このように構成されたオートサンプラによる試料注入の動作シーケンスの一例を以下に説明する。
(1)バルブ2は、図4のポート0−bが連通するポジションにおいて、ニードル7をバイアル8に挿入し、プランジャ3を引いて所定量の試料液を吸い上げる。試料液を吸い上げるときは、インジェクタバルブはe−dが連通するポジションであるので吸い上げた試料液はループ6内に留まり、バルブ2やプランジャ3までは至らない。

従来、ニードル7を介して生じるクロスコンタミネーションを防ぐために、洗浄を強化したりニードル7の表面を研磨平滑化して汚れが付着し難くする等、主として物理的汚染に対する対策がなされていた。しかし、ニードル7の汚れは物理的汚染ばかりではない。ニードル7の材料としてはステンレス鋼が用いられるが、ステンレス鋼は鉄を主材とする合金であるから、ミクロ的に見ればその表面には鉄が露出しており、この鉄の化学的性質によりある種の試料成分が鉄の部分に吸着される。
