Abstract
Description
The present invention relates to an observation apparatus for observing an observation object such as a cultured cell.
For example, phenol red exhibits red to magenta when the medium becomes alkaline (high pH value) and yellow to orange when the medium becomes acidic (low pH value). Therefore, when mold or the like enters the medium and the number of cells decreases, the amount of lactic acid discharged into the medium decreases, and the medium becomes reddish purple. On the other hand, when the number of cells in the medium increases, the amount of lactic acid increases, so the medium turns yellow. When the number of cells in the medium is appropriate, the amount of lactic acid is also moderate, so the medium becomes pink. Therefore, the administrator can keep the cultured cells in a good state by determining the color of the medium at an appropriate timing and, for example, replacing or subculture the medium as necessary so that it becomes pink.
However, in order to determine the color of the culture medium, it is necessary to lift the culture container and hold it over light such as room light. In addition, the color appearance varies depending on the amount of the medium and the type of room light, and further, the judgment person's experience level also affects, so the judgment is not accurate.
Therefore, an object of the present invention is to provide an observation apparatus that can accurately measure the pH value of an object to be observed without contact.

[First Embodiment]
Hereinafter, the first embodiment will be described. This embodiment is an embodiment of an observation system.
First, the overall configuration of this system will be described.
FIG. 1 is a diagram showing the overall configuration of the system. As shown in FIG. 1, the system includes a mounting table 7, a
The culture container as the object to be observed is placed on the placing table 7. There are various types of culture vessels. Here, the dish 1 having a circular outer diameter will be described. The inside of the
The dish 1 is illuminated by the
The
The timing at which the
Further, the
The
And the
Next, the overall flow of the operation of the
FIG. 2 is an operation flowchart of the
Thereafter, when a start instruction is input from the administrator (step S2 YES), the
After execution of this measurement process (step S3), the
Furthermore, the
Thereafter, when a measurement interval (12 hours, 24 hours, etc.) designated in advance by the administrator has elapsed (YES in step S9), the
Further, when the administrator calls the measurement history (step S5), the
Further, when there is a call for a stored color image from the administrator (step S7), the
Next, details of the measurement process (step S3) will be described.
FIG. 4 is an operation flowchart of the
) Is calculated (step S32). At that time, in order to reduce the influence of noise included in the signal, the
Furthermore, the
B / G = a * (pH) ^ 2 + b * (pH) + c (1)
However, “B / G” represents the B / G ratio, “pH” represents the pH value, and “(pH) ^ 2” represents the square of the pH value. That is, Formula (1) expresses B / G ratio with the quadratic function of pH value. When the pH value is actually calculated, a value obtained by solving the equation (1) for the pH is used.
Here, a, b, and c in Equation (1) are all constants, and are set to, for example, a = 0.56, b = −7.48, c = 2.54, and the like.
However, the values of a, b, and c are determined by the characteristics of phenol red, the whiteness of the
Next, the measurement principle of this embodiment will be described in detail.
FIG. 5 is a diagram showing nine color image signals (RGB signals) obtained by placing nine phenol red samples (9 samples having different pH values by 0.2) on the placing table 7 of the present system. is there. The horizontal axis is the pH value of the sample, and the leftmost sample (pH value = 6.8, that is, the acidic side) generally looks orange, and the rightmost sample (pH value = 8.4, that is, alkaline). The side) sample generally looks reddish purple. The vertical axis represents the signal value of the color image signal, and the larger the value, the brighter the signal.
Since the maximum values of the R signal, G signal, and B signal included in the color image signal (RGB signal) are all 255, the maximum value of the RGB signal (added value of the R signal, G signal, and B signal) is 765 (= 255 × 3 colors). As can be seen from FIG. 5, when the pH value changes, the G signal and the B signal change more greatly than the R signal. Therefore, it can be seen that the differences in the colors of phenol red such as orange, red, and magenta as seen from the human eye are only differences in the ratio between the blue component and the green component.
FIG. 6 is a diagram illustrating the ratio of the R signal, the G signal, and the B signal when the RGB signal (added value of the R signal, the G signal, and the B signal) is 100% in FIG. It can be seen that as the pH value increases, the ratio of the B signal increases, and conversely, the ratio of the G signal decreases. On the other hand, the ratio of the R signal does not change as much as the ratio of the G signal and the B signal.
Therefore, for easier understanding, FIG. 7 shows the ratio of the B signal and the G signal when only the B signal and the G signal are focused on and the added value of the B signal and the G signal is 100%. From FIG. 7, it can be clearly seen that the higher the pH value, the higher the B signal ratio and the lower the G signal ratio.
Further, FIG. 8 is a graph in which the ratio of the B signal to the G signal (B / G ratio) is calculated for each pH value, and the pH value is plotted on the horizontal axis. When each point in FIG. 8 is fitted to a quadratic function, the following expression (1 ′) is obtained.
B / G = 0.56 × (pH) ^ 2-7.48 × (pH) +2.54 (1 ′)
B / G is a B / G ratio, pH is a pH value, and (pH) 2 is a square of the pH value. Therefore, the B / G ratio of phenol red changes uniquely depending on the pH value. This equation (1 ′) corresponds to the equation (1) used in the measurement process of this system.
FIG. 9 is a scatter diagram showing the BG coordinates of nine samples. The abscissa is the value of the G signal, and the ordinate is the value of the B signal.
Using this theory, in the measurement process of this system, the pH value is uniquely calculated from the B / G ratio of the color image signal and the equation (1). Therefore, although the present system measures the pH value indirectly from a color image acquired in a non-contact manner, the accuracy of the measurement is high.
By the way, the color image signal of this system is based on the light emitted from the
Next, it is verified that the above theory does not depend on the depth of the culture medium 3.
In the experiment, a culture vessel (flask) containing a certain medium was prepared, and it was intentionally tilted so that a deep portion and a shallow portion were formed in the same medium. Thereby, a plurality of samples having the same pH value but different depths could be prepared.
FIG. 10 is a scatter diagram showing BG coordinates on a BG plane of a plurality of samples having the same pH value but different depths. In FIG. 10, for comparison, the BG coordinates of the nine samples shown in FIG. 9, that is, samples having the same depth but different pH values are indicated by triangles (reference numeral 33). The BG coordinates of a plurality of samples having the same pH value but different depths are indicated by square marks (reference numeral 34).
Among these, the
From this, it can be seen that a plurality of samples having the same pH value have the same B / G regardless of the depth. That is, the above theory does not depend on the depth of the culture medium 3. Therefore, this system using the above theory can reliably perform the measurement regardless of the depth of the culture medium 3.
(Supplement 1 of the first embodiment)
Note that the
According to the flowchart of FIG. 11, when the
In addition, for a color image that has been determined as NO in step S32 ″ and the pH value could not be calculated, the
The reason why the above determination (step S32 ″) is effective is as follows.
For example, when the culture medium 3 is too shallow, the amount of light incident on the
As an extreme example, when the medium 3 is not present, the light emitted from the
On the other hand, when the medium 3 is too much, light emitted from the
Therefore, the determination criterion in step S32 ″ is set, for example, in the range of 25 to 240. At this time, the

Note that it is desirable that a program (calibration program) for causing the administrator of the system to perform the above calibration is installed in advance in the
(Supplement 3 of the first embodiment)
In the above description, the dish 1 has been described as a culture container. However, culture containers having various shapes and sizes are commercially available according to the application. There are plates (provided with a plurality of wells), flasks (with a breathable cap), and the like. In this system, any culture vessel can be used.
In the above description, only one culture vessel (dish 1) was placed on the mounting table 7. However, the size of the mounting table 7 is sufficiently large, and two or more culture containers are arranged on the mounting table 7. It may be placed.
However, when a well plate is used or when two or more culture vessels are placed side by side, since the number of culture media becomes plural, the pH value needs to be measured for each culture media. In that case, the administrator may confirm the positions of a plurality of culture media on the color image as shown in FIG. 14A and designate the positions of the culture media as measurement points. At this time, the
(Supplement 4 of the first embodiment)
The illumination system (such as the illumination unit 6) of this system is a type (transmission type) illumination system that illuminates the object to be observed (dish 1), but as shown in FIG. ) Reflection type illumination system (reflection type) may be used. In that case, the material or the structure of the mounting table 40 does not transmit light. For example, the mounting table 40 is made of a white plate (for example, a white plastic plate) that reflects light. Moreover, the
The illumination light composed of white light emitted from the
When such a reflective illumination system is used, a, b, and c in the above formula (1) are not only the characteristics of phenol red, the whiteness of the

Further, in this system, the timing for designating the measurement point and the timing for calculating the pH value are not limited to those described above. For example, the calculation of the pH value may be performed after acquisition of all color images during the culture period.
In this system, phenol red is used as a pH indicator, but other pH indicators such as chlorophenol red and bromcresol purple may be used. In any case, the pH value can be calculated from the color image by previously obtaining a relational expression between the color of the pH indicator and the pH value in an experiment using a plurality of samples having different pH values.
[Second Embodiment]
The second embodiment will be described below. This embodiment is an embodiment of an incubator.
