Abstract
Description
本発明は、電子顕微鏡観測やバイオ分子・物質の検出、診断等に使用される、特定物質を選択的に吸着することが可能な吸着性粒子やそれに用いられる複合粒子、およびそれらの分散体に関するものである。 The present invention relates to an adsorbent particle capable of selectively adsorbing a specific substance, a composite particle used therein, and a dispersion thereof, which are used for electron microscope observation, biomolecule / substance detection, diagnosis, and the like. Is.
A material used in the field of detection, diagnosis, or treatment targeting a biological substance is required to specifically act on a target site or substance or to exist independently from the external environment. However, it generally has the property of acting and adsorbing substances other than the target, which are not actually targeted. Such a property of adsorbing almost all materials called proteins (non-specific adsorptivity) is a detection and diagnostic system for selectively extracting and detecting only proteins having a specific molecular structure. This hindered high performance and high sensitivity.
各種アッセイや診断においてターゲットとなる特定物質を検出するためには、微粒子への吸着を用いた方法が幅広く適用されている。用いられる微粒子は数百μm〜数nmサイズの微小粒径で、比表面積が大きいので、非特異吸着性も大きくなる。その結果、ターゲット物質本来の検出が妨げられ、定性・定量がよく行えない状況であった。 In order to detect a specific substance as a target in various assays and diagnoses, a method using adsorption on fine particles is widely applied. The fine particles used have a small particle size of several hundred μm to several nm and a large specific surface area, so that nonspecific adsorption is also increased. As a result, the original detection of the target substance was hindered, and qualitative and quantitative analysis could not be performed well.
例えば、生物由来の物質を検出する方法の一つである固相免疫測定法では、ターゲットである標識抗体の固相への非特異吸着のため、バックグランドが高くなり、識別感度が鈍くなる。また、免疫電子顕微鏡法で生体材料中にあるターゲット物質を観測する場合、標識材料として使用される金属粒子分散液中の金属粒子へのタンパク質の非特異吸着のため、ターゲット物質の正確な標識が妨害され、システムの精度が劣るという問題点があった。 For example, in a solid-phase immunoassay method, which is one of the methods for detecting a biological substance, the background becomes high due to non-specific adsorption of the labeled antibody as a target to the solid phase, and the identification sensitivity becomes low. In addition, when observing a target substance in a biological material by immunoelectron microscopy, accurate labeling of the target substance is not possible due to nonspecific adsorption of proteins to metal particles in a metal particle dispersion used as a labeling material. There was a problem that the accuracy of the system was inferior due to obstruction.
従来より、この非特異吸着を防ぐために牛血清アルブミンを固相にコーティングする方法や、ポリエチレングリコール(PEG)誘導体、短鎖のエチレンオキサイドを固相表面に付着させる方法が提案されてきたが、不十分であった(特許文献1)。 Conventionally, in order to prevent this non-specific adsorption, a method of coating bovine serum albumin on a solid phase and a method of attaching a polyethylene glycol (PEG) derivative or a short-chain ethylene oxide to a solid surface have been proposed. It was sufficient (Patent Document 1).
Thus, the ability to selectively extract and detect only a substance having a specific molecular structure is hindered due to the property of adsorbing non-target substances that are not targeted.
本発明は、上記の問題点に鑑みてなされたものであり、その目的は、タンパク質を吸着しない複合粒子や、特定のタンパク質等を選択的に吸着することが可能な吸着性粒子、およびその分散体を実現することにある。 The present invention has been made in view of the above-mentioned problems, and the object thereof is composite particles that do not adsorb proteins, adsorbent particles that can selectively adsorb specific proteins, and dispersions thereof. また、本発明に係る分散体は、上記の構成に加えて、上記媒体が水系または有機溶媒または樹脂であることを特徴としている。 The dispersion according to the present invention is characterized in that, in addition to the above configuration, the medium is an aqueous or organic solvent or resin.
この構成により、上記複合粒子や吸着性粒子を、使用に即した安定した形態で提供できる。 With this configuration, the composite particles and the adsorptive particles can be provided in a stable form suitable for use.
In order to produce the composite particles and adsorbent particles of the present invention, first, particles of a core substance are prepared from a compound containing the substance by a chemical reaction such as reduction, or commercially available fine particles are purchased, A colloidal dispersion is performed in an organic solvent, and then an acrylamide-containing monomer solution or a dispersion is added to the dispersion and polymerized to obtain composite particles in which the surface of the core particle is coated with polyacrylamide. Further, this composite particle is added to a solution or dispersion of a compound having two or more functional groups (functional group capable of reacting with polyacrylamide amide reaction site and specific protein) and mixed and dispersed to obtain a composite particle surface. It is obtained by reacting and fixing to the amide reaction site of polyacrylamide. The composite particles and the adsorptive particles can be provided as particles, but it is preferable to disperse them in alcohols such as methanol and ethanol or ethanol / water mixed solvents because they can be easily applied to the next step such as coating.
以上のように、本発明に係る複合粒子は、上記被覆層が、タンパク質を吸着しない物質で形成されている構成である。 As described above, the composite particle according to the present invention has a configuration in which the coating layer is formed of a substance that does not adsorb protein.
これにより、種々の基板等をこの複合粒子でコーティングすれば、その基板にタンパク質が吸着するのを効果的に防ぐことができるという汚染防止効果を奏する。 As a result, if various substrates are coated with the composite particles, it is possible to effectively prevent the protein from adsorbing to the substrate.
また、本複合粒子の表面に官能基含有物質を結合し、特定物質吸着性を付与すれば、特定のタンパク質等を選択的に吸着することが可能な吸着性粒子を実現することができる。 In addition, if a functional group-containing substance is bonded to the surface of the composite particle to impart specific substance adsorptivity, an adsorbent particle capable of selectively adsorbing a specific protein or the like can be realized.
吸着性は、上記複合粒子の表面に、特定物質の吸着性を有する反応性サイトを有する構成で付与できる。 The adsorptivity can be imparted by a configuration having a reactive site having the adsorptivity of a specific substance on the surface of the composite particle.
これにより、特定のタンパク質等を選択的に吸着することが可能な吸着性粒子を実現することができ、コア粒子の光学的特性を利用して、特定タンパク質等に対する標識物質となることができ、特定タンパク質等を感度良く検出できる。 As a result, adsorptive particles capable of selectively adsorbing specific proteins and the like can be realized, and using the optical properties of the core particles can be a labeling substance for specific proteins and the like. Specific proteins can be detected with high sensitivity.
図2に示すように、無機微粒子であるコア粒子11の表面はタンパク質17等の生体物質と非特異吸着をするので、バイオセンサーなどの種々の検出用途において、感度が鈍い。
As shown in FIG. 2, since the surface of the

The property of reacting with a specific substance can directly utilize an amide group which is a side chain of polyacrylamide. In addition, the amide group can be converted to an amino group and used by a simple operation. In addition, a functional group that reacts with a specific substance can be introduced by using a substance having a functional group to be introduced and bonding the functional group to polyacrylamide. Or it can also introduce | transduce through the copolymerization of the monomer used as the main component of a polyacrylamide principal chain, and a functional group containing monomer. The amount introduced can be controlled by the copolymerization ratio. Specifically, acrylamide, acrylic acid, allylamine, and other unsaturated compounds such as an allyl group having a functional group in the side chain and a vinyl group are copolymerized to introduce the functional group.
ここで示す官能基とは、アミノ基、カルボキシル基、マレイミド、スクシンイミド系、チオール、ジスルフィド、イミドなど、特定物質に対し反応性を有するものである。 The functional group shown here has reactivity with a specific substance such as amino group, carboxyl group, maleimide, succinimide, thiol, disulfide, and imide.
The acrylamide includes acrylamide and its derivatives, and polyacrylamide can also be crosslinked, especially if it is a bifunctional monomer such as N, N'-methylenebisacrylamide. It is also possible to control the size of the composite particles by changing the mixing ratio of the monomers, particularly the crosslinking agent monomer. Crosslinking is possible even after formation of the composite particles. The polyacrylamide may or may not be crosslinked. In order to obtain better effects, ease of handling and stability, it is desirable to crosslink.
PEG誘導体を表面に有する複合粒子は、従来、分散体として他の媒質中に分散した形での提供に留まっていたが、本発明のポリアクリルアミドで被覆された複合粒子は、安定な粉末としても活用できる。そのため、本発明の複合粒子は、表面のアミド基、官能基、官能基を使って化学修飾させた置換基の効果などにより、水、有機溶媒、高分子等の媒質に自由に分散利用でき、使用用途範囲を拡大できる。 Conventionally, composite particles having a PEG derivative on the surface have only been provided in the form of a dispersion dispersed in another medium. Therefore, the composite particles of the present invention can be freely dispersed and utilized in a medium such as water, an organic solvent, or a polymer due to the effect of the surface amide group, functional group, a substituent chemically modified using the functional group, etc. The range of usage can be expanded.
さらに、PEG誘導体は、分子量分布等から粒径制御をすることが困難であるが、本発明の複合粒子は、粒径の制御が容易にできるので、目的に合わせたサイズの微粒子を提供することができる。 Furthermore, it is difficult to control the particle size of the PEG derivative due to the molecular weight distribution or the like, but the composite particle of the present invention can easily control the particle size, and therefore provides fine particles having a size suitable for the purpose. That is, in FIG. 5, A is a case where PAAm is coated (Example 1), and B is a case where PAAm is not coated (Comparative Example 1). Similarly, in FIG. 6, C is a case where PAAm is coated (Example 2), and D is a case where PAAm is not coated (Comparative Example 2). From the above absorption spectrum, it was shown that protein adsorption was suppressed by coating the core particles with PAAm.
Moreover, the protein adsorptivity of the core particles before and after the PAAm coating was evaluated by examining the absorbance at a wavelength of 562 nm. In addition, since absorption of the comparative examples 1 and 2 exceeded the measurement range, each solution was diluted and used. Therefore, the absorbance was calculated by uniformly adjusting the concentration between the comparative example and the example. As a result, the absorbance of Example 1 was 0.0319, and the absorbance of Comparative Example 1 was 1.07. The absorbance of Example 2 was 0.013, and the absorbance of Comparative Example 2 was 0.765. According to this evaluation result, the amount of protein adsorbed is suppressed to several tenths compared to before the PAAm coating. Furthermore, since the reference experiment showed that the slight absorption observed in the sample of Example 1 was due to measurement error and base-up, as a result, the protein adsorption degree of the sample of Example 1 was limited. It was thought that it converged in the range close to zero.
〔金−ポリアクリルアミド吸着性粒子の官能基確認結果〕
上記官能基確認の結果、波長425nmをピークトップとする吸収が観測された。したがって、実施例1、2のサンプル各々にアミノ基が導入されていることが示された。このように、簡単な手法で官能基導入を達成できることが示された。
[Results of functional group confirmation of gold-polyacrylamide adsorbent particles]
As a result of the above functional group confirmation, absorption having a peak top at a wavelength of 425 nm was observed. Therefore, it was shown that an amino group was introduced into each of the samples of Examples 1 and 2. Thus, it was shown that functional group introduction can be achieved by a simple method.
