Abstract
Description
The present invention relates to an optical signal analysis method.
Conventionally, methods as shown in the following documents have been proposed as optical signal analysis methods. In these optical signal analysis methods, when estimating the autocorrelation function, etc., obtain a time-series signal of fluorescence intensity at only one measurement point and estimate the autocorrelation function using the multiple τ method. It was. In addition, for each of a plurality of measurement points, a continuous time series signal of fluorescence intensity was acquired, and an independent single point multiple τ method analysis method was used between the measurement points.
When trying to estimate an autocorrelation function at a certain time interval at a plurality of measurement points in a sample, two methods are generally conceivable. The first method is a method in which work is sequentially performed for all measurement points. That is, first, the autocorrelation function is estimated at one measurement point, and then the operation is moved to another measurement point to similarly estimate the autocorrelation function. Furthermore, the operation of moving to another measurement point and estimating the autocorrelation function is performed sequentially for all measurement points.
However, in the above-described first method, the entire estimation takes time and the work is complicated. In addition, since the measurement points are sequentially moved, the time difference between the measurement points is large and requires a long time.
Further, in order to realize the second method by software, there is a memory limitation and a CPU limitation. Further, a plurality of sets of optical measurement parts are required, and it is difficult to match the optical measurement conditions (measurement region, irradiation intensity, count rate, etc.) of each set, which causes an error. Therefore, it is very difficult to estimate the autocorrelation function by simultaneously measuring the above-described plurality of points.
In other words, measurement time lag (time delay) occurs at each measurement point, and errors in measurement conditions are included, so fast reactions that occur inside and outside the cell, such as protein-protein interactions, intracellular signal transmission, etc. in real time. It could not be measured.
Therefore, in order to overcome the above-described drawbacks, a general analysis method of multipoint FCS using a plurality of measurement points by measuring a plurality of measurement points alternately for a short time and using all data after completion of the measurement has been proposed. In this method, measurement, calculation, and display are realized in order using a multipoint sequential measurement method.
However, since analysis calculation is started after measurement of all data is completed, all data positions are used as calculation channels, and all data is handled and repeated correlation calculation is performed, the time required for imaging is long. Therefore, as the measurement time becomes longer and the number of measurement points increases, it becomes practically impractical to perform measurement, calculation, and imaging in real time.
The present invention has been made paying attention to such a problem, and an object of the present invention is to provide an optical signal analysis method capable of accurately analyzing a plurality of measurement points in real time. .
In order to achieve the above object, according to a first aspect of the present invention, there is provided an optical signal analysis method, wherein a light emitted from a measurement point designated with respect to a sample is detected by a photodetector, and the photodetector The time-series step of time-dividing the time-series signal output by a plurality of time-series signals to extract data and the number of data for each measurement point, and the data and the number of data for each measurement point for different delay times Reconstructing, performing statistical calculation using each reconstructed data, performing statistical calculation using each reconstructed number of data, and using the results of the two statistical calculations Find the nature.
The optical signal analysis method according to the second aspect of the present invention is the optical signal analysis method according to the first aspect, wherein a plurality of points related to the sample are selected, light emitted from each measurement point is detected by the photodetector, By dividing and extracting the time series signal output from the photodetector for each of the plurality of measurement points, and performing analysis for each of the divided time series signals, the physical properties of each of the plurality of measurement points of the sample are obtained. Ask.
Further, the optical signal analysis method according to the third aspect of the present invention is the second aspect, wherein the time series signal output from the photodetector is divided into data for each of the plurality of measurement points and stored. Accordingly, the physical property for each of the plurality of measurement points is obtained by reconstructing the divided data for each of the plurality of measurement points and performing analysis.
According to a fourth aspect of the present invention, in the second aspect, the divided data for each of the plurality of measurement points is reconstructed and analyzed, information on the number of data by the data reconstruction is set as a weight, and the weight And the physical property for each of the plurality of measurement points is obtained by a combination of the divided reconstruction data for each of the plurality of measurement points.
According to a fifth aspect of the present invention, in any one of the first to fourth aspects, the light emitted from the sample is fluorescence, phosphorescence, reflected light, or scattered light.
According to a sixth aspect of the present invention, in any one of the first to fifth aspects, the optical signal is analyzed using an autocorrelation analysis method or a photon counting histogram method.
According to a seventh aspect of the present invention, in any one of the first to fifth aspects, the optical signal is analyzed using a cross-correlation analysis method or a coincidence analysis method.
Further, according to an eighth aspect of the present invention, in any one of the first to seventh aspects, the step of reconfiguring comprises: a) measuring data of each measurement section and the number of data (weight). A step of storing for a certain period of time, b) a step of calculating data and the number of data of a plurality of adjacent measurement sections, and c) a calculation result having a measurement section width equal to the sum of the adjacent measurement sections used in the calculation. Storing as new measurement section data and the number of data; d) storing newly measured measurement section data and the number of data for a predetermined time; and e) a plurality of adjacent new measurement sections. A step of calculating the data and the number of data; and f) calculating the data of the next new measurement section having the next new measurement section width equal to the total time of the new measurement section used for the calculation. And comprising the steps of storing as the number of data, and by repeating steps a predetermined number of times f) from the d), determining the physical properties of the sample.
According to a ninth aspect of the present invention, in any one of the first to eighth aspects, the physical property of the measurement point relating to the sample is a translational diffusion coefficient, a rotational diffusion coefficient of the molecule or particle, or the measurement. The number of molecules or particles present in the region, or the fluorescence lifetime.
According to the present invention, accurate analysis can be efficiently performed in a short time for a plurality of measurement points. In addition, the physical properties of a plurality of measurement points can be analyzed simultaneously, and the order of measurement for the plurality of measurement points can be freely combined to analyze sample reactions, interactions between samples, and the like. In particular, when the present invention is realized as a multiple τ system multipoint FCS, it is possible to observe a plurality of point points at the same time as compared to the multiple τ system single point FCS, and to capture various reactions inside and outside the living cells. be able to.
In the case of the multiple τ method multipoint FCS, the calculation time is shorter and more efficient than a general FCS calculation method for calculating a large amount of data and its channel position. First, the τ position (channel) for data calculation is determined, and all the data are handled for calculation, and various calculation processes are limited to a limited number of channel positions.
In the case of multiple τ method multipoint FCS, an average value calculation method is used to reduce the number of calculation data, particularly in a large bin time region, avoid processing data with a large number of repetitions, and an error due to a smooth effect. We are trying to reduce it.
In the case of the multiple τ method multipoint FCS, since one piece of data is handled as a minimum analysis data unit for data reading and calculation processing, instructions such as display can be executed at any time by an interrupt method. Real-time display enables system hardware adjustment, measurement data abnormality detection, cancellation, sample selection, and adjustment.
Since multiple points are analyzed and observed using a single excitation light source and measurement device, there are no hardware errors between the points, and various reactions inside and outside the living cells can be captured under the same conditions. it can.
Furthermore, in the case of the multiple τ system multipoint FCS, it is possible to aim at simplification and efficiency of programming accompanying an increase in the number of points.
First, the outline of the present invention will be described. In the present invention, in the data analysis of a multiple τ method multipoint FCS using weighting factors, a measurement signal obtained by repeatedly measuring a plurality of measurement points in time series is divided for each point, and the data table and the number of data are meant for each point. A weighting coefficient table to be created. Then, only the table data and the weighting coefficient at each measurement point are extracted to calculate the autocorrelation function. Here, it is possible not only to observe cell reactions at a plurality of points at the same time, but also to observe different parts of cells, interactions, and the like, based on the data of each measurement point extracted and the calculation result using the weighting coefficient.
In the data analysis of multiple τ method multipoint FCS using weighting factors, when calculating the correlation function, the channel for calculating data and weighting factors is the octave method (the initial bin time value is doubled every 8 channels). The calculation of data and weighting coefficients is limited to a few finite channel results, and a plot of calculation results at the same interval is realized.
Furthermore, in the data analysis of multiple τ method multipoint FCS using weighting factors, when calculating the correlation function, the data average value and weighting factor average value corresponding to different delay times are calculated in different τ regions first. To do. Since the correlation calculation uses only a few average data and weighting factors, the time required for the calculation is short.
In the data analysis of the multiple τ method multipoint FCS using the weighting factor, when performing the correlation function, various processes using the data of the extracted measurement points and the weighting factor can be performed using one piece of data or the weighting factor. The minimum unit of calculation. Therefore, the calculation can be interrupted at any time by an interrupt command, and the result drawing state can be entered. Real-time multipoint FCS can be realized by simultaneously executing the calculation and the display.

FIG. 3 is a flowchart for explaining the procedure of the multiple τ method multipoint FCS calculation using the weighting factor. Here, the flow of calculation of the multiple τ method multipoint FCS using the weighting factor of FIG. 3 is as follows.
縦軸はI(蛍光強度)であり、横軸はt(時間)である。 First, in step S0, a time series measurement signal is obtained as measurement data for the sample 50 using the configuration of FIG. FIG. 4 shows a time series measurement signal obtained at this time. The vertical axis is I (fluorescence intensity), and the horizontal axis is t (time). Next, it is determined whether or not there is acquired data (step S1). If YES, the process proceeds to steps S2-1 to 2-4, and the measured number of points and point number are simultaneously determined. The data file or signal of the measurement signal includes information on the number of points and the point number, and this information is used to divide the data for each point and the data number information (create a division table).
For example, if the measurement points are four points as shown in FIG. 2 in step S1 and data measurement is performed in the order of 1 → 2 → 3 → 4 → 1, the time series measurement signal is shown in FIG. As shown, four point signals P1, P2, P3 and P4 are arranged in time series. Here, P0 is a random measurement data range during movement of the

Here, the signal shown in FIG. 5 is compared with a specific data table as shown in FIG. 6 to perform data division for each measurement point. In FIG. 6, the value of Point indicates a measurement point, 1 is

Similarly, in step S4, the weighting factor is divided for each measurement point. The multipoint measurement signal shown in FIG. 5 includes information indicating the number of data (point position of data) in addition to the information indicating the data size as shown in step S3. This information is also divided for each point, but in multipoint FCS using the multiple τ method, this information is used as a weighting factor for calculation.
Hereinafter, as with the above-described data division, a weight coefficient division method will be described with reference to a specific weight coefficient table in FIG. FIG. 9 is based on the contents of the table of FIG. 6, and the value of Data in FIG. 9 indicates the number of data measured at four measurement points, and represents one data (weighting factor = 1). This part is a unique analysis method of the multiple τ system multipoint FCS when compared with the multiple τ system single point FCS, and is represented as a weighting coefficient in the analysis algorithm.
A method for creating a weighting factor division table will be described below. First, only the weighting coefficient for the
Next, in step S5, the horizontal axis reference value (channel) used for the final FCS plot, its number, and the number of stages to which it belongs are calculated. In the multiple τ method, the number of channels is determined by the total number of read data, and a specific calculation method is as shown in FIG. In other words, the bin time τ 0 is used as a reference value, and the initial value of the zero stage is set, but the initial value of each stage is set to twice the initial value of the previous stage. The number of channels in each stage is 16 for the 0 stage and 8 for each subsequent stage. When the bin time is 2 μSec, the channel structure is as shown in FIG.
The above channel values are described for each stage as shown in FIGS. FIG. 14 is a general one using the bin time τ 0 as a reference value, and FIG. 15 is a case where the bin time is 2 μSec. As can be seen, the channel value is determined by the bin time, but the number of channels and the number of stages are determined by the total number of data read. How many channels and how many channels can be calculated can be found by calculating the number of channels and the number of steps based on the total number of data read. For example, when data is measured for 32 seconds with a bin time of 2 μSec, the total number of channels for calculation is about 176 and the number of stages is 21.
Next, in step S6, the number of stages is determined, that is, the processing timing for processing data by the multiple τ method is calculated. When calculating the data and the weighting coefficient by the multiple τ method, the timing of each stage is not always the same, and the calculation timing of each stage may be different. In other words, the time when two new data are upgraded in the upper stage is the timing of data movement, sum calculation, and product-sum calculation in the lower stage. For example, the 0th stage of processing is performed each time new data is read, while the 1st stage of various processes is performed when 2 1 = 2 data are newly read, and the nth stage of various processes. Processing is performed when 2 n pieces of data are newly read. FIG. 16 shows the processing timing of each stage.
If the determination in step S6 is no, the process proceeds to step S17 to count the total number of data read. The count result is used in the correlation value total calculation (autocorrelation function or cross-correlation function calculation) for each stage number (step S16). After step S17, the process returns to steps S2-1 to 2-4.
If the determination in step S6 is yes, the process proceeds to step S7 to perform processing for moving each data on the four data tables to the upper row. In other words, the value of each table is upgraded before reading new data.
The movement of the data in each stage is synchronized with the processing timing shown in step S6, and the movement process is performed on the n-stage data when 2 n pieces of data are newly read. The data movement of the 0th stage and the 1st stage is as shown in FIGS.
Next, in step S8, similarly to the data movement in step S7 described above, a process for moving each value in the four weighting coefficient tables to the upper row is performed. That is, each weight coefficient table value is upgraded before reading new weight coefficient data.
The movement of the weight coefficient data at each stage is synchronized with the processing timing shown in step S6, and the n-stage data is subjected to a movement process when 2 n weight coefficients are newly read. Since the movement of the weight coefficients in the 0th stage and the 1st stage can be considered in the same manner as in FIGS. 17 and 18, the description thereof is omitted here.
Next, in step S9, new data is read. That is, one new data is read in the 0th row and the 0th column. Following the movement of step S7, one new data is read in the 0th row and the 0th column. The newly read data is divided into four data tables according to the data point information. The method of dividing the measurement points into data tables is as shown in step S3.
Next, in step S10, a new weighting factor is read. In other words, the weighting factor is read and divided in accordance with the operation of reading one new data in the 0th row and 0th column of step S9. Further, the newly read weight coefficient is divided and substituted into the weight coefficient table for each measurement point, and the division method is as shown in step S4.
Next, in step S11, data in the 0th column of each stage is calculated. This is one of the important parts of calculating data by the multiple τ method. Since the delay time τ after
When the sum calculation process is performed at a plurality of
Next, in step S12, the weighting coefficient of the 0th column of each stage is calculated. This is one of the important parts of the multiple τ method multipoint FCS calculation. Similarly to step S11, the delay time τ after
Similar to the processing in the measurement data, when the sum calculation processing of the weighting coefficients is performed at the plurality of
The next step S13, which is also included in the multiple τ calculation method, calculates the product sum of certain data and data after τ delay. The calculation uses a data table of the same measurement point. In other words, the data of the 0th column of each stage is multiplied by the data of each channel of the same stage (FIG. 23). The sum of multiplication calculations is performed when the data of the same stage is updated to a new value, but the calculation timing of each stage must be synchronized with the above step S6.
Next, step S14 calculates the sum of products between the weighting factors by a unique calculation method of the multiple τ method multipoint FCS. The calculation uses the same weighting factor table of measurement points as in step S13. That is, similarly to the calculation method in step S13, the weighting coefficient of the 0th column of each stage is multiplied by the weighting coefficient of each channel of the same stage. The sum of the multiplication calculations is performed when the weighting factor of the same stage is updated to a new value, but the calculation timing of each stage must be synchronized with the above step S6.
Next, in step S15, the product sum of the data and the weighting coefficient is calculated. This is a unique method of the multiple τ method multipoint FCS, and uses a data table and a weight coefficient table of the same measurement point. That is, multiplication of the data in the 0th column of each stage and the weighting coefficient of each channel in the same stage (FIG. 24) is performed. Alternatively, the weighting coefficient in the 0th column of each stage is multiplied by the data of each channel in the same stage (FIG. 25).
The sum of multiplication calculations is performed when the data and weighting factors of the same stage are updated to new values, but the calculation timing of each stage must be synchronized with the above step S6.
After step S16, the process proceeds to step S19 to execute imaging. Here, an FCS curve is plotted as a calculation result. Since the plot here is based on a single read data, there is no need to wait for the completion of the calculation of a large amount of data as in the conventional FCS calculation method so far. Can be plotted. Then, the process of this flow is complete | finished.
