塩化カルシウム管、滴下ロート、温度計を付した100mlナスフラスコに5.00gの2,3−ジブロモプロピオン酸5と3.14g(1.05当量)の4−ニトロフェノールを量り取り、25mlのTHFを加え、溶解させた。これに氷浴撹拌下、4.89g(1.10当量)のジシクロへキシルカルボジイミド(DCC)のTHF溶液(10ml)を滴下すると、発熱して結晶が析出してきた。その後、室温で16時間反応した。
反応後、析出した結晶を濾別し、少量のTHFで洗浄して濾液を濃縮すると、11.6gの淡黄色の油状物が得られた。この油状物をHPLCにより定量分析すると、2,3−ジブロモプロピオン酸−4−ニトロフェニルエステル4が、22%含まれていた。収率は34%と計算される。
Synthesis of 2,3-dibromopropionic acid-4-nitrophenyl ester 4 5.00 g of 2,3-dibromopropionic acid 5 and 3.14 g (1.05 equivalents) in a 100 ml eggplant flask equipped with a calcium chloride tube, dropping funnel and thermometer 4-nitrophenol was weighed and 25 ml of THF was added and dissolved. After the reaction, the precipitated crystals were separated by filtration, washed with a small amount of THF, and the filtrate was concentrated to obtain 11.6 g of a pale yellow oil. When this oily substance was quantitatively analyzed by HPLC, it contained 22% of 2,3-dibromopropionic acid-4-nitrophenyl ester 4 . The yield is calculated as 34%.
2,3−ジブロモプロピオン酸−4−ニトロフェニルエステル4の合成
(1)塩化カルシウム管、滴下ロート、温度計を付した100mlナスフラスコに2.00gのアクリル酸と4.05g(1.05当量)の4−ニトロフェノールを量り取り、20mlのTHFを加え、溶解させた。これに氷浴撹拌下、6.30g(1.10当量)のジシクロへキシルカルボジイミド(DCC)のTHF溶液(12ml)を滴下すると、発熱して結晶が析出してきた。その後、水浴中で18時間反応した。
反応後、析出した結晶を濾別し、少量のTHFで洗浄して濾液を濃縮すると、8.20gの黄色の半油状固体が得られた。この濃縮物をHPLCにより定量分析すると、アクリル酸−4−ニトロフェニルエステルが、50.6%含まれていた。収率は77%と計算される。

(2)ジムロート、滴下ロート、温度計を付した50mlのナスフラスコにアクリル酸−4−ニトロフェニルエステルを1.38g量りとり、室温下で、1.15gの臭素を滴下すると発熱して原料は溶解した。内温は20℃から60℃まで上昇した。
反応終了後、反応系をそのまま50℃にて減圧下濃縮し、低沸点物を除くと、淡黄色の油状物として4を2.45g得た。
収 量 :2.45g
重量収率 :97%
性 状 :淡黄色の油状物
2工程収率:74%
Synthesis of 2,3-dibromopropionic acid-4-nitrophenyl ester 4
(1) To a 100 ml eggplant flask equipped with a calcium chloride tube, a dropping funnel and a thermometer, 2.00 g of acrylic acid and 4.05 g (1.05 equivalents) of 4-nitrophenol were weighed and 20 ml of THF was added and dissolved. After the reaction, the precipitated crystals were separated by filtration, washed with a small amount of THF, and the filtrate was concentrated to obtain 8.20 g of a yellow semi-oily solid. When this concentrate was quantitatively analyzed by HPLC, it contained 50.6% of acrylic acid-4-nitrophenyl ester. The yield is calculated as 77%.

(2) A 1.38 g of acrylic acid-4-nitrophenyl ester was weighed into a 50 ml eggplant flask equipped with a Dim funnel, a dropping funnel and a thermometer, and 1.15 g of bromine was added dropwise at room temperature to dissolve the raw material. . The internal temperature rose from 20 ° C to 60 ° C.
After completion of the reaction, the reaction system was directly concentrated at 50 ° C. under reduced pressure to remove low-boiling substances, and 2.45 g of 4 was obtained as a pale yellow oil.

塩化カルシウム管、滴下ロート、温度計を付した100mlナスフラスコに1.00gの3−アミノ−2−ブテン酸メチルエステル2と15mlのトルエンを加え、溶解させた。これに室温撹拌下、1.93g(2.2当量)のトリエチルアミンを加え、続いて3.37g(1.1当量)の2,3−ジブロモプロピオン酸−4−ニトロフェニルエステル4(発熱)を滴下した。同温で30分撹拌した後、60℃の油浴中で2時間反応した。
TLCにて原料消失を確認後、反応系中の沈殿物を濾別し、トルエン、蒸留水で十分に洗浄して単離すると、5−メトキシカルボニル−6−メチル−2−ピリドン3が白色の結晶性粉末として1.07g得られた。
Synthesis of 5-methoxycarbonyl-6-methyl-2-pyridone 3 (active ester method)
To a 100 ml eggplant flask equipped with a calcium chloride tube, a dropping funnel and a thermometer, 1.00 g of 3-amino-2-butenoic acid methyl ester 2 and 15 ml of toluene were added and dissolved. To this was added 1.93 g (2.2 equivalents) of triethylamine with stirring at room temperature, followed by dropwise addition of 3.37 g (1.1 equivalents) of 2,3-dibromopropionic acid-4-nitrophenyl ester 4 (exothermic). The mixture was stirred at the same temperature for 30 minutes and then reacted in an oil bath at 60 ° C. for 2 hours.
After confirming the disappearance of the raw materials by TLC, the precipitate in the reaction system was filtered off, washed thoroughly with toluene and distilled water, and isolated, and 5-methoxycarbonyl-6-methyl-2-pyridone 3 was white. As a crystalline powder, 1.07 g was obtained.

塩化カルシウム管、滴下ロート、温度計を付した100mlナスフラスコに2.00gのβ−アミノ−クロトノニトリル(純度96%)6と50mlのTHFを加え、溶解させた。これに水浴中撹拌下、8.87g(3.6当量)のトリエチルアミンを加え、続いて8.54g(1.4当量)の2,3−ジブロモプロピオン酸クロライド1(発熱)を30℃以下の温度で滴下した。その後20℃で1時間反応した。
反応終了後、反応系中の沈殿物を濾別し、THF、蒸留水で十分に洗浄して単離すると、7が淡黄色の固体として1.71g得られた。また、水層を20mlのクロロホルムで3回抽出し、無水硫酸ナトリウム乾燥後、減圧濃縮すると、7が淡黄色の固体として0.19g得られた。
収 量 :1.90g
重量収率 :61%

To a 100 ml eggplant flask equipped with a calcium chloride tube, a dropping funnel and a thermometer, 2.00 g of β-amino-crotononitrile (purity 96%) 6 and 50 ml of THF were added and dissolved. To this was added 8.87 g (3.6 equivalents) of triethylamine with stirring in a water bath, and then 8.54 g (1.4 equivalents) of 2,3-dibromopropionic acid chloride 1 (exothermic) was added dropwise at a temperature of 30 ° C. or lower. Thereafter, the reaction was carried out at 20 ° C. for 1 hour.
After completion of the reaction, the precipitate in the reaction system was separated by filtration, washed thoroughly with THF and distilled water and isolated to obtain 1.71 g of 7 as a pale yellow solid. The aqueous layer was extracted three times with 20 ml of chloroform, dried over anhydrous sodium sulfate, and concentrated under reduced pressure to obtain 0.19 g of 7 as a pale yellow solid.

塩化カルシウム管、滴下ロート、温度計を付した100mlナスフラスコに2.00gの2,3−ジブロモプロピオン酸と40 mlのエーテルを加え、溶解した。アセトン/ドライアイス浴で反応系を−30℃まで冷却し、1.41g(1.2当量)のクロロギ酸イソブチルを加え、続いて、1.05g(1.2当量)のN−メチルモルフォリンを5分かけて滴下した。内温は−20℃まで上昇し、塩酸塩が析出した。その後、30分間同温で反応させた。
反応後、内温が0℃になるまで自然昇温し、すぐに塩酸塩を濾過、濾液を減圧濃縮して、2.86gの無色透明な油状物の8を得た。

25mlナスフラスコに500mgの3−アミノ−2−ブテン酸メチルエステル2と7.5mlのトルエンを加え、溶解させた。これに水浴中撹拌下、1.05g(2.4当量)のトリエチルアミンを加え、続いて1.73g(1.2当量)の(2−メチルプロポキシ)カルボニル2,3−ジブロモプロピオネート8のトルエン溶液(2.5ml)を10分かけて滴下した。60℃で30分反応後、288mg(0.2当量)の8を追加して同温で3時間反応した。
反応終了後、反応系中の沈殿物を濾別し、少量のトルエン、蒸留水、酢酸エチルで洗浄して単離すると、3が薄い肌色の粉末として475mg得られた。
収 量 :475mg
重量収率 :65%
