioned. These operating states are always displayed in real time on an information output / display unit 61 (specifically, a graphic panel) described later.

Specifically, the inhalation exposure chamber main body 31a is configured such that a conical upper lid portion, a cylindrical body portion, and a bottomed short cylindrical bottom portion are detachably connected to each other. .

As shown in FIGS. 1 and 2, the inhalation
When the
