【発明の属する技術分野】
【0001】
本発明は、新規なモノクローナル抗体、その用途および製造に関し、さらに詳細には、各種ヒト型結核菌およびウシ型結核菌が分泌する多型な蛋白質MPT64および蛋白質MPB64の間で保存された領域を特異的に認識するモノクローナル抗体、その結核症診断用途および製造に関する。

【従来の技術】
結核菌群は分類学的には抗酸菌に属する菌群である。抗酸菌には、ヒト型結核菌(マイコバクテリウム ツベルクローシス Mycobacterium tuberculosis)、ウシ型結核菌(マイコバクテリウム ボビス Mycobacterium bovis)、ネズミ型結核菌(マイコバクテリウム ミクロチ Mycobacterium microti)、トリ型結核菌(マイコバクテリウム アビウム Mycobacterium avium)および冷血動物結核菌などが知られている。しかして、これらの中でヒトの結核症に関与するのは、ほとんど全てヒト型結核菌であるが、稀には、ウシ型結核菌による感染がある。また、ヒト型結核菌、ウシ型結核菌、マイコバクテリウム アフリカナム(Mycobacterium africanum)およびネズミ型結核菌以外のマイコバクテリウム属に属する微生物は、非定型抗酸菌と呼ばれている。

ヒト型結核菌(マイコバクテリム ツベルクローシス Mycobacterium tuberclosis)は、蛋白質MPT64(本明細書においては MPT64蛋白 とも記す)を特異的に産生し、菌体外へ分泌する。一方、BCG菌株のウシ型結核菌(マイコバクテリウム ボビス BCG(Mycobacterium bovis BCG)、以下、 BCG菌 と記す)は、蛋白質MPB64(本明細書においては MPB64蛋白 とも記す)を特異的に産生し、菌体外へ分泌する。そして、MPT64蛋白はMPB64蛋白と同一の物質であることが知られている。

M. Harboeらは、このMPB64蛋白をBCG菌から分離、精製して、その性質などについて研究して報告している(Morten Harboe et. al., Properties of Protein MPB64, MPB70, and MPB80 of Mycobacterium bovis BCG., INFECTION AND IMMUNITY, Vol.52, 293−302, 1986)。また、特開平1−247094号公報も、MPB64蛋白は、ヒトに感染して結核症と酷似した症状を呈する非定型抗酸菌によっては産生されないが、ウシ型結核菌のBCG菌によって産生され、かつMPT64蛋白と同一物質であり、BCG菌およびヒト型結核菌に特異な蛋白質であるとしている。

従って、このことは抗MPB64蛋白抗体は抗原をMPB64蛋白とする抗体であるが、同時にMPT64蛋白の抗体でもあることを意味する。従って、病原性的には無害なBCG菌を培養し、その培養液からBCG菌によって産生されたMPB64蛋白を抽出、精製し、該MPB64蛋白を抗原として抗MPB64蛋白抗体を作成し、その抗体を用いた抗原抗体反応(免疫反応)によって被検体中の結核菌を検出することにより、ヒトの結核症を明確、かつ迅速に判別することができる結核症の簡便な診断法を確立することが可能となる。なお、MPB64蛋白をコードする遺伝子の塩基配列およびアミノ酸配列はR. Yamaguchiらによって既に報告されている(Yamaguchi,R. et al., Cloning and Characterization of the Gene for Immunogenic Protein MPB64 of Mycobacterium bovis BCG, Infection and Immunity, Vol. 57, 283−288, 1989)。

抗MPB64蛋白抗体を使用した結核菌の検査薬および検出方法として、たとえば、特開平7−110332号公報記載の方法が知られている。この方法における免疫学的方法として、逆受身血球凝集反応(RPHA)、逆受身ラテックス凝集反応(RPLA)および固相酵素免疫測定法(ELISA)などが記載されている。前二者においては凝集時間が、たとえば、3時間程度と長く、感度が低く、かつ定量性は全くないなどの不都合な問題がある。後者においては、前二者に比較して試験操作が煩雑で、かつ通常は特殊な機械を使用する関係上、安全キャビネット中での実施が容易ではないなどの欠点がある。

そこで、本発明者らは、特開平11−108931号公報において、MPT64蛋白が混在する被検体を呈色標識抗MPB64蛋白モノクローナル抗体と免疫反応により結合せしめ、これによって生じた抗原抗体複合体を第二抗MPB64蛋白抗体により捕捉することを特徴とするイムノクロマトグラフィー測定法などによる結核菌検出法を提案した。しかしながら、最近、前記の免疫反応において、ヒト型結核菌のうちで陰性を示す或る種の結核菌が見受けられることが明らかとなった。

【発明が解決しようとする課題】
本発明者らは、従来の抗MPB64蛋白モノクローナル抗体が認識しないヒト型結核菌が存在することに鑑み、各種ヒト型結核菌およびウシ型結核菌が分泌する各種のMPT64蛋白およびMPB64蛋白に対して幅広く免疫反応するモノクローナル抗体を提供することにより、より精度の高い結核診断法を提供することを目的とする。

【課題を解決するための手段】
本発明者らは、上述の目的の下に鋭意研究した結果、結核菌の中にはMPT64蛋白をコードする遺伝子のレベルでミューテーションが起きているものがあり、遺伝子に多型が存在することを見出した一方で、そのN末端付近の配列は多型の中でも保存されていることを見出し、本発明を完成するに至った。

すなわち、従来の抗MPB64蛋白モノクローナル抗体が認識しないヒト型結核菌のうち、1つは、配列表の配列番号1で示される塩基配列において、66番目のアミノ酸のロイシン(Leu)から86番目のアミノ酸のプロリン(Pro)までの21個のアミノ酸をコードする遺伝子が欠損していることがわかった。また、他の1つは、配列表の配列番号1で示される塩基配列において、第266番目のシトシン(C)が欠損し、それ以降のコドンがフレームシフトを起こしていることがわかった。また、別の1つは、配列表の配列番号1で示される塩基配列において、512番目から687番目までの176bpの塩基が欠損していることがわかった。

その一方で、上記何れのヒト型結核菌においても、配列表の配列番号1で示される塩基配列において、24番目のアミノ酸のアラニン(Ala)から65番目のアミノ酸のセリン(Ser)までの42個のアミノ酸配列からなるペプチド(以下MPB64Δ(24−65a.a.)ペプチド と記す)をコードする遺伝子(以下 MPB64Δ(24−65a.a.)遺伝子 と記す)が保存されていることがわかった。

かくして、本発明者は、MPB64Δ(24−65a.a.)ペプチドに対するモノクローナル抗体を取得することに鋭意研究した結果、配列表の配列番号1の24番目のアミノ酸であるアラニン(Ala)から41番目のアミノ酸であるシステイン(Cys)までの18個のアミノ酸配列(配列番号3のアミノ酸配列)からなるペプチド(以下 MPB64ΔAペプチド と記すこともある)および42番目のアミノ酸であるグルタミン(Gln)から65番目のアミノ酸であるセリン(Ser)までの24個のアミノ酸配列(配列番号4のアミノ酸配列)からなるペプチド(以下 MPB64ΔBペプチド と記すこともある)のいずれも抗原性が強く、抗原決定基あるいはエピトープを含む領域であるとの知見が得られた。

したがって、本発明の第1の局面によれば、配列番号3または4のアミノ酸配列からなるペプチドに対するモノクローナル抗体が提供される。かくして、MPB64ΔAペプチドとMPB64ΔBペプチドの2種類の抗原のそれぞれに対する抗体が得られたので、変異の有無に拘わらず、これを使用して結核菌を検出することが可能となる。また、これら2種のモノクローナル抗体は、MPB64Δ(24−65a.a.)ペプチドのそれぞれ異なる領域に位置するMPB64ΔAペプチドおよびMPB64ΔBペプチドを認識するので、これら2種のモノクローナル抗体を併用することで、精度の高いサンドイッチ式免疫測定法が実施できる。

したがって、本発明の第2の局面によれば、配列番号3のアミノ酸配列からなるペプチドに対するモノクローナル抗体と、配列番号4のアミノ酸配列からなるペプチドに対するモノクローナル抗体とを併用するサンドイッチ式免疫測定法からなる結核症診断方法が提供される。ここにおいて、配列番号3のアミノ酸配列からなるペプチドに対するモノクローナル抗体及び配列番号4のアミノ酸配列からなるペプチドに対するモノクローナル抗体の何れか一方を担体に固定しておくこともでき、例えば、ELISA測定系やイムノクロマトグラフィー測定法に応用できる。

かくして、本発明の第3の局面によれば、配列番号3のアミノ酸配列からなるペプチドに対するモノクローナル抗体及び配列番号4のアミノ酸配列からなるペプチドに対するモノクローナル抗体の何れか一方を予め所定位置に固定せしめて形成された捕捉部位を備える膜担体を用意し、他方のモノクローナル抗体と所定量の被験試料との混合液を、前記捕捉部位に向けて前記膜担体にてクロマト展開せしめ、前記被験試料中に含まれる結核菌由来の蛋白と前記他方のモノクローナル抗体との複合体を前記捕捉部位に捕捉させることを特徴とするイムノクロマトグラフィー測定法が提供される。ここにおいて、上記他方のモノクローナル抗体は適当な標識物質で標識しておくと好都合である。

また、本発明の第4の局面によれば、配列番号3のアミノ酸配列からなるペプチドに対するモノクローナル抗体と、配列番号4のアミノ酸配列からなるペプチドに対するモノクローナル抗体と、膜担体とを少なくとも備え、前記モノクローナル抗体の何れか一方は前記膜担体の所定位置に予め固定されて捕捉部位を形成し、前記モノクローナル抗体の他方は適当な標識物質で標識され、かつ、前記捕捉部位から離隔した位置で前記膜担体にてクロマト展開可能なように配置されてなる結核症診断用イムノクロマト法テストストリップが提供される。
また、配列番号3および4のアミノ酸配列からなるペプチドがMPT64蛋白またはMPB64蛋白の免疫原性を示すことは従来知られていなかった。したがって、本発明の第5の局面によれば、以下の(a)〜(c)の何れか1つのペプチドが提供される:
(a)配列番号3のアミノ酸配列からなるペプチド、
(b)配列番号4のアミノ酸配列からなるペプチド、
(c)配列番号3または4のアミノ酸配列において1または数個のアミノ酸が欠失、置換または付加されたアミノ酸配列からなり、かつMPT64蛋白またはMPB64蛋白の免疫原性を示すペプチド。
さらに、本発明の第6の局面によれば、以下の(a)〜(c)の何れか1つのペプチドをコードする遺伝子が提供される:
(a)配列番号3のアミノ酸配列からなるペプチド、
(b)配列番号4のアミノ酸配列からなるペプチド、
(c)配列番号3または4のアミノ酸配列において1または数個のアミノ酸が欠失、置換または付加されたアミノ酸配列からなり、かつMPT64蛋白またはMPB64蛋白の免疫原性を示すペプチド。
配列番号3および4には特定の遺伝子配列が記載されているが、上記アミノ酸配列をコードする限り、コドンの縮重に基づく変更が許容され、また、MPT64蛋白またはMPB64蛋白の免疫原性を示す限り、1または数個のアミノ酸の欠失、置換または付加による変更が許容されることは言うまでもない。
また、この遺伝子は、常法により、各種の発現ベクターに組み込むことが可能であり、さらに、当該組換えベクターを適当な宿主に導入して形質転換して上記遺伝子を発現させることにより、MPT64蛋白またはMPB64蛋白の免疫原性を示す組換えタンパク質を製造することができる。宿主は、遺伝子工学で通常使用されている大腸菌などの細菌や酵母などが好適であるが、これらに限定されるものではない。
したがって、本発明のさらに他の局面によれば、上記遺伝子を含有する組換えベクター、当該組換えベクターを含む形質転換体、および、当該形質転換体によって発現された前記組換えベクター由来の、MPT64蛋白またはMPB64蛋白の免疫原性を示す組換えタンパク質が提供される。

【発明の実施の形態】
本発明のモノクローナル抗体の製造および本発明のモノクローナル抗体を使用するヒト結核症の診断法における各ステップは、それぞれ、それ自体、公知の免疫学的手法に準拠して行なわれる。

すなわち、MPB64蛋白に対する2種の特異的モノクローナル抗体は、例えば、Mycobacterium bovis 東京株のようなウシ型結核菌の培養上清から抽出精製したMPB64蛋白を抗原としてマウスのような動物を免疫したのち、この免疫された動物の脾臓細胞とミエローマ細胞とを細胞融合して得られた融合細胞をHAT含有培地でセレクトした後に増殖せしめる。

増殖せしめた株を前記のようにして得られたMPB64蛋白を使用して、たとえば、酵素標識免疫法などにより抗MPB64蛋白抗体産生株を選別する。さらに、MPB64Δ(24−65a.a.)遺伝子、MPB64ΔAペプチドをコードする遺伝子(以下、 MPB64ΔA遺伝子 と記す)またはMPB64ΔBペプチドをコードする遺伝子(以下、 MPB64ΔB遺伝子 と記す)を組み込んだ発現プラスミドを構築し、その発現蛋白を使用して、酵素標識免疫法などにより本発明のモノクローナル抗体産生株を選別する。

別法として、MPB64ΔA遺伝子またはMPB64ΔB遺伝子を組み込んだ発現プラスミド構築し、その発現蛋白を抗原としてマウスのような動物を免疫したのち、免疫されたマウスのような動物の脾臓細胞とミエローマ細胞とを細胞融合して得られた融合細胞使用して、上記と同様の選別を行ってもよい。

MPB64Δ(24−65a.a.)遺伝子、MPB64ΔA遺伝子またはMPB64ΔB遺伝子は、化学合成によって得ることができ、また、MPB64Δ(24−65a.a.)遺伝子を鋳型として適当なプライマーを用いてポリメラーゼ連鎖反応によって増幅させることによって得ることもできる。これらの遺伝子は、両末端に適当な制限酵素部位を付加するなど、常法により適当な発現プラスミドに組み込むことができる。

本発明の2種のモノクローナル抗体はそれぞれMPB64蛋白の2種の異なる抗原決定基あるいはエピトープを認識するので、両者を従来のサンドイッチ式免疫測定法に使用することで、従来よりも精度の高い結核症診断を実施できる。上記2種のモノクローナル抗体の何れか一方を固定しておくことも可能であり、これにより、例えば、イムノクロマトグラフィー測定法を容易に実施できる。

イムノクロマトグラフィー測定法は、公知のイムノクロマト法テストストリップの構成に準拠して容易に実施できる。一般に、イムノクロマト法テストストリップは、抗原の第一の抗原決定基にて抗体抗原反応可能な第一の抗体と、前記抗原の第二の抗原決定基にて抗体抗原反応可能で且つ標識された第二の抗体と、膜担体とを少なくとも備え、前記第一の抗体は前記膜担体の所定位置に予め固定されて捕捉部位を形成し、前記第二の抗体は前記捕捉部位から離隔した位置で前記膜担体にてクロマト展開可能なように配置されて構成される。具体的には、例えば、図3に示されるイムノクロマト法テストストリップが挙げられる。本発明においては、配列番号3のアミノ酸配列からなるペプチドに対するモノクローナル抗体と、配列番号4のアミノ酸配列からなるペプチドに対するモノクローナル抗体との何れか一方を第一の抗体として使用し、他方を第二の抗体として使用できる。

図3において、数字1は粘着シート、2は含浸部材、3は膜担体、31は捕捉部位、4は吸収用部材、5は試料添加用部材を示している。図示の例では、膜担体3は、幅5mm、長さ36mmの細長い帯状のニトロセルロース製メンブレンフィルターで作成されている。該膜担体3には、そのクロマト展開始点側の末端から7.5mmの位置に、抗原の第一の抗原決定基にて抗体抗原反応する第一の抗体が固定され、検体の捕捉部位31が形成される。図示の例では、膜担体3は、ニトロセルロース製メンブレンフィルターを用いているが、被験試料に含まれる検体をクロマト展開可能で、かつ、上記捕捉部位31を形成する抗体を固定可能なものであれば、いかなるものであってもよく、他のセルロース類膜、ナイロン膜、ガラス繊維膜なども使用できる。

含浸部材2は、前記第一の抗原決定基と異なる部位に位置する第二の抗原決定基にて前記抗原と抗体抗原反応する第二の抗体を含浸せしめた部材からなる。当該第二の抗体は、適当な標識物質で予め標識される。図示の例では、含浸部材2として、5mm×15mmの帯状のガラス繊維不織布を用いているが、これに限定されるものではなく、例えば、セルロース類布(濾紙、ニトロセルロース膜等)、ポリエチレン、ポリプロピレン等の多孔質プラスチック布類なども使用できる。

参考例1(MPB64蛋白の精製)
MPB64蛋白は、Mycobacterium bovis BCG TOKYO 172(ATCC 35737)の培養上清からMorten Harboeらの方法(Morten Harboe et. al., Properties of Protein MPB64, MPB70, and MPB80 of Mycobacterium bovis BCG., INFECTION AND IMMUNITY, Vol.52, 293−302, 1986)に準拠して抽出精製した。すなわち、Mycobacterium bovis BCG TOKYO 172を培養後、その培養上清を75%飽和硫安分画し、生じた沈殿物を、DEAE−Sephadex A−50カラムクロマト、次にDEAE−Sepharose CL−6Bカラムクロマト操作により分画した。ゲル電気泳動法により確認したMPB64蛋白相当画分を、限外ろ過濃縮後、さらに、DEAE−Sepharose CL−6Bカラムクロマト、限外ろ過濃縮、さらにSephacryl S−200カラムクロマトを行い、精製した。

参考例2(MPB64蛋白によるマウス免疫とハイブリドーマの作製)
参考例1で得られたMPB64蛋白200μg(50μl)をフロインドの完全アジュバンド50μlと混合してエマルジョンを作製し、これをBalb/cメスマウス皮下に注射した。この免疫操作を3週間後に再度行い、さらに3週間後にタイター価の上昇を確認した後、50μg(50μl)の上記MPB64蛋白そのものを腹腔に注射し、3日後に脾臓を摘出し、その脾臓細胞とマウス・ミエローマ細胞(SPII細胞)とを50%ポリエチレングリコール(分子量1500)溶液を用いて細胞融合した。融合細胞は、一般的方法のHAT(ヒポキサンチン、アミノプテリン、チミジン)含有培養液で選別後、PRMI1640培地(日水製薬(株)製、10%牛胎児血清(FBS(Fetal Bovine Serum))含有RPMI(Roswell Park Memorial Institute)培地)で培養増殖させた。

参考例3(MPB64蛋白特異的抗体産生ハイブリドーマの選別)

MPB64Δ(24−65a.a.)ペプチドを認識する抗体を取得することを目的に、MPB64Δ(24−65a.a.)遺伝子を備えた発現プラスミドを作成し,大腸菌で発現させた。

まず、MPB64Δ(24−65a.a.)遺伝子を含む配列表の配列番号2に示されるDNAの両ストランドを化学合成した。なお、この合成遺伝子では、ベクター挿入後に正常に融合タンパクが発現されるように、MPB64Δ(24−65a.a.)遺伝子のN末端側及びC末端側にそれぞれ7塩基及び6塩基のDNA配列が付加されている。

大腸菌発現用ベクターpRSET A 10μgを10mM Tris−HCl(pH7.5)、7mM MgCl2、60mM NaCl、7mM 2−メルカプトエタノール、PvuII 100単位の総計50μl反応溶液中で37℃、1時間反応させた。反応後、100℃、5分、熱処理をし、2.5倍量のエタノールと1/10量の3M酢酸ナトリウムを加えDNAのエタノール沈殿を行った。沈殿を減圧乾燥後、50mM Tris−HCl(pH8.0)、20単位 子牛胸腺アルカリフォスファターゼの総量50μl反応溶液を加え50℃で30分間反応させた。反応後、マイクロピュア−EZ遠心式酵素リムーバー(ミリポア)でろ過し、子牛胸腺アルカリフォスファターゼを除去した。ろ過液に2.5倍量のエタノールと1/10量の3M酢酸ナトリウムを加えDNAのエタノール沈殿を行った。この沈殿を減圧乾燥後、10μlの滅菌水を加えMPB64Δ(24−65a.a.)遺伝子DNA断片挿入用大腸菌発現ベクター断片とした。

乾燥した沈殿を20μlのTE緩衝液に溶解し、このうち2μlを0.5 ml 容積のチューブに移し、試薬LigationHigh(東洋紡績株式会社から購入)2μlと混ぜ、16℃で15時間、反応させた。凍結XL1−Blue大腸菌(フナコシ株式会社から購入)100μlを1.5 ml 容積のチューブに用意し、これに20倍希釈した2‐メルカプトエタノールを加え、10分間、氷上で時々振り、前記の16℃で反応させたDNA溶液を2μl加え、30分間、氷上で放置後、42℃に保温した恒温漕に45秒漬け、直ぐに氷上へもどし、900μlのLB培地(1 % bacto−tryptone、 0.5 % bacto−yeast extract、1 % 塩化ナトリウム)を加え、37℃で1時間培養し、これを50 mg/mlのアンピシリンを含むLB寒天プレートに接種し、37℃で18時間、保温した。

LB寒天プレートに生じた大腸菌コロニー(それぞれ25個)を50 mg/mlのアンピシリンを含む3mlのLB培地に接種し、37℃で18時間、培養し、この培養液を、9,000rpmで5分間遠心分離し、沈殿させ、菌体を回収した。この菌体からキアゲンプラスミドミニキット(株式会社キアゲンの商品)によりプラスミドを調製した。得られたプラスミドについて ABI PRISM Genetic Analyzer(アプライドバイオシステムジャパン株式会社)によりその塩基配列を調べた。それぞれ25個のプラスミドのうち、MPB64ΔA遺伝子またはMPB64ΔB遺伝子を含む目的の発現プラスミドが数個得られた。

生じた大腸菌コロニー(それぞれ4個)を3mlのLB培地に接種し、37℃で18時間培養した。この培養液を100mlのLB培地(50μg/mlのアンピシリンを含む)に入れ、37℃で培養し、OD600が0.6になったときに、100mM IPTGを1ml加え、さらに3時間培養した後、5,000rpmで20分間遠心分離し、沈殿させ、菌体を回収した。1.5 ml 容積のチューブにて、1ml培地あたりの菌体を200μlの10mMリン酸緩衝液に懸濁した後、200μlの2×SDSPAGE サンプリング液(50 mM トリス、1%ラウリル硫酸ナトリウム、5%2‐メルカプトエタノール)を加え、混合し、95℃で5分間、熱処理をし、このうち10μlをSDSポリアクリルアミド電気泳動およびウェスターン・イムノブロッティングに供与したところ、図1および図2のようにそれぞれのペプチドが発現していることが示された。なお、図1は、ゲルコードブルー((株)フナコシから購入)により蛋白質染色した結果であり、図2は、抗ヒスチジンタグ抗体(マウスIgG)にビオチン化抗マウスIgG抗体を反応させ、VECTASTAIN ABC Standard Kit((株)フナコシから購入)により呈色反応させた結果である。

実施例3で得られた形質転換大腸菌を培養し、IPTGによる蛋白発現誘導を行った後、溶菌操作を行い可溶性蛋白質を回収した。その可溶化画分の中から、MPB64ΔAペプチド含有発現蛋白質およびMPB64ΔBペプチド含有発現蛋白質をニッケル結合カラムを用いて分離精製した。それら精製蛋白質を2μg/mlの濃度でPBSに溶解後、その100μlを96穴ELISAプレートの各ウェルに添加し、固相化した。その後、参考例3に記述した操作方法により、MPB64Δ(24−65a.a.)ペプチドを認識する抗体産生ハイブリドーマ(すなわち実施例2で得られた6種)の培養上清を用いて、その特異性について検討した。

抗原64ΔA:MPB64ΔAペプチド含有発現蛋白質;
