拡張された開裂ルールを有するハンマーヘッドリボザイム
発明の背景
本発明はRNA開裂活性を有する組成物の技術分野にある。
ハンマーヘッドリボザイムはある特定のRNA基質を認識して開裂することが
できる触媒性RNA分子の例である(Hotchinsら、Nucleic A
cids Res.14:3627(1986);KeeseとSymons,V
iroids and viroid−like pathogens(J.J
.Semanchik,CRCプレス、ボカレイトン、フロリダ、1987,1
〜47頁)。ハンマーヘッドリボザイムの触媒中心は3つのステムが側面に位置
し、近接したRNAの配列領域又は多くのヌクレオチドによって互いに分離され
た領域によって形成され得る。図1はそのような触媒活性のあるハンマーヘッド
構造の概略図を示す。ステムはI,II及びIIIと表示されている。ヌクレオ
チドはハンマーヘッドリボザイムの標準的な命名法に準じて数えられる(Her
telら、Nucleic Acids Res.20:3252(1992))
。この命名法では、塩基は、開裂部位の5’側に関するその位置に関連する数に
よって表記される。さらに、ステム又はループ領域に含まれる各塩基は、ステム
又はループ内部のその塩基の位置を定義する追加的な表記(小数点と別の数によ
って表記される)を有する。N11.3という表記は、この塩基が対合領域に含まれ
ていて、コア領域から離れたステムの第3番目の塩基であるということを示す。
ハンマーヘッド由来のリボザイムを記載するこの受け入れられた約定によれば、
酵素のコア部分に含まれているヌクレオチドが、他のすべてのヌクレオチドに関
連して、いつでも同一のナンバーを有するようになる。基質結合又はコア形成に
関わるステムのサイズは任意のサイズ及び任意の配列であり得るし、例えばA9
という位置は、他のコアヌクレオチドのすべてに関して同一であろう。例えば、
N^12又はN9^と表記されたヌクレオチドは、挿入されたヌクレオチドを表し
、数字に関する脱字呼号(^)の位置はその挿入が指定されたヌクレオチドの前
か後かを示す。従って、N^12は、ヌクレオチド12位の前に挿入されたヌクレ
オチ
ドを表し、N9^はヌクレオチド9位の後に挿入されたヌクレオチドを表す。
この触媒コア構造のコンセンサス配列は、RuffnerとUhlenbec
k(Nucleic Acids Res.18:6025〜6029(199
0))に記載されている。一方Perrimanら(Gene 113:157
〜163(1992))は、この構造が変異(例えばN9^及びN^12のような天
然に存在するヌクレオチド挿入)をも含有し得ることを示した。従って、タバコ
輪紋(ring-spot)ウイルスのサテライトRNAのポジティブ(+)鎖
がこの2つのヌクレオチド挿入を1つも含有しないのに対し、ル−セールン・ト
ランジェント・ストリーク(lucerne transient strea
k)ウイルスのウイルソイド(vLTSV)は、活性を失うことなくC又はGへ
突然変異し得るN9^=Uの挿入を含有する(SheldonとSymons,N
ucleic Acids Res.17:5679〜5685(1989))。
さらに、この特殊なケースでは、N7=Aであり、R15.1=Aである。一方、カ
ーネーション楼化病に関連するウイルソイド(−CarSV)のマイナス鎖は、
N^12=A及びN9^=Cという両方のヌクレオチド挿入を含有する点できわめ
て珍しい(Hernandezら、Nucleic Acids Res.20
:6323〜6329(1992))。このウイルソイドでは、N7=Aであり、R15.1
=Aである。さらに、この特別なハンマーヘッド構造は、中心ステムがより
開いているにもかかわらず、大変有効な自己触媒開裂を示す。
ハンマーヘッドリボザイムの可能な使用は、例えば、RNA制限酵素の産生及
び、例えば動物、ヒト又は植物の細胞及び原核生物、酵母及びマラリア原虫にお
ける遺伝子発現の特異的な不活性化を包含する。格別な生物医学的関心は、多く
の形態の腫瘍を包含する多くの疾患が特定遺伝子の過剰発現に関連しているとい
う事実に基づいている。関連するmRNAを開裂してそのような遺伝子を不活性
化することは、そのような疾患を抑制し、最終的には治療するための可能な方法
を具現化する。さらに、抗ウイルス薬、抗菌薬及び抗真菌薬を開発することに対
する高いニーズがある。リボザイムがそのような抗感染症薬として潜在価値を有
するのは、微生物の生存に必須であるRNA分子を選択的に破壊し得るからであ
る。
ハンマーヘッドベースリボザイムの薬剤としての使用における最大の障害の1
つは、通常のゲルラッハ(Gerlach)設計では、及び前もって形成された
半ハンマーヘッド構造をターゲットにするときには、受容されるターゲットのア
ベイラビリティが限定されていることである(WO97/18312を参照のこ
と)。ハンマーヘッドリボザイムの基質は、基質結合のために正確なハイブリダ
イズ配列を必要とすることに加えて、トリプレットN16.2U16.1H17(Nは任意
のヌクレオチド、Uはウリジン、Hはアデノシン、シチジン又はウリジンである
)を強く選好することが示されている(Koizumiら、FEBS Lett
.228,228〜230(1988);Ruffnerら、Biochemis
try 29,10695〜10702(1990);Perrimanら、Ge
ne 113:157〜163(1992))。この基質特異性の一般則を変更す
ると、非機能的な基質が生じるという事実は、上記の必要条件から逸脱した基質
が提供されるときに形成される「非コア適合性」の構造が存在することを示唆す
る。このことに関連した証拠がUhlenbeckと共同研究者によって最近報
告された(Biochemistry 36:1108〜1114(1997))。
17位のGを置換すると、G17とC3間に機能的に致命的な塩基対が形成され、
開裂のための切れやすい(scissile)結合の正確な配向性とC3の相互
作用に必要な三次構造の形成が妨害されることを実証したのである(Murra
yら、Biochem.J.311:487〜494(1995))。同様の理由
で16.1位のUに対する強い選好性が存在し得る。16.1位のU要求性を効
率的に軽減し得るリボザイムを単離することを試みた多くの実験がなされてきた
が、16.1位にU以外の塩基を有する基質を開裂し得るハンマーヘッド型リボ
ザイムを見出そうとする試みは、不可能であることが証明された(Perrim
anら、Gene 113:157〜163(1992))。
開裂領域における要求性が減少又は変更した効率的な触媒分子が強く所望され
るのは、それらが単離されればこのタイプの分子が開裂し得る利用可能なターゲ
ット配列の数を大いに増加させるからである。例えば、N16.2U16.1H17以外の
トリプレットを含有する基質を効率的に開裂し得るリボザイム変異体を得ること
が所望されるのは、それが潜在的なターゲット開裂部位の数を増加し得るから
である。
分子の中央領域にデオキシリボヌクレオチドの塊を含有する化学修飾されたオ
リゴヌクレオチドは、遺伝子発現を特異的に不活性化する薬剤としての潜在可能
性を有する(Gilesら、Nucleic Acids Res.20:76
3〜770(1992))。これらオリゴヌクレオチドは、DNAに結合したRN
A鎖がRNアーゼHによって分解されるハイブリッドDNA−RNA二重鎖を形
成し得る。そのようなオリゴヌクレオチドは、RNAの開裂を促進すると考えら
れ、RNA開裂活性を有するか又はRNA分子を開裂するものとして(開裂する
のはRNアーゼHである)は特徴づけられない。これらオリゴヌクレオチドのi
n vivo応用に対する重要な欠点はその低い特異性であり、ハイブリッド形
成及び開裂がRNA分子上の所望されない部位でも起こり得ることである。
適切な遺伝子で細胞をトランスフェクトすることによって触媒活性のあるRN
A分子を細胞内で組換え技術により発現させようとしたかつての試みは、特定の
RNA基質を不活性化するのに非常に高い発現量が必要とされるのであまり有効
でないことが証明された。さらに、今日市販されているベクター系は概して応用
し得ない。さらに、非修飾リボザイムは、RNアーゼによる分解に対するRNA
の感受性及びそのタンパク質との相互作用のために直接投与することはできない
。従って、ハンマーヘッドリボザイムを体外から投与するためには、化学的に修
飾した活性物質を使用しなければならない(例えば、Heidenreichら
、J.Biol.Chem.269:2131〜2138(1994);Kieh
ntopfら、EMBO J.13:4645〜4652(1994);Paol
ellaら、EMBO J.11:1913〜1919(1992);及びUsm
anら、Nucleic Acids Symp.Ser.31:163〜16
4(1994)によって論じられている)。
米国特許第5,334,711号は、核酸ターゲット配列を開裂するのに好適
で所望により置換された1〜10の炭素原子を有するアルキル、アルケニル又は
アルキニル基をリボースの2'−O原子に含有する修飾ヌクレオチドを含有する
、35〜40ヌクレオチド長の合成触媒オリゴヌクレオチド構造に基づいた、そ
のような化学修飾した活性物質を記載する。これらオリゴヌクレオチドは修飾ヌ
ク
レオチドの構築ブロックを含有し、ハンマーヘッド構造に似た構造を形成する。
これらオリゴヌクレオチドは特定のRNA基質を開裂することができる。
ハイブリダイゼーションアーム又は活性中心に数多くのデオキシリボヌクレオ
チドを使用すると、所望のターゲット配列に関連した配列も開裂され得るので、
RNアーゼHの活性化により特異性の欠失につながる場合がある。さらに、触媒
性DNAオリゴマーは、タンパク質と相互作用し、ヌクレアーゼ分解に対する抵
抗性が無いために、in vivo応用に特に適してはいない。
従って、RNAを開裂する組成物を提供すること、特に、高い安定性、活性及
び特異性を同時に有するRNA開裂オリゴマーを提供することが本発明の目的で
ある。
N16.2U16.1H17以外の開裂部位トリプレットを有するRNA基質を開裂する
組成物を提供することも本発明のもう1つの目的である。
発明の概要
開示されるのは、RNA開裂活性を有する組成物、並びにin vitro及
びin vivoでRNA基質を開裂するためのその使用である。この組成物は
、活性中心(このサブユニットはヌクレオチド及び/又はヌクレオチドアナログ
から選択される)及びRNA基質との特異的ハイブリダイゼーションの形成に寄
与するフランキング(flanking)領域を含有する。好ましい組成物は、RNA基
質と組合わさって、ハンマーヘッド構造に似た構造を形成する。開示組成物の活
性中心は、C16.1を有するRNA基質の開裂を可能にするI15.1の存在によって
特徴付けられる。
図表の簡単な説明
図1は、ハンマーヘッド構造及び対応する命名法(SEQ ID NO:1)
の概略図である。H17とN1.1の間で開裂が起こり、H17に2’,3’−サイク
リックリン酸を産生する。
図2は、RNA基質を開裂するオリゴマー(SEQ ID NO:2)の1例
と結合したRNA基質(SEQ ID NO:3)の概略図である。このオリゴ
マーと基質によって形成される構造はハンマーヘッドリボザイムの構造に似てい
る。このケースでは、基質がステムI及びIIIの半分を構成し、ループI及び
IIIは存在していない。開裂はH17の3’で起こる。
図3は、ハンマーヘッドリボザイム(上)におけるA15.1−U16.1塩基対の相
互作用及び、A15.1−U16.1塩基対を置換したI15.1−C16.1塩基対(下)の予

図4Aは、短い5’−フルオレセインで標識されておりGCA部位を含有する
オリゴリボヌクレオチド基質(SEQ ID NO:8)を、N7の位置に様々
なヌクレオ塩基(U、C、A及びG;それぞれSEQ ID NO:18、22
、23及び24である)をそれぞれ含有する4種の2’−O−アリル化5−リボ
触媒オリゴマーの変異体で、開裂したときの時間(分)に対する開裂生成物分画
のグラフである。
図4Bは、短い5’−フルオレセインで標識されておりGCA部位を含有する
SEQ ID NO:8のオリゴリボヌクレオチド基質を、N7の位置に様々な
ヌクレオ塩基又は塩基アナログ(U、5−ニトロインドール、I及びキナゾリン
−2,4−ジオン;それぞれSEQ ID NO:18、27、25及び26で
ある)をそれぞれ含有する4種の2’−O−アリル化5−リボ触媒オリゴマーの

発明の詳細な説明
開示されるのは、RNA開裂活性を有する組成物、並びにin vitro及
びin vivoでRNA基質を開裂するためのその使用である。この組成物は
、活性中心(このサブユニットはヌクレオチド及び/又はヌクレオチドアナログ
から選択される)及びRNA基質との特異的ハイブリダイゼーション形成に貢献
するフランキング領域を含有する。好ましい組成物は、RNA基質と組合わさっ
て、ハンマーヘッド構造に似た構造を形成する。開示組成物の活性中心は、C16 .1
を有するRNA基質の開裂を可能にするI15.1の存在によって特徴付けられる
。
天然に存在するハンマーヘッドリボザイムはすべてA15.1−U16.1塩基対を有
する。さらに、コンセンサスハンマーヘッド配列に基づいたリボザイムの基質
は、N16.2U16.1H17トリプレット(H17はグアノシンではない)を含有する基
質を強く選好することが知られている(Koizumiら、FEBS Lett
.228,228〜230(1988);Ruffnerら、Biochemis
try 29,10695〜10702(1990);Perrimanら、Ge
ne 113:157〜163(1992))。16.1位のU要求性を効率的に
軽減し得るリボザイムを単離することを試みた多くの実験がなされてきたが、1
6.1位にU以外の塩基を有する基質を開裂し得るリボザイムを見出そうとする
試みは不可能であることが証明された(Perrimanら、Gene 113
:157〜163(1992);Singhら、Antisense and N
ucleic Acid Drug Development 6:165〜1
68(1996))。
しかしながら、最近公表されたX線結晶構造(Pleyら、Nature 3
72:68〜74(1994)、scottら、Cell 81:991〜100
2(1995)及びScottら、Science 274:2065〜2069
(1996))を検討すると、このA15.1−U16.1相互作用はアデノシンの環外ア
ミン(N6)とウリジンの4−オキソ基とが単一の水素結合をした非標準的な塩
基対であることが理解された。次いで(結晶構造に基づいた)モデリング研究か
ら、この野生型A15.1−U16.1塩基対の相互作用は、イソ構造的な配向性をとり
、C16.1の2−ケト基とウリジンターンのA6との要求される接触を保存するI1 5.1
−C16.1塩基対での置換によって空間的に模倣し得ることが発見された。こ
のモデルでは、15.1位と16.1位との間の安定化される水素結合の極性が
I15.1−C16.1相互作用において逆転しているが、この結合付近にある塩基の正
確な配向性は維持される。
15.1位にイノシンを含有するゲルラッハ型リボザイムアナログがN16.2C16.1
H17トリプレットを含有するRNA基質を容易に開裂することが発見されて
いる。このことに基づいて、N16.2C16.1H17トリプレットを含有する核酸のタ
ーゲット配列を開裂する組成物、好ましくは合成オリゴマーが開示される。H17
はグアノシンでないことが好ましい。N16.2C16.1X17トリプレットを有する基
質を開裂する能力により、開示されるタイプの組成物により開裂されるタ
ーゲットの数は効果的に倍となる。
RNA開裂活性を有する組成物
特に開示されるのはRNA基質を開裂する組成物であり、ここでその組成物は
成分(a)及び(b)を包含し、成分(a)は5’−Z1−Z2−3’という構造
を包含し、成分(b)は5’−Z3−Z4−3’という構造を包含する。成分(a
)及び(b)は別個の分子であり得るか又は共有結合され得る。成分(a)及び
(b)における要素Z1及びZ4は、それぞれヌクレオチド、ヌクレオチドアナロ
グ又は両者の組み合わせからなるオリゴマー配列であるか、又はオリゴヌクレオ
チドアナログである。要素Z1及びZ4のオリゴマー配列は、RNA基質と好まし
くはハイブリダイゼーションによって特異的に相互作用する。
これらの好ましい組成物において、要素Z2は以下のいずれかの構造を有し、
5’−X3X4X5X6X7X8X9−3’又は
5’−X3X4X5X6X7X8X9X9^−3’
要素Z3は以下のいずれかの構造を有する:
5’−X12X13X14X15.1−3’又は

これらの好ましい組成物における要素Z2及びZ3は、ヌクレオチド、ヌクレオ
チドアナログ又は両者の組み合わせからなる。要素Z2及びZ3におけるヌクレオ
チド及びヌクレオチドアナログはそれぞれ以下の構造を有する。
構造式(I)において、各Bは、アデニン−9−イル、シトシン−1−イル、
グアニン−9−イル、ウラシル−1−イル、ウラシル−5−イル、ヒポキサンチ
ン−9−イル、チミン−1−イル、5−メチルシトシン−1−イル、2,6−ジ
アミノプリン−9−イル、プリン−9−イル、7−デアザアデニン−9−イル、
7−デアザグアニン−9−イル、5−プロピニルシトシン−1−イル、5−プロ
ピニルウラシル−1−イル、イソグアニン−9−イル、2−アミノプリン−9−
イル、6−メチルウラシル−1−イル、4−チオウラシル−1−イル、2−ピリ
ミドン−1−イル、キナゾリン−2,4−ジオン−1−イル、キサンチン−9−
イル、N2−ジメチルグアニン−9−イル又はそれらの機能的同等物であり;
各Vは、O、S,NH又はCH2基であり得る。
各Wは、−H、−OH、−COOH、−CONH2、−CONHR1、−CON
R1R2、−NH2、−NHR1、−NR1R2、−NHCOR1、−SH、−SR1、
−F、−ONH2、−ONHR1、−ONR1R2、−NHOH、−NHOR1、−
NR2OH、−NR2OR1、置換されているか又は置換されていないC1〜C10の
直鎖又は分岐アルキル、置換されているか又は置換されていないC2〜C10の直
鎖又は分岐アルケニル、置換されているか又は置換されていないC2〜C10の直
鎖又は分岐アルキニル、置換されているか又は置換されていないC1〜C10の直
鎖又は分岐アルコキシ、置換されているか又は置換されていないC2〜C10の直
鎖又は分岐アルケニルオキシ、及び置換されているか又は置換されていないC2
〜C10の直鎖又は分岐アルキニルオキシであり得る。W基の置換基は、独立して
ハロゲン、シアノ、アミノ、カルボキシ、エステル、エーテル、カルボキサミド
、ヒドロキシ又はメルカプトである。R1及びR2は置換されているか又は置換さ
れていないアルキル、アルケニル又はアルキニル基であり得て、ここで置換基は
独立してハロゲン、シアノ、アミノ、カルボキシ、エステル、エーテル、カルボ
キサミド、ヒドロキシ又はメルカプトである。
D及びEは、隣接したヌクレオシド又はヌクレオシドアナログとの間にホスホ
ジエステル又はホスホロチオエートジエステル結合を一緒に形成するか又はヌク
レオシド間結合のアナログを一緒に形成する残基である。
Bは、X15.1においてはヒポキサンチン−9−イル又はその機能的同等物であ
り;X5、X8及びX12においてはグアニン−9−イル、ヒポキサンチン−9−イ
ル又は7−デアザグアニン−9−イルでありえ;X6、X9、X13及びX14におい
てはアデニン−9−イル、2,6−ジアミノプリン−9−イル、プリン−9−イ
ル又は7−デアザアデニン−9−イルでありえ;X4においてはウラシル−
1−イル、ウラシル−5−イル、チミン−1−イル又は5−プロピニルウラシル
−1−イルでありえ;X3においてはシトシン−1−イル、5−メチルシトシン
−1−イル又は5−プロピニルシトシン−1−イルでありえ;X7、X9^及びX
^12においてはアデニン−9−イル、シトシン−1−イル、グアニン−9−イル
、ウラシル−1−イル、ウラシル−5−イル、ヒポキサンチン−9−イル、チミ
ン−1−イル、5−メチルシトシン−1−イル、2,6−ジアミノプリン−9−

ラシル−1−イル、4−チオウラシル−1−イル、2−ピリミドン−1−イル、
キナゾリン−2,4−ジオン−1−イル、キサンチン−9−イル、N2−ジメチ
ルグアニン−9−イル又はそれらの機能上の同等物でありえる。X15.1のBは、
好ましくはヒポキサンチン−9−イル、又はこの塩基の2位における任意の基と
C16.1の2−オキソ基との間に水素結合を形成し得ない類似体である。好ましく
は、BはX15.1においてグアニン−9−イルではない。
X3、X4、X5、X6、X7、X8、X9、X12、X13及びX14におけるBは、ハ
ンマーヘッドリボザイムの触媒中心に存在する機能的に同等なヌクレオ塩基でも
あり得る。例えば、ハンマーヘッドリボザイムのC3、U4、G5及びA6は、tR
NAのウリジンターンに酷似した構造を形成する(Pleyら、Nature
372:68〜74(1994))。他のヌクレオチド基もウリジンターンを形成
し得るので、ヌクレオチドX3、X4、X5、X6は、ウリジンターンに似た構造を
形成する潜在可能性を有するヌクレオチド又はヌクレオチドアナログにより1つ
のグループとして置換される可能性がある。同様に、ハンマーヘッドリボザイム
の触媒コアにあるせん断された塩基対は、他の非標準的な塩基対の相互作用に似
ていてもよい相互作用を有する。ハンマーヘッドリボザイムの触媒コアに関する
結晶構造の知識は、非標準的な塩基対がイソ構造的な非標準塩基対で置換された
ゲルラッハ型のハンマーヘッドリボザイムが活性であるという発見とともに、同
様のイソ構造的な塩基対置換が触媒コアのどこかで可能であることを示している
。
定義
本明細書で使用されるように、オリゴマーとは、同一のクラス(ヌクレオチド
のような)又は異なるクラス(ヌクレオチドとエチレングリコールのような)の
混合物であり得るサブユニットからなるオリゴマー分子を呼ぶ。開示されるオリ
ゴマーは、オリゴマー配列、非ヌクレオチドリンカー又はオリゴマー配列と非ヌ
クレオチドリンカーとの組み合わせであることが好ましい。開示されるオリゴマ
ーはオリゴマー配列であることがより好ましい。オリゴマー配列は、サブユニッ
トのそれぞれが塩基特異的なやり方で他のオリゴマー配列と相互作用し得るヌク
レオ塩基(即ち、ヌクレオチド又はヌクレオチドアナログの塩基部分)を包含す
るオリゴマー分子である。核酸鎖のハイブリダイゼーションはそのような塩基特
異的な相互作用の好ましい例である。オリゴマー配列は好ましくはヌクレオチド
、ヌクレオチドアナログ又はその両方から構成されるか又はオリゴヌクレオチド
アナログである。
非ヌクレオチドリンカーは、オリゴマー配列ではなくて、オリゴマー配列と共
有結合し得る任意の分子であり得る。好ましい非ヌクレオチドリンカーは非ヌク
レオチドサブユニットから形成されるオリゴマー分子である。そのような非ヌク
レオチドリンカーの例は、LetsingerとWu(J.Am.Chem.S
oc.117:7323〜7328(1995))、Benselerら(J.Am
.Chem.Soc.115:8483〜8484(1993))及びFuら(J.
Am.Chem.Soc.116:4591〜4598(1994))によって記
載されている。好ましい非ヌクレオチドリンカー又は非ヌクレオチドリンカーの
サブユニットは、置換されているか又は置換されていないC1〜C10の直鎖又は
分岐アルキル、置換されているか又は置換されていないC2〜C10の直鎖又は分
岐アルケニル、置換されているか又は置換されていないC2〜C10の直鎖又は分
岐アルキニル、置換されているか又は置換されていないC1〜C10の直鎖又は分
岐アルコキシ、置換されているか又は置換されていないC2〜C10の直鎖又は分
岐アルケニルオキシ、及び置換されているか又は置換されていないC2〜C10の
直鎖又は分岐アルキニルオキシを包含する。これらの好ましい非ヌクレオチドリ
ンカー(又はサブユニット)の置換基は、ハロゲン、シアノ、アミノ、カルボキ
シ、エステル、エーテル、カルボキサミド、ヒドロキシ又はメルカプトであり得
る。
本明細書で使用されるように、ヌクレオシドとは、アデノシン、グアノシン、
シチジン、ウリジン、2’−デオキシアデノシン、2’−デオキシグアノシン、
2’−デオキシシチジン又はチミジンを呼ぶ。ヌクレオシドアナログとはヌクレ
オシドの塩基又は糖部分のどこかの部位に化学修飾を含有するヌクレオシドの化
学修飾した形態である。本明細書で使用されるように、ヌクレオシドアナログと
いう用語は、例えばイノシンやシュードウリジンのような天然に存在する修飾さ
れたヌクレオシドに基づいたヌクレオシドアナログと、糖の2’位への修飾のよ
うな他の修飾を有するヌクレオシドアナログの両方を網羅する。本明細書で使用
されるように、ヌクレオチドとは上記のようなヌクレオシドのリン酸誘導体をよ
び、ヌクレオチドアナログは上記のようなヌクレオシドアナログのリン酸誘導体
である。ペプチド核酸のようなオリゴヌクレオチドアナログのサブユニットもヌ
クレオチドアナログであると考えられる。
本明細書で使用されるように、リボヌクレオチドは、2’ヒドロキシル基を有
するヌクレオチドである。同様に、2’−デオキシリボヌクレオチドは、2’水
素のみを有するヌクレオチドである。従って、本明細書で使用されるリボヌクレ
オチド及びデオキシリボヌクレオチドとは、化学修飾されていないヌクレオシド
成分のアデノシン、グアノシン、シチジン及びウリジン、又は2’−デオキシア
デノシン、2’−デオキシグアノシン、2’−デオキシシチジン及びチミジンを
それぞれ有する天然に存在するヌクレオチドをよぶ。リボヌクレオシド、デオキ
シリボヌクレオシド、リボヌクレオシドアナログ及びデオキシリボヌクレオシド
アナログは、ヌクレオチド及びヌクレオチドアナログに見出されるリン酸基又は
リン酸基のアナログを欠いたものとして同様に定義される。
本明細書で使用されるように、オリゴヌクレオチドアナログとは、配列依存的
なハイブリダイゼーションのような核酸様の性質を有し、標準的なRNA又はD
NAヌクレオチドから離れる修飾を1つ又はそれ以上の部位に含有する、核酸様
物質のポリマーである。オリゴヌクレオチドアナログの好ましい例はペプチド核
酸である。
本明細書で使用されるように、塩基対とは1つ又はそれ以上の水素結合を介し
て相互作用するヌクレオチド又とはヌクレオチドアナログの対を呼ぶ。塩基対と
いう用語は、ワトソン−クリック型塩基対として一般に特徴付けられる相互作用
に限定されず、非標準的な又はせん断された塩基対の相互作用を包含する(To
palとFresco、Nature 263:285(1976);Loman
tとFresco、Prog.Nucl.Acid Res.Mol.Biol
.15:185(1975))。従って、ヌクレオチドA15.1及びU16.1はハンマ
ーヘッドリボザイムにおいて塩基対を形成する(図1参照)が、この塩基対は非
標準的である(図3参照)。
2つのヌクレオシドのヌクレオシド間連結は、ホスホジエステル結合又はホス
ホロチオエート基又は、例えば米国特許第5,334,711号に記載された他
の結合のような修飾されたホスホ結合によって達成され得る。
フランキング要素Z1及びZ4
(要素Z2及びZ3によって形成される)活性中心に隣接する要素Z1及びZ4の
モノマーサブユニットは、好ましくはヌクレオチド及び/又はヌクレオチドアナ
ログである。要素Z1及びZ4は、所定のRNA基質と好ましくはハイブリダイゼ
ーションによって特異的に相互作用し、活性中心のZ2及びZ3とともにRNA基
質を特異的に開裂する構造(好ましくはハンマーヘッドリボザイムに似た構造)
を形成するように設計される。
一方、要素Z1及びZ4のサブユニットは、リボヌクレオチドであり得る。しか
しながら、リボヌクレオチドが存在するとオリゴマーのin vivo安定性が
減少するので、リボヌクレオチドの数は可能な限り少ないことが好ましい。要素
Z1及びZ4(及びまた活性中心のZ2及びZ3)は、ピリミジンヌクレオ塩基を含
有する位置に好ましくはリボヌクレオチドを1つも含有しない。そのような位置
は、好ましくはヌクレオチドアナログを含有する。
要素Z1及びZ4における多数のデオキシリボヌクレオチドの使用もより好まし
くないのは、タンパク質との望ましくない相互作用が起こり得るか、又は意図し
ないRNアーゼH感受性のDNA−RNAハイブリッドが形成し得るためである
。従って、要素Z1及びZ4は、それぞれ好ましくは(1)リボヌクレオチドを含
有せず、(2)3つより多い連続したデオキシリボヌクレオチドの配列を含有し
ない。
要素Z1及びZ4のサブユニットは、好ましくはヌクレオチド、ヌクレオチドア
ナログ又はその組み合わせである。好ましくは、要素Z1及びZ4におけるヌクレ
オチド及びヌクレオチドアナログはそれぞれ以下の構造を有する。

D及びEは、隣接したヌクレオシド又はヌクレオシドアナログとの間に、ホス

ヌクレオチド及び/又はヌクレオチドアナログを有する構造式(I)の要素Z1
及びZ4にとっては、各Wは置換されているか又は置換されていないC1〜C10
の直鎖又は分岐アルコキシ、C2〜C10の直鎖又は分岐アルケニルオキシ又はC2
〜C10の直鎖又は分岐アルキニルオキシであることが好ましい。
さらに、フランキング要素Z,及びZ1は4ペプチド核酸(ペプチド性核酸とも
言及される;例えば、Nielsenら、Science 254:1497〜
1500(1991)及びDueholmら、J.Org.Chem.59:5

含有し得る。この場合、各サブユニットのカップリングは、例えば、酸アミド結

。
フランキング要素Z1及びZ4のサブユニットは、RNA分子において天然に存
在するヌクレオ塩基とハイブリダイズするか又は相互作用し得るヌクレオ塩基又
はヌクレオ塩基アナログを含有する。ヌクレオ塩基は、天然に存在する塩基(即
ち、アデニン、グアニン、シトシン、チミン及びウラシル)並びに2,6−ジア
ミノプリン、ヒポキサンチン、5−メチルシトシン、シュードウラシル、5−プ
ロピニルウラシル及び5−プロピニルシトシンのような、ターゲットRNAとの
特異的な結合を可能にするヌクレオ塩基アナログから好ましくは選択される。
RNA基質と要素Z1及びZ4との間の強くて配列特異的な相互作用(即ち、R
NA基質とオリゴマーとの間のより安定なハイブリッド)が好ましい。この目的
のために、要素Z1及びZ4のオリゴマー配列において、標準的なヌクレオ塩基に
代わる以下のヌクレオ塩基アナログが使用されることが好ましい:アデニンの代
わりに2,6−ジアミノプリン;ウラシルの代わりにチミン又は5−プロピニル
ウラシル;及びシトシンの代わりに5−メチルシトシン又は5−プロピニルシト
シン。2−アミノ−2’−O−アルキルアデノシンもまた好ましい(Lammら

)。さらに、二本鎖の安定性を高め得るフェノキサジンのようなヌクレオ塩基ア
ナ口グを産生するために、ウラシルの4位及び5位に芳香族系を連結し得る(L

開示組成物による開裂に好ましいRNA基質は以下の構造を有する:
5’−Z4’−C16.1−X17−Z1’−3’
ここで、Z1’及びZ4’は、それぞれZ1及びZ4と相互作用し、C16.1はシチジ
ンであり、X17はアデノシン、グアノシン、シチジン又はウリジンである。開裂
はX17の3’で起こる。好ましくは、X17はアデノシン、シチジン又はウリジン
であり、より好ましくは、X17はアデノシン又はシチジンであり、最も好ましく
は、X17はアデノシンである。好ましくは、X16.2(即ち、Z4’における3’
ヌクレオシド)はアデノシン又はグアノシンである。開示組成物によって開裂さ
れ得る基質のターゲット部位は、これまでのハンマーヘッドリボザイムでは基質
の16.1位にウリジンが要求されるので、従来のハンマーヘッドリボザイムの
ターゲット部位とは異なっている。
Z4’に存在する3’位であるN16.2位は、グアノシン、アデノシン、シチジ
ン又はウリジンのいずれかであり得る。N16.2はグアノシン、アデノシン又はシ
チジンのいずれかであることが好ましい。N16.2はグアノシン又はアデノシンの
いずれかであることがより好ましい。N16.2はグアノシンであることが最も好ま
しい。開示化合物による開裂に好ましい基質は、N16.2がグアノシン、アデノシ
ン又はシチジンであり、X17がアデノシンであるものである。開示化合物による
開裂により好ましい基質は、N16.2がグアノシン又はアデノシンであり、X17が
アデノシンであるものである。開示化合物による開裂に最も好ましい基質は、N16.2
がグアノシンであり、X17がアデノシンであるものである。
フランキング要素Z1及びZ4は、それぞれ独立に、好ましくは3〜40、より
好ましくは5〜10のヌクレオチド又はヌクレオチドアナログを含有する。Z1
及びZ1’が相互作用して少なくとも3つの塩基対のステムを形成し、Z4及びZ4
’が相互作用して少なくとも3つの塩基対のステムを形成することが好ましい
。これらステムがそれぞれZ2及びZ3に隣接していることがより好ましい。Z1
及びZ1’が相互作用して3つより多い塩基対のステムを形成し、Z4及びZ4’
が相互作用して3つより多い塩基対のステムを形成することが最も好ましい。
好ましいRNA基質は、RNAのターゲット領域に関連した阻害性の二次構造
をほとんど有さないものである。RNA分子のどの領域が二次構造を含有し、そ
れ故より好ましくないか、RNA分子のどの領域が二次構造をほとんど有さず、
それ故より好ましいかを決定するための多くの方法がある。単鎖RNAの領域を
決定するための好ましい方法は、単鎖RNAの領域と選択的に反応させるか又は
それを認識することによってこの領域をマッピングするものである。ヌクレオチ
ドのワトソン−クリック面にある窒素と反応する多くの化学物質(硫酸ジメチル
(DMS)、ジエチルピロカーボネート(DEPC)、カトキサール(CMCT)、カ
ルボジイミド)がある。ワトソン−クリック型の塩基対合に関与するヌクレオチ
ドは、関与しないヌクレオチドよりもこれらの化学物質に対してより少ない反応
性を示す。化学修飾したRNAとの(逆転写酵素、RNアーゼT1、コブラ毒ヌ
クレアーゼ、ヌクレアーゼS1、ヌクレアーゼV1を使用する)酵素反応から、
その長さが化学反応の起こった塩基に依存する、短縮されたオリゴヌクレオチド
が創出される。化学反応はRNAの単鎖領域で選好的に起こるので、これらの技
術はRNAの二次構造がどこにあるかを示す。例えば、硫酸ジメチル及び逆転写
酵素マッピングのような方法は、二本鎖RNAの領域がどこにあるかを示す。逆
転写酵素は、適切な条件下では、二本鎖RNAの領域を複製し得ないので、ポリ
アクリルアミドゲル電気泳動法(PAGE)によって分析したときに、複製途中
で停止した(abortive)転写産物が存在し、これがどこに強い二次構造
のRNA領域があるかを示す。この方法の諸例は、Kamarら、Bioche

RNAのどの領域が単鎖であるかを評価する別の方法はRNアーゼHマッピン
グである。この方法では、短いランダムDNAオリゴヌクレオチドが合成され、
タ一ゲットRNAと混合される。容易にアクセスできる領域がこの短いDNA分
子とハイブリダイズされる。次いでこのDNA−RNAハイブリッド領域がRN
アーゼHによって開裂される。次ぎに標識化したRNA分子がPAGEによって
分析され得る。開裂された分子を配列決定の梯子(sequencing la
dder)と比較することにより、単鎖DNAの領域が推定され得る。この方法
の諸例は、Hoら、Nature Biotechnology 16:59〜
63(1988)及びMcSwiggenの米国特許第5,525,468号に
記載されている。
RNAのアクセス可能な単鎖領域を決定するために、in vitro選別実
験(Szostak、TIBS 19:89〜93(1992))も実施され得る
。例えばターゲットRNAは、PCR増幅のために使用され得るプライマー結合
領域を含有するランダムDNAオリゴヌクレオチドと混合され得る。増幅及び再
選別を繰り返すことで、RNAと結合し得るDNA配列を濃縮することが可能に
なる。これによってオリゴヌクレオチドハイブリダイゼーションのためにアクセ
スし得るRNAの領域が同定される。この選択又は濃縮の工程は任意のサイズ選
別技術であるか又は二本鎖核酸の分離技術であり得る。例えば、セファデックス
カラムクロマトグラフィーでは、大きい、結合性のDNA:RNA複合体と小さ
な
非結合性のDNA分子が分離され、またニトロセルロース濾過法では結合性のR
NAが保持される一方、非結合性のDNA分子は素通りする。
所定の基質に対するオリゴマーを最適化するための方法も数多くある。例えば
、特定の配列要求性を持たないオリゴマーのN7位を変更し、所定のターゲット
配列のために設計されたオリゴマーのなかに所望しない二次構造が発生する可能
性を最小化することに役立てることができる。また、7−デアザグアノシン又は
7−デアザアデノシンのような特定の塩基修飾は、所望しないプリン:プリン相
互作用を防ぐために、数多くのグアノシン又はアデノシンを有する領域に利用さ
れ得る。同様の置換は、例えば触媒オリゴマーのZ1又はZ4領域に存在し得るグ
アノシンリッチ領域へイノシンを導入することによって達成され得る。
触媒コア
要素Z2及びZ3は、開示組成物の触媒コアを形成すると考えられ、好ましくは
ヌクレオチドアナログ及び少数のリボヌクレオチドから構成される。要素Z2及
びZ3においては、(構造式(I)の)各Wは、C1〜C5の直鎖又は分岐アルキ
ル、C2〜C5の直鎖又は分岐アルケニル、C2〜C5の直鎖又は分岐アルキニル、
C1〜C5の直鎖又は分岐アルコキシ、C2〜C5の直鎖又は分岐アルケニルオキシ
、及びC2〜C5の直鎖又は分岐アルキニルオキシであることが好ましい。また、
各X3、X4、X7及びX^12においては、WはNH2、OH−置換C1〜C4のアル
キル、OH−置換C2〜C4のアルケニル、OH−置換C1〜C4のアルコキシ又は
OH−置換C2〜C4のアルケニルオキシであることが好ましい。各X3、X4、X7
及びX^12においては、WはNH2、メトキシ、2−ヒドロキシエトキシ、アリ

要素Z2及びZ3におけるサブユニットは、好ましくはリボヌクレオチドと弱く
だけハイブリダイズし得るヌクレオチドアナログである。そのようなサブユニッ
トの例は、リボースの2’位に好ましくは1〜5の炭素原子を有する置換されて
いるか又は置換されていないアルキル、アルケニル、アルキニル、アルコキシ、
アルケニルオキシ又はアルキニルオキシ基を含有するヌクレオチドアナログであ

要素Z2及びZ3は、触媒構造の形成を可能にするようなやり方で相互作用する
。好ましい組成物では、Z2及びZ3は、ハンマーヘッド触媒構造に似た触媒構造
の形成を可能にするようなやり方で相互作用する。Z2及びZ3が相互作用して触
媒構造を形成し得る1つの方法は、Z2及びZ3を構成するヌクレオチド及び/又
はヌクレオチドアナログの相互作用を介してである。開示組成物はRNアーゼH
から独立したRNA開裂活性を有する。即ち、開示組成物は、RNアーゼHに関
わらずにRNA基質の開裂を起こすことができる。開示組成物はRNアーゼHに
よるRNAの開裂を促進し得る可能性があるが、そうしないことが好ましい。
Z2とZ3との所望の相互作用は、好ましくはZ2の3’末端及び/又はZ3の5
’末端へ要素をカップリングすることによって強められる。単一の要素(本明細
書においてZ5と呼ばれる)は、要素Z2及びZ3をこのように共有結合させるた
めに使用され得る。開示組成物のそのような形態の構造は、5’−Z1−Z2−Z5
−Z3−Z4−3’となろう。別個の要素(本明細書においてZ6及びZ7と呼ば
れる)は、Z2及びZ3にカップルされ得て、好ましくは相互作用(非共有的に)
して、要素Z2とZ3の相互作用を安定化又は別のやり方で増強し得る。この形態

要素Z5、Z6及びZ7は、オリゴマー配列、非ヌクレオチドリンカー又はオリ
ゴマー配列と非ヌクレオチドリンカーの組み合わせであることが好ましい。要素
Z5、Z6及びZ7は、オリゴマー配列であることがより好ましい。これらのオリ

要素Z5は、Z2の3’末端をZ3の5’末端へカップリングする共有リンカー
として役立ち得る。要素Z5は、好ましくはポリエチレングリコールのような非
ヌクレオチド分子、又はヌクレオチド、ヌクレオチドアナログ及びオリゴヌクレ
オチドアナログを包含するオリゴマー配列、又はヌクレオチド、ヌクレオチドア
ナログ及びオリゴマーヌクレオチドアナログの組み合わせのいずれかから構成さ
れる。要素Z5の好ましい形態は、分子内で相互反応してステム−ループ構造を
形成し得る、ヌクレオチド、ヌクレオチドアナログ、オリゴヌクレオチドアナロ
グ、又はヌクレオチド及びヌクレオチドアナログの組み合わせから構成される。

開示組成物の好ましい形態は、Z2の3’末端にGがついたものである。Ta
iraと共同研究者(AmontovとTaira、J.Am.Chem.So
c.118:1624〜1628(1996))は、ハンマーヘッド様リボザイム
のR9に並置したグアノシンから得られるスタッキングエネルギーが触媒構造の
形成を安定化させることを示した。従って、Z6の5’ヌクレオチドはGである
ことが好ましい。
開示組成物の3’末端は、例えば、リボース糖の3’位が(例えば、上記に定
義したような置換されているか又は置換されていないアルキル、アルコキシ、ア
ルケニル、アルケニルオキシ、アルキニル又はアルキニルオキシ基を包含するこ
とによって)修飾されたヌクレオチドアナログを使用することによって、エクソ
ヌクレアーゼによる分解に対して保護され得る。開示組成物はまた、3’−3’
−連結ジヌクレオチド構造(Ortigaoら、Antisense Rese
arch and Development 2:129〜146(1992))
及び/又は2つのホスホロチオエート結合のような2つの修飾されたホスホ結合
を包含することによって、エクソヌクレアーゼによる3’末端での分解に対して
安定化され得る。
開示組成物はまた、その細胞への取込みを向上させる及び/又は特定の細胞内
局在化を可能にするために、補欠分子族へ連結され得る。そのような補欠分子族
の例は、ポリアミノ酸(例えば、ポリリシン)、脂質、ホルモン又はペプチドであ
る。これら補欠分子族は通常直接的に又は適切なリンカー(例えば、6−アミノ
ヘキサノール又は6−メルカプトヘキサノールに基づいたリンカー)によってオ
リゴマーの3’又は5’末端を介して連結される。これらリンカーは市販されて
いて、補欠分子族をオリゴマーへ連結させるのに好適な技術は当業者に知られて

ハイブリダイゼーション速度を高めることが開示組成物の生物学的な活性にと
って重要であり得るのは、こうすれば低濃度の組成物でより高い活性を達成する
ことが可能になるからである。このことは短命のRNA基質又はあまり頻繁には
生じないRNA基質にとって重要である。ハイブリダイゼーションの実質的な加
速は、例えば、(例えばいくつかのリジン残基を含有する)正電荷のペプチドを
オリゴヌクレオチドの末端へカップリングさせることによって達成され得る(C
orey,J.Am.Chem.Soc.117:9373〜9374(199
5))。開示化合物は上記のリンカーを使用するこのやり方で平易に修飾され得る
。別のやり方では、ハイブリダイゼーション速度は、スペルミニル残基を含有す
るサブユニットを取込むことによっても高められ得る(SchmidとBehr
、Tetrahedron Lett.36:1447〜1450)。開示化合
物のそのような修飾は二次構造を有するRNA基質に結合する能力も向上させる
。
オリゴマーの合成
開示化合物は任意の適切な方法を使用して合成され得る。多くの合成法が知ら
れている。開示化合物の合成には以下の技術が好ましい。プリンリボヌクレオチ
ド残基を含有し、転換したチミジン残基のような適切な3’末端(Ortiga
oら、Antisense Research and Developmen
t 2:129〜146(1992))又は3’−エクソヌクレアーゼによる起こ
り得る分解を防ぐ3’末端の2つのホスホロチオエート連結を有する、2’−O
−アリル修飾オリゴマーが、任意の市販DNA/RNA合成機を用いた固相β−
シアノエチルホスホロアミダイト化学によって合成され得る(Sinhaら、N
ucleic Acids Res.12:4539〜4557(1984))。
好ましい方法は、リボヌクレオチドのための2’−O−tert−ブチルジメチ
ルシリル(TBDMS)保護戦略(Usmanら、J.Am.Chem.Soc
.109:7845〜7854(1987))であり、必要とされる3’−O−ホ
スホロアミダイトはすべて市販されている。さらに、アミノメチルポリスチレン
の
支持物質としての使用がその有利な性質のために好ましい(McCollumと
Andrus,Tetrahedron Letters 32:4069〜4
072(1991))。市販のフルオレセインホスホロアミダイトを使用して、合
成中に基質RNAの5’末端へフルオレセインを付けることができる。一般に、
標準的なRNAサイクルを使用して所望のオリゴマーが合成され得る。アセンブ
リーが完了したら、密封バイアルにて濃縮水性アンモニア水/エタノール(3:
1v/v)で55℃、8時間処置することにより、不安定な塩基保護基をすべて
除去する。エタノールは2’−O−TBDMSが早めに除去されるのを抑制する
が、そうしないと脱保護の基本条件下で得られるリボヌクレオチドの位置でかな
りの鎖開裂が起こってしまう(Usmanら、J.Am.Chem.Soc.1
09:7845〜7854(1987))。凍結乾燥した後に、TBDMS保護化
オリゴマーは、60℃で2時間、トリエチルアミントリヒドロフルオリド/トリ
エチルアミン/N−メチルピロリドンの混合物で処置し、中性的な条件下でシリ
ル保護基を迅速的かつ効率的に除去する(Wincottら、Nucleic
Acids Res.23:2677〜2684(1995))。次いで、完全に
脱保護化したオリゴマーは、CathalaとBrunelの方法(Nucle
ic Acids Res.18:201(1990))によりブタノール沈殿さ
れ得る。精製は、変性ポリアクリルアミドゲル電気泳動、又はイオン交換HPL
C(Sproatら、Nucleosides and Nucleotide
s 14:255〜273(1995))と逆相HPLCの組み合わせのいずれか
により実施され得る。細胞内で使用するには、合成されたオリゴマーをアセトン
中の過塩素酸ナトリウムで沈殿してそのナトリウム塩へ変換することが好ましい
。次いで残った微量の塩は、好ましくは市販の小さな使い捨てゲル濾過カラムを
使用して除去される。最終工程として、この単離オリゴマーの真性をマトリック
ス介助レーザーデソープション質量分析法(Pielesら、Nucleic
Acids Res.21:3191〜3196(1993))及びヌクレオシド
塩基組成分析によって確認する。さらに、対応する化学合成された短いオリゴリ
ボヌクレオチド基質に対するこのオリゴマーを用いた開裂機能テストも好ましい
。
RNA基質の開裂
RNA切断は細胞の核または細胞質に存在するタンパク質因子により促進され
るので、開示された化合物は非常に高いインビボ活性を持っている。ハンマーヘ
ッドリボザイムの活性を増大できるそのようなタンパク質因子の例は、例えば、
HIV1のヌクレオカプシドタンパク質NCp7(Muller et al.,J.Mol.Biol
.242:422−429(1994))および異種核リボ
核タンパク質A1(Heidenreich et al.,Nucleic Acids Res
.23:2223−2228(1995))である。従って、
長いRNA転写体は開示された化合物により細胞内で効率的に切断される。
開示された化合物は、活性物質として一つまたはいくつかのオリゴマーおよび
、随意に医薬として受容可能な補助物質、添加物および担体を含んでいる医薬組
成物に使用できる。そのような医薬組成物は、真核生物、原核生物およびウイル
ス、特に細胞中の腫瘍遺伝子またはウイルス遺伝子またはRNA分子のようなヒ
ト遺伝子の発現を特異的に不活性化する薬剤の製造に適している。応用のさらな
る範囲は植物遺伝子または昆虫遺伝子発現の不活性化である。従って、開示され
た組成物はヒトおよび動物のための薬剤ならびに植物のための殺虫剤に使用でき
る。
細胞へ開示された化合物を送達するためには種々の方法が利用可能である。例
えば、一般的に開示された化合物はマイクロパーティクル内または上に取り込む
ことができる。本明細書で使用される場合、マイクロパーティクルには合成およ
び/または天然ポリマーで形成されたリポソーム、ビロソーム、マイクロスフェ
アおよびマイクロカプセルが含まれる。マイクロカプセルおよびマイクロスフェ
アを作成する方法は当業者には既知であり、溶剤蒸発、溶剤鋳造、噴霧乾燥およ
び溶剤伸長が含まれる。種々のマイクロパーティクル内へ取り込むことができる
有用なポリマーの例としてはポリサッカライド、ポリ無水物、ポリオルトエステ

カチオンリポソームまたはマイクロカプセルは、開示された化合物のような陰
性に荷電した化合物を送達するのに特に有用であるマイクロパーティクルであり
、化合物はリポソームの陽性に荷電した外側表面へイオン的に結合できる。Fe

されたリポソームまたはマイクロカプセルが、陰性に荷電した化合物をイオン的
に結合するであろう正味の陽性電荷を持つように、カチオンリポソームまたはマ
イクロカプセルは十分な量の一つまたはそれ以上のカチオン側鎖を含んでいる脂
質からなる混合物を用いて製造できる。カチオンリポソームを製造するために使

開示された組成物送達のためのマイクロパーティクルの好適な形はヘムを持つ
マイクロパーティクルである。これらのマイクロパーティクルにおいて、ヘムは
マイクロパーティクルの外側表面内へインターカレートされているかまたは共有
結合で結合されている。ヘムを持つマイクロパーティクルは、肝細胞のようなヘ
ムレセプターを発現する細胞により優先的に結合および取り込まれるので、有効
投与量に必要とされる薬剤量が著しく少なくてもよいという利点を持っている。
そのような標的化送達はまた、非標的化送達法において相対的に高薬剤濃度使用
する場合に起こりうる全身的副作用も減少させる。ヘムを持つマイクロパーティ
クルの形成に好適な脂質は1,2−ジオレオイルオキシ−3−(トリメチルアン
モニウム)プロパン(DOTAP)およびジオレオイルホスファチジルエタノー

開示された組成物はまたカチオンリポソームによりカプセル化またはカチオン

開示された組成物を含んでいるリポソームは、標的細胞への開示された化合物
送達のための有効量で、例えば、静脈内または腹腔内投与により全身的に投与で
きる。他の可能な経路には、適切なマイクロパーティクルとともに使用された場
合の経皮または経口が含まれる。一般的に、個体に投与されるリポソーム結合オ
リゴマーの総量は、同一の所望されるまたは意図される効果のために投与されな
ければならない非結合オリゴマー量よりも少ないであろう。
ポリ乳酸およびポリグリコール酸コポリマー、ポリエチレンおよびポリオルト
エステルのような種々のポリマーを含んでいる組成物および開示された組成物は
、カテーテルまたは注射器を使用して適切な細胞に局所的に送達できる。そのよ
うな組成物を細胞へ局所的に送達する他の手段には、注入ポンプの使用(例えば
、Alza Corporation,Palo Alto,Californ

aからのもの)または移植物に隣接した領域への治療組成物の徐放性放出を達成
できるポリマー性移植物内への組成物の取り込み(例えば、Johnson a

治療的応用には、活性物質は好適には0.01から10,000μg/kg体
重、より好適には0.1から1000μg/kg体重の濃度で投与される。投与
は、例えば、注射、スプレーとして吸入(例えばエアロゾルとして)、経口的(例
えば、錠剤、カプセル、被覆錠剤その他として)、局所的または直腸的(例えば
座薬として)により実施できる。
開示された組成物は遺伝子発現の特異的不活性化のための方法に使用でき、組
成物が細胞内に存在する前もって決められたRNA分子を切断するように(切断
は好適には触媒的に起こる)、活性濃度の組成物が細胞に取り込まれる。米国特
許第5,334,711号に記載されている類似の組成物はマウスでの遺伝子不
活性化に成功している(Lyngstadaas et al.,EMBO J.1
4:5224−5229(1995))。この方法は培養細胞に対してインビトロ
で、ならびに生きている生物体(原核生物またはヒト、動物または植物のような
真核生物)に対してインビボで実施できる。
開示された組成物はまた、RNA分子を切断するためのRNA制限酵素として
使用できる(例えば、細胞なしのインビトロ反応)。開示された組成物はまたRN
A分子の制限切断のための反応キットにも使用でき、例えばそれには、オリゴマ
ーおよび適した緩衝化物質が含まれている。この場合、オリゴマーおよび緩衝化
物質は溶液、懸濁液または散剤または凍結乾燥物のような固形物の形で存在でき
る。試薬は一緒に、お互いに分離されてまたは随意に適した担体上にも存在でき
る。開示された組成物はまた、診断試薬としてまたは未知遺伝子の機能を同定す
るためにも使用できる。
本発明は以下の非制限的な実施例を参考にしてさらに理解されるであろう。実施例 実施例1:15.1位でのイノシン置換は効果的にN16.2C16.1H17を切断でき ることを示している切断反応
N16.2C16.1H17モチーフ(式中、H17はグアノシンではない)の各々の順列
を含む12の基質の組が合成された。切断アッセイに使用されたオリゴマーおよ
び対応する基質は表1に示されている。基質の各々は5’末端がフルオレセイン
で標識されており、および3’末端に逆方向チミジンキャップが使用された。四
つの触媒オリゴマーの組が合成され、基質の各々に対して適切に一致した触媒オ
リゴマーを提供している。これらの触媒オリゴマーの各々は15.1位にイノシ
ンを持っている。基質の16.1位にUおよび触媒オリゴマーの15.1位にA
がある対照基質および触媒オリゴマーも合成された。

上記の基質および触媒オリゴマーは、切断のために16.1位のUを必要とす
ることを克服するための15.1位のイノシンの能力を決定するための切断反応
で使用された。すべての反応は以下のプロトコールを使用して実施された。反応
は典型的には100μlで行われ、反応液には蒸留およびオートクレーブ処理さ
れたH2O、10mM MgCl2、10mM トリス−HCl pH7.4、5
μMリボザイムおよび0.25μM基質が含まれている。触媒オリゴマー、基質
および緩衝液は一緒に加えられ、95℃で5分間加熱された。5分以上かけて室
温まで冷却後、反応液を10mM MgCl2に加え、混合して37℃に置かれ
た。特定の時間間隔(10、30、60および120分)で10μlを採り、反
応を停止させるために3μlの充填緩衝液(95%ホルムアミド、100mM
EDTA pH8.0、0.05%ブロモフェノールブル−)に加えた。試料は
20%ポリアクリルアミドゲル電気泳動により分析された。ゲルはMolecu
lar Dynamics Fluorescence Imagerで分析さ
れた。表1に示された基質および触媒オリゴマーを使用したこの型の切断反応の
結果は表2に示されている。 数字は示された時間点(反応を開始して0,10、30、60および120分
後)で切断された基質のパーセントを示している。結果は、16.1位にCを持
つ基質は15.1位にIを含んでいる触媒オリゴマーにより切断されうることを
示している。120分の時点で種々の基質間に相違はあるものの、16.1位に
Cを持つ基質は15.1位にIを持つ触媒オリゴマーに関して、すべての場合で
効率よく切断できることをデータは示しており、15.1位でのIの置換はN16 .2
C16.1H17部位を含んでいる適当な基質の切断を実際に可能にすることを示し
ている。
C16.1を持っている12の基質およびU16.1を持っている対照基質の対応する
触媒オリゴマー(すべて表1に示されている)による切断初速度は単一ターンオ
ーバー速度論を用いて決定された。単一ターンオーバー速度論は2.5μlの1
00μMリボザイム溶液、2.5μlの10μM 5’フルオレセイン標識基質
溶液および10μlの100mMトリス−HCl pH7.4溶液を混合するこ
とにより算定された。混合物は90μlの最終容量に希釈し、95℃に5分間加
熱した後、37℃に冷却した。10μlの100mM MgCl2溶液を添加す
ることにより反応を開始させた。反応成分の最終濃度は、250nM基質、2.
5μMリボザイムおよび10mM MgCl2であった。種々の時間に10マイ
クロリットルの試料を採り、反応を停止させるために10μlの100mM E
DTA、ブロモフェノールブルー溶液と混合した。切断生成物はPAGEにより
未反応基質から分離し、Molecular Dynamics Fluore
scence Imagerで定量された。

は速い速度で切断されることを示している。15.1位にAを持っている対照触
媒オリゴマーとの比較(G16.2U16.1C17トリプレットを持つ基質の切断)
は、A16.2C16.1H17およびG16.2C16.1H17トリプレットを持つ基質(15.
1位にIを持つ触媒オリゴマーにより切断される)が、標準A15.1−U16.1塩基
対を持つ反応体を含む対応する対照反応よりも速い切断初速度を持っていること
を示している。実施例2:リボヌクレオチドのみを含んでいる触媒オリゴマーによるGCHおよ びACHトリプレットでの切断速度
GCHおよびACHトリプレットを切断するために設計された、すべてリボヌ
クレオチドからなるオリゴマー(修飾なし)による基質RNAの切断が評価され
た。使用されたオリゴマーは配列ID番号:17(ACHトリプレット後を切断
)(表1)および配列ID番号:18(GCHトリプレット後を切断)(表1)
であった。対応する短い蛍光標識基質、配列ID番号:4、5および6(ACH
トリプレットを含んでいる)(表1)および配列ID番号:7、8および9(GC
Hトリプレットを含んでいる)(表1)、を各々配列ID番号:17および配列I
D番号:18の触媒オリゴマーと共に使用した。
反応は単一ターンオーバー速度論条件下、2.5μM触媒オリゴマーおよび2
50nM基質を使用し、10mM Mg2+存在下のpH6.0で、および1mM
Mg2+存在下のpH7.4の両方で実施された。他のすべての条件は実施例1
で使用されたものと同一であった。
データは実施例1のように分析された。全リボヌクレオチド触媒オリゴマーお
よび対応する基質の6つの組み合わせのk2値は表4に示されている。
GCA、GCC、ACAおよびACC部位を標的とするすべてのリボヌクレオチ
ドオリゴマーは、GCUまたはACU部位を標的とするすべてのリボヌクレオチ
ドオリゴマーよりも速い切断速度を持っている。さらに、切断活性は1mM M
g2+存在下で非常に速いことを表4は示している。このレベルのMg2+はインビ
ボにおけるMg2+濃度に類似している。実施例3:6つのみのリボヌクレオチドを含んでいる触媒オリゴマーによるGC HおよびACHトリプレットでの切断速度
GCHおよびACHトリプレットを切断するように設計された6−リボヌクレ
オチドオリゴマーによる基質RNAの切断が評価された。これらのオリゴマーに
おいて、リボヌクレオチドはU4、G5、A6、G8、G12およびI15.1位に存在し
ており、他のすべての位置は2’−O−アリル−リボヌクレオチドであった(番

7、8および9(GCHトリプレットを含んでいる)(表1)、が各々配列ID番
号:17および配列ID番号:18の触媒オリゴマーと共に使用された。
反応は単一ターンオーバー速度論条件下、2.5μM触媒オリゴマーおよび2
50nM基質を使用し、10mM Mg2+存在下のpH6.0で、および1mM
Mg2+存在下のpH7.4の両方で実施された。他のすべての条件は実施例1
で使用されたものと同一であった。
データは実施例1のように分析された。6−リボ触媒オリゴマーおよび対応す
る基質の6つの組み合わせにおいて計算されたk2値は表5に示されている。
これらの結果は、U4、G5、A6、G8、G12およびI15.1位以外のすべてのリ
ボヌクレオチドが修飾されている場合は、配列ID番号:17(ACHトリプレ
ット後を切断)(表1)および配列ID番号:18(GCHトリプレット後を切断
)(表1)のような触媒オリゴマーに活性が残っていることを示している。これら
の結果はまた、触媒オリゴマーは生理学的濃度のMg2+で基質を切断できること
も示している。実施例4:U4位の糖修飾が異なっている触媒オリゴマーによるGCAトリプレ ットでの切断速度
