Amorphous Compound In this specification, a compound is “amorphous” means N- {2-chloro-4-[(6,7-dimethoxy-4-quinazolinyl) oxy] phenyl} -N′-. It means a solid state in which propylurea molecules are assembled without forming crystals having a regular spatial arrangement. Specifically, it means a case where the amorphous state can be confirmed by conventional powder X-ray diffraction analysis. For example, when X-ray powder diffraction using Cu—Kα radiation is performed to create a graph showing the intensity with respect to the diffraction angle (2θ), in this graph, crystalline N- {2-chloro-4- [ (6,7-Dimethoxy-4-quinazolinyl) oxy] phenyl} -N′-propylurea shows a sharp diffraction peak pattern. On the other hand, in the case of an amorphous compound, it does not substantially have a sharp diffraction peak pattern as in the case of a crystalline compound. This means that in the present invention, the N- {2-chloro-4-[(6,7-dimethoxy-4-quinazolinyl) oxy] phenyl} -N′-propylurea compound is usually amorphous. means.
より具体的には、化合物が「非晶質」であるとは、例えば、後述する評価試験の試験1に記載したような方法にしたがって測定される粉末X線回折図において、そのグラフが鋭いピークを実質的に持たない所謂ハローパターンを示す場合をいう。さらに具体的には、回折角2〜40°の間において、そのグラフは、単一でなめらかなベル型状のハローパターンを示す場合をいう。例えば、該グラフは、図1の(a)で示される粉末X線回折パターンを実質的に示すものである。
More specifically, when the compound is “amorphous”, for example, in a powder X-ray diffraction diagram measured according to the method described in
According to a preferred embodiment of the present invention, the amorphous compound has a diffraction angle between 2 and 40 ° in the diffraction graph showing the intensity with respect to the diffraction angle (2θ) obtained by X-ray powder diffraction using Cu-Kα radiation. The height of the maximum peak existing in is 1000 cps or less, more preferably 500 cps or less, and still more preferably 100 cps or less.
In the present specification, the “maximum peak” means a peak including a point (maximum value) at which the intensity is maximum in the diffraction graph. The “maximum peak height” is the line between the straight line connecting the end points of the peak of the peak shape of a peak and the intensity at the maximum peak (straight line (β)) shown as “h” in FIG. Means a difference. Therefore, the “maximum peak height” does not mean the difference between the baseline (straight line (α)) and the intensity at the maximum peak (straight line (β)) indicated by “A” in FIG. .
In the present specification, N- {2-chloro-4-[(6,7-dimethoxy-4-quinazolinyl) oxy] phenyl} -N′-propylurea compound is almost the compound that is “amorphous”. In addition to the case where it is present as completely amorphous, as long as it exhibits the above-mentioned properties as “amorphous” as an amorphous compound, N- {2-chloro- in a partially refined crystalline state 4-[(6,7-dimethoxy-4-quinazolinyl) oxy] phenyl} -N′-propylurea may be included.
In the present invention, “crystalline” N- {2-chloro-4-[(6,7-dimethoxy-4-quinazolinyl) oxy] phenyl} -N′-propylurea is N- {2-chloro- It means that substantially all of the 4-[(6,7-dimethoxy-4-quinazolinyl) oxy] phenyl} -N′-propylurea molecule is in a crystalline state with an ordered spatial arrangement. In the present invention, this crystalline compound can be easily obtained by those skilled in the art, for example, by the method described in WO03 / 008388.

According to the present invention, the N- {2-chloro-4-[(6,7-dimethoxy-4-quinazolinyl) oxy] phenyl} -N'-propylurea compound, which is amorphous, is a crystallization solvent. N- {2-Chloro-4-[(6,7-dimethoxy-4-quinazolinyl) oxy] phenyl} -N'-propylurea dissolved in the solution was crystallized by grinding the resulting crystals. It can be produced by a process comprising qualitative.
Here, the “crystallization solvent” can dissolve N- {2-chloro-4-[(6,7-dimethoxy-4-quinazolinyl) oxy] phenyl} -N′-propylurea, There is no particular limitation as long as crystals can be crystallized from the solution. Preferably, the crystallization solvent is chloroform.
In order to crystallize N- {2-chloro-4-[(6,7-dimethoxy-4-quinazolinyl) oxy] phenyl} -N'-propylurea dissolved in the crystallization solvent from the solution. The solution can be cooled and the crystallized by crystallization.
結晶化により得られた結晶は、必要に応じて、粉砕処理前に、さらに乾燥処理に付してもよい。乾燥処理としては、例えば、真空乾燥処理が挙げられる。 The crystals obtained by crystallization may be further subjected to a drying treatment before the pulverization treatment, if necessary. Examples of the drying process include a vacuum drying process.
Here, “pulverization” refers to crystalline N- {2-chloro-4-[(6,7-dimethoxy-4-quinazolinyl) oxy] phenyl} -N′-propylurea that is crushed and ground to form fine particles. Any apparatus may be used as long as it can be converted into an amorphous state and can be made amorphous, but typically, a pulverizer is used.
Here, the crusher is not particularly limited as long as it has a function of mechanically crushing and grinding to form fine particles, and examples thereof include a ball mill, a rod mill, a hammer mill, a pin mill, and a micros. In the present invention, it is preferable to make the crystal amorphous by grinding with a ball mill.
非晶質化に要する粉砕時間は、使用する粉砕機の機種、粉砕動力の大きさ、対象物の全重量、添加剤の種類と添加割合等により変動し、当業者であれば適宜選択することができる。例えば、結晶性のN−{2−クロロ−4−[(6,7−ジメトキシ−4−キナゾリニル)オキシ]フェニル}−N’−プロピルウレア25.0gを、ボールミルで粉砕して非晶質化する場合に要する時間は、少なくとも約10時間である。 The pulverization time required for amorphization varies depending on the type of pulverizer to be used, the size of the pulverization power, the total weight of the object, the type and addition ratio of additives, etc. Can do. For example, 25.0 g of crystalline N- {2-chloro-4-[(6,7-dimethoxy-4-quinazolinyl) oxy] phenyl} -N′-propylurea is pulverized with a ball mill to become amorphous. The time required to do this is at least about 10 hours.
Amorphous Composition In this specification, “amorphous” in an amorphous composition refers to N- {2-chloro-4-[(6,7-dimethoxy-4-quinazolinyl) oxy] phenyl}- This means that the N′-propylurea molecule does not form crystals with a regular spatial arrangement, but forms an aggregated solid state with a pharmaceutically acceptable additive compound. This means that the amorphous state can be confirmed by X-ray diffraction analysis. In contrast, the amorphous composition does not substantially have a sharp diffraction peak pattern as in the case of the crystalline compound. In the present invention, this usually means that the “amorphous composition” is amorphized.
