本発明は、DNA断片間の相補的相互作用を利用して、DNAの構造を特定するために用いられる光学的DNAセンサに関するとともに、前記DNAセンサを用いたDNA読取装置に関する。

【従来の技術】
近年、医療分野、農業分野等の幅広い分野で生物の遺伝子情報が利用されるようになってきているが、遺伝子の利用に際しては、DNAの構造解明が不可欠である。DNAは螺旋状によじれあった二本のポリヌクレオチド鎖を有し、それぞれのポリヌクレオチド鎖は4種の塩基(アデニン:A、グアニン:G、シトシン:C、チミン:T)が一次元的に並んだヌクレオチド配列を有し、アデニンとチミン、グアニンとシトシンという相補性に基づいて一方のポリヌクレオチド鎖の塩基が他方のポリヌクレオチド鎖の塩基に結合している。

DNAの構造解明とは、ヌクレオチド配列を特定することであるが、DNAのヌクレオチド配列を特定するためにDNAマイクロアレイ及びその読取装置が開発されている。従来では、DNAマイクロアレイ及びその読取装置を用いて次のようにしてサンプルDNAのヌクレオチド配列を特定する。

まず、既知のヌクレオチド配列を有した複数種類のプローブDNA断片をスライドガラス等の固体担体に整列固定させたDNAマイクロアレイを準備する。次に、サンプルDNAを一本鎖のDNA断片に変性して、変性したサンプルDNA断片に蛍光物質等を結合させる。

次に、サンプルDNA断片をDNAマイクロアレイ上に添加すると、サンプルDNA断片は、ハイブリダイゼーションによってDNAマイクロアレイ上に固定される。つまり、サンプルDNA断片の塩基は、複数種類のプローブDNA断片のうち相補性を有するDNA断片の塩基と水素結合して、二本鎖が生じる。一方、サンプルDNA断片は、相補性を有しないプローブDNA断片とは結合しない。サンプルDNA断片に蛍光物質でマーキングを施しているため、サンプルDNAと結合したプローブDNA断片が蛍光を発することになる。例えば、TCGGGAAというヌクレオチド配列を有するサンプルDNA断片は、AGCCCTTというヌクレオチド配列を有するプローブDNA断片とのみ結合し、そのプローブDNA断片が蛍光を発する。

次いで、DNAマイクロアレイを読取装置にセッティングして、読取装置にて解析する。読取装置は、DNAマイクロアレイ上の蛍光強度分布を計測するものである。

読取装置は、大きくわけて二種類あり、面状光方式と共焦点レーザ方式(例えば、特許文献1参照。)がある。
面状光方式の読取装置は、励起光(主に紫外線)をDNAマイクロアレイに面照射し、DNAマイクロアレイをレンズでCCDイメージセンサに結像し、DNAマイクロアレイで発した蛍光をCCDイメージセンサで受光し、DNAマイクロアレイの面内の蛍光強度分布をCCDイメージセンサで計測するようになっている。

共焦点レーザ方式の読取装置は、レーザーダイオードから発した励起光をコリメーターレンズで収束し、レーザ光線をDNAマイクロアレイに対して二次元走査し、レーザ光線の二次元走査と共に顕微鏡及びフォトマルも走査させ、レーザ光線により発した蛍光を顕微鏡を介してフォトマルで受光し、蛍光強度をフォトマルで計測し、二次元走査によってDNAマイクロアレイの面内の蛍光強度分布を計測するようになっている。

何れの方式でも、DNAマイクロアレイ上の蛍光強度分布は二次元の画像として出力される。出力された画像内で蛍光強度が大きい部分には、サンプルDNA断片のヌクレオチド配列と相補的なヌクレオチド配列を有したプローブDNA断片が含まれていることを表している。従って、二次元画像中のどの部分の蛍光強度が大きいかによってサンプルDNA断片のヌクレオチド配列を確定することができる。

【発明が解決しようとする課題】
しかしながら、共焦点レーザ方式の読取装置は、顕微鏡と、レーザ光線及び顕微鏡をDNAマイクロアレイに対して走査する機構とを必要とし、そのため装置全体が大きいという問題がある。面状光方式の読取装置は、DNAマイクロアレイをCCDイメージセンサに結像するレンズを必要とし、同様に装置が大型である。

読取装置の大型化を抑えるために、文献公知の発明ではないが、本出願人は、固体撮像デバイスの表面に複数種のプローブDNA断片を配列して固定してなる光学的DNAセンサを開発している。この光学的DNAセンサを用いてDNAを特定するには、蛍光標識したサンプルDNA断片を固体撮像デバイス上の相補的なプローブDNA断片とハイブリダイゼーションさせ、この光学的DNAセンサを読取装置にセッティングする。そして、読取装置の光源から固体撮像デバイスの表面に励起光を照射すると、サンプルDNA断片と結合したプローブDNA断片が蛍光を発し、その部分が明るくなっている蛍光強度分布の画像を読取装置が固体撮像デバイスで読み取る。この光学的DNAセンサは、固体撮像デバイスの表面にプローブDNA断片を固定化しているため、レンズや顕微鏡が無くとも蛍光強度分布を表す二次元画像を固体撮像デバイスで鮮明な像を撮像することができ、読取装置の小型化を図れるという効果を奏する。

しかしながら、この光学的DNAセンサには、次のような問題点もある。即ち、固体撮像デバイスが蛍光のみならず励起光に対しても感度を示すため、サンプルDNA断片と結合していないプローブDNA断片の部分も明るくなってしまい、固体撮像デバイスで得られた画像のコントラストが低い。そのため、固体撮像デバイスで得られた画像中のどの部分の強度が大きいかを特定することが困難となる。

そこで、本発明の目的は、蛍光強度分布を表したコントラストの高い画像を取得することができるようにすることである。

【課題を解決するための手段】
以上の課題を解決するために、請求項1に記載の発明は、例えば図1〜図4に示すように、
蛍光体励起光の被照射によって蛍光を発するサンプルDNA断片を特定するために用いられる光学的DNAセンサにおいて、
固体撮像デバイス(例えば、固体撮像デバイス2)と、
前記固体撮像デバイスの表面に成膜された酸化チタン層(例えば、酸化チタン層34)と、
既知の塩基配列を有するとともに前記酸化チタン層上に配列されて固定された複数種のプローブDNA断片(例えば、スポット60,60,…)と、を備えることを特徴とする。

〔第一の実施の形態〕
図1は、本発明が適用された光学的DNAセンサを示した斜視図であり、図2は、この光学的DNAセンサの平面図であり、図3は、図2のI−I破断線で破断して矢印方向に見て示した断面図である。

酸化チタン層34は、後述のDNA同定方法に用いられる蛍光物質を励起させる蛍光体励起光(主に紫外線であり、特に308nmを中心波長とした帯域の紫外線。)を吸収し、蛍光体励起光によって励起された蛍光物質から発する蛍光(主に可視光線であり、特に520nmを中心波長とした帯域の可視光線)を透過する性質を有する。
吸収を特性づける光学物性パラメータである消光係数k(>0)は複素屈折率Nとの間で下記に示す式(1)の関係が成り立つ。

式(1)においてiは、虚数単位である。ここでnは所定方向に進む光の波の位相速度を決め、消光係数kは波の振幅の大きさを波の進行方向とともに減衰させる働きを有する。光の進行方向をzとし、その光の強度をIとすると、両者の間で下記に示す式(2)の関係が成り立つ。

ルチル型結晶の酸化チタン層34は、正方晶であり、チタン原子の配置から体心立方構造である。この結晶は光軸がC軸にある1軸結晶であり、厳密には入射光の電場ベクトルとC軸のなす角度によって複素屈折率Nは異なるが、平均的には、300nm程度の紫外線の消光係数kは2であり、440nm程度の可視光線では、消光係数kは0.06になり、460nmではk=0とみなせる。
図5(b)に、酸化チタン層34の厚さと、308nm波長の蛍光体励起光及び530nm波長の蛍光の透過率との関係を対数グラフで示す。図5(b)に示すように、酸化チタン層34の厚さが厚くなるにつれて、蛍光体励起光の透過率が低くなり、酸化チタン層34の厚さが100nm以上であれば、蛍光体励起光の透過率は1.0×10-3以下となる。一方、蛍光の透過率は蛍光体励起光ほど低くなく、酸化チタン層34の厚さに関わらず50%以上となっている。

以上のように、導電層32とトップゲート電極30のうちの少なくとも一方のITOの光吸収端は、キャリア密度の増加とともにエネルギーの大きい方へシフトしているからキャリア密度を小さくさせてより短波長の光を吸収させることができる。そして、図5(c)に、第一実施形態の構造で光学的DNAセンサ1の導電層32又はトップゲート電極30のITOのキャリア密度を1.0×1019〔1/cm3〕とし、ITOの光学定数N(308nm)=2.2−0.34i(ここでのiは虚数単位)としたときにおける、酸化チタン層34の厚さと308nm波長の蛍光体励起光及び530nm波長の蛍光の透過率との関係を対数グラフで示す。導電層32及びトップゲート電極30の両方のキャリア密度が1.0×1019〔1/cm3〕を越えた場合の図5(b)と比較しても、308nm波長の蛍光体励起光が導電層32又はトップゲート電極30において更に遮蔽されていることがわかる。

第二実施形態の光学的DNAセンサの製造方法は、第一実施形態の光学的DNAセンサ1の製造方法とほぼ同じであるが、トップゲート電極30のITO層、導電層32を成膜する際に、そのキャリア密度が1.0×1020〔1/cm3〕以下になるように成膜速度及び雰囲気中の酸素濃度を調整する。なお、成膜速度が一定である場合には、ITO成膜反応炉内の酸素分圧を高くして酸素濃度が増加するに従ってITO中の酸素欠陥が減り、キャリア密度が減らすことができる。また、ITO成膜反応炉内の酸素分圧、つまり反応炉雰囲気中の酸素濃度が一定である場合には、成膜速度が遅くなるに従ってITO中の酸素欠陥が減り、キャリア密度が減らすことができ、酸素分圧が高い状態でITOの成膜速度を遅くすることが望ましい。

なお、誘電体多層膜35は、二種類の誘電体層が蛍光体励起光の中心波長の1/4の光学膜厚で交互に積層されたものに限らず、屈折率の異なる三種類以上の誘電体層が蛍光体励起光の中心波長の1/4の光学膜厚で周期的に積層されたものであっても良い。

【発明の効果】
