本発明は、野菜などの農作物における農作物の表面に残留している農薬成分を分析する残留農薬分析方法に関する。

【従来の技術】
多くの殺虫剤や殺菌剤は、生体内に取り込まれた場合に、生体内の本来のホルモン作用に影響を与える外因性内分泌攪乱化学物質(いわゆる環境ホルモン(EDS))となり得る。このことから、近年、食品の安全性に対して消費者の関心が高まっており、特に生で食される農産物等の食品について、残留農薬の低減が望まれている。
そこで、現在、食品等の残留農薬を迅速且つ正確に測定する方法の開発が期待されている。

従来はクロマトグラフ等により残留農薬量の測定が行われていた。しかし、従来の測定法では、残留農薬を測定するために数時間、場合によっては数日を要していた。このため、すべての食品等について残留農薬を測定するのは不可能で、消費者の要望に十分応えることができなかった。更に、クロマトグラフやその他の生物測定法では、多くの複雑な化学処理が必要であった(非特許文献1)。

そこで本発明者らは、これまでに、全反射減衰赤外分光法(ATR赤外分光法)を用いた農産物の残留農薬測定方法を開発している(非特許文献2〜6)。この測定方法によると、10分程度で複数の残留農薬の量を測定することが可能である。

【発明が解決しようとする課題】
上記ATR赤外分光法を用いた場合、各試料の測定の度にバックグラウンドスペクトルを測定する必要があった。また、この方法では、果物や穀物等のプリズムの表面に密着させることのできないものは測定することができなかった。

本発明はこのような課題を解決するために成されたものであり、赤外分光光度計を利用した拡散反射法を用いることにより、平坦な表面を持たない農産物についても、非破壊で残留農薬濃度の測定を行うことが可能な残留農薬分析方法を提供する。

【課題を解決するための手段】
上記課題を解決するために成された本発明に係る残留農薬分析方法は、農作物の表面に残留している農薬成分の濃度を測定する残留農薬分析方法において、
a)前記農薬成分の残留濃度が既知の種々の値である複数の農作物について、農作物表面における前記農薬成分によるフーリエ変換拡散反射赤外スペクトルのデータを、赤外分光光度計を利用して拡散反射法により採取する第1ステップと、
b)第1ステップにおいて採取されたデータを1次微分スペクトルに変換する第2ステップと、
c)第2ステップにおいて得られたスペクトルに部分最小自乗回帰法を適用して、各農薬濃度を算出する最適モデルを導出する第3ステップと、
d)赤外分光光度計を利用した拡散反射法により、前記農薬成分の残留濃度が未知である分析対象の農作物の表面における前記農薬成分によるフーリエ変換拡散反射赤外スペクトルのデータを採取する第4ステップと、
e)第4ステップにおいて採取されたデータを1次微分スペクトルに変換する第5ステップと、
f)第5ステップで得られた1次微分スペクトルに、第3ステップにおいて得られた最適モデルを適用して、前記分析対象の農作物の表面の前記農薬成分の残留濃度を算出する第6ステップと、
を有することを特徴としている。

【発明の実施の形態】
まず、農薬成分の残留濃度が既知の種々の値である複数の農作物に対し、それらの表面に残留している該農薬成分によるフーリエ変換拡散反射赤外スペクトルのデータが、赤外分光光度計を利用した拡散反射法により採取される(第1ステップ)。すなわち、赤外光を試料に入射し、拡散反射した赤外光を検出器で検出することにより、フーリエ変換拡散反射赤外スペクトルのデータが採取される。次に、このスペクトルデータが1次微分スペクトルに変換される(第2ステップ)。続いて、第2ステップにおいて得られたスペクトルに部分最小自乗回帰法を適用して、各試料の農薬濃度を算出する最適モデルが導出される(第3ステップ)。以後、第4ステップから第6ステップにより、未知の農作物における農薬成分の残留濃度を迅速に測定できる状態となる。更に、第7ステップにより、未知の農作物における農薬成分の残留濃度が許容値以下か否かを迅速に判別することができる。

赤外分光光度計を利用した拡散反射法を用いることにより、平坦な表面を持たないレタス等の農産物試料についても、試料を破壊することなく、短時間で、試料表面に残留する農薬の濃度を測定することができる。

【実施例】
図1に示すように、上向き拡散反射装置11(パイク・テクノロジーズ社)を備えるFT−IR分光光度計10(島津製作所FT−IR8200D)により、レタス試料12(crisp heads:レタス株)のフーリエ変換拡散反射赤外スペクトルを測定する。マイケルソン干渉計(図示せず)からの干渉赤外光13を平面鏡セット(下方鏡及び上方鏡)のうちの上方鏡14を介して楕円鏡15に投射し、その焦点面上に配置した試料12に照射する。試料から発せられた拡散反射赤外光16は楕円鏡15により集光され、平面鏡セットのうちの下方鏡17により反射されて焦電検出器18で検出される。検出器18からは、試料から発せられた拡散反射赤外光に応じた検出信号が出力される。
ランベルト−ベール(Lambert−Beer)の法則によれば、高速フーリエ変換された拡散反射赤外光は、試料の化学的情報を含んでいる。本発明においては、この原理を利用して残留農薬濃度の測定を行っている。

検出器18は、制御部20およびパーソナルコンピュータ(パソコン)30に接続されている。
制御部20は、検出信号をA/D変換して干渉信号データを得る。そして、この干渉信号データをフーリエ変換することによってフーリエ変換拡散反射赤外スペクトルのデータを作製する。実際に拡散反射法により試料12の拡散反射赤外スペクトルを測定する場合は、まず、試料12が無い状態で拡散反射赤外スペクトルを測定する。これがバックグランドスペクトルである。次に、試料12を試料載置部に載置して拡散反射赤外スペクトルを測定する。この試料12の拡散反射赤外スペクトルを前述のバックグランドスペクトルで減算することにより、拡散反射による赤外スペクトルのデータ(以下「スペクトルデータ」という)が得られる。このスペクトルデータに基づき、制御部20内の表示部に拡散反射赤外スペクトルが表示される。また、このスペクトルデータは、インタフェイスを介してパソコン30に転送される。

パソコン30は、クベルカームンク(Kubelka−Munk)変換を行うことにより、透過スペクトルと同じ形態を有するスペクトルに変換する。その後、1640cm−1の水の吸収波数で正規化したスペクトル、及び較正モデルを導出するために、サヴィツキー−ゴーレイ(Savitzky−Golay)法を用いて、1次微分スペクトルに変換する。そして、この1次微分スペクトルに基づき、部分最小自乗回帰法を用いた以下のような手法で、試料の残留農薬濃度を測定するための較正モデルを算出する。そして、この較正モデルのうちの最適モデルに基づき、試料12に残留する農薬の濃度を算出する。

(残留農薬の種類及び参照値)
本実施例においては、そのままの状態のレタス株(intact lettuce heads)を試料として用い、以下の3種の残留農薬を分析した。(1)メソミル殺虫剤(S−メチル N−(メチルカルバモイロキシ)チオアセチミド)、(2)フェンバレレート殺虫剤((RS)−α−シアノ−3−フェノキシベンジル)、(3)ベノミル殺菌剤(メチル 1−(ブチルカルバモイル)−2−ベンジミド−アゾレカルバメート)。
農場から40個のレタス株を試料として採取し、農薬適用手順によって3グループに分けた。第1グループは農薬を全く散布しないもの(以下、「農薬未使用レタス試料」とする)、第2グループは残留レベルが許容値以下となるように通常の手順で農薬を散布したもの、そして第3グループは残留レベルが許容値以上となるようにしたもの(以下、「農薬過使用レタス試料」とする)である。各試料のグループ分けはクロマトグラフ法により確認した。そして、その結果の値をppm(10−6g/g)で表し、それを試料の参照残留農薬濃度とした。

次に、各試料のスペクトルを、試料側面の中央部分において測定した。スペクトル測定の際、波数分解能は2cm−1に、積算回数は100回とした。拡散反射赤外スペクトルは2800cm−1〜800cm−1の範囲で測定した。クベルカームンク(Kubelka−Munk)変換を行うことにより、透過スペクトルと同じ形態を有するスペクトルを得た。その後、1640cm−1の水の吸収波数で正規化したスペクトル、及び、較正モデルを導出するための1次微分スペクトルを得た。

(部分最小自乗回帰法を用いた較正及び検定手法)
試料の残留農薬濃度を測定するための較正モデルは、スペクトルの部分最小自乗回帰(PLSR)により得た。この際、マトラブ・ケモメトリクス・ツールボックス(Matlab chemomemetrics toolbox)(マスワークス社製:Mathworks Inc.)を使用した。PLSRで、参照残留農薬濃度及びスペクトルデータから主成分ベクトルを連続的に抽出した。この作業は、予測誤差が最小になるまで連続して行った。その後、モデルの相互較正を一つ抜き法(leave−one−out method)により行った。PLSRモデルの最適因子数を得るために、各々の抽出因子の各数値について二乗和誤差(PRESS)値を計算した。測定により得たスペクトルデータをクベルカームンク(Kubelka−Munk)変換により変換し、これに対してサヴィツキー−ゴーレイ法により得られる1次微分スペクトルをPLSRの際に使用した。これらのスペクトル加工は、ケモメトリクスソフトウェアであるSHIMADZU Hyper IR(島津製作所製)により行った。

図2に(a)農薬過使用レタス試料、(b)農薬未使用レタス試料、(c)メソミル殺虫剤、(d)フェンバレレート殺虫剤、(e)ベノミル殺虫剤、の拡散反射スペクトル図をそれぞれ示す。図2では、試料のスペクトルを1640cm−1の水の吸収波数で正規化している。図2に示されるように、農薬を全く散布しない試料と農薬残留レベルが許容値以上となるようにした試料では、1800cm−1〜1000cm−1のスペクトルパターンにおいて、農薬の適用状況の特徴が現れている。すなわち、農薬残留レベルが許容値以上となるようにした試料において、各農薬に特徴的なピークがより顕著に現れている。なお、各農薬の標準試料スペクトルの特徴的な吸収バンドは表1に示す通りである。

各試料のスペクトルは、各農薬の特徴的な吸収バンド領域において、大きな差異が見られた。この差異は、正規化されたスペクトルよりも、図3に示す1次微分スペクトルにおいて、より明らかであった。農薬残留レベルが許容値以上となるようにした試料においては、図3において矢印で示された農薬の特徴的な吸収バンドに由来する1次微分スペクトルの主なピークまたは谷が明確になっている。
更に、本発明に係る方法では1時間に24試料の測定を行うことができ、1時間に6試料の測定しか行うことができないATR法と比べて迅速に試料の測定を行うことができる。
また、本発明に係る方法によれば、各々のサンプルの測定を行う度にバックグラウンドスペクトルを測定するという必要がないため、迅速に測定を行えるだけでなく、容易に測定を行える。

図4は1次微分スペクトルにおける一つ抜き法によるPLS法の相互較正を行った結果の、PLS因子数と二乗和誤差の関係を示している。信頼度95%のF検定の結果、メソミル殺虫剤に対するPLSモデルの最適因子数は6、フェンバレレート殺虫剤に対するPLSモデルの最適因子数は5、及びベノミル殺菌剤に対するPLSモデルの最適因子数は4であった。
表2に示される1次微分スペクトルを用いて得られるモデルにより、最適判別法が得られる。

この1次微分スペクトルにより得られるモデルは、正規化スペクトルにより得られるモデルよりも正確であることがわかっている。これは農薬残留レベルが許容値以上となるようにした試料の1次微分スペクトルにおいて、農薬に特徴的な吸収バンドが、正規化スペクトルのバンドと比べてより顕著となっている実験結果からもわかる。これらのモデルは0.9よりも大きい相関係数を有している。

図5にメソミル殺虫剤、フェンバレレート殺虫剤、及びベノミル殺菌剤に対するPLS最適モデルによる残留農薬濃度値と参照残留農薬濃度値の関係を表す散布図を示す。信頼度95%のt検定によるモデルにおいては偏り(bias)及び歪み(skew)のいずれに対しても帰無仮説が成り立ったため、偏りおよび歪みのいずれの補正もこれらのモデルには必要ではなかった。
本発明の方法による測定の結果、フェンバレレート殺虫剤の標準残留濃度は2.0ppm、ベノミル殺菌剤の標準残留濃度は0.8ppmであり、また、これらの残留殺虫剤の測定に対する較正の標準誤差は、フェンバレレート殺虫剤の残留基準値またはベノミル殺菌剤の登録保留基準値といった農薬の安全基準値の10分の1以下であった。このように、本発明に係る残留農薬測定方法は、他の方法で測定を行った場合と比べて高い感度と高いS/N比を有する。このことから、上記モデルによれば、正確な食物の安全性診断が行えるものと思われる。
一方、メソミル殺虫剤のモデルの標準誤差は、メソミル殺虫剤の登録保留基準値0.5ppmに対して、0.62ppmであった。これは、0ppm〜10ppmという広い測定範囲で較正を行ったためであって、0〜2ppmといった狭い範囲で較正を行うことで、標準誤差を実用レベルまで改善することは容易に行うことができると考えられる。

現在、農業生産物について、残留農薬量の迅速な分析法が求められている。本発明に係る方法によれば、拡散反射赤外スペクトル方法を用いることにより、迅速かつ正確な測定を現実に行うことが可能である。本発明に係る方法を用いれば、一つのスペクトル測定にほんの2分程度しかかからない。また、本発明に係る方法を用いることにより、従来の方法と比べてより再現性よく正確に測定を行うことが可能である。また、最適な較正モデルを導出し、残留農薬の許容値を超えないように識別レベルをセットすれば、試料が安全性を満足するものであるか否かの識別作業を、農作物の流通過程で実際に行うことが可能となるものと思われる。
更に、本発明に係る方法は、農作物以外にも、内分泌攪乱物質を含む土壌や水などの測定にも用いることが可能である。

A calibration model for measuring pesticide residue concentrations in samples was obtained by partial least squares regression (PLSR) of the spectra. At this time, a Matlab chemometrics toolbox (Mathworks Inc .: Mathworks Inc.) was used. Principal component vectors were continuously extracted from the reference pesticide residue concentration and spectral data by PLSR. This operation was continuously performed until the prediction error was minimized. Thereafter, the models were cross-calibrated by the leave-one-out method. To obtain the optimal number of factors for the PLSR model, a sum of squared error (PRESS) value was calculated for each value of each extraction factor. The spectrum data obtained by the measurement was converted by Kubelka-Munk conversion, and the first-order differential spectrum obtained by the Savitzky-Golay method was used for the PLSR. These spectral processings were performed by SHIMADZU Hyper IR (manufactured by Shimadzu Corporation) which is chemometrics software.

The spectrum of each sample showed a large difference in the characteristic absorption band region of each pesticide. This difference was more apparent in the first derivative spectrum shown in FIG. 3 than in the normalized spectrum. In the sample in which the pesticide residue level is set to be equal to or higher than the allowable value, the main peak or valley of the first derivative spectrum derived from the characteristic absorption band of the pesticide indicated by the arrow in FIG. 3 is clear. .
Furthermore, the method according to the present invention can measure 24 samples per hour, and can measure samples more quickly than the ATR method, which can measure only 6 samples per hour.
Further, according to the method of the present invention, it is not necessary to measure the background spectrum every time each sample is measured, so that the measurement can be performed not only quickly but also easily.

FIG. 4 shows the relationship between the number of PLS factors and the sum of squares error as a result of performing the cross calibration of the PLS method by the single subtraction method in the first derivative spectrum. The F-test with 95% confidence showed that the optimal number of factors for the PLS model for mesomil insecticide was 6, the optimal number of factors for the PLS model for fenvalerate insecticide was 5, and the optimal number of factors for the PLS model for benomyl fungicide was 4 Met.

It has been found that the model obtained from the first derivative spectrum is more accurate than the model obtained from the normalized spectrum. This can be seen from the experimental result that the absorption band characteristic of the pesticide is more prominent in the first derivative spectrum of the sample in which the residual level of the pesticide is equal to or higher than the allowable value, as compared with the band of the normalized spectrum. . These models have a correlation coefficient greater than 0.9.

FIG. 5 is a scatter diagram showing the relationship between the residual pesticide concentration value and the reference residual pesticide concentration value by the PLS optimal model for the mesomil insecticide, fenvalerate insecticide, and benomyl fungicide. According to the method according to the present invention, it is possible to actually perform quick and accurate measurement by using the diffuse reflection infrared spectrum method. With the method according to the invention, one spectral measurement takes only about two minutes. In addition, by using the method according to the present invention, accurate measurement can be performed with higher reproducibility as compared with the conventional method.
