実施例および比較例で得られた液体クロマトグラフィー用充填剤すなわちエンドキャップされたシリカゲルについて、高分解能500MHz29Si固体NMR(日本電子社製「Jeol ECP−500」)を用いて、シングルパルスマジックアングルスピニング法で、下記条件で、Si種のNMR測定を行い、得られたNMRスペクトルから下記式によりシラノール基残量を算出した。

実施例1〜3および比較例1〜2で得られた液体クロマトグラフィー用充填剤の29Si−NMRチャートを図1、図3、図5、図7および図9にそれぞれ示す。これらから分かるように、実施例1〜3の液体クロマトグラフィー用充填剤では、残存シラノール基(−100ppm付近のピーク(Q3))は検出されなかった。

The 29 Si-NMR charts of the packing materials for liquid chromatography obtained in Examples 1 to 3 and Comparative Examples 1 and 2 are shown in FIGS. 1, 3, 5, 7 and 9, respectively. As can be seen from these, no residual silanol group (peak (Q 3 ) around -100 ppm) was detected in the packing materials for liquid chromatography of Examples 1 to 3.

実施例および比較例で得られた液体クロマトグラフィー用充填剤をステンレススチールカラム(内径4.6mm、長さ150mm)に充填し、アミトリプチリンを標準サンプルに用いて、液体クロマトグラフィー評価試験を行った。アミトリプチリンは抗うつ作用を有する強塩基性の化合物であり(pKa=9.4)、液体クロマトグラフィにおいてテーリングが激しく分析が難しい代表的な化合物である。液体クロマトグラフィー評価試験は、移動相を構成する緩衝液として残存シラノール基の影響を最も受けやすいもの(pH7.0、温度20℃)を用い、下記操作条件で行った。実施例1〜3および比較例1〜6の液体クロマトグラフィー用充填剤のアミトリプチリンの液体クロマトグラフィーチャートを図2、図4、図6、図8、図10、図11、図12、図13および図14にそれぞれ示す。
