【発明の属する技術分野】
本発明は、正イオンと負イオンを空間に放出して空気中の微生物を不活化したり、有害物質を分解・除去したりする環境調整方法及び環境調整装置に関し、例えば、医薬品、食品、化学物質などの製造施設の部屋や病院の集中治療室、半導体製造室あるいはバイオクリーンルームのように、衛生上、あるいは製品品質上、雰囲気中のカビや菌の数を抑えなければならない部屋などの環境調整方法及び環境調整装置に関するものである。

【従来の技術】
医薬品、食品、化学物質などは、消費者の健康に直接影響があるため、その製造施設の部屋は、衛生上、厳しい安全基準が設定されている。例えば、カビや菌の数にも基準があり、この基準を満たすよう、部屋の雰囲気中の菌数を厳しく管理しなければならない。

そのため、従来は、図17に示すように、HEPA(high efficiency particulate air)フィルターや活性炭フィルターなどの高性能なフィルター101を備えた空気浄化装置110を製造施設の部屋20の側壁21などに設けて、この空気浄化装置110を運転することにより、製造施設の部屋20の空気を吸い込んでフィルター101を通過させ、空気中に含まれる無数のカビ、菌などの微生物や、アンモニア、ホルムアルデヒドなどの有害物質を除去して、清浄な空気を送り出し、製造施設の部屋20の環境を清潔に保つようにしていた。

しかし、フィルター101では製造施設の部屋20の雰囲気中に存在するカビや菌を単に捕集するのみであって不活化させることはできず、製造施設の部屋20内に残存するカビや菌は、製造施設の部屋20の側壁21や製造ライン30に配置される製造装置31,32に付着して増殖を始める。ここで、「不活化」とはカビや菌に積極的に働きかけて、その活動を抑えることをいい、広義には「殺菌」や「除菌」という意味も含まれる。

【発明が解決しようとする課題】
したがって、このような製造施設では、製造ライン30を一定期間稼動するごとに、製造施設の部屋20を密閉状態にして、製造ライン30に配置された製造装置31,32をすべて停止し、ホルマリンにより製造施設の部屋20内を薫蒸消毒することなどが行なわれている。しかしながら、ホルマリンによる消毒効果は一時的なものであり、扉23の開け閉めや人や物の出入りによって製造施設の部屋20内に新たにカビや菌が持ち込まれ、製造施設の部屋20の側壁21や製造装置31,32に付着して増殖するので、再び消毒を行なわなければならなくなる。

図18は、製造施設の部屋20の側壁21や製造装置31,32などに付着した細菌の数の変化の一例を示している。製造ライン30の稼動中、製造施設の部屋20の側壁21などに付着したカビや菌が増殖し、その数がG2まで増加すると、製造ライン30を停止してホルマリンにより製造施設の部屋20の薫蒸消毒を行なう。この消毒により菌数をG1まで減らす。

しかしながら、上記のように、従来は製造ライン30の稼動中に、製造施設の部屋20の雰囲気中のカビや菌を不活化することができず、製造ライン30の稼動中に付着菌数がG2に達する時間が比較的短かったため、稼動から消毒までのサイクルt2も短くなり、消毒のたびに生産性が落ち、生産コストも高くなるという問題があった。更に、僅かな浮遊微生物あるいは、粒子の存在により製品が汚染され、その品質が劣化する問題があった。

【課題を解決するための手段】
上記の問題を解決するため本発明では、外部と隔離された環境内に正負イオンを放出して前記環境内の微生物を不活化すること及び/若しくは有害物質を分解及び/若しくは除去することにより、環境を調整するようにした。ここで、環境内の微生物には、環境の雰囲気中に浮遊している微生物と、環境を外部と隔離する壁などに付着した微生物とがある。また、環境内の有害物質とは、環境の雰囲気中に気体及び粒子の形で存在する有害物質を意味している。

図15は、正イオンと負イオンによる空気中に浮遊するクラドスポリウム(黒カビ)の除去試験の結果を示している。クラドスポリウムは、キノコなどの真菌に分類される微生物である。試験は、容積13.5m3の密閉容器内を温度25℃、湿度42%に保ち、この密閉容器内に一定数の生きたクラドスポリウムの胞子を満たした後、正イオンと負イオンを供給することにより行なった。図15のグラフはクラドスポリウムの数の変化をエアサンプラーで追跡したものである。なお、本試験のイオン送出条件としては、風量が4m3/分であり、イオン送風機の吹出し口から10cm離れた位置にて、正負イオン濃度はそれぞれ約4万個/cm3とした。また、図15の試験において対象空間の中央部において、正負イオンの濃度を測定したところ、正負イオン濃度は、それぞれ約2,000個/cm3であった。なお、このイオンの測定条件としては、イオンカウンターの加速電圧を調整し、1cm2/V・s以上の移動度を有するものを選択的に測定対象として、数値化している。

図15に示すように、イオンの供給により残存するクラドスポリウムの数は時間の経過に従って減少していき、供給を開始してから約4時間でクラドスポリウムをほぼ全滅させることができる。したがって、正イオンと負イオンにはクラドスポリウムに代表されるさまざまな環境で生息するカビ類、(例えば、パン酵母、赤パンカビ、青カビ、こうじカビ、水カビなど)の胞子を空中で不活化して、その繁殖を抑える作用があると考えられる。また、本効果を得るための適切なイオン発生条件は以上に限られるものではなく、菌種、希望する清浄度の条件に従って、イオン濃度は自由に変更することが可能である。

また図16は、正イオンと負イオンによる空気中に浮遊する大腸菌の除去試験の結果を示している。大腸菌は、微生物の中でも最も原始的なバチルスなどが属する細菌類に分類されるものである。試験は、容積36m3の密閉容器内を温度25℃、湿度42%に保ち、この容器内に一定数の生きた大腸菌を浮遊させた後、容器内に正イオンと負イオンを供給することにより行なった。図16に示す試験結果は、大腸菌の数の変化をエアサンプラーで追跡したものである。なお、図16の試験も、図15と同じイオン発生条件となるよう設定しており、風量が4m3/分であり、イオン送風機の吹出し口から10cm離れた位置にて、イオン濃度は4万個/cm3としている。また、図16と図15は空間の大きさが異なるため、風の方向等を若干調整することにより、図16の試験における対象空間の中央部において、正負イオン濃度は、それぞれ約2,000個/cm3に保つように設定している。

図16に示すように、イオンの供給により残存する大腸菌の数は時間の経過に従って減少していき、供給を開始してから約6時間で大腸菌をほぼ全滅させることができる。したがって、正イオンと負イオンには、大腸菌に代表されるさまざまな細菌類(例えば、サルモネラ菌、病原性大腸菌O157など)を空中で不活化して、その増殖を抑える作用があるものと考えられる。これらの細菌類には食中毒を引き起こす病原菌もあるが、例えば、冷蔵庫内にイオン発生素子を設けることにより、空中に浮遊する病原菌が食品へ混入することによる食中毒の防止も期待できる。

なお、図15、図16に示す試験においては、電極間に交流高電圧を印加することにより、空気中の酸素ないしは水分が電離によりエネルギーを受けてイオン化した、H+(H2O)m(mは任意の自然数)と、O2 −(H2O)n(nは任意の自然数)が主体のイオンを放出するようにしている。これらH+(H2O)m及びO2 −(H2O)nは、浮遊菌の表面に付着し、化学反応して活性種であるH2O2または・OHを生成する。H2O2または・OHは、極めて強力な活性を示すため、これらにより、空気中の浮遊細菌を取り囲んで不活化することができる。ここで、・OHは活性種の1種であり、ラジカルのOHを示している。

正負のイオンは浮遊細菌の細胞表面で式(1)〜式(3)に示すように化学反応して、活性種である過酸化水素(H2O2)または水酸基ラジカル(・OH)を生成する。ここで、式(1)〜式(3)において、m、m’、n、n’は任意の自然数である。これにより、活性種の分解作用によって浮遊細菌が破壊される。従って、効率的に空気中の浮遊細菌を、不活化、除去することができる。

また、上記式(1)〜式(3)は、空気中の有害物質表面でも同様に作用を生じさせることができるため、活性種である過酸化水素(H2O2)または水酸基ラジカル(・OH)が、ホルムアルデヒドやアンモニアなどの化学物質を酸化若しくは分解して、二酸化炭素や、水、窒素などの無害な物質に変換することにより、実質的に無害化することが可能である。

その他、正イオンと負イオンには、インフルエンザウィルス、コクサッキーウィルス、ポリオウィルス、エイズなどのウィルス類も不活化する働きがあり、これらウィルスの混入による汚染が防止できる。

また、正イオンと負イオンには、臭いの元となる分子を分解する働きがあることも確かめられており、空間の脱臭にも利用できる。

この技術を図17に示す製造施設の部屋20で製造ライン30の稼動中に利用すれば、製造施設の部屋20の雰囲気中のカビや菌を不活化し、浮遊菌数を一定の水準以下に抑えておくことが可能である。正負イオンには製造施設の部屋20の壁や製造ライン30に配置される製造装置31,32へ付着したカビや菌を不活化する作用は小さいが、このように浮遊菌数を抑えることで、カビや菌の付着を抑えることができるため、製造ライン30の稼動開始から製造施設の部屋20のホルマリン消毒作業までのサイクルを従来よりも長くすることができる。このことは、製造ライン30のトータルの稼働休止時間を短くでき経費の節減などが図れることを意味する。

しかしながら、一般家庭の部屋とは異なり、製造施設の部屋は面積が広く高さもあって容積が大きく、また正負イオンの寿命も数秒と短いため、正負イオンによるカビや菌の不活化の効果を挙げるためには、種々の工夫が必要である。例えば、所望のイオン濃度を確保するために、イオン発生装置の設置台数や設置位置などのハード面と、イオン量、温度、湿度などの製造設備の部屋の環境条件の制御、イオン発生装置の運転制御などのソフト面から検討すべき点が多い。

ところで、このような製造施設の部屋は、バイオクリーンルームに設定される場合もある。バイオクリーンルームは、空気の清浄化に加えて、細菌や微生物の制御もその対象になっており、温度や湿度のコントロールも半導体製造室などの産業用クリーンルーム以上に厳しいものが要求されるものもある。このため、バイオクリーンルームは、医薬品製造、食品加工、化学物質製造などの製造業の他、病院の無菌手術室や集中治療室、実験動物を飼育する実験動物施設にも使われている。この他最近注目され始めているのは、バイオハザード(生物災害)分野であり、通常の実験では、クラス100程度であるが、組み換えDNA実験や生菌実験などでは、クラス10程度の清浄度が要求される(2001年2月25日発行「クリーンルームと機器・材料」p.19、発行者「島 健太郎」)。

清浄度クラスとは、1ft3の空気中に含まれる粒径0.5μm以上の粒子数を表示するものであり、この数値が小さいほど、清浄度が上がる。通常のバイオクリーンルームは、クラス100〜クラス100,000の間で制御され、抗生物質や抗ガン物質などの製造プロセスでは、ずっと厳しいクラス10〜クラス100が要求されている。食品加工分野でも、発酵工程や食肉加工工程でクラス1,000程度のクリーン化が進んでいる(2001年2月25日発行「クリーンルームと機器・材料」p.19、発行者「島 健太郎」)。

さらにバイオクリーンルームでは、粒子数に加えて、カビや菌などの浮遊微生物の数についても基準が定められており、一般的には、表2に示す上記清浄度クラスに対応させた米国航空宇宙局(NASA)規格がある(2001年2月25日発行「クリーンルームと機器・材料」p.228、発行者「島 健太郎」)。

表2に示すように、清浄度クラスが上がれば、浮遊菌の数の上限値も小さくなる。そのため、バイオクリーンルームでは、システム内の要所要所に殺菌器を設置して殺菌している他、定期的にタンクや配管などを殺菌・消毒している。殺菌技術には、紫外線利用の殺菌方法、高温加熱殺菌法やオゾン利用の殺菌方法、直接・間接に加熱する殺菌方法などが開発されている。

しかしながら、これらの殺菌方法では、紫外線ランプの近傍を通過する空気やオゾン発生器や熱源の近傍の空気に対してしか殺菌効果が得られないため、これら殺菌器の設置台数を多数確保しなければならないという欠点がある。特に、オゾンは人間に有害であるため、定期消毒の際はオゾン発生器を停止させるなどして、安全性にも配慮する必要があった。

これに対し、正イオンと負イオンを用いる方法は、イオン発生部に空気流を流すことにより、正イオンと負イオンをバイオクリーンルームの全体に送り出すことが可能であるため、イオン発生素子の設置台数をそれほど多くする必要がない。また、これらのイオンは人間にも無害であるため、人間が行なう清掃作業中でも、連続してイオン発生素子を駆動することができる。また、バイオクリーンルームの定期消毒の際でもイオン発生素子を駆動させておくことができ、消毒効果をいっそう高めることができる。

【発明の実施の形態】
本発明の第1の実施形態について説明する。図1は、密閉された医薬品の製造施設の部屋20の模式的な透視斜視図である。製造施設の部屋20内には、医薬品の製造ライン30が形成されている。製造ライン30には、医薬品の製造工程で使用する製造装置31,32が配置されている。医薬品の製造工程は、通常、製造装置によって機械的に自動で行なう工程と、作業員が手作業で行なう工程とがある。ここでは、一例として、全工程がA工程、B工程、C工程の3工程からなり、A工程とC工程は製造装置31,32をそれぞれ使用し、B工程は作業員33が行なう工程とする。

製造施設の部屋20を囲む側壁の1つ(側壁21)には、イオン発生素子1(図2参照)を備えたイオン発生装置10が設置されている。また、側壁21には、作業員33の出入りや医薬品の原料や製品の搬出入のために、扉23が設けられている。側壁21と向かい合う側壁22には、正イオンと負イオンの数を測定するイオンカウンター42と、臭気の強さを検出する臭気センサー43とが設けられている。

図2は、イオン発生装置10の模式的な透視斜視図である。イオン発生装置10は、前面が曲面に形成された筐体11に覆われている。筐体11の前面には、上方に吸込み口12、下方に吹出し口13が開口形成されている。筐体11の内部には、送風通路が形成されており、この送風通路に送風ファン14とイオン発生素子1が設けられている。送風ファン14が駆動されると、製造施設の部屋20の空気が吸込み口12を通って送風通路に吸込まれ、吹出し口13から吹出される。なお、イオン発生装置10内に従来と同様にフィルターを設けてもよい。ただし、その場合は、フィルターの下流にイオン発生素子1を配設するものとする。

図3は、イオン発生素子1の模式的な透視斜視図である。箱型の本体2の上面には矩形の開口部が形成され、この開口部に平板状の誘電体3が設けられている。この誘電体3の上面は本体2の上面と一致している。本体2の内部下方には、高圧パルス駆動回路6が収容されている。誘電体3の上面と下面には、板状の内部電極4と網状の外部電極5が対向配置されている。外部電極5は本体2の上面から外部に露出している。これらの内部電極4と外部電極5には、リード線7を介して高圧パルス駆動回路6が接続されている。高圧パルス駆動回路6を駆動すると、内部電極4と外部電極5の間に正電圧と負電圧からなる高圧パルス電圧が印加され、プラズマ放電が起こる。これによって外部電極5の周辺空気がイオン化され、ほぼ同数の正イオンと負イオンが発生する。

なお、上記電極間に交流高電圧を印加することにより、空気中の酸素ないしは水分が電離によりエネルギーを受けてイオン化し、H+(H2O)m(mは任意の自然数)とO2 −(H2O)n(nは任意の自然数)を主体としたイオンを生成し、これらをファン等により空間に放出させる。これらH+(H2O)m及びO2 −(H2O)nは、浮遊菌の表面に付着し、化学反応して活性種であるH2O2または・OHを生成する。H2O2または・OHは、極めて強力な活性を示すため、これらにより、空気中の浮遊細菌を取り囲んで不活化することができる。ここで、・OHは活性種の1種であり、ラジカルのOHを示している。

正負のイオンは浮遊細菌の細胞表面で式(4)〜式(6)に示すように化学反応して、活性種である過酸化水素(H2O2)または水酸基ラジカル(・OH)を生成する。ここで、式(4)〜式(6)において、m、m’、n、n’は任意の自然数である。これにより、活性種の分解作用によって浮遊細菌が破壊される。従って、効率的に空気中の浮遊細菌を不活化、除去することができる。

以上のメカニズムにより、上記正負イオンの放出により、浮遊菌等の不活化効果を得ることができる。

また、上記式(4)〜式(6)は、空気中の有害物質表面でも同様の作用を生じさせることができるため、活性種である過酸化水素(H2O2)または水酸基ラジカル(・OH)が、有害物質を酸化若しくは分解して、ホルムアルデヒドやアンモニアなどの化学物質を、二酸化炭素や、水、窒素などの無害な物質に変換することにより、実質的に無害化することが可能である。

したがって、送風ファン14を駆動することにより、イオン発生素子1によって発生させた正イオンと負イオンを製造施設の部屋20内に送り出することができる。そして、これらの正イオンと負イオンの作用により製造施設の部屋20の雰囲気中のカビや菌を不活化し、その増殖を抑制することができる。

図7(a)は上記の構成によって、製造施設の部屋20の雰囲気中の菌数を一定の水準以下に抑えることを示している。正イオンと負イオンは、製造設備の部屋20の壁や製造装置31,32に付着したカビや菌を不活化する作用は小さく、その付着菌の増殖を抑える効果が少ないが、こうして雰囲気中の浮遊菌数を抑えることで、図7(b)に示すように、製造設備の部屋20の壁や製造ライン30に配置される製造装置31,32へのカビや菌の付着を抑えることができ、時間経過に伴う累積付着菌数を低減することができる。これにより、ホルマリンによる製造施設の部屋20全体の薫蒸消毒が必要となる菌数G2に達する時間T1を従来のそれ(t1)より延ばすことができる。したがって、製造施設を稼動させておくことができる期間が延びるので、生産性を向上させることができ、生産コストも抑えることができる。

なお、図1のように、イオン発生装置10を製造施設の部屋20の側壁21に1台のみ設置する場合は、製造施設の部屋20全体に早く充分な量の正イオンと負イオンを行き届かせるために、多量のイオンを発生させるイオン発生素子1や風量及び/若しくは風速の大きな送風ファン14を使用し、かつ、側壁21のできるだけ高い位置又は天井にイオン発生装置10を設置することが望ましい。

上記のように1台のイオン発生装置10のみでも効果は見込めるが、製造施設の部屋20は面積が広く高さもあって容積の比較的大きな空間であることを考えると、イオン発生装置10の設置台数は2台以上にするのがよい。図4は、一例として、製造施設の部屋20の対向する側壁21,22にイオン発生装置10A,10Bをそれぞれ設置した場合を示している。このように、2台のイオン発生装置10A,10Bを均等に離して配置することで、空気の循環が良くなり、空気のよどんだ場所、例えば、製造装置31,32の間なども含めて製造施設の部屋20全体により均一に正イオンと負イオンを送り出することができるようになる。

なお、イオン発生装置10の設置箇所は、製造施設の部屋20の側壁21に限られず、天井24や床25にイオン発生装置10を設置してもよい。あるいは、図5に示すように、イオン発生装置10A,10Bを製造装置31,32の内部及び/若しくは外部にそれぞれ設けて、製造ライン30の周辺の正イオンと負イオンの濃度を高くするようにしてもよい。この場合は、製造装置31,32自体にイオンカウンター42、臭気センサー43を設けるとよい。これにより、製造ライン30の周辺空気に含まれるカビや菌を効率よく不活化することができる。さらに、図6に示すように、製造施設の部屋20の側壁21にもイオン発生装置10Cを設置すれば、製造施設の部屋20全体に均一にイオンを供給することができるため、製造装置31,32に付着するカビや菌を一段と減らすことができる。

次に、製造施設の部屋20の環境条件の制御と、イオン発生装置10の運転制御について、図1の場合を例にして説明する。図8は、図1の製造施設の部屋20におけるイオン発生装置10の制御ブロック図である。イオン発生装置10には、筐体11の内部に、駆動回路などの電装部品が設けられている。駆動回路は、イオン発生素子1や送風ファン14の駆動状態を制御するマイクロコンピューターより成る制御部15を備えている。製造施設の部屋20の側壁22に設けたイオンカウンター42,臭気センサー43は制御部15に接続され、検出結果を制御部15に出力するようになっている。

制御部15は、イオンカウンター42と臭気センサー43の出力に基づいてイオン発生素子1の高圧パルス駆動回路6を制御するとともに、送風ファン14のモーター16を制御する。さらに制御部15は、これらの制御のタイミングをタイマー17によって決めている。

図9は、製造施設の部屋20の環境条件の制御とイオン発生装置10の運転制御の一例を示すフローチャートである。製造ライン30の稼動中、イオン発生装置10の運転を開始すると、イオン発生素子1から発生した正イオンと負イオンが送風ファン14によって製造施設の部屋20に放出される。

製造施設の部屋20の雰囲気中(環境)のイオン数や臭気の強さは、イオンカウンター42と臭気センサー43により随時検出され、制御部15に入力される。制御部15は、この入力に基づいて、製造装置の部屋20のイオン濃度を設定値付近で維持するようにイオン発生装置10からのイオン発生量を制御する。これにより、寿命の短い正イオンと負イオンを製造施設の部屋20全体に無駄なく均一に供給することができる。なお、製造施設の部屋20の温度や湿度は、エアーコンディショナー(図示せず)などで、一定の範囲の中に維持されている。
