Abstract
Description
1)抗原及び免疫調節剤
一本鎖及び二本鎖合成RNA18種のパネル(表1参照)をシグマ社(Sigma)から購入し、無菌PBSに溶解した。これらのRNAはプールとして或いは個々に使用した。卵白アルブミン(OVA、低エンドトキシン)はシグマ社(A7641)から購入した。コレラ毒サブユニットB(CTB)はカルビオケム(Calbiochem)(カタログ番号227039)から、完全フロイントアジュバント(CFA)はDIFCO(カタログ番号263810)から、ヒトIgG(hIgG)はシグマ社(カタログ番号I4506)から購入した。コンホメーションエピトープを保ち且つ三量体化する能力を有する組換えgp140 HIV抗原は、停止変異(stop mutation)を導入することにより、IIIB株のgp160エンベロープタンパク質から得た。この抗原は、ベルナルド・モス(Bernard Moss)博士(N.I.H.)の厚意により提供されたワクチニアウィルスベクターによってBS−C−1(ATCC)細胞内で発現し、レンチルレクチンセファロースクロマトグラフ(ファーマシア(Pharmacia)、ニュージャージー州ピスカタウェイ)によって精製した。gp140抗原であることは、フィッツジェラルド(Fitzgerald)(カタログ番号20−HG81)から購入したHIVエンベロープ特異的抗体を用いたウェスタンブロット分析によって確認した。インフルエンザウィルス(A/WSN/32H1N1株)はMDBK細胞で増殖させ、上清からショ糖勾配遠心法によって精製した。ウィルスの不活性化のために、ウィルス粒子を攪拌しながら15分間、短波長UV光に曝露した。不活性化は、許容MDCK細胞へのウィルス滴定により確認した。可変領域内にI−Ed−拘束性ヘマグルチニン由来ペプチドSFERFEIFPKE(IgHA)[配列番号1]を有する組換えマウスIgG2bを得て、先に特徴付けたように精製した。
6〜8週齢のC57BL/6、BALB/c及びTLR4−/−C3H/HeJ雌性マウスを、ジャクソンラボラトリ(Jackson Laboratories、マサチューセッツ州バーハーバー)から購入し、アライアンス・ファーマスーティカル社(Alliance Pharmaceutical Corp.)で特定の病原体条件に収容した。C57BL/6及びBALB/cマウスにおける観察結果の主要なものは、エンドトキシンに対する応答性を欠損したC3H/HeJマウスにおいて再現された。雌性のSprague Dawleyラット(250〜330グラム)はタコニックファーム(Taconic Farms)から購入し、同様の条件下に置いた。
マウスとラットは前述のように各々気管内点滴、噴霧投与によってプライミングし、マウスの場合は、鼻腔内に二週間の間隔で二回追加免疫を行った。大量域寛容の誘導のために、それらのマウスを静注によってプライミングした。最後に、強い免疫応答を誘導するために、マウスをCFAで乳化した抗原の皮下注射により免疫感作した。プライミング、追加免疫、或いは寛容の誘導に用いた抗原の量は、OVA−100μg、HIV gp140−10μg、hIgG−200μg、ショ糖精製UV−WSN−20μgであった。合成RNAは、一用量につき40〜50μgの量を、抗原と共に或いは抗原なしで、SCL COMPLEXESに添加し或いは添加せずに用いた。CTBの一用量当たりの含有量は10μgであった。抗原は、生理食塩水に添加して導入するか、或いは処方の際にSCLマトリクスと適合する不活性ビヒクルであるパーフルオロカーボン(パーフルブロン(perflubron)(未希釈なパーフルオロオクチルブロマイド)、リクイベント(Liquivent()、アライアンス・ファーマスーティカル社)に添加して導入する(点滴或いは噴霧投与の全量は40〜45μL)。
リン脂質を主な賦形剤として短鎖脂質複合体(「免疫複合体」)を得る技術プロセスは噴霧乾燥であった。ここではこのプロセスのより簡略化したバージョンを用いた。即ち、リン脂質を水にホモジネートし(リポソーム或いはミセルの形成のため)、賦形剤と活性剤を混合し、次いで噴霧乾燥した。詳細には、噴霧乾燥の直前に、2種の調製物AとBを混合して水性組成物を調製した。調製物Aは、0.14gのジオクタノイルホスファチジルコリン(アバンティポーラーリピッズ社(Avanti Polar Lipids))を23mLの温DI水に溶解したミセル調製物を含んでいた。このリン脂質ミセル調製物に、0.0357gのCaCl2・2H2Oと0.714grのラクトースを溶解した。調製物Bは、20mgの卵白アルブミン(シグマ社)と5mLのPBSに溶解した4mgのpA:pU(エンドトキシンは含まず)とを含んでいた。フィード用混合物(2mL調製物A+調製物B)を標準B−191ミニ噴霧乾燥機によって次の条件で噴霧乾燥した。入口温度70℃、出口温度43℃、アスピレータ90%、ポンプ2.2mL/分、窒素流量2400L/時間。得られた複合体のPL:OVA:pApU:CaCl2・2H2O:ラクトースの重量比は12:20:4:3:61であった。
抗体応答はELISAによって測定した。即ち、ウェルを抗原(2μg/mLのgp140、8μg/mLのショ糖精製ウィルス、10μg/mLのhIgG又は10μg/mLのOVA)でコーティングし、シーブロック(SeaBlock)(ピアース社(Pierce)、イリノイ州ロックフォード、カタログ番号37527)でブロッキングした。血清と気管支肺胞洗浄液の段階希釈液は、室温で少なくとも2時間インキュベートした。洗浄後、アルカリホスファターゼ(シグマ社、カタログ番号A7434)結合抗マウスIgG抗体を用いたアッセイを行い、基質(pNPP、シグマ社、カタログ番号N2765)を添加し、ソフトマックス(SoftMax)ソフトウェアを備えた自動ELISAリーダー(モルキュラーデバイス社社(Molecular Devices)、ThermoMax)で測定した。
前日に合成RNAで処置したマウスと対照マウスの肺におけるケモカインの発現量を、次に記述するDNAアレイ技術を用い測定した。全RNAをRNeasyキット(キアゲン社(Qiagen)、カリフォルニア州バレンシア)を用いて肺から単離した。このRNAをRNase−フリーDNaseI(ストラタジーン社(Stratagene)、カリフォルニア州サンディエゴ)で処理し更に精製した。DNAアレイはスーパーアレイ社(SuperArray Inc.、メリーランド州ベサダ)のNonrad−GEArrayキットを用い行った。即ち、cDNAプローブをビオチン−16−dUTPを含有するdNTPmixと共にMMLV逆転写酵素を用いて合成した。GEArray膜を68℃で1〜2時間プリハイブリダイゼーションした。ハイブリダイゼーションは、ビオチン標識cDNAと共に膜をインキュベートすることにより行った。ハイブリダイズした膜を、2×SSC−1%SDSで2回、0.1×SSC−0.5%SDSで2回洗浄した。膜をアルカリホスファターゼ抱合型ストレプトアビジン(バイオソース社、カリフォルニア州カマリロ)と共に更にインキュベートし、最終的に、CDP−Star化学発光基質を用い現像した。シグナルの強度は、Gel−Proソフトウェアを備えたImage−Pro解析システム(メディアサイバネティックス社、メリーランド州シルバースプリングス)によって測定した。
前日に合成RNA又は生理食塩水を用い処理をしたマウスの肺から得た細胞浮遊液を、前述のように、コラゲナーゼ消化とフィコール勾配遠心によって調製した。細胞を1%(v:v)マウス血清(シグマ社、カタログ番号M5905)と1%(w:v)ウシアルブミン(フラクションV、シグマ社、カタログ番号A3059)を含むリン酸緩衝生理食塩水に再懸濁し、PE標識ラット抗マウスCD11b(ファーミンジェン社、カタログ番号01715A)、PE標識ハムスター抗マウスCD11c(ファーミンジェン社、カタログ番号09705A)、或いは適切なPE標識アイソタイプ対照(ファーミンジェン社、カタログ番号11125A又は11185A)を、106個の細胞に対して1μgの抗体となる濃度で氷上で40分間染色した。分析はベクトンディッキンソンFacsCalibur装置によって行った。ヨウ化プロピジウムを用い、生育不能細胞をゲートアウトした。
CD11c+樹状細胞等のプロフェッショナルAPCを、BALB/cマウスの脾臓から、ラット抗マウス抗CD11c抗体(ミルテニイバイオテック社(Miltenyi Biotech))を結合させ磁気ビーズを用いて分離した。即ち、単一細胞浮遊液を、MACS緩衝液(BSAとEDTA)に107細胞/mLで再懸濁し、磁気ビーズと共に氷上で15’インキュベートし、洗浄し、磁気カラムに通した。溶出前にカラムを3回洗浄し、次いで2回連続して洗浄し、50μg/mLのRNAモチーフ又は5ng/mLのrIL−12(バイオソース社、カリフォルニア州カマリロ)と共に、又はなしで100μg/mLのIgHAで一夜インビトロパルシングした。あるいは、ラット抗マウスCD40モノクロナール抗体(BDファーミンジェン社)を事前にコーティングしたウェル上で、細胞をIgHAと共に一夜インキュベートした。細胞を洗浄し、平衡無菌生理食塩水に再懸濁し、皮下注射によってナイーブBALB/cマウスに養子免疫細胞移入した(2.5×105APC/マウス)。前述のようにHAクラスII拘束性ペプチドによって刺激した後、T細胞応答を第14日にIL−2 ELISPOT分析によって測定した。
免疫応答の大きさは、値の正規分布と等分散を仮定し、t検定を用いて比較した。
1)免疫反応を調節するRNAモチーフの系統的定義
ウィルス感染の際には、一過性の非通常RNA種が生成され、「危険」シグナルとして活動する。従って、複数の多様なRNAモチーフが生来の免疫細胞によって認識され、適応免疫応答を大きく調節すると仮定された。この仮定を系統的な原理から検証するために、合成一本鎖及び二本鎖RNAモチーフのライブラリーを、経気道投与タンパク抗原(OVA)に対する特異的IgG応答を調節する能力に関してスクリーニングした。プロセスを簡略化するために、スクリーニングを次の2ラウンドに設定した。(1)ラウンド1はRNA種のプールを用い(表1)、(2)第2ラウンドでは免疫応答に最大の影響を与えるプール内の要素を詳細に調べる。
ssRNA:ポリヌクレオチド(ポリA、ポリU)は、ヌクレオチドとポリヌクレオチド−ホスホリラーゼを用い酵素的に調製されるが、調製プロセスには動物由来の材料は入らない。dsRNA:ポリアデニル酸(ポリA、pA)をポリウリジル酸(ポリU、pU)と共にアニール。一般に、本発明のdsRNA及びssRNAはホモポリマーであり、dsRNAの場合は、単一の塩基或いはヌクレオチド(例えば、アデニン)が一本の鎖を一貫して形成し、その相補が他の鎖を一貫して形成する。ssRNAの場合、一本鎖は同一のヌクレオチドで一貫して形成される。しかしながら、混合ヌクレオチド(dsRNAの場合はその相補も)で形成されたdsRNA或いはssRNA組成物の使用は、本発明の範囲内である。本発明のdsRNA及びssRNA組成物は、塩基/ヌクレオチドであるアデニン(A)、グアニン(G)、シトシン(C)、ウラシル(U)、及びイノシン(I)を含む。表1及び図8AのRNA組成物は、実施例で用いられる各種RNA組成物を説明するものである。本発明のRNA組成物は、実施例27で調製され精製された。
各RNAプール(表1)が適応免疫に与える影響を、OVAの経気道同時免疫感作C57B1/6マウスを用いて測定した。「材料及び方法」の章に記載のように、抗体反応は、IgG終点力価(n=4/群)における平均±SEMとして示す。対照として、無菌PBSに添加したOVA、コレラ毒サブユニットB(CTB)と組み合わせたOVA、更にPBSだけのものを用いた。図1Aに示すように、dsRNAに対応するプールが、抗体反応に最大の影響を与え、この特定の免疫を実質的に増強した。一方、互いに相補的な一本鎖種の混合物については、この増強は部分的なものにとどまった。
RNAプールの代わりに個々のdsRNAを用いた以外は前記実施例と同様に「材料及び方法」の章に記載の方法で実験を行った。結果を図1Aと同様の仕方で図1Bに示す。この結果は、独立した2種類の実験を体現しているものである。INSET:OVAに対する平均IgG2a力価と平均IgG1力価の比。左から右への順序はメインパネルのものと同じである(PBS OVA、CTB OVA、pC:pG OVA、pI:pC OVA及びpA:pU OVA)。この2ラウンド目のスクリーニングにおいては、図1Bに示すように、二本鎖RNAとして構成された2種類のモチーフ、即ちpA:pU及びpI:pCが、C57BL/6マウスにおける特定の抗原に対するIgG応答の発現に大きな影響を与えることが示されている。エンドトキシンに対する応答性が低いTLR4−/−C3H/HeJマウスでも同様の結果が得られた(図示せず)。
ELISPOT分析により得られた結果は、脾臓1個当りのIFN−γ及びIL−4スポット形成コロニー(SFC)の数の平均±SEMで示す(n=4/群)。図1Cに示すように、pA:pUとpI:pC(40μg、「材料及び方法」参照)の両方が特定のT細胞応答に対する増幅効果を有していた。驚くべきことに、特定の抗原に対するT細胞応答のプロフィールはRNA残基の性質に依存しており、これは、免疫系がRNA関連危険モチーフを区別できることを示している。A残基及びU残基を含有するRNAモチーフは、I及びCを含有するモチーフよりも、C57BL/6マウスにおけるIFN−γ−産生Th1細胞(図1C)の分化を方向付けることができる。この結果は、IgGアイソタイプの誘導が異なることを反映しており、Thプロフィール(図1B−INSET)と一致している。
図2Aに示すように、HIV gp140(10μg、「材料及び方法」参照)断片に対する抗体反応に及ぼす影響を、気道を抗原のみで感作した或いは抗原と共にpA:pU(40μg、「材料及び方法」参照)で感作したC57BL/6マウスを用いて測定した。結果はIgG終点力価(n=3/群)の平均±SEMで示す。
実施例4に示すように、UV不活性化WSNウィルス(UV−WSN)(20μg、「材料及び方法」参照)のみによる粘膜免疫感作、或いは該ウィルスと一緒に各dsRNAモチーフ(50μg)を用いて粘膜免疫感作を行った後、インフルエンザウィルス特異IgG抗体を測定した。対照として、同じ株のインフルエンザウィルス(n=4/群)による感染後の抗体反応を用いた。結果は、図2BにIgG終点力価の平均±SEMとして示す。
全インフルエンザウィルスに対するT細胞応答を、脾臓細胞のELISPOT分析により測定し、バックグラウンドを引いたIFN−γ、IL−4及びIL−2 SFC(平均±SEM、n=4/群)で示す。pA:pUを用いた場合の抗原に対するT細胞応答を、抗原のみで免疫感作を行った後の応答、或いはインフルエンザウィルス感染後の応答と比較した(図2C参照)。
dsRNA関連危険モチーフ(pA:pU、pI:pC等)は自然免疫成分を介して間接的にT細胞応答に影響を及ぼすと仮定した。この仮説を検証するため、RNA投与後の肺リンパ球様組織におけるケモカイン遺伝子の発現を決定した。
RNAモチーフによるケモカイン遺伝子の局所的アップレギュレーションを、経気道処理の1日後、DNAアレイ技法により測定した(材料及び方法「ケモカイン遺伝子発現の測定」参照)。結果を、非処理マウスの肺組織で測定された発現レベルに対する増加倍率で示す。dsRNAにより惹き起こされたケモカインの発現を、LPSにより誘導されたものと比較した。Th1細胞及びTh2細胞と選択的に結合するケモカインはそれぞれ、実線及び破線で囲って示した(図3A)。
dsRNA処理したマウスの肺におけるプロフェッショナルAPCの増加を、処理1日後、FACS分析により評価した。結果を、肺間質組織における全細胞数に対するCDllc+細胞数及びCDllb+細胞数の百分率(n=4/群)として図3Bに示す。CD11b+(単球)はpA:pUとpI:pCとにより増加するが、これはケモカインのアップレギュレーションと一致する(実施例7参照)。対照的に、CD11c+樹状細胞の増加を有効に誘導したのはpI:pCだけであった。dsRNAモチーフは投与部位におけるAPCを増加すると結論される。
dsRNAモチーフによるプロフェッショナルAPCの活性化を次の方法で確認した。まず、ex vivoで抗原と共にdsRNAを用いてCD11c+細胞のパルシングを行い、ナイーブBALB/cマウスに養子移入実験を行い、T細胞応答を測定した(図3C)。対照として用いたものは、抗原パルスAPC、或いはrIL−12と抗CD40mAbとで共刺激した抗原付加(loaded)であった。結果は、ELISPOT分析で見積もられる脾臓中のIL−2SFCの数として図3Cに示す。
dsRNAモチーフにより刺激されるクロスプライミング(APCが、クラスI拘束性T細胞を感染なしでプライミングする能力を得る特別な状況をいう)を、BALB/cマウスを用いて検討した。マウスは、人工的に組換えを行ったHIV gp140抗原(10μg)と共にpA:pUで処理して用いた。検討は、MHCクラスI拘束性コグネイトペプチドを用いたin vitro刺激によるELISPOT分析により行った(「材料及び方法」参照)。対照として、用量を合わせたgp140抗原を用いた。結果は、脾臓1個当りのIFN−γ及びIL−4SFCの数の平均±SEM(n=4/群)として図3Dに示す。
dsRNAモチーフにより刺激されるクロスプライミングをC57BL/6マウスを用いて検討した。マウスは、全OVAと共にpA:pUで処理して用いた。検討はMHCクラスI拘束性ペプチドを用いたin vitro刺激によるELISPOT分析により行った(「材料及び方法」参照)。対照として、用量を合わせたOVA抗原又は無菌PBSを用いた。結果は、脾臓1個あたりのIFN−γ及びIL−4SFCの数の平均±SEM(n=4/群)として図3Eに示す。
危険分子は、無害の抗原と、感染プロセスに関連する抗原とを区別するのに関与している。高用量の場合、非感染性精製タンパク抗原は不応答即ち免疫寛容を誘導する。自己或いは無害の抗原に対して寛容を獲得する中心的な方法は「免疫無視(immunological ignorance)」或いは「免疫寛容」である。前者では、抗原は、空間的分離によりAPCにアクセスできない。後者では、抗原はAPCにアクセスでき、細胞内に取り込まれてプロセシングされるが、共刺激が少ない場合、エピトープが提示される。正味の結果が、T細胞のこのレベルにおける免疫不応答即ち寛容となり得る。感染や免疫攻撃の場合、「免疫無視」や「寛容」を防ぐメカニズムが存在する。このようなメカニズムは、共刺激分子や炎症性ケモカイン、炎症性サイトカインの発現の活性化を伴う、APCの新規な移動パターンの誘導により発生する。その結果、「危険分子」が存在することによって生じた環境において、免疫系が曝されている全ての抗原に対して、無視や寛容よりもむしろ強い免疫応答が生じる。例えば、腫瘍関連抗原は、免疫エフェクターにより無視されたり寛容原性との関連において提示されることも多い。このような抗原に対する免疫受容能を回復するための手段の直接的実用化は、抗癌治療において有意義である。pA:pUモチーフ及びpI:pCモチーフの危険シグナルとしての適格性を調べるため、hIgGの静脈接種による寛容モデルを用いた。
まず、マウスに、標準的な寛容原性用量(200μg)のhIgGのみを静注し(黒印)、或いはこれと共にpI:pC或いはpA:pU(40〜50μg)を静注し(白印;図4A参照)、その後、免疫原性用量(50μg)のhIgGをCFAに乳化させたものを皮下注射し追加免疫を行った。hIgGに対する抗体の力価を、追加免疫後、異なる時間間隔でELISAにより測定した。対照として、CFAに添加したhIgGで免疫感作したマウスを用いた。最大力価(破線)を示す。結果は、終点力価の平均±SEM(n=5/群)として図4Aに示す。
危険モチーフは、免疫防御の防御体制を即座に高める能力を有すると仮定した。従って、pA:pUとpI:pCが、インフルエンザウィルスの初期感染の進展にどのような影響を与えるかについて調べた。
非定式化サブユニットワクチン(一般には精製したタンパク抗原)に対する免疫応答は最小規模のものであるため、同じ微小構造に含まれるAPCを抗原とdsRNA危険モチーフとに生体内で同時接触させ、更に良い結果が得られるかを調べた。この試験のため、プロトタイプの抗原(OVA)をpA:pU或いはpI:pCと組合せ調製し、賦形剤として生体適合性があり且つ免疫学的に不活性なリン脂質(ジオクタノイルホスファチジルコリン等)及び乳糖を加えて短鎖脂質複合体(「SCL」)の形態とした(「材料及び方法」参照)。短鎖脂質複合体に調製したOVA+pA:pU或いはpI:pCをC57BL/6マウスの気道に投与した際の抗体反応を測定した所、この抗体反応は、生理食塩水中の非定式化抗原で免疫感作したマウス、或いはdsRNAモチーフなしで調製しSCL複合体とした抗原で免疫感作したマウスの抗体反応に比べ非常に高かった(図5A)。
短鎖リン脂質からなり、モデル抗原(OVA)のみをロードした短鎖脂質複合体、及びモデル抗原(OVA)とdsRNAモチーフとをロードした短鎖脂質複合体を作成し、C57BL/6マウスを用いて試験した(図5A)。抗体反応はELISAで測定した。結果は、経気管免疫感作2週間後に行ったIgG終点力価の平均±SEM(n=4/群;データは独立した2種類の測定結果である)で示す。対照として、PBS中のOVAと、OVAをCTB(コレラ毒B)と組合せ調製して短鎖脂質複合体(ジオクタノイルホスファチジルコリン)としたものとを用いた。この結果、RNAモチーフの免疫特性を保存し抗原及びdsRNAを含む分子複合体が作成され、これは実用に耐えるものであることが分かった。
図5Bは、C57BL/6マウスにおける、全OVA抗原又はクラスI拘束性優性OVAペプチドに対する局所(肺)T細胞応答及び全身性(脾臓)T細胞応答を測定した結果を示す。マウスは、OVAのみを短鎖脂質複合体(ジオクタノイルホスファチジルコリン)としたもの、或いはOVAとpA:pUとを短鎖脂質複合体(ジオクタノイルホスファチジルコリン)としたもので免疫感作した。分析はELISPOTにより実施し、結果はIFN−γSFC/器官(平均±SEM;n=4/群)として示した。
OVAとdsRNAとを組合せ調製した脂質複合体でラットの免疫感作を行った。対照として、抗原を含まないSC脂質複合体、OVAは含むがdsRNAモチーフは含まないSCL複合体、及び用量を合わせたOVAをそれぞれ生理食塩水に添加し用いた。結果を、ELISAにより測定した、血清(図5C)及び35日目の気管支肺洗浄液(図5D)に含まれるOVA特異的抗体の終点力価(平均±SEM;n=4/群)として示す。
dsRNA等の危険モチーフを含まない抗原は、免疫原性が弱いか、高用量の場合は免疫寛容を誘導する。しかしながら、dsRNAモチーフは、免疫系が抗原を認識する方法を変化させる。即ち、このようなモチーフは、弱い応答性や寛容ではなく、共存する抗原に対し強く応答するように適応(T細胞及びB細胞)免疫を導くと共に、免疫寛容を阻害する或いは阻止する。従って、dsRNAモチーフの認識による自然免疫は、B及びT細胞適応免疫のマスタースイッチとして作用する(図7)。
(A)マウスを各RNAプールで気管内免疫感作し、経鼻注入による追加免疫を2週間毎に2回行った。抗体反応はELISAにより行い(図8B)、結果はIgG終点力価の平均±SEMとして示す(n=4/群)。対照として、OVAを無菌PBSに添加したもの、OVAをコレラ毒サブユニットB(CTB)と一緒に用いたもの、PBSのみをそれぞれ用量を合わせて用いた。即ち、ウェルを抗原(10μg/mLのOVA)でコーティングし、シーブロック(ピアース社、イリノイ州ロックフォード、カタログ番号37527)でブロッキングした。血清と気管支肺胞洗浄液の段階希釈液は、室温で少なくとも2時間インキュベートした。洗浄後、アルカリホスファターゼ(シグマ社、カタログ番号A7434)結合抗マウスIgG抗体を用いたアッセイを行い、基質(pNPP、シグマ社、カタログ番号N2765)を添加し、ソフトマックスソフトウェアを備えた自動マイクロタイタープレートリーダー(モルキュラーデバイス社、ThermoMax)で測定した。
(A)dsRNAモチーフにより刺激されるクロスプライミングをBALB/cマウスを用いて検討した。マウスは、10μgの人工的に組み換えたHIV gp140抗原とpA:pUとで処理(プライミング+2回の追加免疫)して用いた。応答は、実施例19に記載したように、V3ドメイン由来のMHCクラスI拘束性コグネイトペプチドR10Kによるin vitro刺激を用いたELISPOT分析により測定した。対照として、用量を合わせたgp140抗原を用いた。結果を、IFN−γ及びIL−4 SFCの数/脾臓(n=4/群)の平均±SEMとして図9Aに示す。

I RNAモチーフの標識:
1.次のプロトコルにより、各RNAモチーフをULYSIS Alexa488標識でタグ化した。
1.C57BL/6マウス(雌性、4匹)から脾臓細胞及び肺細胞を単離;
・肺細胞は、脾臓細胞とは異なり、切り刻んだ後、コラゲナーゼ緩衝液中37℃で30分間インキュベートしてから、次のステップに進むことを要する;
・70umファルコン細胞ストレーナを通す;
・洗浄後、再度MACS緩衝液に懸濁させる:
2.推奨プロトコルに従い、CD11b特異的又はCD11c特異的MACSビーズで標識する;

・酢酸ナトリウム及びエタノールを用いて−70℃でpA:pUを沈澱させた。
・pA:pUを熱変性させ、Alexa Fluor488試薬により90℃で標識した;
・反応を停止させ、標識されたpA:pUをエタノール沈澱させた。
THP−1細胞を2×106細胞/mLの濃度で懸濁させた;
この懸濁液(50μL、5×104細胞)を12×75mmチューブに入れた;
非タグ化pA:pU又はpI:pCを20又は100μg/mLの濃度でTHP−1細胞に添加し、15分間インキュベートした;

Claims (21)
- 二本鎖RNAを含む組成物を対象に投与することを含む、対象の抗原に対するT細胞応答を増強する方法。
- 前記組成物と前記抗原とを同時投与することを更に含む、請求項1に記載の方法。
- 対象に二本鎖RNAを投与した後に、抗原を投与若しくは接触させる、請求項1に記載の方法。
