本発明は、個別に識別情報が付与された微粒子を用いて、微粒子に固相化されたプローブによる被検体中の生体物質を検出する検査方法及びその検査システムに関する。

【従来の技術】
一般に、公知な被検体中の多数の物質を同時に測定するマイクロアレイと称する手法が知られており、広く利用されている。この手法では、測定信号を分離するために空間的に分離した多数のプローブをガラス等の担体上にスポット状に固相している。その後、ハイブリダイゼーションを行った後、各プローブにおける蛍光色を読み取り、識別を行っている。

また、マイクロアレイと同様な技術として、特許文献1及び特許文献2には、ビーズアレイが提案されている。このビーズアレイは、個別のビーズを識別するために1または2種類の蛍光物質を形成させている。但し、ビーズアレイを用いた場合、識別精度が低いため、誤測定が生じやすい欠点がある。

そこで、特許文献3には、これらのビーズをバーコード形状に加工し、識別の情報を持たせたものが開示されている。この方法においては、フォトリソグラフにより加工したバーコード形状の粒子に、タンパク等を結合させ、フローサイトメトリーにて検出する。

また、特許文献4には、多孔質構造を持つ捕捉部材、例えばフィルタ上にプローブを固相化し、捕捉部材面に対して検体溶液を上下方向に駆動させることで、反応を高速化する技術が知られている。

【発明が解決しようとする課題】
前述したマイクロアレイを用いた方法は、位置の確定のために多くの画像処理が必要であり、それらの機器が高価となっている。また、設計時にマイクロアレイ上の所定位置へ一度固定したプローブは後から動かせないので、項目設計の自由度が低い。
さらに、マイクロアレイは、それぞれを個別に作製しなければならないので、同じ仕様でもマイクロアレイ間における品質が一定していないため、原理的に抜き取り検査ができず、全品検査という手間が掛かっている。

また、ビーズアレイによる方法においても、識別装置が大掛かりになり、高価となっている。また、識別精度が低いため、誤測定が生じやすい欠点がある。さらに、蛍光物質は長期保管において劣化してしまい、取り扱いに問題がある。

前述した特許文献3に開示されているビーズをバーコード形状に加工した技術は、フローサイトメトリーでの検出は、反応と検出が別々の工程となる等、操作が煩雑であることや、装置自体の調整や解析に熟練を要すること、また反応を高速化することが難しい。この手法においても、装置が大掛かりとなる。

このように、マイクロアレイの作製には大掛かりな製造装置が必要であり利用者がマイクロアレイを製造することは非常に困難であり、利用の自由度や、用途の拡大に限界があった。例えば、プローブの選択・プローブの設計を誤って、マイクロアレイを作製した場合には、ひとつのプローブの作成ミスのために、マイクロアレイ全体を作製し直す必要があった。

また、特許文献4においては、反応の高速化が図れる反面、捕捉部材上にプローブを固相化しなければならないため、マイクロアレイと同様な課題があり、また、手間が掛かる作業となっている。

そこで本発明は、個々に識別情報が与えられ、プローブの交換が容易にできる微粒子を用いた簡素な構成により、高精度な識別精度を有し、プローブ情報の高速処理が可能な微粒子を用いた生体物質の検査方法及びその検査システムを提供することを目的とする。

【課題を解決するための手段】
本発明は上記目的を達成するために、個々に識別情報が付与され、所定の検体に反応するプローブが含まれる溶液中で該プローブが固相化された多種多数の微粒子と、該溶液を透過可能で、かつ、上記微粒子を捕捉可能な捕捉部材を有し、該微粒子と検体を含み流動する溶液とを接触させ、前記捕捉部材により捕捉された微粒子から、上記識別情報と上記プローブによる検体の結合量の情報との両者を検出する検体の検査方法を提供する。

以上のような構成の検体の検査方法によれば、検体溶液が流動されて、それに伴い微粒子が移動しつつ、プローブと検体溶液中の検体との反応が促進されて高速化される。また、検体溶液の流動に伴い捕捉部材表面に微粒子が一時的且つ重なり無く捕捉され、この状態を撮像した画像データの解析によりフィルタ表面上に存在する微粒子を検出して、目的とする反応物のみが有効に測定される。

さらに、個々に識別情報が付与され、所定の検体に反応するプローブが含まれる溶液中で該プローブが固相化された多種多数の微粒子と、上記多種多数の微粒子と検体を含む溶液とが混入される容器と、上記溶液を透過可能で、かつ、微粒子を捕捉可能な捕捉部材と、上記溶液を所望する温度に制御する温度制御手段と、上記溶液を流動させる駆動手段と、上記微粒子を画像データとして取り込む検出手段と、付与された上記識別情報と固相化された上記プローブとを関連づけて、上記検出手段により得られた画像データから上記識別情報と反応したプローブ情報とを解析する解析手段とを備える検査システムを提供する。

以上のような構成の検体の検査システムは、前述した検査方法と同様な作用効果に加えて、従来のマイクロアレイを利用する装置の構成よりも小型で簡易で且つ低コストであり、ビーズアレイを利用するによる方法においても、識別装置よりも、簡易な作業で識別精度が高く、正確な測定を行うことが出来る。

【発明の実施の形態】
以下、図面を参照して本発明の実施形態について詳細に説明する。
まず、本発明の生体物質の検査方法及び検査装置に係る実施形態について説明する。ここでは、この検査方法に用いられるプローブが固定された微粒子について説明する。
この検査方法では、予め識別情報を持たせた複数の微粒子にプローブを固定して用いる。これらの微粒子の識別情報としては、サイズ、形状の差、色の差又はID情報等のいずれか、又はこれらの組合せを利用することが可能であり、それぞれに適した検出部を設ける。例として、微粒子にマイクロファブリケーションの技術を用いて、固有の形状を持たせることや、微粒子自体に微細加工技術により、数値、形状を付加することや、暗号、バーコードなどの情報を持たせることで、光学的手段、あるいは取得画像の演算処理によって識別が可能となる。

さらに、微粒子としては、識別情報を付加・認識が容易だけではなく、目的とする検出物に対するプローブが容易に固相化される物質が適している。両者を同時に実現できるものが好ましいが、場合によっては、2次的な加工により、プローブの固相化を行っても構わない。例えば、樹脂製、ガラス製のビーズなどを有効に用いることができる。

これらの微粒子には、予めプローブが結合しやすくするための処理を施す。この処理としては、微粒子表面の電荷をマイナスにチャージすることで、プラスチャージを持つ核酸と結合しやすくする様な方法や、微粒子表面に官能基を有する分子を結合させて、化学修飾を施した核酸プローブと結合しやすい状態にする等がある。具体的な例として、核酸をプローブに用いる場合であれば、プローブにアミノ基、アルデヒド基、チオール基、ビオチンなどの修飾を行い、微粒子表面をアミノ基、アルデヒド基、チオール基、アビジンなどで修飾し、それぞれの反応を利用して結合させることや、または、2価性若しくは、1価性の架橋剤を用いて微粒子とプローブを結合させることができる。勿論、これらに限定されるものではない。

このような識別情報を持つ微粒子を複数種用意し、これらの微粒子を反応溶液中で分散させプローブと反応させることにより、プローブが固相化された微粒子を作成する。このプローブと微粒子の反応は、ピペット操作により非常に簡便に行うことが可能である。また、後に行われる解析のために、この時に微粒子における識別情報と固相化されたプローブの種別を関係づけたデータを作成する。

このような微粒子を用いることにより、マイクロアレイ製造装置のような大掛かりな装置は必要なく、作製の自由度が高い。例えば、プローブの設計を誤った場合であっても、そのプローブを有する微粒子のみをピペット操作にて作り直して、入れ替えることで対応することができる。また、予め多種の識別情報を施した微粒子を多数用意しておき、必要なプローブ数に応じて、これらの中から選択して利用することもできる。

また、本発明による検査方法においては、微粒子を後述するような光学的な検出を行う検出部を用いて識別することが可能である。従って、プローブとの反応量を測定するための検出部にあわせて、微粒子自体の識別方法を選択することができる。例えば、検体を蛍光標識し、蛍光顕微鏡を検出部とした場合には、微粒子の検出も蛍光検出とすれば、全体構成を簡略化することが可能にある。但し、検出部は、これに限定されるものではない。

次に図1には、前述した微粒子を用いた検査システムを概念的に示し、図2は、この検査システムのブロック構成を示す。
この検査システムは、検体溶液が透過可能な基材により形成された2枚の捕獲部材(例えば、フィルタ等)1a、1bと、これらの捕捉部材1a、1bが間隔をあけて嵌め込まれ、反応キャピラリーとしても機能する透明なチューブ4と、捕捉部材1a、1b間に封止された微粒子(プローブ)3と、これらの捕捉部材1a、1b間に配置され、反応した微粒子3から識別情報及びプローブ情報等を光学的に読み取るための撮像部を含む検出部5とを備えている。このチューブ4は、ガラスや樹脂等の透明部材により形成され、検体溶液が入れられている。温度・液駆動部7により、このチューブ4内に入れられた検体溶液は、所定温度に保たれ、矢印(図1)に示すように流動される。従って、捕捉部材1a、1b間に封止されている微粒子3は、この流れに乗って移動されている。捕捉部材1a、1b、チューブ4及び検体溶液により、微粒子3のプローブに対する反応部6を構成する。また、検出部5は、固体撮像素子(CCD等)により撮像するカメラを搭載し、撮像レンズ5aがチューブ4に近接して配置されている。

さらに、温度・液駆動部7及び検出部5を制御し、検出部5で検出された識別情報やプローブ情報を解析し、その結果を例えば、液晶表示装置等の表示部9に表示させる制御・解析用コンピュータ8が設けられている。また微粒子3へ可視光を照射する照明部10が設けられ、微粒子3の輝度の検出を蛍光で行うことができる。尚、図示していないが、制御・解析用コンピュータ8にインターフェース機能を搭載して、ネットワーク等を介して外部装置例えば、ホストコンピュータと接続して、通信によりデータや制御信号(外部装置からの制御)のやり取りを行ってもよい。

このようなシステム構成において、検体溶液を捕捉部材面に対して直交する方向に流動させることにより、微粒子3のプローブと検体溶液中の検体との反応を高速化することが可能である。つまり、検体溶液の流動(攪拌)により、プローブ分子と検体分子の衝突頻度が高められる。このため反応が促進され、一般的な検体溶液を静止状態で反応されるものに対して、飛躍的に反応速度(反応時間)を速めることが可能となる。

この原理を本実施形態の微粒子3による反応に適用すると、反応が高速化されるだけでなく、検体溶液が流れ込む(吸引)側の捕捉部材表面に複数の微粒子3が一時的に捕捉され、この時の捕捉部材で捕捉された微粒子3を検出することで、目的とする反応物のみを有効に測定することができる。

また本実施形態の検査システムは、従来のマイクロアレイを利用する装置の構成よりも簡易であり、ビーズアレイを利用するによる方法においても、識別装置よりも、簡易な作業で識別精度が高く、正確な測定を行うことが出来る。

この捕捉部材1a、1bにおける検体溶液が流れ抜ける粗さ(孔径)としては微粒子3のサイズよりも細かいものを利用することで、微粒子3が捕捉部材1a、1bを透過することなく、捕捉部材で捕捉することができ、これを検出することでB/F分離が可能となる。

この際に、捕捉部材1a、1b上へのプローブの固相化が不要であることから、固相化反応に適した捕捉部材基材を選択する必要がないため、捕捉部材1a、1bの基材の選択範囲の制限が少なく、検出部5にあわせて、好適する捕捉部材基材を利用すればよい。例えば、検体を蛍光標識した場合には、捕捉部材1a、1bの基材に自家蛍光の少ないものを用いることが好ましい。但し、捕捉された微粒子3をその捕捉部材面と対面する方向以外から観察し、捕捉された微粒子3のみを検査するような場合には、捕捉部材1a、1bの自家蛍光により制約されることはない。

また、検出部5のカメラによる画像の取得は、広い視野に広がる複数の粒子の画像を一括取得することができ、効率よく検査画像を取得することが可能となる。尚、検出部5は、カメラだけでなく顕微鏡を応用した検査装置が考えられる。捕捉部材に対する溶液の駆動によって検体溶液中の微粒子3は捕捉部材面上に展開される。検体溶液が吸引されている状態では捕捉部材表面には、微粒子3だけが散在するため、これらを顕微鏡に付属して設けられている検査装置によって、画像として取得する。さらに、検出部5は、カメラを利用した画像の取得に限らず、スキャニングなどにより取り込むことも可能である。

また、この際に、B/F分離をより確実に行うために検体溶液を抜き取り、洗浄を行うことはS/Nの向上に有効である。この場合には、検体溶液を吸引して廃棄し、新たに洗浄溶液を添加し、再度、検体溶液の流動を繰り返す。また、必要に応じて同様な洗浄を複数回繰り返すことによって、S/Nをより向上させることが可能となる。

本実施形態における微粒子3は、球形状に限定されず、楕円体、直方体、円柱等のいかなる形状であっても問題ない。また、微粒子3の材質としては、限定されるものではないが、ガラス・金属などの無機物が微細加工や化学的な修飾、自家蛍光によるバックグラウンドの問題に対して有利であり、樹脂製のものは、安価で用意に粒径の揃った微粒子3を用意することが可能であり、用途に応じて使い分けることが望ましい。

また、検体溶液中への分散の度合いは、限定されるものではないが検査面に対する微粒子3の締める面積が20〜80%程度となることが望ましい。20%より少ないと、検査の精度が低下し、また、80%を超えると捕捉部材表面で微粒子3の重なりが発生し、正確に検査することが困難となる。

このように微粒子3の重なりを防止するために、種々の方法が考えられる。

例えば、捕捉部材面に対する溶液の駆動により微粒子3が捕捉されるが、この際に溶液の駆動方向とは直交する面方向に系全体を振盪することで、捕捉部材表面上に均一に分散させることが可能となる。他にも、超音波を用いて分散させる方法や、微粒子3自体に電荷や磁気を持たせることで、外部からの電圧印加や、磁力により分散状態を制御し、均一に捕捉部材表面上へ分散させることができる。

また、図3(a)に示すように捕捉部材1表面の全面に亘り微粒子3を均一に捕捉できるように、微細加工により多数の凹部2を形成することも有効である。この場合には、図3(b)に示すように微粒子3は、それぞれの凹部2に捕捉され、重なりを防止することが可能である。

さらに、図4(a)、(b)に示すように1表面の全面に亘り微粒子3を均一に捕捉できるように、微細加工により多数の凹凸部11を形成してもよい。この凹凸部11により、図4(c)に示すように、微粒子3が、それぞれの凹凸部11の凹部分に捕捉され、重なりを防止することが可能である。

また図5に示すように、チューブ4を縦方向に配置して、上下に捕捉部材1a、1bを嵌め込み、検体溶液を上下に流動させつつ、微粒子3を挟み込むようにすることにより、分散状態を制御することができ、微粒子3の重なり合いによる検査精度の低下を抑えることが可能となる。尚、図1に示した構成において、捕捉部材表面を撮像するのではなく、捕捉部材1a、1bの間のチューブ4部分を撮像する場合には、捕捉部材1a、1bには、前述したような表面加工等を施す必要はない。また図5に示すように縦方向にチューブ4を配置した場合で上方向から撮像する場合には、上側の捕捉部材が透明か又は、微粒子3の蛍光を透過するように構成される。

また、検体溶液に含まれる検体への標識は、蛍光で行うことが好ましいがこれに限定されるものではない。蛍光は、生体由来の検体に対する標識や取り扱いが容易であること、比較的安価であることから生体由来の検体の検査に広く利用されている。例えば、検体にcDNAを利用する場合には、例えばRNAよりの逆転写反応の際に、蛍光標識基質として取り込ませたり、プライマの末端に蛍光標識することで、PCRによる増幅により標識を行ったりと、通常の分子生物学的な手法にて行われている方法にて容易に検体に修飾を行うことができる。

また、微粒子3に固相化するプローブは、核酸のみならず生体由来の物質として、例えば、タンパク質や抗体といったものを固相化することが可能であり、幅広いアッセイへの応用が可能である。

検出部5で検出された画像から特定の微粒子3を識別するためには、その画像に対して種々の処理を施す。例えば、取得された画像を、微粒子3の散布状態に応じて、複数の領域に分割する。それぞれの分割された領域に対し、微粒子3の識別情報を検出し、同時にその強度を測定する。この識別情報と結合強度の関係をもって、結果の出力値とする。

ここで、画像の個々の微粒子3は、画像上で重なり合わさらないようにことが好ましい。検体溶液の流動を利用して捕捉部材表面上にトラップされた微粒子3の展開状態は、検体溶液が往復する毎に変わるため、検体溶液が流動するごとに画像を取得し、複数の画像から個々の微粒子3に最適な状態を結果として採用することや、また、複数の結果の平均値などを評価することで、結果を導くことが可能となる。勿論、積極的に散布することも可能である。

このような反応系に最適な微粒子3の粒径は、10μm〜100μm程度の直径が有効であり、これ以上微小な場合には、微粒子3の加工や識別情報の付加が難しくなる。また、これ以上大きい場合には1つの系内に導入するプローブの種類を多くした場合には、微粒子3の数量が多くなり、検出が困難となる。経験的には、上記程度の粒子径が適当であるが、勿論、これに限定されるものではない。

以下に、前述した検査システムを用いた生体物質の検査方法を実施した第1の例について説明する。

微粒子3として、ポリスチレン製からなる4種のサイズの球形状を用意した。それぞれ平均粒径は、25μm、50μm、75μm、100μmとする。それぞれの微粒子3の粒径は非常に精度よく揃っており、バラツキは±5μm程度であり、そのサイズによる種の識別は容易である。尚、サイズのバラツキが少ない程、使用できる種別の数が多くなる。

夫々の粒径の微粒子3の表面にアミノ基を導入し、またプローブの合成核酸の末端をアミノ基修飾し、それぞれをBis[Sulfosuccinimidyl]suberateを用いて結合させた。本例ではDNAの変異を解析することを目的として、それぞれの微粒子3に1塩基ずつ異なる4種のプローブとした。

これらのプローブを固相した微粒子3をFITCにて蛍光標識済みのK−rasガン遺伝子(K−rasコドン12を増幅するプライマーを利用してPCRによって増幅しておいたもの)と混合した。混合した検体溶液を、陽極酸化により得られた多孔質薄膜上に滴下し、図2に示した構成による検出システムを用いて65℃でのハイブリダイズ反応を行った。尚、微粒子3の構成(組合せ)により照明部10及び検出部5を適宜、変更できるような構成となっている。一例として、微粒子3の形状を可視光の落射照明により行い、微粒子3の輝度の検出を蛍光で行うものを示す。

照明部10にはメタルハライド光源を用い、落射光学系にはOLYMPUS製の蛍光落射ユニットを用いた。可視光の落射には、OLYMPUS製のBFLの捕捉部材セットを用いた。また、蛍光画像の測定に関しては、各種の蛍光色素に対応した捕捉部材セットをソフトウエアより制御し変更可能とした。ここでは蛍光標識をFITCにて行い、その検出には、OLYMPUS製WIBA捕捉部材セットを用いた。

同時にカメラによる撮影と溶液の駆動を連動して制御できる構成とし、検体溶液の流動を往復させる反応と連動して、蛍光画像及び、明視野画像を取得した。これを画像処理のソフトウエアにて、明視野画像より各微粒子3のサイズを、蛍光画像よりその蛍光強度を測定した。これにより、以下の結果が得られた。結果はカメラの出力階調(輝度)から計算し、検出値の最大を100%として表示したものである。

この結果より、検体はプローブ2とのパーフェクトマッチである変異を持つものであることが確認された。

次に、本発明の生体物質の検査方法を実施した第2の例について説明する。 微粒子3は、シリコン基板にフォトリソグラフィー技術を用いて複数の微小なバーコード状の識別記号を設けた後、これらをダイシングにより小片化して作製される。この微粒子3には、100種の識別情報を付与し、癌関連の100の遺伝子のプローブを固相化した。

プローブ核酸は、末端をアミノ基にて修飾し、微粒子3の表面には、チオール基を有するシランカップリング剤をあらかじめ反応させ、それぞれのプローブ核酸とをN−(4−Maleimidobutyryloxy)succinimideにて架橋させた。得られた微粒子3を等量ずつ混合させた溶液を調製した。サンプルには、正常ヒト繊維芽細胞株TIG−1(以下、TIG−1と略す)由来のtotal RNAと、ヒト大腸癌細胞株COLO320DM(以下、COLO320DMと略す)由来のtotal RNAを各25μg用意し、これらを鋳型としてoligo dT primerを用いて逆転写法によって、FITC標識1本鎖cDNAを作製した。このFITC標識されたcDNAを滅菌蒸留水35μlに溶解させ、95℃で5分間加熱して変性させ、その後、氷水中で急冷した。このcDNA溶液に、20×SSPE溶液を15μl加えて(最終塩濃度6×SSPE溶液)、液体試料とした。それぞれの検体サンプルを平均孔径10μm、膜厚100μmのナイロン製の捕捉部材にて、前述した検査システムを用いて50℃でハイブリダイズ反応を行った。

そして検出部5により検出された明視野画像を制御・解析用コンピュータ8で解析して、各微粒子3の識別情報を判読するとともに、同時に取得された蛍光画像より、各微粒子3の蛍光強度を測定し、各プローブに対する検出シグナルを表にして各プローブのRNAの発現量解析を行った。

このうち、ヒトゲノム中に存在しない遺伝子1個(ネガティブコントロール;NC)、COLO320DMにおいて、特異的に発現が上昇すると予測される遺伝子1個(c−myc)、代表的なハウスキーピング遺伝子1個(β−action)、両細胞間で発現量の変動がないと予測される遺伝子1個(GAPDH:インターナルコントロール;IC)を選択し、それぞれのcDNA配列に特異的な塩基配列に着目し、評価を行った。その結果、NC(ネガティブコントロール)では共に蛍光が認められないことから、ハイブリダイゼーション反応が特異的に進行したことが示された。また、c−mycとβ−actionのシグナルについて観察してみると、TIG−1に比べて、COLO320DMのほうが確かに蛍光強度が強くなっていることが認められた。

また、各遺伝子についての蛍光強度を算出したところ、GAPDH(IC)の輝度を1:1に補正し、各遺伝子の発現量の比を比較したところ、COLO320DMでは、c−mycが5.5倍、β−actionが1.4倍の上昇が認められた。この結果を、RT−PCRの結果と比較したところ、ほぼ同様の結果が得られた。また、他のプローブの結果についても同様に、癌にて発現量が上昇するとされる遺伝子群、及び発現量が低下するとされる遺伝子群のそれぞれにて、COLO320DMでは、リファレンスであるTIG−1に対する発現量が正しく解析されていることが確認できた。

【発明の効果】
