界面活性剤組成物
本発明は界面活性剤に関し、特に金属イオンの存在により安定化されたブロッ
ク共重合体界面活性剤に関する。
ブロック共重合体界面活性剤は多くの産業において様々な用途がある。それら
は食物添加物として使用され、化粧品工業に応用され、また、消泡剤、帯電防止
剤、洗浄剤、ゲル化剤、発泡調節剤、分散剤、および均染剤としても提案されて
いる。しかしながら、その多様性と、外的および内的投与の両方についての生理
学的安全性のため、ブロック共重合体界面活性剤は製薬業において特に重要であ
る。
ブロック共重合体界面活性剤の医療の応用には、薬の受渡システムと製剤にお
ける多くの使用がある。例えば、ブロック共重合体界面活性剤は静脈脂肪乳剤な
どにおける乳化剤、抗生物質などに用いる湿潤剤、濃化剤、錠剤などに用いる被
覆剤、エリキシル剤やシロップ剤の透明性を維持するためなどの可溶化剤、分散
強化剤、および発泡剤として用いられている。これらの重合体は、軟膏や坐薬の
基剤、錠剤結合剤、合成ポリグリコール酸縫糸の潤滑剤としても用いられ、また
微小粒子懸濁液の安定化に使用されることもよく知られている。多くの応用にお
いて、ブロック共重合体界面活性剤の2つ以上の機能を利用することがある。
また、多くのブロック共重合体界面活性剤にはそれらに固有の治療的応用があ
ることが分かっている。例えば、あるブロック共重合体界面活性剤は佐剤または
ハイポレミック剤(hypolemic agent)として作用することが示されているし、ポ
ロキサマー182LFおよびF188(Poloxamers 182LF and F188)にはそれ
ぞれ製薬の補助剤および下剤としての治療的応用がある。また、F188は心筋
梗塞や鎌状血球発症において血流を改善する能力や、やけど治療への応用の可能

ブロック共重合体界面活性剤は固体相および水性または有機溶液中のいずれで
もかなり不安定で、減成を受ける。溶液中では、この減成はより中性のpH下で

て、ミクロモルレベルにおいて長期の調査でFe3+とCu2+がかなりの自動酸化
を起こすことが開示されている。

したがって、酸化減成に対してブロック共重合体界面活性剤組成物を安定化さ
せる方法には特定の必要性がある。これらの化合物を安定化する方法の1つに、
抗酸化剤を添加してラジカル分解反応を阻害することが挙げられる。しかしなが
ら、モノチオグリセロールなどの抗酸化剤を薬の製剤に利用することは望ましく

定化できることを発見した。その金属イオンは、非常に低い金属イオン濃度でブ
ロック共重合体界面活性剤に安定性を与え、また従来の安定化技術よりも安全面

予測されることを明確に示していることを考慮すると、より一層驚くべきことで
ある。金属イオンおよび/またはポリアルキレングリコールは、医療および非医
療のいずれの使用を目的とする組成物を安定化するのにも用いることができると
考えられる。
したがって、一つの様相から見ると、本発明は、ブロック共重合体界面活性剤
と金属イオンとを含む組成物であり、前記金属イオンが前記ブロック共重合体界
面活性剤の酸化減成を阻害するのに十分な量で存在することを特徴とする組成物
を提供し、特に、界面活性剤(および、任意的にその特定の界面活性剤と伝統的
に関連する不純物と減成物)と、金属イオンと、任意的に前記金属イオンの対イ
オンと、任意的に例えば水または有機溶媒、または2種類以上の溶媒の混合物な
どの液体溶媒系とから本質的に成る組成物を提供する。
別の様相から見ると、本発明は、ブロック共重合体界面活性剤組成物に対して
金属イオンを前記ブロック共重合体界面活性剤の酸化減成を阻害するのに十分な
量で添加することを含む、安定化されたブロック共重合体界面活性剤組成物の製
造方法を提供する。
更に別の様相から見ると、本発明は、ブロック共重合体界面活性剤を含む組成
物の製造のための金属化合物の使用であり、前記金属のイオンが前記ブロック共
重合体界面活性剤の酸化減成を阻害するのに十分な量で存在することを特徴とす
る使用を提供する。
更に別の様相から見ると、本発明はヒトまたはヒトでない動物(例えば、哺乳
類や鳥類の動物など、血管を有する動物が好ましい)の体の治療または診断の方
法を提供する。前記方法は、生理学的に許容できるブロック共重合体界面活性剤
と生理学的に許容できる量の金属イオンとを含む組成物を治療的または診断的に
有効な量で前記の体に投与することを含む方法であり、前記金属イオンが前記ブ
ロック共重合体界面活性剤の酸化減成を阻害するのに十分な量で存在することを
特徴とする方法である。
本発明において、前記金属イオンは安定化効果を達成するのに十分な量、例え
ば治療または診断的な投与のために前記組成物を安定化するのに十分な濃度で用
いられる。前記組成物に使用される金属イオンの量は、ブロック共重合体界面活
性剤と存在する他の賦形剤の性質および量、例えば治療または診断的な使用や化
粧的な使用などの前記組成物の使用、前記活性剤の性質などに依存するが、必要
な量は当業者により容易に決定できる。一般に、前記ブロック共重合体界面活性
剤に対する使用される金属イオンの濃度は1×10-6Mから1×10-3Mの範囲
であり、好ましくは10-4から10-5Mの次数である。
本発明の金属イオンはブロック共重合体界面活性剤を安定化させるイオンであ
ればどんなイオンでも良い。特に好ましい金属イオンとしては、安定した低原子
価状態を有するイオン、すなわち遷移金属イオンなどの還元性イオンが挙げられ
、特に、鉄、ジルコニウム、ニッケル、およびコバルトのイオン、例えば、Fe3+
、Zr4+、Ni2+、およびCo2+が挙げられる。また、本発明の組成物は、同
じ組成物に存在する1つ以上のブロック共重合体界面活性剤を安定化するために
1つ以上の金属イオンを利用する組成物を含む。
前記組成物が医療に使用される場合、前記金属イオンは生理学的に許容できる
濃度で存在する。本発明に基づいて有用な前記金属イオンは投与前にキレート剤
に結合できると考えられる。しかしながら、安定化に必要な金属イオンの濃度が
非常に低いことを考えると、これは必要ではなく、このことが本発明の更なる利
点を成す。
本発明のブロック共重合体界面活性剤は、好ましくは非イオン系であり、例え
ばブロックアルキレンオキシド共重合体であってもよく、例えばプルロニック類
(pluronics)(例、エチレンオキシドとプロピレンオキシドとのブロック共重
合体界面活性剤であるプルロニックF68および108)等のポロキサマー類や
、テトロニック類(tetronics)等のポロキサミン類であってもよい。
一般に、安定化される前記ブロック共重合体界面活性剤の分子量は1000か
ら20,000の範囲が都合よく、好ましくは1550から15000であり、
最も好ましくは2000から15000の範囲の原子質量単位である。本発明の
ブロック共重合体界面活性剤は、例えば不活性無水雰囲気中での高温高圧条件下
など、標準の重合方法によって調製できるが、市場で容易に入手できる。
前記組成物に用いられるブロック共重合体界面活性剤の量は、存在する他の賦
形剤の性質と量、例えば治療または診療的な使用や化粧的な使用などの組成物の
使用、活性剤の性質、例えば乳化剤や被覆剤としてなどのブロック共重合体界面
活性剤の特定の使用に依存するが、必要な量は当業者によって容易に決定できる
。
また、驚くべきことに、ポリアルキレングリコール(例、ポリアルキレンオキ
シド)をブロック共重合体界面活性剤組成物に少量添加することにより、組成物
の安定性を増加できることが分かった。
したがって、更に別の様相から見ると、本発明はブロック共重合体界面活性剤
とポリアルキレングリコールとを含む組成物であり、前記ポリアルキレングリコ
ールが前記ブロック共重合体界面活性剤の酸化減成を阻害するのに十分な量で存
在することを特徴とする組成物を提供する。
また更に別の様相から見ると、本発明はブロック共重合体界面活性剤を含む組
成物の製造のためのポリアルキレングリコールの使用であり、前記ポリアルキレ
ングリコールが前記ブロック共重合体界面活性剤の酸化減成を阻害するのに十分
な量で存在することを特徴とする使用を提供する。
前記ポリアルキレングリコールは金属イオンと共に用いることが好ましく、こ
のことが本発明の更に別の様相を成す。
前記ポリアルキレングリコールはブロック共重合体または単独重合体であるこ
とが可能で、C2、C3、またはC4アルキレン単位を有することが好ましく、特
にC2またはC3単位が好ましい。好ましくはポリエチレングリコールである。一
般に、前記ポリアルキレングリコールの分子量は300から10,000の範囲
の原子質量単位であり、好ましくは400から2000の原子質量単位である。
前記組成物に用いられるポリアルキレングリコールの量は、前記ブロック共
重合体界面活性剤および存在する他の賦形剤の性質と量、例えば治療または診療
的な使用や化粧的な使用などの組成物の使用、活性剤の性質などに依存するが、
必要な量は当業者によって容易に決定できる。一般に、使用されるポリアルキレ
ングリコールの量は前記組成物の1〜30重量%、好ましくは5〜20重量%、
最も好ましくは10〜20重量%の範囲である。前記組成物が医療に使用される
場合は、前記ポリアルキレングリコールは生理学的に許容できる量で存在する。
1つ以上のブロック共重合体界面活性剤を安定化するために1つ以上のポリアル
キレングリコール類を使用することも本発明において考えられる。
本発明のポリアルキレングリコール類は標準の重合技術により容易に調製され
市場でも容易に入手できる。

以下、実施例を用いて本発明を更に説明するが、これらの実施例は本発明を限
定するものではない。
実験
ポロキサマー338を分析するために、屈折率検出器を用いるサイズ排除高速
液体クロマトグラフィーの2つの方法が開発された。最初の方法では、ファーマ
シア社(Pharmacia)製のスパロース12コラム(Superose 12 column)(30cm×
7.8mm)を流速0.8mL/minで使用した。注入体積は10μlで、移動相
には10%メタノールと25mMのNaClを含んだ。溶液試料は注入前に2%
以下の濃度まで水で典型的に希釈した。
2つ目のHPLC方法では、最初に試料をメタノールで溶解し、更に注入前に
80%メタノールで適当な濃度まで希釈した。この方法では、スパルコ社(Supel
co)製のTSK G2000SWXLコラム(30cm×7.8mm)と、6部の
メタノールと4部の62.5mMのNaClを含む移動相を使用する。
図1aは、NF等級のポロキサマー338と精製ポロキサマー338の典型的
なクロマトグラムを示す。BASF社から得たポロキサマー338は、名目の分
子量がそれぞれ14.6Kと3Kである高分子量(HMW)の親ピークと低分子
量の不純物/減成物ピークを示した。明示のように、膜ダイアフィルトレーショ
ンにより精製すると低い分子量ピークは著しく減少した。図1bは、40℃での
貯蔵下で異なる範囲に減成された溶液中のポロキサマー338試料の典型的なク
ロマトグラムを示す。溶液中でのポロキサマー338の減成は、最初の親ピーク
の減少と、対応する低分子量ピークの増加から自明である。新たなピークは見ら
れなかった。低分子量ピークの広がりは非常に遅い段階でのみ起こり、その段階
では親ピークがほとんど消滅している。図2aはポロキサマー338の親ピーク
面積対濃度(% w/v)の標準曲線を示す。図2bは、ポリエチレングリコール分
子量標準とウォーターズ社(Waters)のGPCソフトウェアを用いた、分子量決定
のためのSE−HPLC(サイズ排除高速液体クロマトグラフィー)システムの
検量線を示す。ポロキサマー338の減成は親ピークの徐々の減損に明示されて
いるので、安定性試料のHPLCデータは親ピーク下の面積の百分率として表さ
れる。蒸気滅菌
「乾燥品」サイクルを用い、キャッスルM/C 3500(Castle M/C 3500)
またはETCプロジェネシス・ラボST−9−6(ETC Progenesis Lab ST-9-6)
のいずれかの蒸気滅菌器によって蒸気滅菌を行った。直立位置を維持するために
ガラス瓶またはアンプルに入れた試料をアルミニウム製バスケットに置いた。そ
の曇り点の106〜108℃よりも高い温度でポロキサマー338を溶液から相
分離し、析出してガラス瓶の底に濃縮層を形成した。放置して冷却すればその濃
縮層は硬化してゲル状の層になる。従って、蒸気滅菌のサイクルが完了するとす
ぐに、まだ熱いうちにガラス瓶を静かに振り、析出した界面活性剤を再分散させ
た。実施例1 溶液中でのポロキサマー338の安定性
様々な溶液安定性の調査において試料の調整は同様に行われた。約1.5mL
の3%(w/v)ポロキサマー338溶液を各2mLの透明なガラスアンプル(キン
ブル社(Kimble)製、あらかじめ処理および洗浄済み)に移した。アンプルは酸素
火炎で密封した。試料に窒素を流し、その上部空き高に流された後すぐに密封し
、試料に酸素が漏れないことを確実にするためにチャンバを通して窒素ガスをゆ
っくりと流しながら、40℃の真空炉内で貯蔵した。各時点において、必要なら
ば、HPLC分析前にガラス瓶をいくつか回収して水で希釈した。試料は通常1
回のみ注入したが、2回注入した試料もあり、その結果を平均した。また、視覚
的にも試料を調べ、pH値を記録した。溶液安定性−pH安定性−37℃および40℃にて
様々なpHの3%ポロキサマー338溶液の試料を40℃で2ヶ月貯蔵した後
分析した。試料は最初に0.22μmフィルタを通過させた。その結果は親ピー
クの百分率で表され、下記の表に示し、図3にプロットした。 前記データから分かるように、ポロキサマー338は酸性(pH<4)および
アルカリ性(pH>8)条件下では安定性がより高く、その間のpH範囲では安
定性がより低いようである。実施例2
ポロキサマー338の3%水性溶液と2×10-4M(11〜12ppm)の金
属イオンを用いてポロキサマー338の安定性に対する金属イオンの効果を調べ
た。二重の試料をpH8の40mMトリス緩衝液で処理し、滅菌ろ過し、1,2
ヶ月間40℃にさらした。そのSE−HPLCの結果を下記の表に示す。
上記の通り、対照は1ヶ月後40℃で著しく減成した。Co+2イオンとFe+3
イオンは溶液中のポロキサマー338を安定化することが分かった。実施例3
Co+2イオンの安定化効果を確認するために同濃度の金属イオンを用いて別の
研究を行ったが、信頼水準を上げるために試料は三重にした。前記試料を40℃
にさらし、親ピーク面積の百分率で表されるSE−HPLCの結果を下記の表に
示す。
今回も、本調査の結果はFe+3とCo+2が減成に対してポロキサマー338を
安定化できることを確認したが、Fe+2 とCu+2には著しい効果がなかった。N
i+2は前調査では安定化効果を示さなかったが、本調査では著しい安定化効果を
示した。実施例4
好ましい対照としてのCo+2イオンと共に、ZrCl4塩を用いて3%(w/v)ポ
ロキサマー338水性溶液の安定性に対するジルコニウムイオンの効果を調べた
。試料は40℃にさらし、親ピーク面積の百分率で表されるSE−HPLCの結
果(3重の試料)を下記の表に示す。 前記データは、10-5Mでの効果はCo+2ほど強くはないが、Zr+4もポロキ
サマー338の安定性を著しく改善できることを証明している。実施例5 PEG 1450の効果
50℃および蒸気滅菌条件下において、水溶液中の精製ポロキサマー338
(95.5%)を用いて安定化の調査を行った。試料を50℃で3週間貯蔵した
後、以下の結果が得られた。
上記の通り、10%のPEG1450を添加した試料は対照より安定性が高か
った。実施例6
ポロキサマー338を用いて50℃で15日間貯蔵し、別の安定化調査を行
った。全ての試料はpH9の40mMトリス緩衝液中で調製した。以下に結果を
示す。
本調査でも、10%PEG1450を添加した試料は対照より少し安定性が高
かった。PEG1450はポロキサマー338に対して何らかの安定化効果を示
