【発明の詳細な説明】
5HT2C受容体アンタゴニストとしてのインドリン誘導体
本発明は、薬理活性を有するインドリン誘導体、それらの製造方法、それらを
含有する組成物およびCNS障害の治療におけるそれらの使用に関する。
PCT/EP96/00368(WO96/23783)、PCT/EP97
/03156(WO97/48699)およびPCT/EP97/03157(
WO97/48700)は、5HT2C/2B受容体アンタゴニスト活性を有すると
記載されたインドリン誘導体を開示している。この度、PCT/EP96/00
368の包括的な範囲内にあるが、その明細書中に詳しく開示されていない一連
の新規な化合物が見いだされ、それは意外にも強化された5HT2C受容体活性を
示すことが判明した。5HT2C受容体アンタゴニストは不安、鬱病、鬱病、強迫
障害、偏頭痛、アルツハイマー病、睡眠障害、食欲不振や病的飢餓のような食障
害、恐慌発作、コカイン、エタノール、ニコチン、アルコールおよびベンゾジア
ゼピンなどの濫用物質の使用中止に伴う作用、精神***症、さらに水頭病のよう
な脊髄外傷および/または頭部損傷に関する障害といったようなCNS障害の治
療にて有用である可能性があると考えられる。本発明の化合物はまた緑内障、I
BS(過敏性結腸症候群)のような特定のGI(胃腸内)障害ならびに黄斑浮腫
や網膜症などの微小血管系障害の治療に有用であると考えられる。
それゆえに本発明は、第一の態様として、式(I):

R1は水素またはC1-6アルキルであり;
R2、R3およびR4基は独立して水素、ハロゲンまたは1個またはそれ以上のフ
ッ素原子によって置換されていてもよいC1-6アルキルであり;
R5およびR6基は独立して水素またはC1-6アルキルであり:
XおよびYは、独立してCHまたは窒素を意味する;ただし、Yが窒素で、R5
およびR6が共に水素であるとき、Xは窒素である。]
で示される化合物またはその塩を提供する。
C1-6アルキル基、単独あるいは他の基の一部であるかどうかにかかわらず、
直鎖状または分枝状であってもよい。
好ましくはR1は水素である。
適当にはR2、R3およびR4基は、独立して水素、ハロゲンまたは、所望によ
り1個またはそれ以上のフッ素原子によって置換されてもよいC1-6アルキルで
ある。好ましくは、R2は1個またはそれ以上のフッ素原子によって置換された
C1-6アルキル、特にCF3であり、R3はC1-6アルキル、特にメチルである。好
ましくはR4は水素またはC1-6アルキル、好ましくはR4は水素である。
R5およびR6基は、独立して水素またはC1-6アルキル、特にメチルである。
本発明の個々の化合物は:
5−メチル−1−[4−(ピリジン−2−イルメチルオキシ)フェニルカルバモ
イル]−6−トリフルオロメチルインドリン、
5−メチル−1−[2−(ピラジン−2−イルメチルオキシ)ピリジン−5−イ
ル−カルバモイル]−6−トリフルオロメチルインドリン、
5−メチル−1−[2−(1−ピリジン−2−イルエトキシ)ピリジン−5−イ
ル−カルバモイル]−6−トリフルオロメチルインドリン、
およびその医薬上許容される塩を包含する。
式(I)の化合物は通常医薬上許容される酸、たとえばマレイン酸、塩化水素
酸、臭化水素酸、燐酸、アセチル酸、フマル酸、サリチル酸、クエン酸、乳酸、
マンデル酸、酒石酸およびメタンスルホン酸のような酸と酸付加塩を形成するこ
とができる。
式(I)の化合物はまたN−オキシドまたは水和物のような溶媒和物を形成す
る
こともあり、本発明はまたそれらの形も包含する。本明細書で言及する場合、「
式(I)の化合物」なる語はそれらを形を含むと解される。
式(I)のある種の化合物は、鏡像異性体を含む立体異性体の形で存在可能で
あり、本発明はこれら各立体異性体にまで及び、ラセミ化合物を含むその混合物
に及ぶ。異なった立体異性体の形は、通常の方法で一の化合物を他の化合物から
分離することもでき、あるいはいずれかの所定の異性体を立体特異的または不斉
合成によって得ることができる。本発明はまた互変体の形およびその混合物に及
ぶ。
本発明の化合物は、PCT/EP96/00368(WO96/23783)
に記載されているような通常の方法を用いて、例えば式(II):
[式中、R1、R4、R5、R6、XおよびYは、式(I)の記載と同意義である]
で示される化合物を、式(III):
[式中、
R2およびR3は式(I)の記載と同意義であり、AおよびBは、結合した場合に
基−NHCOを形成するのに不可欠な適当な官能基を含有する]
で示される化合物と結合させ、その後、所望によりその医薬上許容される塩を形
成することにより、製造することができる。
AおよびB基の適当な例は:
(i) Aが−N=C=Oであって、Bが水素であり、
(ii) Aが−NHCOLであって、Bが水素であり
(iii) Aが−NH2であって、BがCOLであるか、または
(iv) Aがハロゲンであって、Bが−CONH2である;ここでLは脱離基で
ある。適当な脱離基Lには例えば塩素または臭素のようなハロゲン、イミダゾー
ルまたは所望により、例えば、1個またはそれ以上のハロゲンと置換されていて
もよいフェノキシまたはフェニルチオがある]
式(II)および式(III)の化合物は、既知の方法またはそれに類似した方法
例えば、WO95/01976およびPCT/EP96/00368(WO96
/23783に記載されている方法によって調製することもできる。
式(II)の新規な中間生成物もまた、本発明の一部を形成する。
医薬上許容される塩は、通常適当な酸または酸誘導体との反応によって調製す
ることができる。N−酸化物は、通常過酸化水素または過カルボン酸と反応させ
て形成することができる。
式(I)の化合物およびそれらの医薬上許容される塩は、5HT2C受容体アン
タゴニスト活性を有しており、不安、鬱病、癲癇、強迫障害、偏頭痛、アルツハ
イマー病、睡眠障害(概日リズム障害を含む)、食欲不振や病的飢餓のような食
障害、恐慌発作、コカイン、エタノール、ニコチンおよびベンゾジアゼピンなど
の濫用物質の使用中止に伴う作用、精神***症、さらに水頭病のような脊髄外傷
および/または頭部損傷に関連する障害といったようなCNS障害の治療または
予防に有用である可能性があると考えられる。本発明の化合物はまた緑内障、I
BS(過敏性結腸症候群)のようなある種のGI(胃腸内)障害ならびに黄斑浮
腫や網膜症などの微小血管系障害の治療に有用であると考えられる。
そういうわけで、本発明はまた、特に上記障害の治療や予防における治療物質
としての使用のための式(I)の化合物またはその医薬上許容される塩を提供す
る。
本発明はさらに、ヒトを含む哺乳動物において、式(I)の化合物またはその
医薬上許容される塩の治療上の有効量を患者に投与することからなる、上記の障
害の治療または予防方法を提供する。
さらなる態様は、本発明は上記障害の治療または予防のための医薬組成物の製
造における、式(I)およびその医薬上許容されている塩の使用を提供する。
本発明はまた、式(I)の化合物またはその医薬上許容されている塩、および
医薬上許容される担体を含む医薬組成物提供する。
適当には、周囲温度および大気圧で混合することにより製造される本発明の医
薬組成物を、通常経口、非経口または直腸投与に適合され、それ自体が錠剤、カ
プセル、経口液体製剤、散剤、顆粒、ロゼンジ、復元可能な散剤、注射および注
入可能な液剤または懸濁液または坐薬の形態であってもよい。一般的に、経口投
与用組成物が好まれる。
経口投与用錠剤およびカプセルは、単位剤形であってもよく、結合剤、充填剤
、錠剤化剤、潤滑剤、崩壊剤および許容できる湿潤剤等の通常の賦形剤を含有し
ていてもよい。当該分野において周知の方法にしたがって、錠剤をコーティング
してもよい。
経口液体製剤は例えば、水性あるいは油性懸濁液、溶液、乳剤、シロップ剤ま
たはエリキシルの形態であってよく、または使用前に水または他の適当なビヒク
ルで復元可能な乾燥製品の形態であってもよい。かかる液体製剤は、懸濁剤、乳
化剤、非水性ビヒクル(食用油を含んでもよい)、保存料および、所望により、
慣用的な香味料および着色料などの慣用的な添加物を含有してもよい。
非経口投与の場合、本発明の化合物またはその医薬上許容される塩および滅菌
ビヒクルを用いて流体単位剤形を調製する。ビヒクルおよび使用される濃度に応
じて、その化合物をビヒクルに懸濁させることも溶解させることもできる。液剤
の調製においては、該化合物を注射するために溶かし、適当なバイアルまたはア
ンプルに充填する前に濾過滅菌して密封する。局所麻酔薬、保存料および緩衝液
のような添加物もまたビヒクルに溶かすことが都合がよい。安定性を強化するた
めに、組成物をバイアルに充填後凍結させ、真空下で水を除去する。非経口用懸
濁液は、化合物を溶解する代わりにビヒクルで懸濁し、滅菌処理を濾過により行
なうことができないことを除いては実質的に同様の方法で調製される。かかる化
合物は滅菌ビヒクルで懸濁する前にエチレンオキシドに曝すことで滅菌すること
ができる。有利には、界面活性剤および湿潤剤を組成物に含め、本発明の化合物
の均質な分配を容易にする。
投与方法によるが、組成物は0.1重量%ないし99重量%、好ましくは重量
の
10ないし60重量%の有効成分を含有してもよい。
前記した障害の治療における化合物の使用用量は通常の方法にて、障害の重度
、患者の体重やその他の類似した要因に応じて変化するであろう。しかしながら
、一般的指針として通常単位投与量は、0.05ないし1000mg、より適当
には0.05ないし20.0mg、例えば0.2ないし5mg;またかかる単位
投与量を一日一回以上、例えば一日2、3回投与し、そうすることで一日の投与
量をおよそ0.5ないし100mgの範囲で投与してもよい。またかかる治療を
数週間から数ヶ月にわたり継続してもよい。
本発明に従って薬を服用した場合、本発明の化合物から許容できない毒素学的
作用は考えられない。
次の記載例および実施例を用いて本発明の化合物の調製を説明する。
記載例1
1−メトキシ−4−ニトロ−2−トリフルオロメチルベンゼン(D1)
ナトリウム(11.78g、0.512mol)を乾燥メタノール(1l)中
で溶解し、その得られた溶液にメタノール(100ml)中の1−クロロ−4−
ニトロ−2−トリフルオロメチル−ベンゼン(96.22g、0.427mol
)溶液を加えた。その反応混合物を3時間還流し、次いで冷却し真空下で蒸発さ
せた。残渣を水(500ml)とジクロロメタン(3×400ml)間に分配さ
せた。合した有機抽出物を乾燥させ(Na2SO4)、ついで蒸発させ、白色固体
の標記化合物(93.76g、99%)を得た。
記載例2
(5−メトキシ−2−ニトロ−4−トリフルオロメチルフェニル)アセトニトリル
(D2)
1−メトキシ−4−ニトロ−2−トリフルオロメチルベンゼン(D1)(93
g、0.421mol)および4−クロロフェノキシアセトニトリル(77.5
5g、
0.463mol)の乾燥DMF(500ml)中混合物を−10℃で0.75
時間にわたって、KOt−BU(103.85g、0.927mol)の乾燥D
MF(400ml)中攪拌溶液に滴下した。添加終了後、得られた紫色の溶液を
1時間−10℃に保もち、次いで氷/水(1.51)および5Mの水性HCl(
1.51)の混合物中に注いだ。その得られた混合物をジクロロメタンで抽出し
た(3×11)。その合した抽出物を水(3l)で洗浄し、乾燥させ(Na2SO4
)および減圧の下で蒸発させた。残渣を、溶出液として10−40%酢酸エチル
/石油エーテルを用いて溶離するシリカ上のクロマトグラフィーに付して粗生成
物を得、それを酢酸エチル/石油エーテルから再結晶して白色固体、融点103
−104℃の標記化合物を得た。
記載例3
5−メトキシ−6−トリフルオロメチルインドール(D3)
エタノール/水(9:1、1.61)および氷酢酸(16ml)中の(5−メ
トキシ−2−ニトロ−4−トリフルオロメチルフェニル)アセトニトリル(D2)
(85g、0.327mol)を室温で0.5時間50psiで炭素上で10%
パラジウム(50g)で水素化した。反応混合物を濾過し、真空下で蒸発させた
。残渣を水性K2CO3(1l)およびジクロロメタン(2×11)の間に分配し
、合した有機抽出物を乾燥し(Na2SO4)、蒸発させて灰色固体として標記イ
ンドール(67.63g、96%)を得た。
記載例4
5−メトキシ−6−トリフルオロメチルインドリン(D4)
氷酢酸(500ml)中のインドール(D3)(67.63g、0.315m
ol)をシアノホウ水素化ナトリウム(40g、0.637mol)と室温で攪
拌しながら少しづつ処理した。室温で3時間後反応混合物を水(500ml)で
希釈し、次いで40%水性水酸化ナトリウムで冷却しながら塩基性にした。混合
物は次いで
ジクロロメタン(3×500ml)で抽出され、合した抽出物を乾燥し(Na2
SO4)、蒸発させて、灰白色固体の標記化合物(67.73g、99%)を得
た。
記載例5
5−ヒドロキシ−6−トリフルオロメチルインドリン(D5)
5−メトキシ−6−トリフルオロメチルインドリン(D4,7.5g,34.3
mmol)およびヨードトリメチルシラン(12.5ml,89.3mmol)
の乾燥クロロホルム(70ml)中混合物を還流下で6.5時間加熱した。次い
で冷却した混合物にメタノールを攪拌させながら注意深く加え、ついで溶媒和物
を真空下で除去した。残渣を、飽和状態の重炭酸ナトリウム溶液および水で塩基
性になるまで処理し、ジクロロメタン/メタノールで抽出した。有機抽出物をブ
ラインで洗浄し、乾燥して、蒸発させた。残渣をソックスレー抽出器中でエーテ
ルで抽出し、得たれた溶液を濃縮し、3個の産物(合計2.85g,41%)、
融点>180℃分解の標記化合物を得た。
記載例6
1−アセチル−5−ヒドロキシ−6−トリフルオロメチルインドリン(D6)
インドリン(D5、2.84g、14mmol)および無水酢酸(1.32m
l、14mmol)の乾燥ジクロロメタン(50ml)中混合物を、3時間室温
で攪拌し、次いで、蒸発させた。残渣を注意深く飽和状態の重炭酸ナトリウム溶
液で処理し、次いで、その固形生成物を濾過し、水で洗浄し、乾燥させて、標記
化合物(3.28g、96%)、融点244−7℃を得た。
記載例7
1−アセチル−6−トリフルオロメチル−5−トリフルオロメチルスルホニルオ
キシ−インドリン(D7)
0℃のアセチルインドリン(D6、1.19g、4.9mmol)の乾燥ピリ
ジン(10ml)中溶液に、トリフルオロメタンスルホン酸無水物(1.52g
、5.4mmol)を加えた。次いで該混合物を室温にまでゆっくりと加温しな
がら一晩中攪拌した。混合物を部分的に蒸発させ、残りの液体を水で充分に希釈
し、沈澱物は濾過した。粗生成物はジクロロメタンに溶解し、溶液を1N塩酸お
よびブラインで洗浄し、乾燥、蒸発させて標記化合物(1.77g,96%)を
得た。
記載例8
5−メチル−6−トリフルオロメチルインドリン(D8)
トリフルオロメチルスルホニルオキシインドリン(D7、1.77g、4.6
9mmol)、塩化リチウム(0.60g、14.1mmol)およびビス(ト
リフェニルホスフィン)パラジウム(II)塩化物(0.10g、0.14mmo
l)の乾燥ジメチルホルムアミド(15ml)中混合物にテトラメチルスズ(0
.72ml、5.2mmol)を加えた。混合物を3.5時間110℃で加熱し
、次いで冷却、蒸発させた。残渣をジクロロメタンおよび水間に分配させ、有機
相をブラインで洗浄し、乾燥、蒸発させた。粗生成物をエタノール(30ml)
に溶解し、10%水性水酸化ナトリウム溶液(7.5ml)および固体水酸化ナ
トリウム(1g)を加え、その混合物を還流下で一晩中加熱した。エタノールは
真空下で除去され、残渣を水で希釈し、ジクロロメタンで抽出した。有機抽出物
をブラインで洗浄し、乾燥、蒸発させた。残渣をシリカゲル(50g)上のクロ
マトクラフィーに処し、吸引下、2:1のエーテル/石油エーテルで溶出し、標
記化合物(0.70g、74%)、
融点43−4℃を得た。
記載例9
2−(4−ニトロフェノキシ)メチルピリジン(D9)
2−ピリジルカルビノール(5.45g、50mmol)を水素化ナトリウム
(油中80%、1.65g、55mmol)の乾燥ジメチルホルムアミド(DM
F、40ml)中−20℃の攪拌懸濁液に滴下した。混合物を同温度で2時間攪
拌し、次いでDMF(10ml)中の4−フルオロニトロベンゼン(7.05g
、50mmol)に加えた。混合物を室温にまでゆっくりと加温しながら一夜攪
拌した。DMFは真空下で除去され、残渣はジクロロメタン/メタノールおよび
水の間に分配した。有機相は水およびブラインで洗浄され、乾燥、蒸発させた。
残渣をエーテルから再結晶し、標記化合物(3.55g、31%)融点37−8
℃を得た。
記載例10
2−(4−アミノフェノキシ)メチルピリジン(D10)
ニトロフェノキシメチルピリジン(D9、3.54g、15.4mmol)の
メタノール(250ml)中溶液を塩化錫(II)(14.4g、73.2ml)の
溶液と濃HCl(25ml)中溶液と2.5時間60℃で処理した。エタノール
を真空下で除去し、残渣を10%水性水酸化ナトリウムおよびジクロロメタンの
間に分配した。有機抽出物を水およびブラインで洗浄し、乾燥、蒸発させてガム
状の標記化合物(2.76g、90%)を得、それを放置して固体(融点62−
5℃)を得た。
記載例11
5−ニトロ−2−(ピラジン−2−イルメチルオキシ)ピリジン(D11)
乾燥ジメチルホルムアミド(50ml)中のピラジンメタノール(2.9g、
0.
026mole)を−20℃まで冷却し、アルゴン下、水素化ナトリウムの鉱油
中80%分散液(1.03g、0.034mole)で処理した。混合物を2時
間−20℃で攪拌した。2−クロロ−5−ニトロピリジン(3.34g、0.0
21mole)を加え、混合物を0.5時間−20℃で攪拌され、次いで室温ま
で加温し、2時問攪拌した。水(2ml)を滴下し、真空下で溶媒を除去した。
残渣をジクロロメタン中に溶解し、10%水性水酸化ナトリウム溶液で洗浄し、
ついで水で洗浄し、乾燥させ(Na2SO4)、真空下で蒸発させて固体残渣を得
、それを溶出液として酢酸エチルを用いてシリカのプラグを通して濾過し、オレ
ンジ色の固体として標記化合物を得た。
記載例12
5−アミノ−2−(ピラジン−2−イルメチルオキシ)ピリジン(D12)
エタノール(80ml)中の5−ニトロ−2−(ピラジン−2−イルメチルオ
キシ)ピリジン(D11、2.0、0.0086mole)を濃HCl(8ml
)中の塩化錫(8.3g、0.044mole)で処理した。混合物を1時間6
0℃で加熱した。室温まで冷却した後、混合物を水で希釈し、10%水性水酸化
ナトリウム溶液で塩基性にし、ジクロロメタンに抽出し、乾燥させ(Na2SO4
)、真空下でガム状にまで蒸発させ、それをシリカゲル上の酢酸エチルで溶出す
るクロマトグラフィーに付して、標記化合物(0.74g、45%)を淡黄色固
体として得た。
記載例13
N−[2−(ピラジン−2−イルメチルオキシ)ピリジン−5−イル]カルボン酸フ
ェニル(D13)
ジクロロメタン(40ml)中の5−アミノ−2−(ピラジン−2−イルメチ
ルオキシ)ピリジン(D12、0.73g、0.0036mole)をアルゴン
下で−78℃まで冷却した。トリエチルアミン(0.5ml、0.0036mo
le)を加え、つづいてクロロギ酸フェニル(0.5ml、0.0040mol
)を滴下し、および混合物を2時間かけて室温にまで加温し、水性Na2CO3(
50ml)中に注ぎ、ジクロロメタン(3x50ml)で抽出した。合した有機
抽出物を乾燥させ(Na2SO4)、真空下で蒸発して固体の標記化合物(1.0
5g)を得、それをさらに精製することなく用いた。
記載例14
5−ニトロ−2−(1−ピリジン−2−イルエトキシ)ピリジン(D14)
乾燥ジメチルホルムアミド(40ml)中の1−ピリジン−2−イルエタノー
ル(2.0g、0.016mole)を−10℃まで冷却し、アルゴン下で水素
化ナトリウムの鉱油中80%分散液(0.5g、0.017mole)で滴下処
理した。混合物を−10℃で2時間撹拌した。2−クロロ−5−ニトロピリジン
(2.58g、0.016mole)を添加し、混合物を−10℃で0.5時間
攪拌し、次いで室温まで加温して2時間攪拌した。水(2ml)を滴下し、溶媒
を真空下に除去した。残渣をジクロロメタンで溶解し、乾燥させ(NO2SO4)
、真空下で蒸発させて固体残渣を得、それを溶出液として酢酸エチルを用いるシ
リカ上のクロマトグラフィーに付して、標記化合物(1.15g、29%)をオ
レンジ色固体として得た。
記載例15
5−アミノ−2−(1−ピリジン−2−イルエトキシ)ピリジン(D15)
エタノール(30ml)中の5−ニトロ−2−(1−ピリジン−2−イルエト
キ
シ)ピリジン(D14、1.142g、0.0047mole)を濃HCl(8
ml)中の塩化錫(II)(4.32g、0.023mole)で処理した。混合
物を1時間で60℃まで加熱した。室温に冷却した後、混合物を水で希釈し、1
0%水性水酸化ナトリウム溶液で塩基性にし、ジクロロメタンに抽出し、乾燥さ
せ(Na2SO4)、真空下で蒸発させてガム状物質を得、それを酢酸エチルで溶
出するシリカゲル上のクロマトグラフィーに付し、標記化合物(0.54g、5
4%)を淡黄色油状物質として得た。
実施例1
5−メチル−1−[4−ピリジン−2−イルメチルオキシ)フェニルカルバモイル
]−6−トリフルオロメチルインドリン
2−(4−アミノフェノキシ)メチルピリジン(D10、0.30g、1.5
mmol)、クロロギ酸フェニル(0.19ml、1.5mmol)およびトリ
エチルアミン(0.20ml、1.5mmol)の乾燥ジクロロメタン(20m
l)中混合物を室温で2.25時間攪拌し、次いで蒸発させた。残渣を乾燥アセ
トニトリル中に懸濁した。インドリン(D8、0.30g、1.5mmol)お
よびトリエチルアミン(0.20ml、1.5mmol)を加え、混合物を1.
5時間100℃で加熱した。冷却後、混合物を水中に注ぎ、沈澱物を濾過して、
水で洗浄し乾燥させた。粗生成物をジクロロメタンから再結晶することで、標記
化合物(0.35g、55%)を得た。融点178−8℃、M.S.m/z=4

実施例2
5−メチル−1−[2−(ピラジン−2−イルメチルオキシ)ピリジン−5−イ
ル−カルバモイル]−6−トリフルオロメチルインドリン(E2)
インドリン(D8、0.73g、3.61mmo1)、カルバミン酸フェニル
(D13、1.05g)およびトリエチルアミン(2ml)の乾燥ジメチルホル
ムアミド(25ml)中混合物を3時間100℃で加熱した。冷却後、溶媒を真
空下で除去し、残渣をジクロロメタンに溶解し、10%水性水酸化ナトリウム溶
液で洗浄し、
乾燥させ(Na2SO4)、真空下で蒸発させた。得られた溶液を酢酸エチルで溶
出するシリカゲル上のクロマトグラフィーに付し、標記化合物(0.52g、3
3%)を白色固体として得た。融点215−218℃。
実施例3
5−メチル−1−[2−(1−ピリジン−2−イルエトキシ)ピリジン−5−イ
ル−カルバモイル]−6−トリフルオロメチルインドリン(E3)
ジクロロメタン(30ml)およびトリエチルアミン(0.4ml)中の5−
アミノ−2−(1−ピリジン−2−イルエトキシ)ピリジン(D15、0.54
g、0.0025mol)を−30℃でクロロギ酸フェニル(0.31ml、0
.0025mol)で滴下処理し、該混合物を2時間かけて室温まで加温し、水
