【0001】

The reaction of the optically active 3- (N, N-dimethylamino) -1- (2-thienyl) -propan-1-ol with a halogenated formate is carried out by using a solvent containing such a base (for example, toluene). ), Preferably by heating and refluxing. Note that the time required for this heating and reflux can be appropriately selected by those skilled in the art.
[0035]
In this way, the optically active 3- (N, N-dimethylamino) -1- (2-thienyl) -propan-1-ol is demethylated and optically represented by the following formula (II) Active N, O-bisalkoxycarbonyl derivatives:

[0037]
(Wherein R is defined as above and * represents an asymmetric center) is generated and isolated using means well known to those skilled in the art. The optical purity of the N, O-bisalkoxycarbonyl derivative represented by the formula (II) obtained above is determined by the starting material (3- (N, N-dimethylamino) -1 used for this production. It almost depends on the optical purity of-(2-thienyl) -propan-1-ol).
[0038]
Next, the N, O-bisalkoxycarbonyl derivative represented by the above formula (II) is hydrolyzed under basic conditions.
[0039]
In order to produce optically active 3- (N-methylamino) -1- (2-thienyl) -propan-1-ol in a higher yield and more efficiently, in the hydrolysis step, the above formula (II The N, O-bisalkoxycarbonyl derivative represented by) is preferably 3- (N-ethoxycarbonylN-methylamino) -1-ethoxycarbonyl- (2-thienyl) propane.
[0040]
In this invention, sodium hydroxide is mentioned as an example of the base used for this hydrolysis. In the hydrolysis, a mixture of alcohol and water is used as a solvent. Examples of preferred alcohols include methanol, ethanol, isopropanol, and n-butanol. The mixing ratio of alcohol and water is not particularly limited, but is preferably 1: 1 to 1: 5 (volume ratio).
[0041]
Hydrolysis is performed under reflux. For example, when using a mixture of ethanol and water as the solvent, the mixture is heated to a temperature of about 82 ° C. and refluxed.
[0042]
As a result, two alkoxycarbonyl groups in the N, O-bisalkoxycarbonyl derivative represented by the above formula (II) are hydrolyzed to produce optically active 3- (N-methyl) represented by the formula (IV). Amino) -1- (2-thienyl) -propan-1-ol:

[0044]
Is generated. The resulting optically active 3- (N-methylamino) -1- (2-thienyl) -propan-1-ol is isolated using means well known to those skilled in the art.
[0045]
In this way, optically active 3- (N-methylamino) -1- (2-thienyl) -propan-1-ol is produced in high yield. The optical purity of 3- (N-methylamino) -1- (2-thienyl) -propan-1-ol obtained by the method of the present invention is the same as that of N, O-bisalkoxy used for this production. It almost depends on the optical purity of the carbonyl derivative.

[0061]
In a reaction vessel, the compound (2) ((S) -3- (N-ethoxycarbonyl N-methyl) amino-1-ethoxycarbonyloxy- (2-thienyl) propane obtained in Example 1: 1.40 g 4.4 mmol) was dissolved in a mixture of ethanol (3 ml) and water (8 ml). To this solution was then added 48% NaOH (3.1 g, 37 mmol) and stirred at 82 ° C. for 4 hours. After the reaction product was cooled to room temperature, MTBE (methyl tert-butyl ether: 30 ml) was added and liquid-separated to obtain an MTBE layer. This MTBE layer was washed with brine (20 mL twice), dried (MgSO 4 ), and concentrated under reduced pressure to give a yellow oil.
[0062]
The 1 H-NMR spectrum was measured for this yellow oil. The obtained results are shown in Table 3.
[0063]
[Table 3]
[0064]
From the results, it was confirmed that the obtained yellow oil was the title compound (4) (title compound (4) 0.61 g, yield 80.0%).
[0065]
Subsequently, the optical purity of the obtained title compound (4) was analyzed by HPLC under the conditions described in Table 4. Similarly, the optical purity of starting material (1) (3- (N, N-dimethylamino) -1- (2-thienyl) -propan-1-ol) used in Example 1 is also shown in Table 4. Analysis by HPLC under the conditions described.
[0066]
[Table 4]
[0067]
The title compound (4) obtained had an optical purity of 95.8% ee. The optical purity of the starting material (1) was almost the same.
[0068]
From this, the optical purity of 3- (N-methylamino) -3- (2-thienyl) propan-1-ol (title compound (4)) produced by the method of the present invention is used as a starting material. It can be seen that it almost depends on the optical purity of (3- (N, N-dimethylamino) -1- (2-thienyl) -propan-1-ol).
[0069]
【The invention's effect】
According to the present invention, optically active 3- (N-methylamino) -3- (2-thienyl) propan-1-ol can be produced in high yield. Furthermore, in the present invention, the optical purity of the starting material (3- (N, N-dimethylamino) -1- (2-thienyl) -propan-1-ol) is obtained by 3- (N-methylamino) obtained. ) -3- (2-Thienyl) propan-1-ol. As a result, the optical purity of the resulting 3- (N-methylamino) -3- (2-thienyl) propan-1-ol improves as the starting material with higher optical purity is used.
