動物における体重に関連する病気の診断及び治療方法
この出願は、1997年1月24日に出願された米国仮出願第60/035,
896号の利益を要求する。
米国政府は、この発明の支払い済ライセンスもっており、限られた情況におい
て、特許所有者に、国立衛生研究所、国立心臓、肺及び血液研究所により与えら

発明の背景
肥満及び甚大な体重喪失を含む体重の異常は、多くの人を悩ませている。肥満
は、過剰の体脂肪がある状態である。それは、しばしば、糖尿病、高血圧及び高
コレステロールレベルを含む他の病気と関係している。肥満の指標としての体重
インデックスに基づいて、米国人の成人の59%が健康な体重を超過していると
いうことが最近報告された。甚大な体重喪失は、癌、AIDS、組織消耗、神経性食
欲不振、慢性感染症又は胃腸病を含む様々な起源から生じ得る。幾つかの遺伝子
(ob、db、tub、Ay及びfat)が、以前に、マウスにおける肥満の制
御に関与すると報告された。
発明の要約

与える病気を指示するものである。
好適具体例において、評価のステップは、ICAMレセプター又はβ2インテ
グリンポリペプチドを有する生物学的試料を、ICAMレセプター又はβ2イン
テグリンポリペプチドに特異的に結合する抗体と、その抗体及びICAMレセプ
ター又はβ2インテグリンポリペプチドを含む反応複合体の形成を与える条件下
で接触させることを含む。この抗体及びICAMレセプター又はβ2インテグリ
ンポリペプチドを含む反応複合体の形成を検出する。形成された反応複合体の量
を評価する(この量は、生物学的試料中のICAMレセプター又はβ2インテグリ

細胞外部分を用意する。結合性分子又はその細胞外部分を用意する。この薬剤、

重と関連した病気の治療を必要とする動物を用意する。ICAMレセプター又は
β2インテグリンの代謝又は構造の情況を変えることのできる薬剤を用意する。
この薬剤をこの動物に、治療上有効な量で、体重と関連した病気の治療が起きる
ように投与する。

方法である。ICAMレセプター又はβ2インテグリンポリペプチドを用意する

詳細な説明
この発明は、動物が体重異常の危険にあるかどうかを測定する方法を提供する
。動物を用意する。ICAMレセプター又はβ2インテグリンの代謝又は構造の
状況をその動物において評価する。ICAMレセプター又はβ2インテグリンの
代謝又は構造における異常は、体重異常の危険にあることの診断となる。
動物とは、ヒト並びに非ヒト動物を意味する。非ヒト動物には、例えば哺乳動
物例えばサル、チンパンジー、エイプ、ゲッ歯類、ブタ、ウサギ、ヤギ、ウシ又
はガチョウが含まれる。動物は又、トランスジェニック非ヒト動物をも包含する
。用語トランスジェニック動物は、外来DNA即ち部分的に又は完全に異種のD
NAのその細胞のDNA中への導入により;又は傷害例えばイン・ビトロで誘導
した突然変異例えば欠失その他の染色体の再配列のその細胞のDNAへの導入に
より;又は相同DNAのその細胞のDNA中への導入(DNAを挿入された細胞
のゲノムを変化させる、例えば、天然の遺伝子と異なる位置に挿入するか又は挿
入がノックアウトを生じるような仕方での導入)により新たな遺伝情報を獲得し
た動物を包含することを意味している。この動物は、トランスジーンを、生殖系

体重異常は、例えば、肥満又は体重喪失例えば甚大な体重喪失を包含すること
を意味している。肥満は、過剰の体脂肪がある状態である。最も一般的な肥満の
指標は、体重インデックス(BMI)であり、これは、個人の体重(キログラム)を
その身長(メートル)の平方で除したものである。BMIは、体脂肪と高度に相関
している。国立衛生研究所及び米国健康協会による1995年度のガイドライン
は、健康な体重をBMIが25未満と定めている。肥満は、しばしば、他の病気

体重喪失は、減じた食物摂取の結果として又は食餌制限の欠如(正常の又は過
剰の食物摂取時)において生じ得る状態である。正常又は過剰の食物摂取におい
て生じる体重喪失は、例えば、インシュリン依存性の真性糖尿病、甲状腺中毒症
又は食物の吸収不良により生じ得る。減じた食物摂取から生じる体重喪失は、例
えば癌、AIDS、組織消耗、神経性食欲不振、慢性感染症又は胃腸病により生じ得
る。例えば、癌の悪液質は、腫瘍を有する宿主の栄養摂取及び代謝の異常の結果
として発生する臨床的症候群であり、一般に進行した悪性のものである。この症
候群は、食欲不振、体重喪失、重大な組織消耗、無力症及び臓器機能不全を包含
する。悪性疾患を有する個体は、一般に、脂質代謝に異常を有する(体脂肪貯蔵
の加速された異化を伴う増大された脂質分解を含む)。グルコース投与の期間中
、正常な個
体は、脂質代謝を抑制し、優先的にグルコースを酸化する。しかしながら、癌を
有する個体は、内生の脂質代謝を抑制することができず、脂肪酸の持続的酸化を
生じる。特異的脂質の割合の相対的変化は、更なる身体の組成及び代謝の混乱に
寄与する。慣用の栄養サポートは、一般に、癌患者における罹病率を減じない。
ICAMレセプター(細胞間接着分子)とは、接着レセプターのICAMファミ
リーのメンバーである任意のレセプターを意味する。ICAMレセプターは、白
血球の接着を媒介する。ICAMレセプターには、例えば、ICAM−1、IC
AM−2及びICAM−3が含まれる。好適なICAMレセプターは、ICAM
−1である。ICAM−1は、例えば、白血球、内皮細胞、上皮細胞、肝細胞、
含脂肪細胞、筋細胞及び繊維芽細胞において発現される。ICAM−1は、炎症

β2インテグリンとは、任意のβ2白血球インテグリンを意味する。β2インテ
グリンは、例えば、Mac−1及びLFA−1を包含する。好適なβ2インテグ
リンは、Mac−1(αMβ2又はCD11b/CD18とも呼ばれる)である。M
ac−1は、例えば、内皮細胞への接着、ファゴサイトーシス及び好中球アポト
ーシスを含む幾つかの好中球機能に関与する。

抗体とICAMレセプター又はβ2インテグリンとの間で形成された反応複合
体の検出を、当業者に公知の標準的方法により達成することができる。例えば、
これらの抗体を、例えば放射性標識又は非放射性標識(例えば、蛍光標識)を用い
て標識することができる。

突然変異は、優性であっても劣性であってもよい。突然変異は、ホモ接合であっ
てもヘテロ接合であってもよい。
ICAMレセプター又はβ2インテグリン代謝の如何なる情況も評価すること
ができる。ある具体例において、代謝情況は、ICAMレセプターのβ2インテ
グリンへの結合である。任意の体重に関係するパラメーターを評価することがで
きる。ある具体例において、パラメーターは、脂質代謝(例えば、脂肪酸酸化の
調節)である。
薬剤は、例えば、任意の物質(例えば、体重獲得のための薬物又は体重喪失の
ための薬物)を包含することを意味する。この発明の薬剤は、好ましくは、IC
AMレセプターのβ2インテグリンの代謝情況を変えることができる。かかる変

テグリン及び/又は結合性分子の適当なコンホメーションの折り畳みの阻止又は
減少;ICAMレセプター又はβ2インテグリン及び/又は結合性分子の結合特
性の調節;ICAMレセプター又はβ2インテグリン及び/又は結合性分子を活
性化し又は不活性化するのに必要なシグナルの干渉;ICAMレセプター又はβ2
インテグリン及び/又は結合性分子の間違った時点での活性化又は不活性化;
又は他のレセプター、リガンド若しくはICAM−1若しくはMac−1及び/
若しくは結合性分子の正常の合成若しくは機能に必要な他の分子の干渉。この結
合性分子は、ICAM又はβ2インテグリンと同じ又は異なる細胞上にあってよ
い。
薬剤の例には、ICAMレセプター若しくはβ2インテグリンポリペプチド又

ICAMレセプター又はβ2インテグリンは、体重の異常の治療において脂肪
代謝を調節するための天然の又は人工のリガンドの探索を可能にする。ある具体
例において、この薬剤は、ICAMレセプター又はβ2インテグリンに対する天
然のリガンドである。ある具体例において、この薬剤は、ICAMレセプター又
はβ2インテグリンに対する人工のリガンドである。
アナログとは、天然のICAMレセプター又はβ2インテグリンと、アミノ酸
配列において又は配列に関係しない点において或はこれらの両方において異なっ
ている化合物を意味する。この発明のアナログは、一般に、天然のICAMレセ
プター又はβ2インテグリン配列の20アミノ酸残基の好ましくは40アミノ酸

り又は1つ以上の非保存的アミノ酸置換、欠失若しくは付加(これらは、ICA

この発明の範囲内の他のアナログには、例えば、ペプチド安定性を増大させる
改変を有するものが含まれる。かかるアナログは、例えば、ペプチド配列中に少
なくとも1つの非ペプチド結合(ペプチド結合に置き換わったもの)を含むことが
できる。例えば天然のL−アミノ酸以外の残基(例えば、D−アミノ酸又は非天

ICAMレセプター若しくはβ2インテグリン又は生物学的に活性なこれらの
断片若しくはアナログ、又は結合性分子又は生物学的に活性なその断片若しくは
アナログは、例えば、その同起源の分子と、相補的分子上の結合部位について競
争し、それにより、ICAMレセプター又はβ2インテグリンと結合性分子との
間の結合を減じ又は排除することができる。ICAMレセプター若しくはβ2イ
ンテグリン又は結合性分子を、例えば、天然のICAMレセプター若しくはβ2
インテグリン又は結合性分子の精製若しくは分泌により、組換えのICAMレセ

それ故、アナログ及び断片を生成する方法及びそれらを活性について試験する
方法は、当業者に公知である。
薬剤は又、アンチセンス分子として用いられる核酸であってもよい。アンチセ
ンス療法は、例えば、ICAMレセプター若しくはβ2インテグリンポリペプチ
ド又はその変異物をコードする細胞性mRNA及び/又はゲノムDNAと細胞条
件下で特異的にハイブリダイズ(例えば、結合)するオリゴヌクレオチド又はその
誘導体を、コードされる蛋白質の発現を例えば転写及び/又は翻訳を阻止するこ
とにより阻止するために、投与し又はイン・シトゥーで生成させることを包含す
ることを意味する。この結合は、慣用の塩基対の相補性によるものであってよく
、又は、例えば、二本鎖DNAへの結合の場合には、二重らせんの主溝内での特
異的相互作用によるものであってよい。
ある具体例においては、アンチセンス構築物が、ICAMレセプター又はβ2
インテグリン遺伝子の天然の配列(例えば、その遺伝子の発現に関与する配列)に
結合する。これらの配列には、例えば、開始コドン及び終止コドンが含まれる。
他の具体例においては、アンチセンス構築物は、野生型遺伝子中に存在しない
ヌクレオチド配列に結合する。例えは、アンチセンス構築物は、外因性の非野生
型配列の挿入を含むICAMレセプター又はβ2インテグリン遺伝子の一領域に

発現も阻害することなく、阻止するという利点を提供する。
本発明のアンチセンス構築物は、例えば、細胞中で転写されたときに、ICA
Mレセプター又はβ2インテグリンポリペプチドをコードする細胞性mRNAの
少なくとも一のユニーク部分に対して相補的であるRNAを生成する発現プラス
ミドとして送達することができる。或は、アンチセンス構築物は、エキス・ビボ
で生成されたオリゴヌクレオチドプローブであり、これは、細胞中に導入された
ときにICAMレセプター又はβ2インテグリン遺伝子のmRNA及び/又はゲ
ノム配列とハイブリダイズすることにより発現の阻止を引き起こす。かかるオリ

ンチセンスオリゴヌクレオチドの例には、ICAM−1メッセージのAUG翻訳
開始コドン又は3’非翻訳領域内の配列を標的とするホスホロチオエートアンチ
センスオリゴヌクレオチド(Chiang等,J.Biol.Chem.266:18162-18171(1991));
ICAM−1の3’非翻訳領域に対するアンチセンスオリゴヌクレオチド(Kumas
ka等,J.Clin.Invest.97:2362-2369(1996))が含まれる。
模倣物とは、形状及び/又は電荷分布において、ICAMレセプター若しくは
β2インテグリン又は結合性分子に似ている分子を意味する。この模倣物は、ペ
プチドであってもペプチド以外であってもよい。模倣物は、ICAMレセプター
又
はβ2インテグリンの結合性分子への結合を競争的に阻害するので、治療剤とし
て作用することができる。例えば、結合性分子への結合に関与する特定のICA
Mレセプター又はβ2インテグリンポリペプチドのアミノ酸残基をマップするた
めのスキャニング突然変異誘発例えばアラニンスキャニング突然変異誘発、リン

性分子への結合においてそれらの残基を真似、それ故、ICAMレセプター又は

抗体は、直接又は間接にICAMレセプター又はβ2インテグリンの代謝に影
響を与える任意の部位に対する抗体を包含することを意味する。これらの抗体は
、例えば、ICAMレセプター若しくはβ2インテグリン又は結合性分子、又は
これらのサブユニット若しくは断片に対するものであってよい。例えば、抗体は

薬剤は又、ICAMレセプター若しくはβ2インテグリン及び/又は結合性分
子の合成、翻訳後修飾、又は機能に必要な分子のインヒビター、又はICAMレ
セプター若しくはβ2インテグリン及び/又は結合性分子の合成又は機能を阻害
する分子のアクチベーターをも包含する。薬剤には、例えば、サイトカイン、成
長因
子、ホルモン、シグナリング構成要素、キナーゼ、ホスファターゼ、ホメオボッ
クス蛋白質、転写因子、翻訳因子及び翻訳後因子又は酵素が含まれる。薬剤は又
、例えばICAM−1若しくはMac−1及び/又は結合性分子を少なくとも部
分的に不活性化し又は破壊することのできる電離線、非電離線、超音波及び毒物
を包含することも意味する。
薬剤は又、完全にはICAMレセプター又はβ2インテグリン特異的でない薬
剤を包含することも意味する。例えば、薬剤は、他の脂質代謝関連遺伝子又は蛋
白質を変えることができる。かかる重複する特異性は、更なる治療上の利点を提
供することができる。
この発明は又、体重の調節において有用であるとして同定された薬剤をも包含
する。
この発明は又、薬剤を、動物の体重を調節する能力について評価する方法をも
包含する。薬剤を用意する。ICAMレセプター、ICAMレセプターの細胞外
部分、β2インテグリン又はβ2インテグリンの細胞外部分を用意する。この薬剤
を、ICAMレセプター、ICAMレセプターの細胞外部分、β2インテグリン
又はβ2インテグリンの細胞外部分と接触させる。その薬剤がICAMレセプタ

細胞外部分と相互作用するかどうかを測定する。もし相互作用が見出されれば、
この薬剤を、更に、試験用動物に治療上有効な量で投与する。試験用動物の体重
に対する薬剤のイン・ビボでの効果を評価する。
ある具体例において、相互作用が起きたかどうかを測定することは、薬剤と化
合物との間で結合が起きたかどうかを測定することを含む。ある具体例において
は、この試験的薬剤を試験用動物に2回以上投与する。
この発明は又、薬剤を、ICAMレセプター若しくはβ2インテグリン又はこ
れらの細胞外部分の結合性分子への結合の変化を測定することにより、動物の体
重を調節する能力について評価する方法をも包含する。薬剤を用意する。ICA
Mレセプター若しくはその細胞外部分又はβ2インテグリン若しくはその細胞外
部分を用意する。結合性分子又はその細胞外部分を用意する。この薬剤、ICA
Mレセプター若しくはその細胞外部分又はβ2インテグリン若しくはその細胞外

及び結合性分子若しくはその細胞外部分を合わせる。ICAMレセプター若しく
はその細胞外部分及び結合性分子若しくはその細胞外部分を含む複合体の形成、
又はβ2インテグリン若しくはその細胞外部分及び結合性分子若しくはその細胞
外部分を含む複合体の形成を検出する。この薬剤の存在時の複合体の形成におけ
るこの薬剤の不在時と比較しての変化は、ICAMレセプター若しくはその細胞
外部分又はβ2インテグリン若しくはその細胞外部分の結合性分子への結合を変
化させる薬剤を指示する。もし複合体の形成に変化が起きたならば、更に、試験
用動物を用意して、この薬剤を、更に、その試験用動物に治療上有効な量で投与
する。この薬剤の試験用動物の体重に対するイン・ビボでの効果を評価する。
結合を変化させることは、例えば、結合を阻害し又は促進することを包含する
。ある具体例において、結合性分子は、β2インテグリンである。ある具体例に
おいて、結合性分子は、ICAMレセプターである。薬剤の効力を、例えば、様
々な濃度の薬剤を用いて得られたデータから投与量応答曲線を生成することによ
り評価することができる。複合体の形成を測定する方法は、標準的であり、当業
者に公知である。

この薬剤を、この動物に、治療上有効な量で、体重に関係する病気の治療が起き
るように投与する。
体重に関係する病気は、例えば、肥満又は体重喪失であってよい。ある具体例
において、肥満は、糖尿病、高血圧又は高コレステロールレベルと関係している
。ある具体例において、体重喪失は、癌、AIDS、組織消耗、神経性食欲不振、慢
性感染症、胃腸病、インシュリン依存性真性糖尿病、甲状腺中毒症又は食物の吸
収不良により生じる。
治療は、例えば、体重に関係する病気の予防、治療、症状の軽減、又は治癒を
包含することを意味する。
薬剤は、例えば、ICAMレセプター若しくはβ2インテグリンポリペプチド
又は生物学的に活性なこれらの断片若しくはアナログ;ICAMレセプター若し
くはβ2インテグリンポリペプチド又は生物学的に活性なこれらの断片をコード
する核酸;ICAMレセプター若しくはβ2インテグリンの調節配列又は生物学
的に活性なこれらの断片をコードする核酸;ICAMレセプター又はβ2インテ
グリンの核酸に対する結合性分子(このICAMレセプター又はβ2インテグリン
の核酸は、例えば、ICAMレセプター又はβ2インテグリンの調節領域を含む
核酸であるか又はICAMレセプター若しくはβ2インテグリン又は生物学的に
活性なICAMレセプター若しくはβ2インテグリンの断片の構造領域を含む核
酸である);アンチセンス核酸;ICAMレセプター若しくはβ2インテグリン又
は結合性分子の模倣物;ICAMレセプター若しくはβ2インテグリン又は結合
性分子に対する抗体;代謝産物;又は阻害性の炭水化物若しくは糖蛋白質であっ
てよい。ある具体例においては、この薬剤は、アンタゴニスト、アゴニスト又は
スーパーアゴニストである。
肥満を生じるICAMレセプター又はβ2インテグリンの過少発現又は無発現
を含む体重異常に関して、直接又は間接に、更なるICAMレセプター又はβ2
インテグリンを加えることは、好ましい。例えば、ICAMレセプター又はβ2
インテグリンの追加は、ICAMレセプター又はβ2インテグリンポリペプチド
の追加により、又はICAMレセプター又はβ2インテグリン遺伝子の追加によ
り、又はICAMレセプター又はβ2インテグリン遺伝子のアップレギュレーシ
ョンにより、又はICAMレセプター又はβ2インテグリンのアゴニストの追加
により生じ得る。ある具体例において、ICAMレセプター又はβ2インテグリ
ン遺伝子を、一層高い発現を与えるように改変することができる。かかる改変は
、イン・ビトロ又はイン・ビボで、当業者に公知の標準的技術によってなされ得
る。
体重喪失異常を生じるICAMレセプター又はβ2インテグリンの過剰発現を
含む体重異常に関して、直接又は間接に、ICAMレセプター又はβ2インテグ
リンを阻害することは、好ましい。例えば、ICAMレセプター若しくはβ2イ
ンテグリンのインヒビター例えばICAMレセプター若しくはβ2インテグリン
に対するアンタゴニスト例えば抗体、ICAMレセプター若しくはβ2インテグ
リンポリペ
プチドの断片若しくはアナログ、ICAMレセプター若しくはβ2インテグリン
の模倣物、ICAMレセプター若しくはβ2インテグリンのアンチセンス分子、
又はICAMレセプター若しくはβ2インテグリンの発現(例えば過剰発現)を誘
導する分子(例えば、サイトカイン)のインヒビターの追加。例えば、ICAMレ
セプター又はβ2インテグリンのある断片又はアナログは、同起源の分子と、相
補的分子上の結合部位について競争し、それにより、ICAMレセプター又はβ2
インテグリンとそれらの結合性分子との間の結合を減じ又は排除することがで
きる。薬剤の投与は、その薬剤を標的細胞に到達させる任意の方法によって達成
することができる。これらの方法には、例えば、注射、付着、移植、座剤、経口
摂取、吸入、局所投与、又は薬剤の標的細胞への到達が得られる他の任意の投与
方法が含まれる。注射は、例えば、静脈注射、皮内注射、皮下注射、筋肉注射又
は腹腔内注射であってよい。移植は、移植可能な薬物送達システム例えばミクロ

テム例えば圧縮された、融合された若しくは部分融合されたペレットを挿入する
ことを含む。座剤は、グリセリン座剤を包含する。経口摂取投与は、腸溶性被覆
されていてよい。吸入は、吸入器内のエアゾールを伴う薬剤の投与(単独で又は
吸収し得るキャリアーに付着しての何れか)を包含する。
薬剤の投与は、単独でも、他の治療剤と組み合わせてもよい。ある具体例にお
いては、薬剤を適当なキャリアーと合わせ、リポソームに取り込ませ、又はポリ
マー放出システムに取り込ませることができる。
この発明のある具体例においては、幾らかの期間(例えば、数時間、数日、数
週間、数ヶ月又は数年間)にわたって薬剤に対する順次的露出を生じるように投
与をデザインすることができる。これは、上記の方法の1つによる薬剤の反復投
与により或は、制御された放出のための送達システム(薬剤を、反復投与するこ
となく、長期間にわたって動物に送達するシステム)により達成することができ
る。制御された放出のための送達システムとは、投与時に直ちにすべての薬剤の
放出が起きることはなく、幾らかの期間にわたって放出されるように遅延される
ことを意味する。放出は、一気に起きても、徐々に継続的に起きてもよい。かか
るシステム
の投与は、例えば、長期間作用する経口投与形態、ボーラス注射、経皮的パッチ
又は皮下インプラントによるものであってよい。
放出が一気に起きるシステムの例には、例えば、薬剤が、ポリマーマトリクス
に封入されたリポソームに捕捉されたシステム(これらのリポソームは、特異的
な刺激例えば温度、pH、光又は分解酵素に対して感受性である)及び、薬剤が
、マイクロカプセルコア分解酵素と共にイオン性被覆されたマイクロカプセルに
封入されたシステムが含まれる。薬剤の放出が徐々に継続的であるシステムの例
には、例えば、薬剤がマトリクス内にある形態で含まれるエアゾールシステム及
び、薬剤が例えばポリマーを通して制御された速度で浸透する拡散システムが含
まれる。かかる持続的放出システムは、例えば、ペレット又はカプセルの形態で
あってよい。
薬剤は、脂質に、例えば溶解形態又はコロイド形態で懸濁させることができる
。この液体は、溶剤、不完全溶剤又は非溶剤であってよい。多くの場合に、水又
は有機性液体を用いることができる。
薬剤は、体重に関係する病気の徴候が現れる前でも、現れた後でも投与するこ
とができる。ある具体例においては、この薬剤を、家族に体重に関係する病気の
病歴のある患者に、又は体重に関係する病気に対する疾病素質を示す表現型を有
する患者に、又は体重に関係する病気にかかりやすい遺伝子型を有すると診断さ
れた患者、又は体重に関係する病気に関係する病気を有する患者に投与する。
薬剤は、治療上有効な量で動物に投与する。治療上有効な量とは、体重に関係
する病気を少なくとも部分的に予防し又は逆行させることのできる量を意味する
。治療上有効な量は、個体ベースで決定することができ、少なくとも部分的には
、動物種、動物のサイズ、動物の年齢、使用する薬剤、用いる送達システムの型
、体重に関係する病気の徴候の開始からの投与のタイミング及び単一、多数又は
制御された放出投薬養生法を採用するかどうかの考察に基づくことになろう。治
療上有効な量は、かかる因子を用い、日常的実験を用いて、当業者の一人によっ
て決定することができる。
好ましくは、薬剤の濃度は、体重1kg当たり約0.1〜1000mgの、一
層好ましくは約0.1〜500mgの、尚一層好ましくは約0.1〜100mg
の、最も好ましくは約0.1〜5mgの投与量である。特異的濃度は、部分的に
、使用する特定の薬剤に依存する(幾つかのものは、他のものよりも効果的であ
るので)。現実に投与される薬剤の投薬濃度は、少なくとも部分的に、作用部位
で望まれる終濃度、投与方法、特定の薬剤の効力、特定の薬剤の寿命、及び体重
に関係する病気の徴候の開始からの投与のタイミングに依存する。好ましくは、
投薬形態は、実質的に動物に有害な影響を与えないようなものである。この投薬
は、当業者の一人により、かかる因子を用いて、日常的実験によって決定するこ
とができる。
ある具体例においては、種々の遺伝子構築物を遺伝子治療プロトコールの部分
として用いて、例えば、ICAMレセプター若しくはβ2インテグリンポリペプ
チド又はそれらの結合性分子のアゴニスト又はアンタゴニスト形態の何れかをコ
ードする核酸を送達することができる。発現ベクターを、非野生型のICAMレ
セプター又はβ2インテグリンが発現されている細胞において、ICAMレセプ
ター又はβ2インテグリンポリペプチドの機能を再構成し、或は排除するために
、特定の細胞型におけるICAMレセプター又はβ2インテグリンポリペプチド
のイン・ビボトランスフェクション及び発現に利用することができる。ICAM
レセプター又はβ2インテグリンポリペプチドの発現用構築物及びその変異物を
、任意の生物学的に有効なキャリアー例えばICAMレセプター又はβ2インテ
グリンの遺伝子を細胞にイン・ビボで有効に送達することのできる任意の配合物
又は組成物にて投与することができる。アプローチは、例えば、主題の遺伝子の
、ウイルスベクター(例えば、組換えのレトロウイルス、アデノウイルス、アデ
ノ随伴ウイルス及び1型単純ヘルペスウイルス、又は組換えの細菌又は真核生物
のプラスミド)中への挿入を包含する。ウイルスベクターは、細胞を直接トラン
スフェクトし;プラスミドDNAは、例えばカチオン性リポソーム(リポフェク
チン)の助けによって送達され又は誘導体化され得る(例えば、抗体と結合体化)(
ポリリジン結合体、グラミシジンS、人工ウイルスエンベロープ又は他のそのよ
うな細胞内キャリアー並びに遺伝子構築物の直接的注射又はイン・ビボ若しくは
イン・ビトロで行われるCaPO4沈殿)。上記の方法は当業者に公知であり、過
度の実験を必要とせずに実施することができる。適当な標的細胞の形質導入は遺
伝子治療の決定的な
第1のステップに相当するので、特定の遺伝子送達システムの選択は、意図され
る標的の表現型及び投与経路(例えば、局所投与か全身投与か)等の因子に依るこ
とになろう。投与は、当業者に公知の方法によって、治療上有効であるように、
一種以上の細胞型に向けることができ、又、ある細胞型内の1個以上の細胞に向
けることができる。好適具体例においては、薬剤を、動物の肝臓又は脂肪組織に
投与する。他の好適具体例においては、薬剤を、動物の幹細胞へ投与する。例え
ば、遺伝子工学処理したICAMレセプター又はβ2インテグリンを、ウイルス
ベクターによって、肝臓又は造血幹細胞に投与する。ある具体例においては、投
与を、出生前動物又は胎児の細胞において行う。ICAMレセプター又はβ2イ
ンテグリン発現のイン・ビボでの形質導入において用意された特定の遺伝子構築
物は、細胞のイン・ビトロでの形質導入(例えば、上記の診断アッセイにおける
使用)にも有用であるということは認められよう。
この発明は又、体重と関係する病気の危険にある動物の治療方法をも包含する
。体重と関係する病気の危険にある動物を用意する。ICAMレセプター又はβ2
インテグリンの構造又は代謝の状況を変えることのできる薬剤を用意する。こ
の薬剤を、この動物に、治療上有効な量で、この動物の治療が起きるように投与
する。体重に関係する病気の危険にあることは、例えば、体重に関係する病気の
家族の病歴、体重に関係する病気にかかりやすい表現型の徴候、体重に関係する
病気にかかりやすい遺伝子型、又は体重に関係する病気と関係する病気を有する
ことから生じ得る。
この発明は又、動物におけるICAMレセプター又はβ2インテグリンポリペ
プチドの上昇した又は減少したレベルと関連する体重に影響を与える病気の治療
を監視する方法をも包含する。ICAMレセプター又はβ2インテグリンポリペ
プチドの上昇した又は減少したレベルと関連する体重に影響を与える病気の治療
を受けている動物から種々の時点で得られた多数の生物学的試料におけるICA
Mレセプター又はβ2インテグリンポリペプチドのレベルを評価する。
この発明は又、治療上有効な量の薬剤(この薬剤は、動物における体重に関係
する病気の治療を生じるようにICAMレセプター又はβ2インテグリンの代謝
又は構造の状況を変えることができる)及び製薬上許容し得るキャリアーを含む
、動物
の体重に関係する病気を治療するための医薬組成物をも包含する。
ある具体例においては、この薬剤は、ICAMレセプター又はβ2インテグリ
ンのアンタゴニスト例えばICAMレセプター若しくはβ2インテグリンに対す
る抗体、ICAMレセプター若しくはβ2インテグリンの断片若しくはアナログ
、ICAMレセプター若しくはβ2インテグリンの小分子アンタゴニスト、IC
AMレセプター若しくはβ2インテグリンの模倣物、ICAMレセプター若しく
はβ2インテグリンに対するアンチセンス分子、又はICAMレセプター若しく
はβ2インテグリンに対する結合性分子である。
ある具体例においては、この薬剤は、ICAMレセプター又はβ2インテグリ
ンのアゴニスト又はスーパーアゴニストである。
この発明は又、動物の体重を調節するように、アンタゴニスト又はアゴニスト
活性を有するICAMレセプター又はβ2インテグリンポリペプチドの作製方法
をも包含する。ICAMレセプター又はβ2インテグリンポリペプチドを用意す
る。このポリペプチドのアミノ酸配列を変化させる。この変化させたポリペプチ
ドを、ICAMレセプター又はβ2インテグリンの代謝の状況に対する効果につ
いて試験する(ICAMレセプター又はβ2インテグリンの代謝状況の変化は、動
物の体重を調節するようにアンタゴニスト又はアゴニスト活性を有するICAM
レセプター又はβ2インテグリンを指示する)。これらのポリペプチドを生成させ
て、ICAMレセプター又はβ2インテグリンの代謝状況に対する効果について
、例えば、ここに記載したような当業者に公知の方法によって試験することがで
きる。ある具体例においては、変化させたポリペプチドを、更に動物において試
験して、この変化させたポリペプチドが動物の体重を調節するかどうかを測定す
ることができる。
この発明は又、動物の肝臓の脂肪含量を増大させる方法をも包含する。動物を
用意する。その動物から得られた可溶性ICAMレセプター又はβ2インテグリ
ンを、その動物の肝臓の脂肪含量を増大させるようにその動物に投与する。好適
具体例において、この動物は、ガチョウである。ある具体例においては、この動
物を遺伝子工学処理してICAM−1を欠乏させる。かかる方法は、食品産業に
おいて利用して、例えば、パテ用の脂肪の多いガチョウの肝臓を生成することが
で
きる。
この発明は又、動物により分泌されるミルク中の脂肪含量を増大させる方法を
も包含する。ミルクを分泌することのできる動物を用意する。ICAMレセプタ
ー又はβ2インテグリンの構造又は代謝の状況を変えることのできる薬剤を用意
する。この薬剤を、この動物に、その動物のミルクの脂肪含量を増大させるよう
な量で投与する。好ましくは、この動物は、家畜のミルク産出動物である。ある
具体例においては、この動物は、非ヒトトランスジェニック泌乳動物である。好
適な動物は、ヤギである。ある膜蛋白質は、分泌されたミルク中の脂肪塊のエン
ベロープ上に見出されるので、ミルク中の脂肪生成を増大させることにより、分
泌される脂肪上の膜蛋白質の量は増大される。それ故、かかる方法は、例えば、
望まれる膜蛋白質の動物当たりの収量を増大させるのに有用である。
下記の非制限的実施例は、本発明を更に説明するものである。
実施例 実施例1
:野生型及びICAM−1−/−マウスの正常なチャウ食餌(脂肪5%) での成長
この実施例は、16週齢後において、ICAM−1−/−マウスが、正常のチ
ャウ食餌(脂肪5%)で、野生型マウスよりも一層体重を獲得して、自発的に肥満
に至ったことを示す。
用いた動物は、C57BL/6バックグラウンドのICAM−1−/−マウス
であった。これらのマウスを、暗12時間、明12時間のサイクルに維持した。
水と食物は、自由に摂取させた。これらのマウスの食餌は、5.0%(wt/w
t)の脂肪、5%(wt/wt)の繊維及び22%(wt/wt)の蛋白質を含むマ
ウス用チャウ(Prolab3000:PMIFeeds,Inc.ミズーリ、St.Louis在)であった。
正常なチャウ食餌での野生型とICAM−1−/−マウスの成長曲線を測定し
た。図1を参照されたい。各グループには、13〜14匹の雄のC57BL/6
マウスが含まれた。データは、類似の結果を示す2つの独立した実験の結果を合
わせたものである。限定した数の雌のマウスを試験して、ICAM−1−/−と
野生型の間で同様の成長の差異が示された(12週齢では、ICAM−1−/−
で
23.57±0.64gであるのに対して、野生型では23.95±0.44g
であり、p=0.64;24週齢では、39.87±2.54g対33.02±
1.2gであり、p<0.05;45週齢では、54.10±6.20g対36
.70±1.20gであった、p<0.03。*p<0.004;**p=0.0
5)。
正常なチャウ食餌において、ICAM−1−/−マウスは、16週齢までは、
野生型動物に匹敵する体重を維持した。その後、ICAM−1−/−マウスが、
対照マウスよりも一層体重を獲得するということが認められた。他の遺伝的に肥
満のマウスと比較すると、ICAM−1−/−マウスの成長速度は、Tubbyマウ
スのそれと類似した(但し、Tubbyマウスの体重獲得は、もっと早く(9〜12週
齢で)起きる)。24週齢において、ICAM−1−/−及びTubbyマウスの平均
体重は、同様であった(Tubbyマウスで46gであるのに対してICAM−1−/
−の雌では44gであった;野生型対照は36g)。しかしながら、徐々に対照
よりも多くの食餌を食べたTubbyマウスと異なり、ICAM−1−/−及び対照
マウスによるチャウ食餌の消費は同等である(16〜24週齢において、2週間
当たり20〜25g)ことが見出された。
ICAM−1−/−マウスにおける過剰体重の殆どは、増大した脂肪パッドの
重量によるものであった(表2参照)。データは、平均値±平均値の標準誤差とし
て与えてある。統計的有意さを、スチューデントの試験により評価した。
各グループには、12〜14匹の雄の24週齢のC57BL/6マウスが含まれ
た。マウスには、標準的チャウ食餌を与えた。体重インデックス(BMI)を、体
重(g)を身長(肛門〜鼻の長さ、cm)の平方で除したものとして計算した。白色
脂肪パッドは、皮下の、鼠蹊部の、大網の及び後部腹膜の脂肪パッドを包含した
。茶色脂肪パッドは、肩甲骨間の部位から採取した。
白色脂肪における5.2gの過剰(ICAM−1−/−マウスにおいて8.8
1±0.47gであるのに対して野生型マウスでは3.66±0.36g、p<
0.0001)は、主として、皮下脂肪よりなった(54%)。肩甲骨間の茶色脂
肪パッドの重量も又、ICAM−1−/−マウスにおいて有意に増大していた。
茶色の脂肪組織は、機能的エネルギー費の重要な部位であり、肥満を防止するよ
うに機能する。それ故、肥満のICAM−1−/−マウスにおける茶色脂肪組織
の増大は、更なる白色脂肪沈着を防止する補償因子として、増大した白色脂肪組
織重量に従属して起き得るものであり、又はそれは、一層多くの脂質液胞の蓄積
による細胞の拡大を生じる茶色脂肪組織の機能障害を反映し得る。24週齢のI
CAM−1−/−マウスの腎臓、脾臓及び心臓の重量は、野生型マウスのものと
同様であった。しかしながら、ICAM−1−/−マウスは、少し重い肝臓を有
する傾向があった(1.82±0.13g対1.53±0.09g、p=0.0
6)。ヘマトキシリン染色し、オイル−レッド−Oで対比染色した肝臓切片は、
ICAM−1−/−マウスの血液細胞における異常な脂質沈着を示し、これは、
これらのマウスが脂肪肝を発症していることを示している。実施例2
:野生型及びICAM−1−/−マウスのウエスタン型食餌(脂肪21 %)での成長
この実施例は、21%の脂肪を含むウエスタン型食餌を与えた場合に、ICA
M−1−/−マウスが野生型マウスよりも一層多くの体重を急速に獲得し、従っ
て、食餌により誘導される肥満に感受性であることを説明する。
7週齢のICAM−1−/−マウス及び野生型対照用マウスに、21%の脂肪
を含むウエスタン型食餌を与えた。このウエスタン型食餌(Harlan Teklad Adjus
ted Calories Western-Type Diet No.88137,ウィスコンシン、Madison在)は、21%(
w
t/wt)の脂肪(42%のカロリー)、49.2%(wt/wt)の炭水化物及び
19.8%(wt/wt)の蛋白質を含んだ。図2を参照されたい。成長曲線(a
、b)及び2週間当たりの体重獲得(c、d)を、ウエスタン型食餌において、野
生型及びICAM−1−/−マウスについて測定した。a及びcは、雄マウスで
あり、b及びdは、雌マウスである。各グループの9及び11匹のC57BL/
6マウスに、7週齢において、ウエスタン型食餌を与え始めた。体重獲得(グラ
ム/マウス)を2週間の間隔で測定した。「0−2」は、食餌の開始から給餌の
2週目までの給餌期間をいい、以下同様である。雌のグループの実験を、繰り返
して、同様の結果を得た。aは、2〜18週(p<0.02)であり;bは、4〜
18週(p<0.01)であり;c及びdは、*p<0.05である。
雄及び雌の両方の変異型マウスは、対照よりも一層多くの体重を、急速に獲得
した(図2a及びb)。これは、ICAM−1−/−マウスが食餌に誘導される
肥満になりやすいことを示している。
雄と雌のICAM−1−/−マスの間には、それらのウエスタン型食餌により
誘導される体重獲得を調節する能力に差異があった。ウエスタン型食餌での6週
目以後に、ICAM−1−/−雄マウスは、もはや、対照より多くの体重を獲得
しなかった(図2c)。それ故に、雄のICAM−1−/−と野生型マウスとの間
の体重の最大の差異は、給餌の6週目までに達成された。対照的に、雌のICA
M−1−/−マウスは、ウエスタン型食餌において、14週目までに、対照より
も一層多くの体重を獲得した(図2d)。14週目において、雌のICAM−1−
/−マウスは、対照よりも、14.6gだけ重かったが;雄の場合には、この差
異は、7.6gでしかなかった。これらの結果は、体重の調節におけるICAM
−1依存性の機構が、雌において、雄におけるよりも、一層重要な役割を演じて
いることを示す。実験の最後に、雄における過剰な白色脂肪は、主として、皮下
脂肪(56%)よりなった。雌においては、主要な構成成分(64%)は、顕著な腹
部肥満を生じる腹腔内脂肪(大網+後部腹膜)であった(図3)。腹腔内脂肪は、他
の脂肪貯蔵物よりも、脂肪肝、高血糖症及びアテローム性動脈硬化症の一層高い
危険と一層よく相関する。かかる相関は、雌において認められる増大した肝臓重
量及び血漿グルコースを説明することができる。表3を参照されたい。 自発的な肥満のICAM−1−/−マウスと同様に、ウエスタン型食餌で、雄
と雌のICAM−1−/−マウスは、両方とも、対照よりも大きい茶色脂肪沈着
を有した(表3参照)。更に、雄のICAM−1−/−マウスは、対照より重い心
臓を得た(0.312±0.02g対0.249±0.02g、p<0.05)。
有意に多い体重と脂肪組織塊の獲得にもかかわらず、雄と雌のICAM−1−/
−マウスは、両方とも、図2に示した2週間の期間中に対照と同量のウエスタン
型食餌を消費した(約40g)。自発的かウエスタン型食餌により誘導されたかに
よらず、ICAM−1−/−マウスで認められた肥満は、調節されない食餌摂取
によるのではなかった。それ故、ICAM−1−/−マウスにおいて認められた
体重調節欠損が、マウスにおける食餌摂取を調節することが知られているレプチ
