本発明は、例えば蛍光相関分光法(FCS)による解析等に供される液浸対物レンズを有する光学顕微鏡を用いる光分析装置に関する。

【従来の技術】
一般に、この種の光分析装置としては、特開平10−39222号公報に開示されるものが知られている。この公報には、顕微鏡のステージの対物レンズ側に、対物レンズの方向に開放された液溜を設け、標本の液浸と同等の屈折率を持つ透明な液体を滞留させ、対物レンズの先端をこの液体に接触させることが記載されている。

【発明が解決しようとする課題】
しかしながら、上記従来技術では、液溜に滞留された液体に気泡が混じることがあり、気泡によって液溜に滞留された液体の屈折率が変化して、良好な像が得られなくなる問題がある。

本発明は、上記の事情に鑑みてなされたもので、構成簡易にして、対物レンズと試料の間に配置された液体中に気泡が混じるのを効果的に防止し得るようにした光分析装置、その運転方法及び顕微鏡を提供することを目的とする。

【課題を解決する手段】
本発明は、試料に光を照射するための光源と、前記試料に照射された光の透過光または反射光を取得する顕微鏡と、前記顕微鏡で得られた光を電気信号に変換する光電気信号変換手段と、前記光電気信号変換装置で変換された電気信号に基づいて前記試料の各種特性を求めるデータ処理手段とを具備する光分析装置において、前記顕微鏡を、前記試料を収容する容器を支持する容器支持部材と、前記試料に対向して配置される対物レンズと、前記対物レンズと前記容器との間に液体を供給する液体供給手段と、前記液体供給手段で供給された液体中の気泡を除去する気泡除去手段とを備えて構成した。

上記構成によれば、容器支持部材が試料を収容する容器を支持し、液体供給手段が対物レンズと容器との間に液体を供給し、気泡除去手段が供給された液体中の気泡を除去することにより、気泡による光の反射や散乱が減少され、液体中における屈折率が均一化される。従って、対物レンズの焦点合わせを精度良く行うことができ、しかも、液体中における光の損失が少なくなるため、焦点位置で試料への光の照射を効率よく行うことが可能となり、且つ、対物レンズに入る光の損失が液体中において軽減することができる。これにより、高精度な光の測定データを光電気信号変換手段で取得することができて、試料の精度良い分析結果を得ることが可能となる。

【発明の実施の様態】
以下、本発明の実施の形態に係る光分析装置、その運転方法及び顕微鏡について、図面を参照して説明する。

(第一の実施の形態)
図1は、本実施例の第一の実施の形態に係る光分析装置である蛍光分析装置100を示す。即ち、この蛍光分析装置100には、光源5、共焦点光学系を用いた倒立型蛍光顕微鏡1、該倒立型蛍光顕微鏡1で取得される蛍光標識された試料23(図2参照)が発する蛍光を、電気信号に変換する光電気信号変換手段である光電気信号変換部2、該光電気信号変換部2で求めた測定データに基づいて試料23の特性を求めるデータ処理手段であるデータ処理部3及び該データ処理部3で求めた試料23の各種特性を表示する表示部4が設けられる。また、この蛍光分析装置100には、上記各部を制御する制御部6(図3参照)が設けられる。

なお、上記光源5は、例えばレーザー光発生装置で構成され、上記光電気信号変換部2は、例えばフォトマルチプライヤやアバランシェフォトダイオードで構成される。

上記対物レンズ11の上方には、試料プレート12が配置される。この試料プレート12は、図2に示すように容器支持部材であるステージ13に支持されて、上記対物レンズ11の上方に配置される。ステージ13には、振動子21が設けられ、振動子21には、上記振動制御部22が接続されている。

ここで、上記制御部6について説明する。即ち、制御部6は、図3に示すように上記光源5及び光経路16,17の光学系を制御する光源・光学系制御部7、上記ステージ13の移動を制御するステージ制御部8、上記液体供給部14を制御する液体供給制御部9、上記気泡除去部15を制御する気泡除去制御部10、上記対物レンズ11の移動や焦点合わせを制御する対物レンズ制御部28、及び予め定められた蛍光分析装置の運転手順に従って各制御部7〜10、28に命令を与える中央制御部29を有する。

上記制御部6は、試料プレート12がステージ13にセットされると、中央制御部29が予め決められた位置にステージ13を移動させるようにステージ制御部8に命令する。ステージ制御部8は、命令に従ってステージ13を駆動させる。

ここで、上記液体供給部14により供給される液体24中の気泡を排除する方法について図4を参照して説明する。

まず、ステップS40において、試料プレータ12がステージ13にセットされ、このセット状態において、液体供給部14が駆動されて液体調整部20が対物レンズ11上に予め定められた量の液体24を供給ノズル19から吐出させて供給する(ステップS41)。

続いて、ステップS42に移行して、対物レンズ11を試料プレート12方向に移動させ、試料プレート12の底面18と対物レンズ11との間を液体24で満たす。その後、振動制御部22は、振動子21を駆動して、ステージ13を振動させる(ステップS43)。ここで、振動は、ステージ13及び試料プレート12を介して液体24に伝わり、液体24中に浮遊する気泡、試料プレート12の底面18に付着している気泡、対物レンズ11の表面に付着している気泡、が液体24から排除される。

このように、上記光分析装置は、液体供給部14で対物レンズ11と試料プレート12との間に液体24を供給し、この液体供給部14で供給された液体24に対して振動を与えて該液体24中の気泡を気泡除去部15で除去し、試料プレート12上に載置した試料23から対物レンズ11に入射した光を測定し、測定された光のデータに基づいて試料23の特性を求めるように構成した。

これによれば、液体24中の気泡が除去されることにより、気泡による光の反射や散乱が減少され、液体24中における屈折率が均一化されることにより、対物レンズ11の焦点合わせを精度良く行うことが可能となる。また、液体24中の気泡が排除されたことにより、光源5からの照射光の散乱による損失が少なくなることで、焦点位置で試料23への光の照射が効率よく行われる。さらに、対物レンズ11に入る蛍光の損失が液体24中において少なくなり、精度良い蛍光の測定データを、光電気信号変換部2で得ることが可能となる。

なお、上記実施の形態では、ステージ13に気泡除去部15の振動子21を設けて液体24中の気泡を除去するように構成した場合で説明したが、これに限ることなく、その他、図5に示すように振動子21を試料プレート12に接触させて配するように構成したり、あるいは図6に示すように振動子21を供給ノズル19に配するように構成することも可能である。これら図5及び図6の構成においても、同様の効果が期待される。

(第二の実施の形態)
図7は、本発明の第二の実施の形態を示すもので、上述した第一の実施の形態と同様の効果が期待される。但し、図7においては、上記図一及び図2と同一部分について、同一符号を付して、その説明に付いて省略する。

即ち、第二の実施の形態では、気泡除去手段として上記第一の実施の形態における気泡除去部15の振動子21に替えて音波発生器25を用いて構成される。この音波発生器25は、例えば超音波発生素子等で構成され、対物レンズ11の近傍に支持部材26で配置される。そして、この音波発生器25には、音波の発生を制御する音波制御部27が接続されている。

(第三の実施の形態)
図9は、第三の実施の形態に係る光分析装置である蛍光分析装置300を示すもので、上述した第一の実施の形態と同様の効果を期待することができる。但し、図9においては、上記図1と同一部分について、同一符号を付して、その説明を省略する。即ち、第三の実施の形態においては、例えば倒立型蛍光顕微鏡1の対物レンズ11、ステージ13、液体供給部14の供給ノズル19を減圧室30内に収容配置する。この減圧室30には、減圧ポンプ31が連結される。そして、この減圧ポンプ31は、減圧制御部32の接続され、この減圧制御部32を介して駆動制御される。

上記構成において、液体24中の気泡を排除するには、例えば図10あるいは図11に示す手順で実行される。
