The present invention relates to devices and systems for handling microscale fluids. Even more particularly, the invention relates to a fluidic interface port for providing bi-directional fluid interfacing in a microfluidic system.
BACKGROUND OF THE INVENTION In the chemical, biomedical, biological sciences and pharmaceutical industries, it is increasingly desirable to perform the majority of chemical operations such as reactions, separations, and subsequent detection steps in a highly parallel manner. . High-throughput synthesis, screening, and analysis of (bio) chemical compounds enables the economic discovery of new drugs and drug candidates and the realization of high-performance medical diagnostic devices. Increased speed, enhanced reproducibility, reduced consumption of expensive samples and reagents, and reduced waste are crucial for improving the chemical operations required for these applications.
Microfluidic devices and systems provide improved methods for performing chemical, biochemical and biological analysis and synthesis. Microfluidic devices and systems allow for the implementation of multi-step, diverse chemical operations in chip-based microchemical analysis systems. Chip-based microfluidic systems generally include conventional “microfluidic” elements that are particularly capable of handling and analyzing chemical and biochemical specimens. Typically, the term microfluidic in the art refers to a system or device having a network of processing nodes, chambers and reservoirs connected by channels having typical cross-sectional dimensions ranging from about 1.0 μm to about 500 μm. Say. In the art, channels having such cross-sectional dimensions are referred to as “microchannels”.
By implementing chemical operations in a microfluidic system, a number of desirable improvements described above can potentially be realized. Due to the smaller dimensions, diffusion processes such as seed heating, cooling and passive transport (diffusion material transport) can proceed more rapidly. One example is liquid heat treatment, a step typically required in chemical synthesis and analysis. Compared to the heating and cooling of liquid in a beaker, which is performed in a conventional laboratory environment, the heat treatment of the liquid is accelerated by the short diffusion distance in the microchannel. Another example of the efficiency of a microfluidic system is the mixing of species dissolved in a liquid, which is a diffusion-controlled process. The miniaturization of the typical dimensions of the mixing chamber reduces the typical distance overcome by diffusive material transport and consequently shortens the mixing time. As with heat treatment, mixing dissolved chemical species, such as reagents, with precursors for a sample or synthesis step is an operation required for virtually all chemical synthesis and analysis processes. Thus, the ability to reduce mixing time provides significant advantages for most chemical synthesis and analysis processes.
Chem. 65: 2637-2642 10月(1993)、Effenhauserら、Anal. Chem. 66: 2949-2953 9月(1994)、Jacobsonら、Anal. Chem. Chem. Furthermore, the reduction in size enhances the separation operations used in chemical synthesis and analysis processes. One example is capillary electrophoresis, which is a separation technique based on the movement of dissolved charged species through a capillary filled with liquid by application of a longitudinal electric field. By reducing the cross-sectional size of the capillary, the separation efficiency can be greatly improved, thereby speeding up the separation. For example, Effenhauser et al., Anal. Chem. 65: 2637-2642 October (1993), Effenhauser et al., Anal. Chem. 66: 2949-2953 September (1994), Jacobson et al., Anal. Chem. 66: 4127-4132 See December (1994) and Jacobson et al., Anal. Chem. 66: 1114-1118 April (1994).
Another aspect of dimensional reduction is the reduction in the required volume of samples, reagents, precursors and other chemicals that are often very expensive. Milliliter-sized systems typically require milliliter volumes of these materials, while microliter-sized microfluidic systems only require microliter volumes. The ability to perform these processes using smaller volumes greatly reduces costs and allows for economical operation of chemical synthesis and analytical operations. As a result of the reduction in required capacity, the amount of chemical waste produced during the chemical operation is correspondingly reduced.
It can be concluded that the reduction in dimensions associated with microfluidic systems can accelerate important chemical operations, while at the same time leading to compound consumption and chemical waste reduction.
There are numerous uses for microfluidic systems. To interfacing fluids such as samples, analytes, reagents, precursors for synthesis, and buffers toward, within, or between microfluidic systems A number of methods have been described. In conventional microfluidic systems, the structures and methods used to introduce samples and other fluids into the microfluidic substrate limit the capabilities of the microfluidic system. For example, a conventional microfluidic system may include a separate sample introduction channel for introducing a sample into a microchannel for processing. The sample is first introduced into the sample channel and transported through the sample channel to the microchannel. Another method for introducing fluid involves the use of a sample well or reservoir in communication with the microchannel to hold a relatively large supply of sample. A reservoir is a structure that contains a significantly larger volume of fluid than a microfluidic channel. A relatively small amount of feed sample in a sample well or reservoir is introduced into the microchannel.
The total number of samples and other fluids that can be processed on the microfluidic substrate is currently limited by the size and / or number of reservoirs through which these fluids are introduced into the microfluidic system Has been. The disadvantages of known structures and methods for introducing fluids into microfluidic systems are the use of much larger volumes of fluid than required for microfluidic analysis and the significant size inefficiency and sample loss. It is a move. Furthermore, with conventional methods of introducing fluid into a microfluidic system, it is difficult to control the amount of sample introduction that is ultimately introduced into the microchannel after passing through the sample channel or reservoir.
The method of fluid communication with the microfluidic system is by mechanical micropumps and valves, see US Pat. No. 6,033,191 and US Pat. No. 5,529,465. A major drawback of this approach to fluidic interconnection is the complex configuration and operation of these micropumps and valves. Another drawback is the relatively large size and internal capacity when compared to the internal capacity of the microchannel. Often there are multiple orders of magnitude differences between these two capacities, and the resulting inconsistencies make micropumps unattractive to interconnect with many small sized microchannels. .
US Pat. No. 5,173,163 describes a method and apparatus for introducing a fluid sample into a microcapillary for electrophoretic separation. In this method, the liquid is introduced into the capillary by introducing one end of the separating capillary into the tube containing the liquid to be introduced. The combination of external pressure and voltage applied causes liquid to be transported from the tube into the capillary. This proposed method has drawbacks. The size of the device makes it impossible to interconnect with the majority of microchannels and necessitates cleaning the device during successive injections, significantly reducing throughput.
Another method for introducing substances into microfluidic devices is disclosed in US Pat. No. 6,042,709. In this approach, electromotive force is used to move charged compounds through the receiving opening of the microfluidic device. The disadvantage is that the exact amount of liquid and material injected depends on the majority of factors that are difficult to control. One important parameter is the surface potential of the microchannel wall that determines the flow of the liquid along with the applied voltage. This surface potential depends on the pH of the liquid being pumped, and even on the ionic composition and the type of ions present in the liquid. For every injection port, a separate high voltage supply is required along with the associated liquid channel to provide a closed electrical circuit, thus preventing efficient interconnection with the majority of different liquids. That is also a drawback.
A method and apparatus for performing electrophoresis experiments in a highly parallel manner is disclosed in US Pat. No. 6,103,199. Here, a plurality of separating capillaries are arranged in a two-dimensional array with associated wells for receiving liquid chemicals. Chemicals are dispensed from the microtiter plate to these wells by an interconnect method using a pressurized chamber associated with the wells to be filled. The disadvantage is that only a very small fraction of the applied liquid is actually introduced into the target microchannel and the majority of the applied droplet remains in the well by capillary forces. As a result, most of the liquid is discarded and is not available for subsequent chemical processing steps. Only a small portion of the liquid that is actually transported has the effect of being introduced into the target portion of the microfluidic system as “injection efficiency”, i.e., the amount of liquid required for a particular chemical operation in a portion of the microfluidic system. It can be said that the ratio between the volume and the total volume of liquid required for the introduction operation.
In this particular disclosure, electrophoretic separations (ie, chemical operations) require sub-nanoliter quantities, but many microliter samples are removed from microtiter plates (ie, introduction operations), starting from 0.001 A much lower injection efficiency results. Low injection efficiency is a disadvantage as it indicates inefficient use of chemicals and increased waste production.
US Pat. No. 6,090,251 provides a microfabricated structure to facilitate the introduction of fluid into a microfluidic device. Fluid is introduced into these receiving wells that are in direct communication with the associated microchannels by using a pressurized gas to drip liquid into the plurality of receiving wells. In addition to the complexity of the required fluid manifolds and pressurization systems, the disadvantage here is also the injection because only a very small fraction of the applied liquid is actually used in the experiment. The efficiency is essentially low.
Chem. 70: 684-688 2月(1998)、Jacobsonら、Anal. Chem. 68: 720-723 3月(1996)、Jacobsonら、Anal. Chem. Chem. Electrokinetic injection is generally applied to introduce a liquid into a capillary electrophoresis column implemented on a chip-like device. Woolley et al., Anal. Chem. 70: 684-688 February (1998), Jacobson et al., Anal. Chem. 68: 720-723 March (1996), Jacobson et al., Anal. Chem. 66: 2369-2373 July (1994) and Effenhauser et al., Anal. Chem. 67: 2284-2287 July (1995). In this method, by applying a high driving voltage between the first well and the second well disposed downstream, the liquid flows from the first well in the microchannel direction for electrophoretic separation. It is pumped out. The charged inner surface of the microchannel wall induces an electroosmotic liquid flow that draws liquid from the first well into the target microchannel. This method is called “electrokinetic injection” and has several drawbacks. One drawback is that the majority of wells need to be integrated onto the microfluidic device if, for example, the majority of liquids need to be handled in high throughput synthesis and screening applications. Compared to microchannels (approximately 50 μm in diameter) that perform the actual chemical manipulation, typical wells occupy a relatively large area (approximately 5 mm in diameter) and occupy a dominant portion of the chip surface (US Pat. No. 6,143,152) And US Pat. No. 6,159,353). Since the cost of microfluidic chips is highly dependent on the chip surface, the required well integration makes this liquid injection scheme unattractive for high-throughput synthesis and screening applications.
Another disadvantage of conventional systems is that separate electrodes are required with the electronic circuit design to apply the drive voltage for all wells. This requirement makes the device complex and expensive.
Another specific disadvantage of the electrokinetic injection, into the driving voltage applied to the electrodes, hydroxide ions (OH -) by electrolysis of water, hydrogen ion (H +), hydrogen gas (H 2) and oxygen Gas (O 2 ) is formed. The formed ions affect the acidity (ie, pH) of the liquid pumped from the first well, while the generated gas forms gas bubbles within the microfluidic system, which allows experiments and microfluidic devices. May also be destroyed. In addition to electrolysis of an aqueous medium, any electroactive species present can be decomposed by an electrochemical reaction on the electrode surface. For example, the presence of chloride ions, ions present in most liquid media, is thought to form chlorine gas, interacting with fragile (biochemical) compounds present in the injected liquid, There is a possibility of destruction. The injected liquid may contain electroactive components that can be destroyed by electrochemical processes associated with electrokinetic injection. These shortcomings can be grouped together and referred to as “electrochemical contamination”.
Another disadvantage of electrokinetic injection is that the wells need to be thoroughly cleaned during successive experiments to prevent cross contamination. The need for this washing step reduces throughput and makes online monitoring difficult. Another drawback is subjecting the injected liquid to a high voltage. This aspect without galvanic separation makes it virtually impossible to use electrokinetic injection for in vivo or near-vivo applications due to the risk of electric shock. Another disadvantage is that the mentioned electrokinetic injection method is manufactured through microfabrication techniques, i.e. by using expensive equipment and methods that are also applied to the manufacture of computer chips. It can be applied only to the system. These methods are known to be expensive. It would be desirable to provide an interconnect method that is also applicable to current non-chip based capillary systems. Yet another disadvantage is the low injection efficiency of electrokinetic injection. About 10-50 μl of liquid is required to fill a typical well, but certain chemical operations require sub-nanoliter quantities.
US Pat. No. 6,130,098 uses a pressure generated by heating a volume of air directly connected to a microchannel to move a volume of liquid into and through the microchannel Is disclosed. The disadvantage of this fluid interconnection method is that a pressure generating air chamber along with an electronic heating element needs to be integrated into the microfluidic system for accurate and efficient operation. This complicates the device and is associated with increased costs.
Current methods and systems for fluid interconnection with microfluidic devices require that the integration of a large number of interconnected chemical manipulation nodes (ie, electrochromatography columns, reactors, etc.) is difficult and necessary Has certain drawbacks with respect to the relatively large liquid volume, low injection efficiency, electrochemical sample contamination, long wash time between analysis steps, galvanic separation, and required microfabrication techniques Can be concluded based on the above. In addition to these drawbacks, none of the above-described interconnection methods allow for bidirectional fluid interconnection, i.e. transport of liquid towards and from the microfluidic system. There is a need in the art to provide a suitable bidirectional fluid interconnect structure that allows the implementation of a wider range of chemical manipulation steps in microfluidic systems.
SUMMARY OF THE INVENTION The present invention provides methods, devices, and systems for interconnecting fluids in microfluidic systems. By forming openings in the microchannel sidewalls, fluid interconnect ports for direct interconnection to the microfluidic channel network and the surrounding environment are provided in the microchannel of the microfluidic system. When the microchannel is filled with liquid, the opening forms a virtual wall. The opening has a suitable cross-sectional dimension so that capillary forces retain the liquid in the microchannel. The virtual wall is defined by a liquid meniscus in the opening that essentially replaces the side wall of the microchannel so that it does not substantially affect or affect the flow of fluid through the microchannel.
The filling of the microchannel can be achieved through openings in the side wall of the microchannel. To fill the microchannel, the user forms a first liquid droplet and directs the droplet in the direction of the opening, so that the first liquid droplet flows into the microchannel and the microchannel Fill the channel.
Fluid introduction and / or fluid removal can be accomplished through virtual walls formed on the side walls of the microchannel. The process of introducing a fluid sample into the microchannel includes forming a fluid droplet and ejecting the droplet toward a virtual wall. The droplets flow through the virtual wall directly into the microchannel without the need for an intermediate reservoir or channel. An imaginary wall formed on the side wall of the microchannel can also define a fluid discharge port for discharging fluid from the microchannel in the form of droplets.

According to one embodiment, the microchannel of the present invention may include a fluid interconnect port. As used herein, “fluid interconnect port” refers to a structure in a microfluidic system, such as an opening formed in a microchannel, that provides fluid access between the inside and outside of the microchannel. The fluid interconnect port is used to introduce fluids and other materials into the microchannel and / or to remove fluids and / or other materials from the microchannel. The fluid interconnect port is, among other applications, a filling port for filling the carrier liquid into the microchannel, a sample introduction port for introducing the sample into the microchannel, and an exhaust for discharging the fluid from the microchannel. Port can be included.
According to exemplary aspects, the microchannel is defined by a sidewall having any suitable shape that seals at least a portion of the interior of the channel. A fluid interconnect port is formed in the side wall of the microchannel by removing a portion of the side wall that defines the opening. An exemplary embodiment fluid interconnect port is formed by an opening in the sidewall of a microchannel having a diameter of about 0.1 μm to about 200 μm, preferably about 25 μm to about 125 μm, and most preferably about 50 μm to about 100 μm. The openings that form the fluid interconnect ports can have any suitable shape including, but not limited to, a cylinder, a disk, a conical shape, an elliptical shape, and a cubic shape. A microchannel sidewall or wall may be formed by two or more components surrounding the entire microchannel volume. The inner wall of the opening that forms the fluid interconnect port can be coated with a hydrophobic material. According to a first aspect of the invention, the fluid interconnect port is used to fill the microchannel with the first liquid. The first liquid includes a carrier fluid for transporting a sample, reagent or other suitable liquid for performing one or more chemical operations such as reactions and separations. Fluid flow can be induced in the microchannel via any suitable means including, but not limited to, pressurization means and electroosmosis means.
As illustrated in FIGS. 2a-7, the
FIG. 2a is a plan view of the
3a and 3b illustrate the process of filling the
The
According to another embodiment, a
According to one aspect, as shown in FIG. 4b, the
The opening used to apply the
According to yet another embodiment, as shown in FIGS. 5 a and 5 b, the
According to yet another aspect, the cross-sectional dimensions of the
Or, as shown in FIG. 6, the
After filling the microchannel through the
According to another aspect, as shown in FIG. 8, a
According to another aspect of the present invention, a second fluid sample can be introduced into the

The liquid droplet is a U.S. Provisional Patent Attorney Registration No. CVZ-002-1, filed 25 September 2001, the contents of which are incorporated herein by reference, entitled `` 2-pin liquid sample fraction ''. Droplet dispensing system in `` Two-Pin Liquid Sample Dispensing System '', as well as U.S. Provisional Application No. 60 / 325,040, entitled `` Droplet Dispensing '' Any suitable
According to an exemplary embodiment, the lateral dimension of the
The size of the opening and the hydrophobicity of the fluid interconnect port determine the size of the dead volume. For example, the microchannel shown in FIG. 9a has a dead volume of 0, ie, no liquid is retained in the
The relatively small dead volume provided by the
FIG.9b shows a vertical cross-sectional view of the
FIG. 9c shows a cross-sectional view of the
According to one aspect of the present invention, as shown in FIG. 9d, the second liquid introduced into the
An exemplary manner of injecting liquid into a microfluidic system through a virtual wall and handling it sequentially is particularly preferred for performing liquid-liquid extraction. Liquid-liquid extraction is a well-known technique that is widely used in numerous chemical analysis and synthesis steps, especially in drug discovery. In liquid-liquid extraction, two immiscible liquids are brought into close proximity to enable extraction of specific components from a liquid containing a specific substance (ie, the source liquid) into the extraction medium (ie, the target liquid) . A suitable example of liquid-liquid extraction is the extraction of water-soluble substances synthesized in an organic solvent that is immiscible with water. An organic solvent containing the water-soluble synthetic product of interest is mixed with water to form small micelle-like droplets of the solvent in the aqueous phase. The water soluble material diffuses from the solvent and is recovered in the aqueous phase. After a predetermined time, the aqueous phase can be recovered due to the immiscibility of the two substances. In this method, the water-soluble substance is thus extracted from the organic liquid layer. The total contact surface between both liquids is critical. The larger the exchange surface provided by the droplet shape, the faster the extraction process.
Using the microfluidic system of the present invention, liquid-liquid extraction can be carried out between a water-soluble substance and water-immiscible organic liquid phase as follows. To perform a liquid-liquid extraction between a water soluble material and an organic liquid phase, an
According to another embodiment, as shown in FIGS.9e and 9f, the
As shown in FIG. 9f, the
FIG. 9g illustrates an embodiment in which microchannels and
According to another aspect, as shown in FIG. 9h, the microchannel fluid interconnection ports are formed in
According to yet another aspect of the invention, a plurality of openings are disposed in the side wall of the microchannel to allow introduction or drainage of liquid through virtual walls at a plurality of locations in the microchannel. For example, as shown in FIG. 10a, to define multiple virtual walls that allow the simultaneous introduction of multiple liquids, the microchannel is a number of fluid interconnection ports positioned in the lateral direction of the

Fluorescent labels are a preferred class of detectable labels because, in part, multiple luminescence can be obtained by irradiating the fluorescent label with light. Thus, a single label can provide multiple measurable events. The detectable signal can also be provided by chemiluminescent and bioluminescent sources. Chemiluminescent sources include compounds that can be excited electronically by a chemical reaction and then emit a light beam that acts as a detectable signal or donates energy to a fluorescent acceptor. Different families of compounds have been found to provide chemiluminescence under different conditions. One compound family is 2,3-dihydro-1,4-phthalazinedione. The most common compound is luminol, which is a 5-amino compound. Other members of the family include 5-amino-6,7,8-trimethoxy- and dimethylamino [ca] benz analogs. These compounds can emit light using alkaline hydrogen peroxide or calcium hypochlorite and a base. Another family of compounds is 2,4,5-triphenylimidazole, which has lophine as the generic name for the parent product. Chemiluminescent analogs include para-dimethylamino and -methoxy substituents. Chemiluminescence can also be obtained under basic conditions using oxalate esters, usually oxalyl active esters such as p-nitrophenyl and peroxides such as hydrogen peroxide. Other useful chemiluminescent compounds are known and available, including -N-alkylacridinum esters (basic H 2 O 2 ) and dioxetanes. Alternatively, luciferin can be used with luciferase or lucigenin to provide bioluminescence. Illustrative examples of on-chip labeling using fluorescent moieties can be found in Harrison et al., Sensors and Actuators B, 33, pp. 105-09 (1996).
Other label moieties can be non-covalently bound to the molecule of interest. Generally, a ligand molecule (eg, biotin) is covalently bound to the polymer. The ligand then binds to an anti-ligand (eg, streptavidin) molecule that is inherently detectable or covalently attached to a signal system such as a detectable enzyme, fluorescent compound or chemiluminescent compound. A number of ligands and anti-ligands can be used. If the ligand has natural anti-ligands such as biotin, thyroxine and cortisol, it can be used with a labeled anti-ligand. Alternatively, any haptenic or antigenic compound can be used in combination with an antibody. The label can also be conjugated directly to the signal generating compound, for example by conjugating with an enzyme or fluorophore. Enzymes of interest as labels are mainly hydrolases, in particular phosphatases, esterases and glycosidases, or oxidoreductases, in particular peroxidases. Fluorescent compounds include fluorescein and its derivatives, rhodamine and its derivatives, dansyl, umbelliferone and the like. Chemiluminescent compounds include luciferin and 2,3-dihydrophthalazinedione, such as luminol. Means for detecting labels are well known to those of skill in the art. When the label is a fluorescent label, the fluorophore is excited with light of an appropriate wavelength, and the resulting fluorescence is detected by, for example, a microscope, visual observation, photographic film, an electronic detection device such as a digital camera, a charge coupled device ( CCD), or detection by use of a photomultiplier tube, a phototube or the like. Fluorescent labeling and detection techniques are preferred, especially microscopy and spectroscopic methods. Similarly, the enzyme label is detected by providing a suitable substrate for the enzyme and detecting the resulting reaction product. Finally, simple colorimetric labels are often detected by simply observing the color associated with the label. For example, conjugated gold often exhibits a pink color, while various conjugated beads exhibit a bead color.

The coating can be applied automatically using an industrial spray coater or a manually operated airbrush. The coating itself is not limited to the above product, which is merely an example. However, the requirements are: the small droplet size of the spray, preferably less than 10 micrometers in diameter, the good wetting properties of the coating liquid, the sharp spray angle for “self-masking” (ie the spray reaches into the channel) Not) and rotation of the chip / wafer to cover the entire wall.
Another aspect is the use of a shadow mask for spraying, so that a portion of the tip is not coated.

The use of virtual walls in the microchannel sidewalls to form bidirectional fluid interconnects in the microfluidic system provides significant advantages over conventional fluid interconnects. Illustrative Example of the Invention Example 1: Fabrication and Use of Virtual Wall Microfluidic Chip A microchannel structure with a fluid interconnect port is formed on a 1.1 mm thick glass wafer with a width of 100 micrometers, a height of 50 micrometers, and a length of 20 mm. A semi-open channel was fabricated by isotropic etching. A buffered hydrogen fluoride (HF) solution was used as an etchant and a photo patterned etchant resistant silicon nitride mask layer was applied to define the microchannel region to be etched. At each end of each etched half-open microchannel, the channel was accessed by powder blasting a 1 mm diameter hole through the glass wafer.
A 50 micrometer thick dry resist film layer (LAMINAR® 5000, Shipley, Birkenfeld, Germany) was applied to the top surface of the etched wafer. A fluid interconnect port was incorporated by photopatterning a circular opening of 50-150 micrometers in the dry resist film, whereby the opening was aligned and extended into the underlying microchannel. Finally, the wafer was cut into individual chips and placed in a holder to connect the microchannel structure to external systems such as fluid pumps and power supplies.

It has been observed that microchannels are automatically filled by capillary forces in response to application to a liquid fluid interconnect port. Liquid droplets having a diameter of about 50 micrometers were made using a piezoelectrically activated droplet dispenser system (MicroDrop GmbH, Norderstedt, Germany). The droplets were made from a buffer solution containing a fluorescent dye (50 mM bicarbonate buffer, pH 9.0, 1 mM fluorescein) and led to an interconnect port with a diameter of 100 micrometers. The injection process was monitored using an inverted microscope under UV irradiation to excite the fluorescent dye and obtain a bright yellow-green color.
