Similarly, IPS-1 overexpression activated the IP-10, RANTES and ISRE promoters controlled by the IRF family in a dose-dependent manner (FIG. 3a). Next, IFNα promoter activation was analyzed using IFNα4 and IFNα6 promoter Luc constructs. Overexpression of IPS-1 alone slightly induced IFNα4 and IFNα6 promoter activation (FIG. 3b). However, co-expression of IRF7 synergistically activated both IFNα4 and IFNα6 promoters (FIG. 3b), indicating that IPS-1 enhanced IRF7-mediated transcriptional activity. Furthermore, it was found that Myc-IRF3 and Myc-IRF7 migrate slowly when co-expressed with Flag-IPS-1 (FIG. 3c). This migration is due to phosphorylation. This is because phosphatase treatment abolished slow migration (data not shown) and revealed that IPS-1 induces phosphorylation of IRF3 and IRF7.

Induction of endogenous type I IFN and IFN-inducible genes after IPS-1 expression was analyzed. Overexpression of Flag-IPS-1 in HEK293 cells stimulated the induction of endogenous IFNβ (Ifnb) and IFN-inducible genes such as IP-10 (Cxcl10) and GARG16 (Ifit1) (FIG. 4a). Furthermore, IFNα production in the culture supernatant was also observed in HEK293 cells transfected with IPS-1, and this induction was enhanced by co-expression of IRF7 (FIG. 4b). Thus, IPS-1 expression leads to endogenous promoter activation of these genes. Next, control or IPS-1 transfected HEK293 cells were infected with VSV, and the virus titer was measured 24 hours after infection. Viral titer was significantly reduced in IPS-1-expressing cells compared to control cells (FIG. 4c). These results suggested that expression of IPS-1 alone was sufficient to confer an antiviral response, possibly by producing antiviral cytokines such as type I IFN.

To examine whether IPS-1 expression activates NF-κB, HEK293 cells were transfected with IPS-1 transiently with the ELAM1 promoter Luc construct. Expression of IPS-1 led to NF-κB activation in a dose-dependent manner (FIG. 5a). Furthermore, this activation was inhibited by the co-expression of catalytic negative IKKβ (K44A) (FIG. 5b). Neither IPS-1N nor IPS-1C activated NF-κB (FIG. 5c). Furthermore, IL-8 production was observed in the culture supernatant of HEK293 cells transfected with the IPS-1 expression plasmid, but not in HEK293 cells transfected with the control plasmid. This suggests that IPS-1 also activates the endogenous NF-κB promoter (FIG. 5d). Unlike IFNα and IFNβ promoter activation, overexpression of IPS-1 induced ELAM1 activation in TBK1 / IKKi double-deficient MEFs (FIG. 5e). Furthermore, in TRAF6-deficient MEFs, NF-κB and IFNβ promoter activation by IPS-1 was continuously observed. These results indicate that NF-κB activation by IPS-1 is IKKβ-dependent but independent of TRAF6 and TBK1 / IKKi.

The virus yield of the culture supernatant collected from HEK293 cells infected with VSV was measured. BHK cells infected with serial dilutions of recovered virus were covered with DMEM containing 1.0% low melting agarose. Plaques were counted after 24 hours of incubation.

Ifnb;5’−CAGCAATTTTCAGTGTCAGAAGCT−3’(配列番号3)及び5’−TCATCCTGTCCTTGAGGCAGTAT−3’ (配列番号4)、Cxcl10;5’−TGACTCTAAGTGGCATTCAAGG−3’(配列番号5)及び5’−GATTCAGACATCTCTTCTCACCC−3’(配列番号6)、Ifit1;5’−CCTGCTGGTGGTGGACAAAT−3’(配列番号7)及び5’−TGCGGCCCTTGTTATTCC−3’(配列番号8),Gapdh;5’−CTGGGCTACACTGAGCACCAG−3’(配列番号9)及び5’−CCAGCGTCAAAGGTGGAG−3’(配列番号10)。
(Northern blot analysis and RT-PCR)
The 32 P-labeled full-length human IPS-1 probe was hybridized with a human multi-tissue northern blot (manufactured by CLONTECH), washed, and visualized by autoradiography. For RT-PCR analysis, total RNA was isolated using Trizol reagent (Invitrogen) and reverse transcribed using SuperScript III reverse transcriptase (Invitrogen) according to the manufacturer's instructions. PCR was performed continuously using the following primers.
Ifnb; 5'-CAGCAATTTTCAGTGTCAGAAGCT-3 '(SEQ ID NO: 3) and 5'-TCATCCTGTCCTTGAGGCAGTAT-3' (SEQ ID NO: 4), Cxcl10; (SEQ ID NO: 6), If1; 5′-CCTGCTGGTGGGGACAAAAT-3 ′ (SEQ ID NO: 7) and 5′-TGCGGCCCCTGTTTATTCC-3 ′ (SEQ ID NO: 8), Gapdh; 5′-CTGGGCTACACTGAGCACCAG-3 ′ (SEQ ID NO: 9) and 5 '-CCAGCGTCAAAGGTGGAG-3' (SEQ ID NO: 10).
(免疫共沈降及びイムノブロット分析)

(Co-immunoprecipitation and immunoblot analysis)
One million HEK293 cells were seeded in 100 mm dishes. After 12 hours, the cells were transiently transfected with an empty plasmid or a total amount of a predetermined plasmid of 6.0 μg using Lipofectamine 2000 (Invitrogen). Immunoprecipitation and immunoblotting were performed as described in the literature (Nat Immunol. 5, 1061-1068, 2004).

siRNA実験用に、21ヌクレオチドを有するセンス及びアンチセンスオリゴヌクレオチドからなる二本鎖RNAデュプレックスを、ダーマコンリサーチ(Dharmacon research)により合成した。ヒトIPS−1を標的とするのに用いたRNAオリゴヌクレオチドは、次の通りである。Ips−1−1センス;5’−UAGUUGAUCUCGCGGACGAdTdT−3’(配列番号11)、Ips−1−1アンチセンス;5’−UCGUCCGCGAGAUCAACUAdTdT−3’(配列番号12)、Ips−1−2センス;CCGUUUGCUGAAGACAAGAdTdT−3’(配列番号13)、Ips−1−2アンチセンス;UCUUGUCUUCAGCAAACGGdTdT−3’(配列番号14)。HEK293細胞又はHela細胞を、60mm皿(5×105)に、トランスフェクションの12時間前に播種した。100nMのsiRNAを、Lipofectamine 2000又はOligofectamine(Invitrogen社製)を製造者の指示に従って用いて、それぞれHEK293細胞又はHela細胞にトランスフェクトした。トランスフェクションの48時間後、細胞を新たな実験に使用した。RT−PCRにより、IPS−1mRNAのノックダウンを5’−ATGCCGTTTGCTGAAGAC−3’(配列番号15)及び5’−CTAGTGCAGACGCCGCCG−3’(配列番号16)のプライマーで確認した。
(RNA interference)
For siRNA experiments, double-stranded RNA duplexes consisting of sense and antisense oligonucleotides with 21 nucleotides were synthesized by Dharmacon research. The RNA oligonucleotides used to target human IPS-1 are as follows. Ips-1-1 sense; 5'-UAGUUGAUCUCCGCGACGAdTdT-3 '(SEQ ID NO: 11), Ips-1-1 antisense; 5'-UCGUCCGCGAGAUCAACUAdTdT-3' (SEQ ID NO: 12), Ips-1-2 sense; CCGUUGUGCUGAGACAGAGAT 3 ′ (SEQ ID NO: 13), Ips-1-2 antisense; UCUUGUCUCAGCAAACGGGdTdT-3 ′ (SEQ ID NO: 14). HEK293 cells or Hela cells were seeded in 60 mm dishes (5 × 10 5 ) 12 hours before transfection. 100 nM siRNA was transfected into HEK293 cells or Hela cells, respectively, using Lipofectamine 2000 or Oligofectamine (Invitrogen) according to the manufacturer's instructions. Cells were used for new experiments 48 hours after transfection. By RT-PCR, knockdown of IPS-1 mRNA was confirmed with primers of 5′-ATGCCGTTTGCTGAAGAC-3 ′ (SEQ ID NO: 15) and 5′-CTAGTGCAGACGCCGCCG-3 ′ (SEQ ID NO: 16).
(IPS−1ノックアウトマウスの作製)
(1)キメラマウスの作製
IPS−1の生理学的役割を検討するため、ジーンターゲティングによりIPS-/-マウスを作製した。IPS−1-/-マウスを作製するためのターゲティングベクターは、pMC1−neo(Stratagene社製)からのネオマイシン耐性遺伝子で、エクソン1及びエクソン2を置換し、負の選択マーカーとして単純ヘルペスウィルスチミジンキナーゼ(HSV−TK)を挿入することにより構築した(図8a参照)。次に、当業者に公知の方法に従い、エレクトロポーション法で、構築されたターゲッティングベクターをES細胞に導入した。G418とガンシクロビアに二重に耐性を持つコロニーを選択し、PCRにより相同組換えが起きた細胞を選択した後、相同組み換えの認められた陽性クローン(ES細胞)を胚盤胞に導入して仮親に移植することによりキメラマウスを作製した。
(2)ヘテロ欠損(IPS−1+/−)マウスの作製
こうして得られたキメラマウスの交配によってF1マウスを作製した。F1マウスの遺伝型をPCR及びサザンブロット法で解析したところ生殖系列への変異の導入(germ−line化)が確認され、ヘテロ欠損マウス(IPS−1+/−マウス)が作製された。
(3)更に、IPS−1+/−マウスどうしを交配させて、IPS−1遺伝子ホモ欠損マウス(IPS−1−/−マウス)を作製した。
(Preparation of IPS-1 knockout mouse)
(1) Production of Chimeric Mouse In order to examine the physiological role of IPS-1, IPS − / − mice were produced by gene targeting. The targeting vector for generating IPS-1 − / − mice is a neomycin resistance gene from pMC1-neo (Stratagene), replacing exon 1 and exon 2, and herpes simplex virus thymidine kinase as a negative selectable marker. It was constructed by inserting (HSV-TK) (see FIG. 8a). Next, the constructed targeting vector was introduced into ES cells by electroporation according to a method known to those skilled in the art. After selecting colonies that are double resistant to G418 and ganciclovir and selecting cells that have undergone homologous recombination by PCR, a positive clone (ES cell) in which homologous recombination has been observed is introduced into the blastocyst and the parental parent. (3) Furthermore, IPS-1 +/− mice were crossed to produce IPS-1 gene homo-deficient mice (IPS-1 − / − mice).
