本発明者らは、これらの課題を達成すべく鋭意研究を重ねた。その結果、トコフェロール類に、アスコルビン酸類、リボフラビン類、ピリドキシン類、コバラミン類、葉酸、イノシトールヘキサニコチネート、パンテチン、カルニチン類、ユビデカレノンから選ばれる1種を含有する組成物が、LDLを顕著に低下させ、かつ動脈硬化指数も顕著に低下させることを見出した。

(5)リボフラビン酸類が、酪酸リボフラビンである、(1)又は(2)に記載のLDL低下剤及び/又は動脈硬化抑制剤組成物であり、

(2)製法
上記成分及び分量をとり、日局製剤総則「シロップ剤」の項に準じてシロップ剤を製した後、褐色ガラス瓶に充填してシロップ剤を製する。
(試験例)
(1)被験物質
酢酸d−α−トコフェロールは理研ビタミン(株)製造のものを、アスコルビン酸は日本ロッシュ(株)製造のものを、酪酸リボフラビンは三菱ウエルファーマ(株)製造のものを、塩酸ピリドキシンは第一ファインケミカル(株)製造のものを、塩酸ヒドロキソコバラミンはヘキスト・マリオン・ルセル社製造のものを、葉酸及び塩化カルニチンは金剛化学(株)製造のものを、イノシトールヘキサニコチネートは白鳥製薬(株)製造のものを、パンテチンは相互薬工(株)製造のものを、ユビデカレノンは日清ファルマ(株)製造のものを使用した。
(2)動物
試験動物としては、Covance Research Products Inc.からビーグル犬雄を5箇月齢で購入し、約1箇月間の検疫および馴化飼育後に使用した。
(3)投与剤形、製剤の調整方法および製剤の保存方法
試験動物毎の体重をもとに算出した必要量の被験物質を、TORPAC社のゼラチンカプセル(1/2オンス)に充填した。充填後、カプセルは動物毎に区分されたケースに入れ、投与時まで冷蔵保存した。
(4)投与経路および投与期間
被験物質を充填したカプセルは、1日1回9:00〜12:30の間に、試験動物に強制経口投与した。なお、試験動物は投与前2乃至3時間絶食させた。

(Test example)
(1) Test substance d-α-tocopherol acetate was manufactured by Riken Vitamin Co., ascorbic acid was manufactured by Nippon Roche Co., Ltd., and riboflavin butyrate was manufactured by Mitsubishi Pharma Corporation. Pyridoxine is manufactured by Daiichi Fine Chemical Co., Ltd., Hydroxocobalamin hydrochloride is manufactured by Hoechst Marion Lucer, folic acid and carnitine chloride are manufactured by Kongo Chemical Co., Ltd., and inositol hexanicotinate is manufactured by Shiratori Pharmaceutical. The product manufactured by Co., Ltd., the pantethine manufactured by Mutual Yakuhin Co., Ltd., and the ubidecarenone manufactured by Nisshin Pharma Co., Ltd. were used.
(2) Animals As test animals, beagle dogs were purchased from Covance Research Products Inc. at the age of 5 months and used after quarantine and acclimatization for about 1 month.
(3) Dosage Form, Preparation Method and Storage Method The required amount of the test substance calculated based on the body weight of each test animal was filled into a TORPAC gelatin capsule (1/2 ounce). After filling, the capsules were placed in cases separated by animal and stored refrigerated until administration.
(4) Administration route and administration period Capsules filled with the test substance were orally administered by gavage to test animals once a day between 9:00 and 12:30. The test animals were fasted for 2 to 3 hours before administration.

(5)被験試料の調製
カプセル投与前−14および−7日(投与開始前第2週および第1週)、投与後4日、8日、12日に、橈側皮静脈から約10mL採血した。なお、採血前約18時間、試験動物は絶食させた。

(6)試験方法
検査結果は、HDLはホモジニアス法、LDLは化学修飾酵素法、過酸化脂質は八木法、SODは改良亜硝酸法を用いて測定した。
(試験結果)
各試料の投与における測定値は、投与2週間前および1週間前の値の平均を100として換算して求めた。
