図3及び図4に示されるような微粒子回収装置を具備したマイクロチップを作製した。第1の基板3として、60mmx24mmで厚さ5mmのPDMS基板を使用した。第2の基板5として、60mmx24mmで厚さ5mmのPDMS基板を使用した。第1のPDMS基板3には50μm幅、25μm深さの流路7が形成されており、この流路7に直交するように、35μm幅、35μm奥行、25μm深さの微粒子回収用チャンバー9が形成されており、該チャンバー9の開口部と反対側の他端上部には大気に向かって開口する直径0.5mmの孔17が形成されていた。第2のPDMS基板5には、前記微粒子回収用チャンバー9及び孔17に連通する凹陥部13(500μm幅、500μm奥行、200μm深さ)が形成されていた。図示されていないが、流路7の両端には大気に連通するポートが配設されていた。このポートは液体又は気体の入口及び出口となるものである。前記第1のPDMS基板の流路及び微粒子回収用チャンバー形成面と、第2のPDMS基板の凹陥部形成面とを位置合わせして接着させた。その後、第1のPDMS基板の孔17を遮蔽するためのPDMS製シート19(10mmx3mm、厚さ0.3mm)を第1のPDMS基板上に部分的に恒久接着させた。第1のPDMS基板3の端面から流路7までの間隔は300μmであった。
(2)流下試験
前記(1)で作製された本発明の微粒子回収装置を具備したマイクロチップにおいて、出口側のポートを塞ぎ、PDMS製シート19の自己吸着部分を剥離して孔17を露出させた状態で、入口側のポートから流路7に超純水を流したところ、流路全体及び微粒子回収用チャンバー(凹陥部を含む)全体が極めて短時間のうちに超純水で満たされ、孔17の上面にまで達した時点で超純水の流下を止め、PDMS製シート19を再び第1のPDMS基板3の上面に自己吸着させて孔17を遮蔽した。
(3)捕捉試験
前記(2)で、流路及び微粒子回収用チャンバーが超純水で満たされた本発明の微粒子回収装置を具備したマイクロチップを水平な状態に維持しながら、微粒子回収用チャンバーの開口部と正対する位置にレーザ光源110(Nd:VANレーザ,波長1064nm)を配置した。言うまでもなく、レーザ光源110には、ビームを収束させ、焦点距離を調整するための対物レンズやビーム径を調整するためのビームエキスパンダーなどの公知慣用の光学素子類が組み込まれていた。また、この試験では、流路を流れてくる微粒子を検出・解析するために、蛍光顕微鏡(Axiovert135TV, Carl Zeiss社製)をレーザ操作システムと結合・連動させて用いた。
この試験では、直径2μmの蛍光ラテックスビーズ(Polysciences, Inc.社製)を光圧に応答する微粒子として用いた。該微粒子を超純水に1x105個/mlとなる濃度で懸濁させた液をサンプル液として利用した。該サンプル液は、シリンジポンプ(シリンジフィーダー)を利用して入口側ポートから流路7内に導入した。流路7内を流れるサンプル液の流速は192μm/秒とした。流路7を通過したサンプル液は出口側ポートから回収した。
蛍光顕微鏡で流路を流れてくる微粒子を観察し、微粒子回収用チャンバーの開口部手前付近に達した時点でレーザ光源からレーザビームを照射した。レーザビームは微粒子回収用チャンバーの奥方向に合焦するように照射した。
(4)試験結果
微粒子が微粒子回収用チャンバーの開口部から凹陥部にまで異動され、凹陥部内に集積されたかを確認するため、蛍光ラテックスビーズが経時的に凹陥部内に回収集積できるか否かを調べた。すなわち、レーザビームの照射を連続的に行って、凹陥部内の蛍光強度を経時的に測定した。蛍光強度はビデオテープに記録した画像をコンピュータに取り込んだ後、画像解析ソフト(NIH image: http://rsb.info.nih.gov/nih-image/)を用いて、凹陥部内領域のビデオフレーム毎(30フレーム/秒)の蛍光強度を解析することにより求めた。蛍光強度の測定結果は相対値として示される。得られた結果を図6示す。図6に示される結果から明らかなように、凹陥部内の蛍光強度は60秒間にわたって、時間に比例して増加している。このことから、レーザビームを連続的に照射することによって、流路を通過する標的微粒子の流れを光圧によってチャンバー方向に偏向させ、更に、凹陥部内に標的微粒子を回収蓄積できることが確認された。
(5)凹陥部からの標的微粒子の取出試験
レーザビームの照射を止めた後、PDMS製シート19を剥離し、凹陥部内の液体をマイクロピペットで取り出した。この液体を別の蛍光検出器で分析したところ、取り出した液体中に蛍光ラテックスビーズが含まれていることが確認できた。従って、本発明の微粒子回収装置によれば、凹陥部内に回収蓄積させた標的微粒子を装置外へ簡単に取り出すことが出来ることが確認された。
