糖尿病、異常脂血症(dyslipidemia)および高血圧の治療に有用な
アゾリジンジオン(azolidinedione)
およびそれらを含有する組成物
発明の分野
本発明は、新規な抗糖尿病性化合物、それらの互変異性体、誘導体、類縁体、
立体異性体、多形相、薬学的基準に適合する塩および溶媒和物、並びにそれらを
含有する薬学的基準に適合する組成物に関する。本発明は、特に、一般式(I)
で表される新規なアゾリジンジオン(azolidinedione)、それらの誘導体、類縁体
、互変異性体、立体異性体、多形相、および薬学的基準に適合する塩および溶媒
和物、並びにそれらを含有する薬学的組成物に関する。
また、本発明は、前記新規なアゾリジンジオン組成物、その類縁体、誘導体、
互変異性体、立体異性体、多形相、薬学的基準に適合する塩および溶媒和物、並
びにそれらを含有する薬学的組成物の合成方法に関する。
また、本発明は、新規な中間体、該中間体の合成方法、および該中間体の使用
方法に関する。
本発明において先に規定した一般式(I)で表されるアゾリジンジオンは、高
脂血症、高コレステロール血症、高血糖症、骨粗しょう症、肥満症、グルコース
不耐性、インスリン抵抗性、また、インスリン抵抗性が根元的な病態生理学的機
構である疾患または病気の治療および/または予防に有用である。これら疾患お
よび病気の例としては、II型糖尿病、グルコース耐性低下、異常脂血症、高血圧
、冠状動脈心臓病およびアテローム性動脈硬化を含む他の心臓血管系疾患が挙げ
られる。式(I)で表されるアゾリジンジオンは、肥満および乾癬を伴う
インスリン抵抗性の治療に有用である。式(I)で表されるアゾリジンジオンは
また、糖尿病性合併症の治療に使用することも、また、他の疾患および病気、例
えば、多嚢胞性卵巣症候群(PCOS)、腎性糖尿病、糸球体腎炎、糸球体硬化症
、ネフローゼ症候群、高血圧性腎硬化症、末期の腎臓疾患およびミクロアルブミ
ン尿症を含む腎性疾患、並びにアルドースレダクターゼ阻害剤のようにある種の
摂食異常の治療および/または予防に、また、痴呆の認識機能の改善に使用する
こともできる。
発明の背景
インスリン抵抗性は、広範な濃度に亘ってインスリンがその生物学的作用を発
揮する機能の減弱である。インスリン抵抗性では、この欠陥を補償するために異
常に大量のインスリンを分泌する。そうでない場合には、血漿グルコース濃度が
必然的に上昇し、糖尿病になる。先進諸国のなかでは、真性糖尿病が共通の問題
となっており、これは肥満症、高血圧症、高脂質血症(J.Clin.Invest.,(1985)7
5:809-817;N.Engl.J.Med.(1987)317:350-357;J.Clin.Endocrinol.Metab.,(1988)
66:580-583;J.Clin.Invest.,(1975)68:957-969)および他の腎性合併症(特許出
願WO 95/21608参照)を含む様々な異常につながる。インスリン抵抗性および
相対的高アラニン血症が肥満症、高血圧症、アテローム性動脈硬化症およびII型
真性糖尿病に影響を与えることが徐々に認識されつつある。インスリン抵抗性と
、肥満症、高血圧症およびアンギナとの結合が、中央病原性関連症候群Xのよう
なインスリン抵抗性を有する症候群として記載されている。加えて、多嚢胞性卵
巣症候群(特許出願WO 95/07697)、乾癬(特許出願WO 95/35108)、痴呆(Beha
vioral Brain Research(1996)75:1-11)等もまた、中央病原性の特徴と同様のイ
ンスリン抵抗性を有し得る。最近、チアゾリジンジオン(thiazolidinedione)
が骨の無機物質密度を向上させることも報告されており、故に骨粗しょう症の治
療に有用となり得る(EP−783888)。
多くの分子的欠陥がインスリン抵抗性に関連している。これらはインスリン応
答性細胞の原形質膜上のインスリンレセプタ発現の低下、および、グルコース輸
送とグリコーゲン合成を含む当該レセプタにインスリンが結合した後活性化され
るシグナル導入経路の変質を含む。
インスリン作用の欠陥は、インスリン非依存性真性糖尿病および他の関連する
合併症の発生におけるインスリン分泌の欠損よりも重要であると考えられるため
、これは、完全にインスリン分泌の刺激に基づく抗糖尿病性治療の本質的な適合
性についての疑問を生じさせる。最近、タケダは新種の化合物、すなわち式(II
)の5-(4-アルコキシベンジル)-2,4-チアゾリジンジオンの誘導体(文献、Chem.
Pharm.Bull.1982,30,3580-3600)を開発した。該式(II)において、Vは置換又
は無置換の2価の芳香族基、Bは硫黄または酸素原子、およびUは様々な特許文
献に報告されている種々の基を表す。
具体例としては、Uは以下の基であり得る。
(i)式(IIa)の基。
ここで、R1は水素、もしくは炭化水素残渣または複素環残渣(各々置換
され得る);R2は水素または水酸基で置換され得る低級アルキル;Xは酸
素または硫黄原子;Zはヒドロキシル化されたメチレンまたはカルボニル
;mは0または1の整数;nは1〜3の整数である。
これらの化合物は欧州特許出願第0177353号に開示されている。
これら化合物の例を式(IIb)に示す。(ii)式(IIc)の基。
ここで、R1およびR2は同一または異なり、各々水素またはC1〜C5の
アルキル基;R3は水素、アシル基、C1〜C6アルコキシカルボニル基ま
たはアラルキルオキシカルボニル基:R4、R5は同一または異なり、各々
水素、C1〜C5アルキルまたはC1〜C5アルコキシ、もしくはR4、R5は
共にC1〜C4アルケンジオキシ基;nは1、2または3;WはCH2CO
、CHOR6基(R6はR3について規定したものまたは基のいずれかであ
り、R3と同一でも異なってもよい)を表す。
これらの化合物は、欧州特許出願第0139421号に開示されている。
これらの化合物の例を(IId)に示す。
iii)式(IIe)の基。
ここで、A1は置換もしくは無置換の芳香族複素環基;R1は水素原子、
アルキル基、アシル基、アリール部分が置換もしくは無置換のアラルキル
基、または置換もしくは無置換のアリール基;nは2〜6の範囲の整数を
表す。
これらの化合物は欧州特許第0306228号に開示されている。
この化合物の例を式(IIf)に示す。
iv)式(IIg)の基。
ここで、YはNまたはCR5;R1、R2、R3、R4およびR5は水素、ハ
ロゲン、アルキル等;およびR6は水素、アルキル、アリール等;nは0
〜3の整数を表す。
これらの化合物は欧州特許出願第0604983号に開示されている。
この化合物の例を(IIh)に示す。
v)式(IIi)の基。
ここで、RはC1〜C6アルキル基、シクロアルキル基、フリル基、チ
エニル基、置換もしくは無置換のフェニル基;Xは水素、メチル、メトキ
シ、クロロまたはフルオロである。
これらの化合物は米国特許第5037842号に開示されている。 これらの化合物の例を式(IIj)に示す。
(vii)式(IIk)の基。
ここで、A1は置換もしくは無置換の芳香族複素環;R1は水素原子、ア
ルキル基、アシル基、アリール部分が置換もしくは無置換のアラルキル基
、または置換もしくは無置換のアリール基:nは2〜6の範囲の整数を表
す。
これらの化合物は、特許出願WO92/02520号に開示されている。
これらの化合物の例を(IIl)に示す。
発明の概要
少ない投与量に比して薬効が優れ、且つ少ない毒性で効き目が優れるII型糖尿
病[インスリン非依存性糖尿病(NIDDM)]の治療用新規化合物を開発する
目的の下、本発明者等が安全性を有し、且つより優れた効き目を有する新規化合
物の開発について鋭意研究した結果、上記一般式(I)を有する新規なアゾリジ
ンジオン化合物を開発した。
従って、本発明の主たる目的は、新規なアゾリジンジオン化合物、その類縁体
、誘導体、互変異性体、立体異性体、多形相、薬学的基準に適合する塩および溶
媒和物、並びにこれらを含有する薬学的組成物、またはこれらの混合物を提供す
ることである。
本発明の他の目的は、活性が増大され、毒性のない、あるいは毒性の少ない作
用を有する新規なアゾリジンジオン化合物、その類縁体、誘導体、互変異性体、
立体異性体、多形相、薬学的基準に適合する塩および溶媒和物、並びにこれらを
含有する薬学的組成物、またはそれらの混合物を提供することである。
さらに、本発明の他の目的は、上記式(I)の新規なアゾリジンジオン、その
互変異性体、類縁体、誘導体、立体異性体、多形相、薬学的基準に適合する塩お
よび溶媒和物の合成方法を提供することである。
また、本発明の他の目的は、一般式(I)の化合物、その類縁体、誘導体、互
変異性体、立体異性体、多形相、薬学的基準に適合する塩および溶媒和物、また
はこれらの混合物を、適切なキャリヤー、溶媒、賦形剤、希釈剤およびこの種の
組成物の合成に通常用いられる他の媒質と組み合わせて含有する薬学的組成物を
提供することである。
さらに、本発明の他の目的は、式(III)の新規な中間体を提供することであ
る。
ここで、Gは−CHO、−NO2、−NH2または−CH2CH(J)−
COORである。ここで、Jは、塩素、臭素、ヨウ素のようなハロゲン原
子;RはHまたはC1〜C6アルキル、好ましくはC1〜C3アルキル、より
好ましくはメチル、エチルまたはプロピルのような低級アルキル基を表し
:n、mおよびArは式(I)およびその合成方法において規定した通り
である。
発明の詳細な説明
一般式(I)を有する本発明のアゾリジンジオン.
上記式中、R1、R2、R3、R4、R5およびR6は同一または異なり、水素、ハ
ロゲン、ヒドロキシ、シアノ、ニトロまたは必要に応じて置換される、アルキル
、シクロアルキル、アルコキシ、シクロアルコキシ、アリール、アリールオキシ
、アラルキル、アラルコキシ、ヘテロシクリル、ヘテロアリール、ヘテロアラル
キル、ヘテロアリールオキシ、ヘテロアラルコキシ、アシル、アシルオキシ、ヒ
ドロキシアルキル、アミノ、アシルアミノ、アリールアミノ、アラルキルアミノ
、アミノアルキル、アルコキシカルボニル、アリールオキシカルボニル、アラル
キルオキシカルボニル、アルキルアミノ、アルコキシアルキル、アリールオキシ
アルキル、アルキルメルカプト、アラルコキシカルボニルアミノ、アルコキシカ
ルボニルアミノ、アリールオキシカルボニルアミノ、メルカプトアルキル基、カ
ルボン酸もしくはその誘導体、またはスルホン酸もしくはその誘導体から選択さ
れる基である;R1、R2は共に、それらが結合する炭素原子と共に5〜6員環原
子(必要に応じて置換され得る)を含む芳香環構造もまた表し得る;Xは、
酸素、硫黄またはNR9(R9は、R1〜R6について規定された基である水素、ア
ルキル、アリール、アラルキル、アシル、アルコキシカルボニル、アリールオキ
シカルボニル、アラルコキシカルボニル等である)から選択されるヘテロ原子を
表す;Arは、任意に置換される2価の単環または縮合環系の芳香族基、または
複素環基を表す;R7は、水素原子、ヒドロキシ、アルコキシ、ハロゲンまたは
メチル、エチル、プロピル等のC1〜C6アルキルのような低級アルキル、任意に
置換されるアラルキル基を表し、あるいは隣接する基であるR8と結合を形成す
る;R8は、水素原子、ヒドロキシ、アルコキシ、ハロゲンまたはメチル、エチ
ル、プロピル等のC1〜C6アルキルのような低級アルキル基、任意に置換される
アラルキル基を表し、あるいは隣接する基であるR7と結合を形成する;Bは、
酸素原子または硫黄原子を表す;Yは、酸素原子または硫黄原子を表し、nは1
〜4の範囲の整数であり、mは0または1の整数である。
R1〜R6により表される適切な基は、水素、ハロゲン原子(例えばフッ素、塩
素、臭素またはヨウ素);ヒドロキシ、シアノ、ニトロ;置換もしくは無置換の
C1〜C12アルキル基、特に直鎖または分枝鎖のC1〜C6アルキル基(例えばメ
チル、エチル、n−プロピル、イソプロピル、n−ブチル、イソブチル、t−ブ
チル、n−ベンチル、イソベンチル、ヘキシル等);シクロプロピル、シクロブ
チル、シクロペンチル、シクロヘキシル等のようなシクロアルキル基(シクロア
ルキル基は置換され得る);シクロプロピルオキシ、シクロブチルオキシ、シク
ロペンチルオキシ、シクロヘキシルオキシ等のようなシクロアルキルオキシ基(
シクロアルキルオキシ基は置換され得る);フェニルまたはナフチルのようなア
リール基(アリール基は置換され得る);ベンジルまたはフェネチル、C6H5CH2
CH2CH2、ナフチルメチル等のようなアラルキル(アラルキル基は置換され得
、置換されたアラルキルは、CH3C6H4CH2、ハロゲン−C6H4CH2、CH3
OC6H4、CH3OC6H4CH2CH2等のような基である);ピリジル、チエニル
、フリル、ピロリル、オキサゾリル、チアゾリル、イミダゾリル、オキザジアゾ
リル、テトラゾリル、ベンゾピラニル、ベンゾフラニル等のようなヘテロアリー
ル(ヘテロアリールは置換され得る);アジリジニル、ピロリジニル、モルホリニ
ル、ピペリジニル、ピペラジニル等のような複素環基(複素環基は置
換され得る);ベンジルオキシ、フェネチルオキシ、ナフチルメチルオキシ、フ

ミノ基;アミドのようなカルボン酸またはその誘導体(例えば、CONH2、CO
NHMe、CONMe2、CONHEt、CONEt2、CONHPh等、カルボ
ン酸またはその誘導体は置換され得る);OO
CMe、OOCEt、OOCPh等のようなアシルオキシ基(アシルオキシ基は
任意に置換され得る);SO2NH2、SO2NHMe、SO2NMe2、SO2NH
CH3等のようなスルホン酸またはその誘導体(スルホン酸誘導体は置換され得る
)。
R1〜R6により表されるすべての基は置換され得、該置換基は、R1〜R6によ
り表される同一の基から選択され得、同様に規定される。
R1、R2が結合する炭素原子と共にR1、R2により形成される適切な環状構造
は、5〜6員環、好ましくは、任意に置換されたフェニル、ピリジル、フラニル
、チエニル、ピロリル等を含む;置換基はR1〜R6により表される同一の基から
選択され得、同様に規定される。好適な置換基は、ハロゲン、(C1〜C6)アルコ
キシ、シクロ(C3〜C6)アルキル、シクロ(C3〜C6)アルコキシ、アリール
、アラルキル、アラルコキシ、複素環、ヒドロキシ、アシル、アシルオキシ、カ
ルボキシル、アルコキシカルボニル、アラルコキシカルボニル、アミノ、アルキ
ルアミノ、アシルアミノ、アラルコキシカルボニルアミノ、アミノカルボニル等
である。
好適なXには、酸素、硫黄または上記NR9が含まれる。Xは、好ましくは酸
素または硫黄である。
Arにより表される基には、置換もしくは無置換の2価のフェニレン、ナフチ
レン、ピリジル、キノリニル、ベンゾフラニル、ベンズオキサゾリル、ベンゾチ
アゾリル、インドリル、インドリニル、アザインドリル、アザインドリニル、イ
ンデニル、ジヒドロベンゾフリル、ベンゾピラニル、ヒドロベンゾピラニル、ピ
ラゾリル等が含まれる。Arにより表される基の置換基には、直鎖または分枝鎖
の、任意にハロゲン化された(C1〜C6)アルキル、任意にハロゲン化された(
C1〜C3)アルコキシ、ハロゲン、アシル、アミノ、アシルアミノ、チオ、カル
ボン酸およびスルホン酸、およびそれらの誘導体が含まれる。置換基は、R1〜
R6についての置換基と同様に規定される。
より好ましくは、Arは置換もしくは無置換の2価のフェニレン、ナフチレン
、ベンゾフラニル、インドリル、インドリニル、キノリニル、アザインドリル、
アザインドリニル、ベンゾチアゾリルまたはベンズオキサゾリル基を表す。
さらに好ましくは、Arは、2価のフェニレンまたはベンゾフラニルを表し、
これらは任意にメチル、ハロメチル、メトキシまたはハロメトキシ基により任意
に置換され得る。
適切なR7には、水素、メチル、エチルまたはプロピルのような低級アルキル
基;ヒドロキシ;(C1〜C3)アルコキシ;フッ素、塩素、臭素またはヨウ素の
ようなハロゲン原子;C6H5CH2、C6H5CH2CH2、C6H5CH2CH2CH2
、ナフチルメチル等のようなアラルキル、CH3C6H4CH2、ハロゲン−C6H4
CH2、CH3OC6H4CH2、CH3OC6H4CH2CH2等のような置換されたア
ラルキルが含まれ、あるいはR7はR8と共に結合を表す。
適切なR8は、水素原子、ヒドロキシ、(C1〜C3)アルコキシ;フッ素、臭
素、ヨウ素および塩素から選択されるハロゲン、(C1〜C12)アルキルのよう
な低級アルキル基、C6H5CH2、C6H5CH2CH2、C6H5CH2CH2CH2、
ナフチルメチル等のようなアラルキル、CH3C6H4CH2、ハロゲン−C6H4C
H2、CH3OC6H4CH2、CH3OC6H4CH2CH2等のような置換されたアラ
ルキルであり得、あるいはR8はR7とと共に結合を表す。
好ましくは、R7とR8は水素原子を表し、あるいはR7とR8とは共に結合を表
す。
適切なB基には、OまたはSから選択されるヘテロ原子、好ましくは硫黄原子
が含まれる。
BおよびYを含有する適切な環状構造には、2,4−ジオキソオキサゾリジン
−5−イル、2,4−ジオキソチアゾリジン−5−イル、4−オキサゾリジンジ
オン−2−チオン−イル基が含まれる。Bを含有する好ましい環状構造には、2
,4−ジオギソオキサゾリジン−5−イルおよび2,4−ジオキソチアゾリジン
−5−イル基が含まれる。
より好ましくは、Bを含有する環状構造には、2,4−ジオキソチアゾリジン
−5−イル基が含まれる。
適切なY基はOまたはSから選択されるヘテロ原子である。
適明なmは0〜1の範囲に及ぶ整数である。好ましくは、m=0のとき、リン
カー基(CH2)n−は、任意に置換される2価ベンゾフラニル、ベノキサゾリ
ル、ベンゾチアゾリル、インドリル、インドリニル、ジヒドロベンゾフリル、ま
たはジヒドロベンゾピラニルを表すAr基のヘテロ原子に近接する炭素原子に結
合され、m=1のとき、Arは置換もしくは無置換の2価フェニレン、ナフチレ
ン、ピリジル、キノリニル、ベンゾフラニル、ベンズオキサゾリル、ベンゾチア
ゾリル、インドリル、インドリニル、アザインドリル、アザインドリニル、イン
デニル、ジヒドロベンゾフリル、ベンゾピラニル、ジヒドロベンゾイピラニル、
ピラゾリルを表す。
適切なnは、1〜4の範囲に及ぶ整数であり、好ましくは、nは整数1または
2を表す。
好ましくは、m=1のときnは2を表す。
また好ましくは、m=0のときnは1を表す。
また好ましくは、nが2、mが1を表すとき、Arはフェニルまたはナフチル
基を表す。
本発明の一部を形成する薬学的基準に適合する塩には、アゾリジンジオン部分
の塩、例えばアルカリ金属塩(例えばLi、Na、K塩)、アルカリ土類金属塩
(例えばCaおよびMg塩)、有機塩基の塩(例えばシリン、アルギニン、グアニ
ジン、ジエタノールアミン、コリン等)、アンモニウムまたは置換されたアンモ
ニウム塩が含まれ、適切な場合にはいつでもアルミニウム、アルカリ金属塩、ア
ルカリ土類金属塩、アンモニウムまたは置換されたアンモニウム塩のようなアゾ
リジンジオン部分の塩が含まれる。塩には、酸付加塩が含まれ得、酸付加塩には
硫酸塩、硝酸塩、リン酸塩、過塩素酸塩、ホウ酸塩、水素ハロゲン化物、酢酸塩
、酒石酸塩、マレイン酸塩、クエン酸塩、琥珀酸塩、パルモエ酸塩(palmoate)、
メタンスルホン酸塩、安息香酸塩、サリチル酸塩、ヒドロキシナトエ酸塩(hydro
xynaphthoate)、ベンセンスルホン酸塩、アスコルビン酸塩、グリセリンリン酸
塩、ケトグルタル酸塩等がある。薬学的基準に適合する溶媒は水和物であり得、
あるいはアルコールのような結晶化溶媒を含み得る。
本発明において特に有用な化合物には以下のものが含まれる。

するものである。
ここで、R1〜R6、Ar、X、nおよびmは先に規定したとおりであり
、GはCHOまたはNO2基、あるいは−CH2−CH(J)−COORを
表す。ここで、Jはハロゲン原子(例えば塩素、臭素またはヨウ素);Rを
Hまたは先に規定した通りの低級アルキル基を表す。
本発明の態様において、先に規定した一般式(III)の新規な中間体(式(III
)中、GはCHOまたはNO2基であり、m=1である)は、一般式(IV)の化
合物と、
ここで、R1〜R6、X、nは先に規定した通りであり;L1はハロゲン
原子(例えば、塩素、臭素またはヨウ素)、またはメタンスルホン酸塩、ト
リフルオロメタンスルホン酸塩、p−トルエンスルホン酸塩等のような脱

式(III)の化合物を合成するための式(IV)の化合物と式(V)の化合物と
の反応は、例えばTHF、DMF、DMSO、DME等の溶媒の存在下で行われ
得る。溶媒の混合物も使用され得る。不活性雰囲気は不活性ガス(例えばN2、
ArまたはHe)を使用することにより維持され得る。該反応は、塩基(例えば
K2CO3、Na2CO3、NaHまたはこれらの混合物)の存在下において進行し
得る。反応温度は20℃〜150℃、好ましくは30℃〜100℃の範囲に及び
得る。反応時間は1〜24時間、好ましくは2〜6時間である。
本発明の他の態様では、一般式(III)の新規な中間体(式中、GはCHOま

式(III)の化合物を製造するための、式(VI)の化合物と式(VII)の化合物
との反応は、THF、DMF、DMSO、DME等のような溶媒もしくはこれら

rまたはHe)を使用することにより維持され得る不活性雰囲気下において進行
し得る。該反応は、塩基(例えばK2CO3、Na2CO3、NaHまたはこれらの

先に規定した式(III)の新規な中間体はまた、先に規定した一般式(VI)の

ができる。
一般式(VI)の化合物と一般式(V)の化合物との反応は、ジシクロヘキシル
尿素のような適切なカップリング剤、PPh3/DEADのようなトリアリール
ホスフィン/ジアルキルアザジカルボキシレート等を用いて行われ得る。該反応
は、THF、DME、CH2Cl2、CHCl3、トルエン、アセトニトリル、四
塩化炭素等のような溶媒の存在化において行われ得る。不活性雰囲気は、不活性
ガス(例えばN2、ArまたはHe)を用いることにより維持され得る。該反応
は、DMAP、HOBTの存在下において進行し得、これらは0.05〜2当量
、好ましくは0.25〜1当量の範囲で使用され得る。反応温度は0℃〜100
℃、好ましくは20℃〜80℃の範囲であり得る。反応時間は0.5〜24時間
、好ましくは6〜12時間の範囲に及び得る。

一般式(VIII)の化合物と一般式(IX)の化合物との反応は、無溶媒で、ある
いはDMF、DMSO、CH3CN、EtOH、アセトンまたはこれらの混合物
のような溶媒の存在下において行われ得る。不活性雰囲気は、不活性ガス(例え
ばN2、ArまたはHe)を用いることにより維持され得る。該反応は、K2C

本発明は、一般式(I)(式中、R7およびR8は共に結合を表し;Bは硫黄ま
たは酸素原子を表し;すべての記号は先に規定した通りである)で表される新規
なアゾリジンジオン、その互変異性体、誘導体、類縁体、立体異性体、多形相、
薬学的基準に適合し得る塩および溶媒和物の合成方法を提供する。
該方法は、
一般式(X)の化合物を得るために、一般式(III)の化合物(式中、GはCH
O基である)を、2,4−チアゾリジンジオン、2,4−オキサゾリジンジオン
、またはオキサゾリドン−4−オキソ−2−チオンと反応させる工程
を有する。
ここで、R1〜R6、X、Ar、n、m、BおよびYは先に規定した通り
であり;R7およびR8は共に結合を表し、従来法による反応の間に生成さ
れる水を除去する。
一般式(X)の化合物を得るための、一般式(III)の化合物(式中、GはC
HO基である)と、2,4−チアゾリジンジオン、2,4−オキサゾリジンジオ
ン、またはオキサゾリドン−4−オキソ−2−チオンとの反応は、酢酸ナトリウ
ムの存在下において無溶媒の状態で、またはベンゼン、トルエン、メトキシエタ
ノール、またはこれらの混合物のような溶媒の存在下において行われ得る。反応
温度は、使用される溶媒に依存して80℃〜140℃の範囲に及び、酢酸ナトリ
ウムの存在下で無溶媒の状態で反応が行われるときは80℃〜180℃の範囲に
及び得る。ピペリジニウムアセテートまたはベンゾエート、酢酸ナトリウムのよ
うな適切な触媒、もしくはこれら触媒の混合物もまた使用され得る。酢酸ナトリ
ウムを溶媒の存在下において使用することは可能であるが、無溶媒の状態で使用
することが好ましい。反応中に生成する水は、例えばディーンスターク(Dean St
ark)ウォーターセパレータまたはモレキュラーシーブのような吸着剤の使用に
より除去し得る。式(X)の化合物(式中、Bは酸素原子を;Yは硫黄原子を表
す)を合成するためにオキサゾリジン−4−オキソ−2−チオンを使用する場合
、過酸化水素または過酸(例えばmCPBA)のような試薬を用いての酸化によ
りチオ基をオキソ基に変換し得る。
上記方法により得られる一般式(X)の化合物は、式(XI)の化合物を得る
ための公知の方法により還元され得る。
ここで、R1〜R6、X、Ar、n、m、BおよびYは先に規定した通り
である。一般式(XI)の化合物は、一般式(I)の化合物を表す。ここ
で、R7およびR8は水素原子を表し、他のすべての記号は先に規定した通
りである。
一般式(XI)の化合物を得るための式(X)の化合物の還元は、ガス状水素
およびPd/C、Rh/C、Pt/C、ラネーニッケル(Raney Nickel)等のよ
うな触媒存在下において行われ得る。触媒の混合物も使用され得る。該反応
は、ジオギサン、酢酸、酢酸エチル等のような溶媒の存在下において行われ得る
。溶媒の混合物も使用され得る。大気圧および80psiの間の圧力が用いられ得
る。触媒は5〜10%Pd/Cであり得、使用される触媒量は50〜300%w/
wの範囲であり得る。該反応はまた、メタノール中のマグネシウムまたはメタノ
ール中のアマルガムのような金属溶媒還元を使用することにより行われ得る。該
反応はまた、CoCl2のようなコバルト塩および配位子、好ましくは2,2−
ビピリジル、1,10−フェナントロリン、ビスオキシム等のような二座配位子
の存在下において、LiBH4、NaBH4、KBH4等のようなアルカリ金属ホ
ウ化水素と共に行われ得る。上記方法で得られる一般式(X)および一般式(X
I)の化合物は、従来法により薬学的基準に適合する塩または溶媒和物に変換さ
れ得る。
さらに、本発明の他の態様では、一般式(I)の化合物(式中、mは1であり
、他のすべての記号は先に規定した通りである)は、先に規定した一般式(IV)
と、一般式(XII)の化合物との反応により合成することができる。
ここで、R7、R8、B、YおよびArは先に規定した通りであり、R1 0は水素または反応後に除去される窒素保護基である。
一般式(I)の化合物を合成するための一般式(IV)の化合物と一般式(XII
)の化合物との反応は、THF、DMF、DMSO、DME等のような溶媒、ま
たはそれらの混合物の存在下において行われ得る。該反応は、N2、Arまたは
Heのような不活性ガスを用いることにより維持され得る不活性雰囲気下におい
て行われ得る。該反応は、アルカリ(例えば、水酸化ナトリウムまたは水酸化カ
リウム)、アルカリ金属炭酸塩(例えば、炭酸ナトリウムまたは炭酸カリウム)、
アルカリ金属水素化物(例えば水素化ナトリウム)、有機金属塩基(例えば、n−
ブチルリチウム)、アルカリ金属アミド(例えばナトリウムアミド)のような塩
基、あるいはこれらの混合物の存在下において進行し得る。多数の溶媒および
塩基を使用することができる。塩基の量は、1〜5当量、好ましくは1〜3当量
の範囲に及び得る。反応温度は0℃〜120℃、好ましくは20℃〜100℃の
範囲に及び得る。反応時間は0.5〜24時間、好ましくは0.5〜6時間の範
囲に及び得る。
保護基の除去は、塩酸、トリフルオロ酢酸のような酸、またはKOH、NaO
H、Na2CO3、NaHCO3、K2CO3等のような塩基、またはこれらの混合
物との処理を含む従来の方法により行われ得る。これらの試薬は水溶液として、
またはメタノール、エタノール等のアルコール溶液として使用され得る。脱保護
(deprotection)は、保護基がベンジルまたは置換されたベンジル基の場合は、
Pd/炭素のような触媒の存在下においてガス状水素により、あるいは従来の水
素化移動法により行うこともできる。
一般式(I)の化合物(式中、mは1であり、他の記号は先に規定した通りで
ある)はまた、一般式(VI)の化合物と、一般式(XII)の化合物との反応によ
っても得ることができる。
一般式(I)の化合物を合成するための一般式(VI)の化合物と一般式(XII
)の化合物との反応は、ジシクロヘキシル尿素のような適切なカップリング剤、

本発明の他の態様では、一般式(I)の化合物(式中、R1〜R6、X、n、m
およびArは先に規定した通りであり;R7及びR8は水素を表し;Bは硫黄原子
を;Yは酸素原子を表す)は、一般式(XIII)の化合物と、
ここで、R1〜R6、X、Ar、mおよびnは先に規定した通りであり;
Jは塩素、臭素またはヨウ素のようなハロゲン原子であり;Rは低級アル
キル基である。
チオ尿素との反応、および続いてなされる酸処理により合成することができる。
一般式(XIII)の化合物とチオ尿素との反応は、通常、メタノール、エタノ
ール、プロパノール、イソブタノール、2−メトキシブタノール等のアルコール
性溶媒、またはDMSOまたはスルホランの存在下において行われる。該反応は
20℃以上、使用される溶媒の還流温度以下の温度範囲で行われ得る。NaOA
c、KOAc、NaOMe、NaOEt等のような塩基を使用することができる
。該反応に続いて、通常、塩酸のような無機酸を用いた処理が20℃〜100℃
において行われる。
一般式(XIII)の化合物(式中、Jはハロゲン原子である)は、アルカリ金
属亜硝酸塩を用いての、一般式(XIV)のアミノ化合物のジアゾ化、
ここで、すべての記号は先に規定した通りでる。
次いでハロゲン化水素酸および触媒量の酸化銅またはハロゲン化銅の存在下にお
けるアクリル酸エステルでの処理により合成することができる。
薬学的基準に適合する塩は、式(I)の化合物を、エーテル、THF、メタノ
ール、t−ブタノール、ジオキサン、イソプロパノール、エタノール等のよう
な溶媒中で、1〜4当量の塩基、例えば水酸化ナトリウム、ナトリウムメトキシ
ド、水素化ナトリウム、カリウムt−ブトキシド、水酸化カルシウム、水酸化マ
グネシウム等と反応させることにより合成される。溶媒の混合物も使用され得る
。有機塩基、例えばリシン、アルギニン、ジエタノールアミン、コリン、グアニ
ジンおよびこれらの誘導体等もまた使用され得る。あるいは、例えば以下の酸を
用いた処理により、酸付加塩を合成する。すなわち、酢酸エチル、エーテル、ア
ルコール、アセトン、THF、ジオキサン等のような溶媒中で、塩酸、臭化水素
酸、硝酸、硫酸、リン酸、p−トルエンスルホン酸、メタンスルホン酸、酢酸、
クエン酸、マレイン酸、サリチル酸、ヒドロキシナフトエ酸、アスコルビン酸、
パルミン酸、琥珀酸、安息香酸、ベンゼンスルホン酸、酒石酸等のような酸処理
により酸付加塩が合成される。溶媒の混合物もまた使用され得る。
本出願において用いる、無溶媒で、という用語は、溶媒を使用することなく反
応を行うことを意味する。
本発明の一部をなす化合物の立体異性体は、可能な場合は、その単一の鏡像異
性の形態にある反応物を使用することにより、あるいはその単一の鏡像異性の形
態の試薬または触媒の存在下で反応を行うことにより、あるいは従来法により立
体異性体の混合物を分割することにより調製され得る。好ましい方法には、微生
物による分割の使用が含まれ、マンデル酸、ショウノウスルホン酸、酒石酸、乳
酸等のようなキラル酸またはブルシン、シンコナ(cinchona)アルカロイドおよ

本発明の一部をなす一般式(I)の化合物の種々の多形相は、異なる条件下に
おける式(I)の化合物の結晶化により合成され得る。例えば、再結晶化のため
に一般的に用いられる異なる溶媒またはそれらの混合物の使用;異なる温度にお
ける結晶化;結晶化における様々な冷却モード、すなわち、高速冷却から低速冷
却までの変化である。多形相はまた、化合物を加熱または融解し、次いで序々に
またはゆっくりと冷却することにっても得られ得る。多形相の存在は、固体プロ
ーブNMR分光法、IR分光法、走査型示差熱量測定、粉体X線回折またはこの
種の他の技術により決定され得る。
本発明はまた、先に規定した通りの1または2以上の一般式(I)の化合物、
その互変異性体、誘導体、類縁体、立体異性体、多形相、薬学的基準に適合する
塩、一般的に薬学的に使用されるキャリヤー、希釈剤等と複合した薬学的基準に
適合する溶媒和物を含む薬学的組成物を提供する。これらは、高脂血症、高コレ
ステロール血症、高血糖症、骨粗しょう症、肥満症、グルコース不耐性、インス
リン抵抗性;また、インスリン抵抗性が根元的な病態生理学機構である疾患、例
えばII型糖尿病、グルコース耐性低下、異常脂血症、高血圧、冠状動脈心臓病お
よびアテローム性動脈硬化を含む他の心臓血管系疾患;肥満および乾癬を伴うイ
ンスリン抵抗性の治療および/または予防に、アルドースレダクターゼ阻害剤と
して糖尿病性合併症および他の疾患、例えば、多嚢胞性卵巣症候群(PCOS)、
腎性糖尿病、糸球体腎炎、糸球体硬化症、ネフローゼ症候群、高血圧性腎硬化症
、末期の腎臓疾患およびミクロアルブミン尿症を含むある種の腎性疾患、並びに
ある種の摂食異常の治療に、また、痴呆の認識機能の改善に有用である。
薬学的組成物は通常使用される形態、例えば、錠剤、カプセル、粉末、シロッ
プ剤、液剤、懸濁液等であり得、適切な固体または液体キャリヤーまたは希釈剤
中に、あるいは注射可能な液剤または懸濁液を形成するための適切な無菌媒質中
に香料、甘味料を含有し得る。この種の組成物は、典型的に、1〜25重量%、
好ましくは1〜15重量%の活性化合物を含み、残りの組成物は薬学的に適合し
得るギャリヤー、希釈剤、賦形剤または溶媒である。

成分1〜4は5の水溶液と共に均一に湿らせられ、減圧下において乾燥後、粒
状化される。成分6が添加され、顆粒は錠剤製造機により加圧され、成分1を3
0mg含有する錠剤1000個が調合される。
先に規定した式(I)の化合物は、哺乳動物(人間を含む)に経口的または非
口径的ルートのいずれでも臨床的に投与される。経口的ルートによる投与は、よ
り便利であり、注射の痛みの可能性や刺激を避けることができるので好ましい。
しかし、疾患または他の障害により、患者が薬物を飲み込むことができなかった
り、経口投与後の吸収機能に障害がある。
状態においては、薬物を非経口的に投与することが必須となる。いずれのルート
にせよ、一度で投与される、あるいは分けて投与される一日当たりの投与量は、
患者の体重1kg当たり約0.10〜約200mg/kg、好ましくは約0.1
0〜約50mg/kgである。しかし、治療を受ける患者個人に最適の投与量は
、治療に対する個人的反応により決定され得、一般的に、始めは少量が投与され
、その後増分されて最適の投与量が決定される。
適明な薬学的基準に適合するキャリヤーには、固体充填剤または希釈剤および
無菌水溶液または有機溶液が含まれる。所望される上記投与量を提供するのに充
分な量の活性成分がこの種の薬学的組成物中に存在し得る。従って、経口的投与
のために、該成分は適切な固体または液体キャリヤーもしくは希釈剤と複合され
、カプセル、錠剤、粉末、シロップ剤、液剤、懸濁液等を形成する。所望される
場
合には、薬学的組成物は香料、甘味料、賦形剤等のような添加成分を含有し得る
。非経口的投与のために、該成分は無菌水性または有機媒質と複合され得、注射
可能な溶液または懸濁液を形成する。例えば、ゴマ油または落花生油溶液、水性
プロピレングリコール等、並びに薬学的基準に適合する、該成分の水溶性酸付加
塩もしくはアルカリまたはアルカリ土類金属塩を使用することができる。この方
法により調製された注射溶液は、次いで、ヒトに好ましい筋内投薬と共に静脈内
、腹腔内、皮下、筋肉内に投与され得る。
本発明を以下に挙げる例において詳細に説明する。これらは例示として挙げる
だけであり、故に本発明の範囲は限定して解釈されるべきではない。
調製例1
4-[2-(2,3-ジヒドロ-1,4-ベンズオキサジン-4-イル)エトキシ]ベンズアルデヒド
:
ステップA:2-(2,3-ジヒドロ-1,4-ベンズオキサジン4-イル)エチルメタンスル
ホン酸塩
2-(2,3-ジヒドロ-1,4-ベンズオキサジン4-イル)エタノール(17.0g、9
4.9mmol)のピリジン(200ml)溶液に、メタンスルホニルクロリド
(16.31g、142.0mmol)を0℃で滴下して添加した。反応混合物
を25Cで10時間撹拌した。氷水200mlを加え、混合物を100mlの酢
酸エチルで2回抽出した。複合有機抽出物を75mlの2NHClで3回、75
mlの水、50mlの塩水で洗浄し、Na2SO4で乾燥した。溶媒を減圧下でエ
バポレートして除いた。残渣を酢酸エチルの50%pet.エーテルと共に粉砕
し、融点92〜95℃、茶色の固体の標記化合物(15.0g、61%)を得た
。ステッブB:上記において得られた2-(2,3-ジヒドロ-1,4-ベンズオキサジン4-イ
ル)エチルメタンスルホン酸塩(5g、19.4mmol)、p−ヒドロキシベン
ズアルデヒド(5.56g、29.1mmol)および炭酸カリウム(10.7
5g)を50mlの乾燥ジメチルホルムアミド中において70℃で7時間加熱し
た。反応混合物を室温まで冷却した。水100mlを混合物に加え、100ml
の酢酸エチルで2回抽出した。有機抽出物を50mlの水、50mlの塩水で洗
浄し、Na2SO4で乾燥した。溶媒を減圧下に除去し、シロップ状の標記化合物
(4.0g、72%)を得た。
調製例2
2-(2,3-ジヒドロ-1,4-ベンゾチアジン-4-イル)エチルメタンスルホン酸塩:
テトラヒドロフラン(15ml)中の4−ヒドロキシベンズアルデヒド(1.
8g、14.8mmol)およびトリフェニルホスフィン(4.85g、18.
5mmol)混合物に、2−(2,3−ジヒドロ−1,4−ベンゾチアジン−4
−イル)エタノール(2.39g、12.3mmol)およびジイソプロピルア
ゾジカルボキシレート(3.74g、18.5mmol)のTHF(20ml)
溶液を25℃において加えた。反応混合物を25℃で12時間撹拌した。水10
0mlを加え、混合物を100mlの酢酸エチルで2回抽出した。有機抽出物を
Na2SO4で乾燥し、溶媒を減圧下に置いてエバポレートして除いた。粗生成物
を、溶出液としてメタノールとクロロホルムの混合物(5:95)を使用してシ
リカゲルのクロマトグラフィーにかけ、油状の標記化合物(1.6g、44%)を
得た。
調製例3
4−[2−(フェノキサジン−10−イル)エトキシ]ベンズアルデヒド:
ジメチルホルムアミド(3ml)中の水素化ナトリウム懸濁液(60%鉱油、
0.13g、3.24mmol)に、ジメチルホルムアミド(5mL)中のフェ
ノギサジン(0.5g、2.7mmol)を加えた。反応混合物を25℃で0.
5時間撹拌し、ジメチルホルムアミド(3mL)中の4−(2−ブロモエトキシ
)ベンズアルデヒド(0.74g、3.24mmol)を加えた。反応混合物を
25℃で6時間撹拌した。水25mlを加え、混合物を50mlの酢酸エチルで
2回抽出した。複合有機層を50mlの水、50mlの塩水で洗浄し、Na2S
O4で乾燥した。溶媒をエバポレートして除き、残渣を、溶出液として酢酸エチ
ルとpet.エーテルの混合物(3:7)を用いてシリカゲルのクロマトグラフ
ィーにかけ、シロップ状の標記化合物(0.52g、57%)を得た。調製例4
4−[2−(フェノチアジン−10−イル)エトキシ]ベンズアルデヒド:
ステッアA:2−(フェノチアジン−10−イル)エチルメタンスルホン酸塩:
ジクロロメタン(150ml)中の(フェノチアジン−10−イル)エタノー
ル溶液(20.0g、82.0mmol)にトリエチルアミン(24.9g、2
4mmol)を0℃において加えた。ジクロロメタン50ml中のメタンスルホ
ニルクロリド(18.8g、160mmol)に上記反応混合物を0℃において
滴下した。該反応混合物を25℃で3時間撹拌した。氷水250mlを加え、有
機層を50mlの2N HClで2回洗浄し、75mlの水で洗浄し、Na2SO4
で乾燥した。溶媒を減圧下にエバポレートして除いた。残渣をpet.
エーテルと共に粉砕し、融点96〜97℃、茶色い固体の標記化合物(21g、
80%を得た。
ステップB:調製例1において記述した手順と類似の手順により、上記で得た2
−(フェノチアジン−10−イル)エチルメタンスルホン酸塩(22.0g、6
8.8mmol)、p−ヒドロキシベンズアルデヒド(10.0g、82.61
mmol)および炭酸カリウムから、融点128〜130℃、白色固体の標記化
合物(14.0g、59%)を調製した。
調製例5
5−ホルミル−2−(2,3−ジヒドロ−1,4−ベンズオキサジン−4−イル
メチル)ベンゾフラン:
2,3−ジヒドロ−1,4−ベンズオキサジン(1.07g、7.94mmol)
、2−ブロモメチル−5−ホルミルベンゾフラン(1.9g、7.94mmol)
、炭酸カリウム(5.5g、39.74mmol)およびアリコート(Aliquat
)336を、ジメチルホルムアミド25ml中において65℃で8時間撹拌した
。
該反応混合物に水50mlを加え、50mlの酢酸エチルで2回抽出した。抽出
物を50mlの水、50mlの塩水で洗浄し、Na2SO4で乾燥した。溶媒をエ
バポレートして除いて乾燥し、残渣を、溶出液として酢酸エチル:pet.エー
テル(3:7)の混合物を使用してシリカゲルのクロマトグラフィーにかけ、油
状の標記化合物(2.1g、91%)を得た。
調製例6
5−ホルミル−2−(フェノチアジン−10−イルメチル)ベンゾフラン:
調製例5において既述した手順と同様の手順により、フェノチアジン(2.87
g、140mmol)および2−ブロモメチル-5−ホルミルベンゾフラン(2
3g、9.6mmol)から、シロップ状の標記化合物(1.14g、33%)
を調製した。
調製例7
3−メトキシ−4−[2−(フェノチアジン−10−イル)エトキシ]ベンズア
ルデヒド:
調製例4において既述した手順と同様の手順を使用して、2−(フェノチアジ
ン−10−イル)エチルメタンスルホン酸塩(3.0g、9.3mmol)およ
びバニリン(1.7g、11.2mmol)から、融点130℃、白色固体の標
記化合物(2.5g、71%)を調製した。調製例8
4−[2−(9−オキソフェノチアジン−10−イル)エトキシ]ベンズアルデ
ヒド:
調製例3において既述した手順と同様の手順を用いて、9−オキソフェノチア
ジン(2.0g、9.3mmol)および2−(4−ホルミルフェノキシ)エチ
ルメタンスルホン酸塩(2.7g、11mmol)から、濃厚液体の標記化合物
(2.2g、67%)を調製した。
調製例9
4-[2-(9,9-ジオキソフェノチアジン-10-イル)エトキシ]ベンズアルデヒド:
アセトン中の、調製例4で得られた4−[2−(フェノチアジン−10−イル
)エトキシ]ベンズアルデヒド(0.25g、0.7mmol)および4−メチ
ルモルホリンN−オキシド(0.16g、1.4mmol)の混合物に、オスミ
ウムテトラオキシド(0.02g、0.07mmol)を加え、室温で3時間撹
拌した。この時間の経過後、反応混合物を水で希釈し、50mlの酢酸エチルで
抽出した、酢酸エチル層を25mlのNaHSO3水溶液で洗浄し、乾燥し、濃
縮し、ゴム状の標記化合物(0.23g、65%)を得た。
調製例10
4−[2−[2トリフルオロメチルフェノチアジン−10−イル]エトキシ]ベ
ンズアルデヒド:
調製例3に記述した手順と同様の手順により、2−(トリフルオロメチル)フ
ェノチアジン(1.0g、65%)および4−(2−ブロモエトキシ)ベンズア
ルデヒド(1.6g、7.49mmol)から濃厚液体の標記化合物(1.0g
、65%)を調製した。
実施例1
5-[4-[2-(2,3-ジヒドロ-1,4-ベンズオキサジン4-イル)エトキシ]フェニルメチレ
ン]チアゾリジン2,4-ジオン: 調製例1で得られた4−[2−(2,3−ジヒドロ−1,4−ベンズオキサジ
ン−4−イル)エトキシ]ベンズアルデヒド(1.79g、6.36mmol)
および2,4−チアゾリジンジオン(0.74g、6.36mmol)の混合物
を、ピペリジン(73mg、0.858mmol)および安息香酸(90mg、
0.74mmol)を含有するトルエン(100ml)中で、ディーンスターク
装置を用いて還流下で3時間加熱した。反応混合物を室温まで冷却し、分離した
固体を濾過し、10mlの冷却トルエンで2回洗浄した。クロロホルムメタノー
ルから固体生成物を再結晶化し、融点186〜188℃、黄色固体の標記化合
物(1.92g、79%)を得た。
実施例2
5−[4−[2−(2,3−ジヒドロ−1,4−ベンズオキサジン−4−イル)
エトキシ]フェニルメチル]チアゾリジン−2,4−ジオン:
実施例1で得られた5−[4−[2−(2,3−ジヒドロ−1,4−ベンズオ
キサジン−4−イル]エトキシ]フェニルメチレン]チアゾリジン−2,4−ジ
オン(1.2g、3mmol)およびマグネシウムターニング(1.25g、5
1mmol)の混合物を、メタノール(50ml)中で、25℃で20時間撹拌
した。撹拌終了時に水50mlを加え、10%の塩酸を用いてpHを6.5〜7
.0に調整し、該溶液を75mlの酢酸エチルで2回抽出した。複合有機抽出物
を50mlの水で洗浄し、50mlの塩水で洗浄し、Na2SO4で乾燥し、溶媒
を減圧下に除去した。残渣を、溶出液として酢酸エチルとpet.エーテル(2
:8)を使用してシリカゲルのクロマトグラフィーにかけ、シロップ状の標記化
合物(0.5g、41%)を得た。
実施例3
5−[4−[2−(2,3−ジヒドロ−1,4−ベンズオキサジン−4−イル)
エトキシ]フェニルメチル]チアゾリジン−2,4−ジオン、ナトリウム塩:
実施例2で得られた5−[4−[2−(2,3−ジヒドロ−1,4−ベンズオ
キサジン−4−イル)エトキシ]フェニルメチル]チアゾリジン−2,4−ジオ
ン(0.25g、0.651mmol)およびナトリウムメトキシ℃(0.1g
、1.95mmol)の混合物を、エタノール(10ml)中で、25℃におい
て14時間撹拌した。得られる固体を濾過し、5mlのメタノールで2回洗浄し
、10mlのエーテルで2回洗浄し、70℃、減圧下において乾燥し、融点26
5〜267℃、白色固体の標記化合物(0.23g、87%)を得た。
実施例4
5−[4−[2−(2,3−ジヒドロ−1,4−ベンゾチアジン−4−イル)エ
トキシ]フェニルメチレン]チアゾリジン−2,4−ジオン:
調製例2で得られた4−[2−(2,3−ジヒドロ−1,4−ベンゾチアジン
−4−イル)ニトキシ]ベンズアルデヒド(1.5g、5.01mmol)およ
び2,4−チアゾリジンジオン(0.65g、5.52mmol)から、実施例
1において記述した手順と同様の手順により、融点214〜217℃、黄色固体
の標記化合物(1.7g、85%)を調製した。
実施例5
5−[4−[2−(2,3−ジヒドロ−1,4−ベンゾチアジン−4−イル]エ
トキシ]フェニルメチル]チアゾリジン−2,4−ジオン:
実施例4で得られた5−[4−[2−(2,3−ジヒドロ−1,4−ベンゾチ
アジン−4−イル]エトキシ]フェニル メチレン]チアゾリジン−2,4−ジ
オンから、実施例2において記載した手順と類似の手順により、標記化合物(0
.5g、43%)を調製した。実施例6
5−[4−[2−(2,3−ジヒドロ−1,4−ベンゾチアジン−4−イル]エ

5−[4−[2−(フェノキサジン−10−イル)エトキシ]フェニルメチレン
]チアゾリジン−2,4−ジオン:
調製例3で得られた4−[2−(フェノキサジン−10−イル)エトキシ]ベ
ンズアルデヒド(1.5g、4.5mmol)および2,4−チアゾリジンジオ
ン(0.53g、4.5mmol)から、実施例1において記載した手順と類似
の手順により、融点204〜206℃、黄緑色の固体の標記化合物(1.46g
、75%)を調製した。
実施例8
5−[4−[2−(フェノキサジン−10−イル)エトキシ]フェニル メチル
]チアゾリジン−2,4−ジオン:
実施例7で得られた5−[4−[2−(フェノキサジン−10−イル)エトキ
シ]フェニルメチレン]チアゾリジン−2,4−ジオン(1.2g、2.79m
mol)から、実施例2において記載した手順と類似の手順により、融点170
〜172℃、白色固体の標記化合物(0.72g、60%)を調製した。

5−[4−[2−(フェノキサジン−10−イル)エトキシ]フェニルメチル]
チアゾリジン−2,4−ジオン、ナトリウム塩 実施例8で得られた5−[4−[2−(フェノキサジン−10−イル)エトキ
シ]フェニルメチル]チアゾリジン−2,4−ジオン(0.1g、0.2mmo
l)から、実施例3において記載した手順と類似の手順により、融点260〜2

5−[2−[(フェノチアジン−10−イルメチル)−ベンゾフラン−5−イル]
メチレン]チアゾリジン−2,4−ジオン:
調製例6で得られた5−ホルミル−2−[フェノチアジン−10−イルメチル
]ベンゾフラン(1.5g、4.0mmol)から、実施例1に記載した方法と
同様の方法により、融点199℃、黄緑色の固体の標記化合物(1.7g、89

実施例15
5−[4−[2−(フェノチアジン−10−イル)エトキシ]フェニル メチレ
ン]チアゾリジン−2,4−ジオン:
調製例4で得られた4−[2−(フェノチアジン−10−イル)エトキシ]ベ
ンズアルデヒド(5g、14.4mmol)から、実施例1において記載した手
順と同様の手順により、融点168〜170℃、黄色固体の標記化合物(5.3
g、83%)を調製した。
実施例16
5−[4−[2−(フェノチアジン−10−イル)エトキシ]フェニルメチレン
]チアゾリジン−2,4−ジオン、塩酸塩:
実施例15で得られた、乾燥エーテル中の5−[4−[2−(フェノチアジン
−10−イル)エトキシ]フェニルメチレン]チアゾリジン−2,4−ジオン溶
液に、HClガスを0℃において30分間通した。得られた固体を濾過し、乾燥
して、融点116〜118℃、茶色の固体の標記化合物(0.1g、39%)を
得た。
実施例17
5−[4−[2−(フェノチアジン−10−イル)エトキシ]フェニルメチル]
チアゾリジン−2,4−ジオン: 実施例15で得られた5−[4−[2−(フェノチアジン−10−イル)エト
キシ]フェニルメチレン]チアゾリジン−2,4−ジオンから、実施例2におい
て記載した手順と類似の手順により、融点68〜70℃、茶色い固体の標記化合
物(2.0g、50%)を調製した。

5−[4−[2−(フェノチアジン−10−イル)エトキシ]フェニルメチル]
チアゾリジン−2,4−ジオン、塩酸塩:
実施例17で得られた5−[4−[2−(フェノチアジン−10−イル)エト

実施例17で得られた5−[4−[2−(フェノチアジン−10−イル)エト

実施例3において記載した手順と類似の手順により、融点206〜208℃、白
色固体の標記化合物(0.26g、71%)を調製した。

順と同様の手順により、融点210〜212℃、黄色固体の標記化合物(0.8
g、67%)を調製した。

実施例20で得られた5−[4−[2−(フェノチアジン−10−イル)エト

8℃、茶色の固体の標記化合物(0.35g、66%)を調製した。

5−[4−[2−(フェノキサジン−10−イル)エトキシ]フェニルメチレン
]−4−オキサゾリジノン−2−チオン:
酢酸ナトリウム(0.23g、2.7mmol)を含有する酢酸(5mL)中
の、調製例3で得られた4−[2−(フェノキサジン−10−イル)エトキシ]
ベンズアルデヒド(0.3g、0.9mmol)および4−オキサゾリジノン−
2−チオン(0.13g、1mmol)の溶液を5時間還流下で加熱した。反応
混合物を冷却し、氷水中に注いだ。得られた固体を濾過し、水洗し、乾燥し、融
点184〜186℃、橙色の固体の標記化合物(0.3g、61%)を得た。実施例23
5−[4−(2−(9−オキソフェノチアジン−10−イル)エトキシ]フェニ
ルメチレン]チアゾリジン−2,4−ジオン:
調製例8で得られた4−(2−(9−オキソフェノチアジン−10−イル)エ

した手順と同様の手順により、融点262〜264℃、黄緑色の固体の標記化合
物(1.0g、57%)を調製した。
実施例24
5−[4−[2−(9,9−ジオキソフェノチアジン−10−イル)エトキシ]
フェニルメチレン]チアゾリジン−2,4−ジオン: 調製例9で得られた4−[2−(9,9−ジオキソフェノチアジン−10−イ
ル)エトキシ]ベンズアルデヒド(0.2g、0.53mmol)から、実施例
1において記載した手順と同様の手順により、融点248〜250℃、暗い褐色
の固体の標記化合物(0.14g、56%)を調製した。

似の手順により、融点64℃、黄緑色の固体の標記化合物(0.2g、30%)
を調製した。
実験動物コロニーにける突然変異および食事規制に対する異なる感受性により
、肥満症およびインスリン抵抗性を伴う可能性のあるインスリン非依存性糖尿病
の動物モデルが開発された。マウスのdb/dbおよびob/ob(Diabetes,(
1982)31(1):1-6参照)並びにツッカーラットのfa/faのような遺伝子モデル
が、疾患の病態生理学の理解のために様々な研究所で開発がなされ、新規な抗糖

CH3等のようなスルホン酸またはその誘導体(スルホン酸誘導体は置換され得る

R1〜R6により表されるすべての基は置換され得、該置換基は、R1〜R6によ
り表される同一の基から選択され得、同様に規定される。
R1、R2が結合する炭素原子と共にR1、R2により形成される適切な環状構造
は、5〜6員環、好ましくは、任意に置換されたフェニル、ピリジル、フラニル
、チニニル、ピロリル等を含む;置換基はR1〜R6により表される同一の基から
選択され得、同様に規定される。好適な置換基は、ハロゲン、(C1〜C6)アルコ
キシ、シクロ(C3〜C6)アルキル、シクロ(C3〜C6)アルコキシ、アリール
、アラルキル、アラルコキシ、複素環、ヒドロキシ、アシル、アシルオキシ、カ

ラゾリル等が含まれる。Arにより表される基の置換基には、直鎖または分枝鎖
