発明の名称:治療用の修飾サイトカイン
本発明は治療用修飾サイトカインに関する。免疫機構は、異なる炎症的刺激、
外傷、ウイルス及びバクテリアの感染或いは癌のような細胞の退化のシグナル等
に応答してサイトカイン及び他の体液性因子を生産する。"リンフォカイン"、"
モノカイン"及び"サイトカイン"のような用語は元来、リンパ球、単球、及び非
リンパ球細胞から由来する生産物を区別するために作り出されたものであるが、
その後これらのカテゴリーの間に一種のオーバーラップがでてきた。それ故に、
用語"サイトカイン"は"リンフォカイン"、"モノカイン"の同意語として現在使用
されており、今後はこの認められた用語が使用される。当該分野で知られる殆ど
のサイトカインのリスト及びそれらの生物活性のリストが、Aggarwal B.B.及
びPocsik E.により報告されている。
異なるサイトカインが抗腫瘍、抗ウイルス、及び抗菌活性を示すことはよく知
られている。インビトロ及び動物モデルのインビボで観察されるその様な活性に
基づいて、いくつかのサイトカインが既に人の治療に使用されてきた(De Vita
ら、1995年、"癌の生物学的治療"Lippincott社、Phyladelphia)。例えば、イン
ターロイキン-2(IL-2)及びインターフェロンα(IFNα)のようなサイトカイン
は腎転移性癌、ヘアリー細胞白血病、カポシ肉腫、メラノーマ、多発性骨髄腫等
の異なるタイプの腫瘍を持つ患者に有効な抗腫瘍活性を示してきた。IFNβ、腫
瘍壊死因子(TNF)α、TNFβ、IL-1、4、6、12、15及びコロニー刺激因
子(CFSs)のような他のサイトカインがあるタイプの腫瘍に対しある抗腫瘍活性を
示してきた。従って、これらのサイトカインはさらに研究の対象になっている。
別のサイトカインは感染症の治療に使用されてきた(Aggarwal B.B.及びPo
csik E.)。
一般に、サイトカインの治療的投与はその全身毒性により非常に制限されてい
る。この事は人に治療的に有効な量を使用するときに重大な問題となるので、そ
の治療効果を維持しながらこのクラスの生物学的作動体の毒性を低くすることを
目的として、新しいサイトカイン誘導体や新しい治療方法を開発すべく多くの試
みがなされてきている。
腫瘍壊死因子(TNF)αは象徴的な例である。
TNFは主にマクロファージにより分泌され、ある腫瘍の出血的壊死を誘導する
能力に対し始めて発見されたサイトカインである(Carswellら、1975年)。その
後、TNFが、異なる腫痛系に細胞毒性的及び細胞静的効果を与える以外に、炎症
及び免疫応答の制御に重要な他のいくつかの生物学的効果を与えることができる
ことが示された(Beutler及びCerami、1989年;Fiers、1991年)。
TNFが、それが生産される生物に対し健康的或いは毒性的効果をその濃度、そ
の生産部位、及びその作用部位での持続時間の関数として与えることができると
いう考えが、現在信じられている。例えば、少量のTNFに慢性的にさらすと悪液
質が引き起こされる。一方、TNFの急性高生産は重篤な血管障害、ショック及び
死亡すら引き起こす(Beutler及びCerami、1989年)。
TNFの潜在的抗腫瘍活性は動物及び人で実施された種々の試験で評価されてき
た。その様な試験により、TNFによりインビボで引き起こされる抗腫瘍活性は、
内皮細胞への直接的効果により及び炎症と免疫応答の活性化により腫瘍の血管系
に損傷を誘導する能力にほとんど依存していることが示唆された(Sidhu及びBoll
on、1993年)。逆に、TNFの腫瘍細胞に対する直接的細胞毒性はあまり重要と考え
られなかった。
異なるタイプの腫瘍に対し実施されたTNFの臨床試験(第2相)は顕著な抗腫瘍活
性を示さなかったけれど、他の薬剤(IFγ、IL-2、アルキル化試薬、メルファラ
ン等)と併用してTNFを使用することにより元気づける結果が得られた。しかし
ながら、種々のタイプのTNF誘導毒性効果がその薬理学的有効量を使用すること
を強く限定していることが、一般的に特筆された(Spriggs及びYates、1992年)
。その代わり、全身的毒性効果を減らすように高容量のTNFを局部的な治療(例
えば、メラノーマ患者の脚に還流することにより)に用いることによりかなりの
成功を納めた(Lienardら、1992年)。
別の異なるTNFをベースとする治療法が、抗腫瘍活性を損なうことなしに最高
許容量の増加と全身的毒性効果の減少によりTNFの治療効果を増すことを目的と
して、現在評価中である。抗腫瘍効果を引き起こすのに必要な量を約一桁減らす
とよく許容できるようになる事が推定されてきた。
この事は以下のように試みられた。
a)TNFを腫瘍に届け、その局所的濃度を高めることができる融合蛋白の開発。
例えば、TNFと腫瘍特異的抗体を含む融合蛋白が製造された(Hoogenboomら
、
1991年);
b)抗腫瘍活性を維持し低い全身的毒性を持つTNF変異体の開発。それ故に、選
択的にひとつのリセプターのみを認識することができる変異体(p55または
p75)が既に調製された(Loetscher Hら、1993年)。
c)TNFの抗腫瘍活性を損なう事なしにその毒性作用を低下することができる抗
TNF抗体の使用。その様な抗体は既に報告されている(Rathienら、1992年)。
d)より高い半減期を持つTNF誘導体の使用(例えば、TNFとポリエチレングリコ
ールとのコンジュゲート)。
この様な方法の薬理学的潜在能力は現在評価中である。
得られたデータはTNFの抗腫瘍剤としての使用を推奨するけれども、その全身
的毒性の問題は現在まだ解決されていない。
ビオチン-アビジン相互作用を利用した常磁性または放射性同位体の細胞毒性
試薬のデリバー・システムが最近報告された(EP251494及びEP496074)。
特に、EP251494には診断もしくは治療剤を投与するためのシステムが記述され
ている。この診断若しくは治療剤は、アビジンまたはストレプトアビジンとコン
ジュゲートした抗体、コンジュゲートした抗体と複合体を作れる試薬及びビオチ
ンとコンジュゲートした診断または治療剤を含む化合物とを含み、抗体により認
識される標的細胞上でのビオチン−ストレプトアビジン相互作用により治療また
は診断剤の局在化を可能とするように、連続的にまたは適当に遅延されて投与さ
れる。
上述の治療または診断剤は、金属キレート、特に放射性核種と、シスプラスチ
ン、ドキソルビシン等のような低分子量抗腫瘍剤とのキレートを含む。実際、こ
のシステムは50000ダルトン以上の分子量の化合物には適当ではなく(好ましく
は10,000ダルトン以上ではない)、必然的にTNF(51000)のようなサイトカイン
には適用できないことが明らかに指摘されている。
EP496074には、ビオチン化抗体、アビジンまたはストレプトアビジン、及びビ
オチン化診断または治療剤の連続的な投与を提供する方法が記載されている。又
、このケースではリシン(細胞毒性鎖が約30000ダルトンの分子量を持つ蛋白質
)のような細胞毒性試薬が記述されているが、大部分は放射能標識化合物に関す
る適用が開示されている。
WO95/15979には標的細胞に高度に毒性の試薬(I)を局在化する方法が開示さ
れている。それは、リガンド(L)または抗リガンド(AL)とコンジュゲート
した特異的標的分子を含む第一コンジュゲート(C1)の投与と、引き続き抗リガ
ンド(AL)またはリガンド(L)に結合した毒性剤(I)を含む第二コンジュゲー
ト(C2)の投与に基づくものである。この場合、サイトカインは、L/ALに対する
アビジン/ビオチン系と同様にTNFを含む毒性剤(I)の例として挙げられるけれ
ども、サイトカインが使用されるときはリガンド(L)又は(AL)の投与はサイ
トカインを解離して"フリー"なサイトカインに生物効果を引き起こさせるために
特に好まれるということが推諭される。これは結合したサイトカインがそのリセ
プターとは反応できずさらに目的とする生物学的効果を有効に働かすことができ
ないという事実のためであろう。L又はALの必要量は細胞表面への結合に対し拮
抗するためには比較的高くなるものと思われるが、毒性の問題が出て来る可能性
がある。さらに、L/ALがビオチン/アビジンに対応する時、この相互作用が非常
に安定である事を考慮すると(既知の相互作用の中で最も強い非共有結合、Kd=
10-15M)、この結合は実際は解離不能である(D.Savageら、アビジン−ビオチ
ンの化学:ハンドブック、Pierce Biotech社(Rockford)。
WO95/15979にはクレームの方法にサイトカインの使用をサポートする特別な実
験データは報告されてなく、その方法をこのクラスの物質の局在化に適用するた
めに必要な再生可能で実用的な開示内容を提供するには不十分な一般的記述のみ
報告されている。このクラスの物質は単純な細胞毒性機構によってではなく、直
接的な細胞毒性効果を引き起こさずに抗腫瘍効果を示すことができる複雑な前炎
症的、免疫刺激的、血液凝固的、壊死的機構により作用する。他方、上述の明細
書EP-251494及びEP−496074の試薬を使用することが要求されている。
驚いた事には、WO95/15979に報告されている一般的指示とは反対に、コンジュ
44ゲートサイトカインと標的特異的コンジュゲート成分との相互作用がリガン
ド/
抗リガンドカップルのコンジュゲーションの場合のように直接的ではなく、むし
ろ標的特異的成分とサイトカインの間をブリッジとして結合できる第3の成分に
より仲介されるようなシステムによってのみサイトカインが生物学的に活性な形
で有効に局在化することができることが見出された。
本発明の方法により、サイトカインは結合した形でも機能することができ、リ
ガンド/抗リガンドカップルのメンバーの投与はサイトカインを活性な形にする
のに必要ではない。リガンドにコンジュゲートしたサイトカインは細胞表面上の
膜リセプターと相互作用できる一方、適当なリガンドとコンジュゲートした標的
特異成分との相互作用に含まれる対応する抗リガンドと相互作用することは予期
できなかったので、上記のような結果は思いがけない事と考えることができる。
この様に本発明は連続的な治療剤の使用に対し組み合わせた調剤の形で薬学組
成物を提供するものである。この薬学的組成物は;
a)少なくとも3成分からなるリガンド/抗リガンド/リガンドのシステムのリガ
ンドとコンジュゲートしている抗病理的標的化合物;
b)化合物a)のリガンドと相補的な抗リガンド;
c)抗リガンドb)と相補的なリガンドにコンジュゲートしたサイトカイン。但
し、リガンド/抗リガンド/リガンドの相互作用が、サイトカインとその天然
のリセプターとの間の親和力より少なくとも一桁高い親和力により特徴づけ
られている。
本発明により、化合物a)とサイトカインにコンジュゲートできる化合物の例
としては、ビオチンやヂゴキシケニンのようなハプテンが挙げられる。一方、"
抗リガンド"化合物の例としては抗ハプテン抗体(例えば、抗ビオチン抗体及び
抗−ジゴキシゲニン抗体)または、ビオチンがリガンドとして使用されるときは
、アビジン及びそのアナローグ(例えばストレプトアビジン、ニュートロアビジ
ン)が挙げられる。
好ましくは、化合物a)とサイトカインの両方がビオチンにコンジュゲートさ
れ、一方アビジン(またはその類似化合物)が抗リガンドとして使用される。サ
イトカイン及び抗体のコンジュゲーションに使用される技術は広く知られていて
化学的または遺伝子工学的方法諭により結合されることができる。
抗病理学的標的化合物a)は好ましくは、全体または部分的抗体またはモノク
ローナル抗体である。上述の抗体はすでに報告されており、広く、特に腫瘍抗原
に対する抗体の場合に使用されている。
本発明により使用することのできるサイトカインまたはリンフォカインの例と
しては、腫瘍壊死因子、インターフェロン、インターロイキン、コロニー刺激因
子(CSF)などが挙げられる。リポポリサッカライド(LPS)又はその誘導体
のような内在性サイトカインの局所的放出を亢進できる生物学的応答調節剤を使
用して局所的効果を高め、全身的効果を下げることも可能である。
さらに、サイトカインは時々相加的または相乗的効果を示すことができる事が
知られているので、本発明の基礎となっている方法は、全身レベルでの効果をよ
り少なくして相乗的局所効果を得るために実施することができる。癌の生物学的
治療及び異なるサイトカインの組み合わせの効果は広く記述されており、De Vit
aら(1995年、癌の生物学的治療、Lippincott社、Phyladelphia)によりよく要約
されている。
サイトカインの中でTNFの使用が特に好まれる。
好ましくは、コンジュゲート相互の相互作用または人工リセプターとの相互作
用は、サイトカインの膜リセプターの親和定数よりも少なくとも一桁高い親和定
数と、サイトカインが天然のリセプターと相互作用するときよりも少なくとも一
桁低い速度諭的解離定数により特徴づけられるであろう。
抗標的化合物またはコンジュゲート抗体は局所的に投与することができ、ある
いは血流に注入することができて、過剰の循環化合物または抗体が身体から除か
れるまでインビボで抗原または認識細胞構造と反応することができ、一方有意な
部分が病理学的標的に結合したままである。この時点で抗リガンドb)が投与さ
れ、引き続きコンジュゲートサイトカインが抗体または抗標的化合物との結合が
形成されることができる濃度で投与される。このようにしてサイトカインが標的
細胞に固定され長時間持続することが可能となる。
修飾サイトカインの"人工"リセプター(抗リガンドにより表される)からの解離
時間は、"天然"リセプターからの解離時間と比べて少なくとも一桁長いならば、
このことは達成できる。本発明の応用に関する実施例が実証しているように、腫
瘍抗原特異的ビオチン化抗体、ニュートラビジン及びビオチン−TNF(bio-TNF)
を含むシステムにより本発明は実施できる。このシステムでは、10-15Mに等し
いbio-TNFと人工リセプター(アビジン)との間の親和力、及び10-9Mから10-10
Mに等しいbio-TNFとその天然リセプター(TNF-R1及びTNF-R2)との間の親和力は
、人工的リセプターが提示される場所(腫瘍)でのTNFの固定と長い持続が好ま
れるようになっている。
本発明の好ましい実施形態では、腫瘍抗原に特異的なビオチン化モノクローナ
ル抗体が、病変内または病変側投与、腔内(例えば、膀胱、腹腔)投与、動脈(
肝臓、中枢神経)投与により、または全身レベル、局所または局部での血管還流
(脚の還流液、肝臓、胸部)により腫瘍と接触させられ、引き続きニュートラビ
ジンまたはストレプトアビジン及びTNF−ビオチンのアナログ的連続投与が行
われる。
好ましくは、数時間のインキュベーション時間が各投与の間に挿入され、血管
系が過剰の試薬を除く事が可能となり、それ故に腫瘍標的へのより優れた局在が
得られる。
さらに、アミノ末端領域(1−11残基、VRSSSRTPSDK配列)でビオ
チン化したTNFの利用が好ましい。それにより、TNFとその天然リセプター
との多価結合が妨げられず、その作用が不活性化されない。或いは、既知の技術
で容易にビオチン化できる基を持つアミノ酸(リジン、システイン、チロシン、
ヒスチジン等、Savageらによるアビジン−ビオチン化学:ハンドブック、Pierce
Biotec社を参照)が同領域に遺伝子工学的手法により挿入できる。また。グリコ
シル化シグナルを挿入して、その結果炭水化物残基の特異的ビオチン化が、例え
ばビオチン−ヒドラジドまたはその誘導体を用い得られる。アミノ末端領域での
TNFのビオチン化は、TNFと直接にビオチン化される蛋白質フラグメントとのコン
ジュゲートを発現系により構築する遺伝子工学的手法により同様に容易に得る事
ができる。その様な蛋白質の例としては、E.Coli由来のアセチルCoAカルボキシ
ラーゼ及びビオチン化部位を持つそのC端側領域により代表される。
アルファーアミノ基のビオチン化は、ビオチン−TNFの構造を非ビオチン化TNF
の構造に最も近い形に維持するために好まれる。このことは、ビオチン-6-アミ
ノカプロイル-N-ヒドロキシサクシミドエステルの5.5から7.5の間のpHでの反
応に基づいたビオチン化のプロトコールを用いて達成できる。或いは、アビジン
または膜蛋白質との相互作用の能力が、例えば上述のひとつの方法によりビオチ
ン化したTNF由来のサブユニットと非ビオチン化TNF由来のサブユニットとを混合
することにより維持されるようにビオチン化TNFを得る事ができることが見出さ
れた。
好ましくは、4℃で1:3の割合のビオチン化及び非ビオチン化TNFの混合物を
24から72時間インキュベートして、混合反応が実施される。N-末端でコンジュゲ
ートしたこの様な形のTNFは新しく、さらに程度の進んだ発明の局面を表す。
本発明の組成物を商品化してルーティンに利用できるように、適当な治療物質
を含むキットを調製することができる。好ましくは、本発明によるキットは;
− 5から10mgのビオチン化抗体を含むヴァイアル;
− 5から100mgのアビジン、ストレプトアビジンまたはニュートラビジンを含
むヴァイアル;
− 5から10mgのビオチン化TNFを含むヴァイアル(または他のビオチン化サイ
トカイン)
を含む。
本発明の方法は既知の方法、例えばサイトカインと抗体の直接的コンジュゲー
ションから由来する利点と比べて幾つかの利点を提供する;
1)例えば、必ずしも高い抗体親和力が要求されない。というのは、比較的高い
量の抗体とアビジンが投与され、引き続き相対的に低い量のビオチン-TNFが
投与され、とにかく、腫瘍が効果的にラベルされる事が可能であるからであ
る。他方、抗体の量が常にTNF-量にリンクしている従来の抗体−TNFコンジ
ュゲートの場合はこのことは不可能である。
2)分離した抗体とTNFが腫瘍組織に分散していく可能性は、分子量が低いため
に抗体-TNFコンジュゲートで得られるものより良い。
3)アビジン及びストレプトアビジン(10-15M)のような高い親和力の分子で
腫瘍をラベルすることは、人工リセプター(アビジン)との相互作用が熱力
学的及び速度諭的に好まれ、さらに全身レベル(10-9Mから10-10M)での天
然
リセプターとの相互作用が妨げられるように、ビオチン-TNFの量を選択する
ことが可能である。抗体−TNFコンジュゲートの場合は、10-15Mのオーダー
の親和力で腫瘍抗原に結合できる抗体を得ることは極端に困難である。
本発明は以下の実施例においてさらに説明される。実施例は特に腫瘍病理の治
療に言及しているけれども、同じ方法が、サイトカインによる局所治療が使用で
きる各々別の病理の治療に対しても採用できることは容易に理解できる。
実施例1
この実施例は次のシステムに基づく本発明の応用を示す。

実施例1.1
ビオチン化19E12mAb抗体(bio-19E12)の調製
1000μlの19E12mAbの溶液(1mg/ml、pH8.5の重曹水中)と34μlの硫黄-NHS-L
C-ビオチン溶液(1mg/ml、モル比はmAb/ビオチン:1/24)がピペットでEppendor
fチューブに分注された。混合物は室温(23/24℃)で30分間インキュベートされ
た。インキュベーション後、混合物は4℃で2リッターのPBSに対し終夜透析され
、4℃に維持された。
実施例1.2
ビオチン-TNF(bio-TNF)の調製
この章ではビオチン-TNFコンジュゲートの調製に使用されるいくつかのプロト
コールの実施例が報告される。
実施例1.2.1(bio-TNF cod.#Bの調製)
60μlの2回蒸留水中TNF溶液(0.5mg/ml)、6μlの1M炭酸ソーダ水pH6.8、6
μlのAH-BNHS、DMSO中3mg/ml(モル比TNF/ビオチン:1/66)がピペットでEppend
orfチューブに分注された。混合物は室温(23/24℃)で3時間インキュベートさ
れ、7.5μlの1Mリジン溶液と混合された。さらに1時間室温でインキュベート
した後、240μlのRPMI溶液(10%FCS、2mMグルタミン、100IU/mlペニシリ
ン、100μg/mlストレプトマイシン、250ng/mlアンフォテリシンB)が混合物に
加えられた。混合物は次に4℃で2リッターの0.9%NaClに対し終夜透析され(3回
交換)、−20℃に維持された。
実施例1.2.2(bio-TNF cod.#C、cod.#D、cod.#E、cod.#F、cod.#Gの
調製)
bio-TNF cod.#C生成物は、pH7.8のコンジュゲーションバッファーの使用を
除き実施例1.1.1により調製される。
bio-TNF(cod.#D、cod.#E、cod.#F、cod.#G)の比較溶液は異なるインキ
ュベーションバッファーを用い、種々のTNF/ビオチンのモル比で調製される(表
1参照)。
実施例1.3
ビオチン化TNFのRMA Thy1.1C1.2細胞に対する生物活性の測定
この実験は、RMA Thy1.1C1.2細胞に対する細胞毒性テストにより、異なるコン
ジュケーションの条件(実施例1.1参照)で得られた異なるbio-TNF調製物の特異
的生物活性の測定を目指している。方法:
96穴平底プレート(Costar 3595)の各穴に以下の細胞、物質が添加される;
a)50μlの60000RMA Thy1.1C1.2細胞(マイコプラズマ陰性)のRPMIサスペ
ンジョン溶液、但し5%FCS、2mMグルタミン、100IU/mlペニシリ
ン、100μg/mlのストレプトマイシン、250ng/mlのアンフォテリシンB、50n
M 2-メルカプトエタノール、500μg/mlのG418を含む(完全−RPMI);
b)50μlの標準TNF溶液または50μlの望ましい濃度のサンプルの完全RP
MI溶液;
c)10μlのアクチノマイシンDの完全RPMI溶液(330ng/ml)。
それからプレートが5%CO2下37℃で24時間インキュベートされた。さらに、10
μlのチアゾリルブルー溶液(MTT)(PBS中5mg/ml)が各穴に加えられた
。5%CO2下37℃でさらに4時間インキュベーションした後、100μlのライシス溶
液(33%(v/v)N,N-ジメチルフォルムアミド、20%(p/v)ドデシル硫酸
ナトリウムの水溶液で酢酸によりpH4.7に調整される)が各穴に添加された。溶
液は多チャンネルピペットを用い穴で混合され、37℃で24時間インキュベートさ
れた。次に、各穴の吸収は570と650nm(対照)で多チャンネルマイクロプレー
トリーダーにより読み取られた。
細胞毒性活性は、非ビオチン化TNFを用いて得られる検量線に吸収を内挿する
ことにより計算された。結果 a)非ビオチン化TNF(cod.#A)を用いて得られる検量線を用いて測定された。
実施例1.4
抗体とニュートラビジンで前処理された細胞によるbio-TNF及びTNFの結合、及
び会合と解離の速度の比較
この実験は、ビオチン化抗体とニュートラビジン前標的システムにより腫瘍細
胞上に人工リセプターを構築すると、天然リセプターに結合できる最大値と比較
して細胞に結合できるbio-TNFの全量を顕著に増加する事を実証する事を目的と
している。
さらに、この実験はbio-TNFと、抗体及びニュートラビジンで前処理した細胞
の天然及び人工リセプター(ニュートラビジン)との会合及び解離時間を評価す
ることを目的としている。方法
実験は、実施例1.5及び1.6に記述されているようにMRA thy1.1C1.2細胞を処理
して実施された。全結合TNFは、抗−TNFウサギポリクローナル抗体とウサギ蛍光
ヤギ抗−IgG抗体を使用する事に基づく間接法を使用し、FACS解析により検出さ
れた。会合速度諭
50μlのPBS/FCS 2%中50,000個の細胞が丸底プレートの穴にシードされ、1μ
lのbio-19E12 mAb(PBS/FCS 2%中0.5mg/ml、抗体の最終濃度10μg/ml)と混合
され、氷上で10分間インキュベートされた。
細胞は2回200μl/穴のPBS/FCS 2%の添加と1300rpm、2分間の遠心で洗浄さ
れれた。細胞をボルテックスにより再びサスペンドし、50μl/穴の2%PBS/FCS
と1μlの2.5mg/mlニュートラビジン(PBS/FCS 2%中、ニュートラビジンの最
終濃度50mg/ml)とを混合した。氷上で10分インキュベートした後、細胞を再
び上述したように2%PBS/FCSで2回洗浄した。
次に50μlのPBS/FCS2%、1μlの22.2μg/mlの濃度のTNFまたはbio-TNF(TNF
及びbio-TNFの最終濃度は450ng/ml)が各穴に添加され、サンプルが氷上で1時
間インキュベートされた。
2%PBS/FCSでさらに洗浄した後、50μlのウサギ抗TNFポリクローナル血清(P
BS/FCS 2%中1:1000、Genzyme社、cod.#IP300)が各穴に添加され、氷上で10
分間インキュベートされた。
細胞を2%PBS/FCSの添加(200μl/穴)と遠心により2回洗浄した。その後、蛍
光性色素でコンジュゲートした50μlのヤギ抗ウサギ免疫グロブリン抗血清
(PBS/FCS2%中、ヤギ抗ウサギ-FITC)が各穴に添加され、氷上で10分間インキ
ュベートされた。
2%PBS/FCSで最終洗浄した後、サンプルを200μlの2%PBS/FCSに再サスペンド
し、FACSで解析した。解離速度論
50μlの2%PBS/FCS中100,000個の細胞が丸底プレートの8個の穴にシードされ
、1μlのbio19E12 mAb(2%PBS/FCS中0.5mg/ml,抗体の最終濃度10mg/ml)と混合さ
れ、氷上で10分間インキュベートされた。細胞を2%PBS/FCSの添加(200μl/穴
)と1300rpmの2分間の遠心により2回洗浄した。細胞をボルテックスにより再サ
スペンドし、50μl/穴の2%PBS/FCSと1μlのニュートラビジン(2%PBS/FCS中
2.5mg/ml最終ニュートラビジン濃度50μg/ml)とを混合した。氷上で10分間イ
ンキュベートした後、細胞を上述したように再び2%PBS/FCSで2回洗浄した。
次に、50μlの2%PBS/FCSと1μlのTNFまたはbio-TNF(濃度22.2μg/ml、最
終TNF及びbio-TNF濃度450ng/ml)が各穴に添加され、サンプルが氷上で1時間
インキュベートされた。
8個のサンプルが遠心により2回、200μlの2%PBS/FCSで洗浄された。細胞を5
0μlのRPMI(5%FCS、2mMグルタミン、100 IU/mlペニシリン、100μg/m
lストレプトマイシン、250ng/mlのアンフォテリシンB)に再サスペンドし、
次に0.25%パラホルムアルデヒドで4℃、1時間固定した。
2%PBS/FCSでさらに洗浄した後、50μlのウサギ抗TNFポリクローナル血清(P
BS/FCS2%中1:1000、Genzyme社、cod.#IP300)が各穴に添加され、氷上で10分
間インキュベートされた。
細胞を再び2%PBS/FCSの添加(200μl/穴)と遠心により2回洗浄した。その
後、蛍光色素でコンジュゲートした50μlのヤギ抗ウサギ免疫グロブリン抗血清
(2%PBS/FCS中1:12000、ヤギ抗ウサギFITC)が添加され、氷上で10分間インキ
ュベートされた。
2%PBS/FCSで最終洗浄した後、サンプルを200μlの2%PBS/FCSに再サスペンド
し、FACSで解析した。結果
ニュートラビジンを予め細胞に対し標的させておくと、単なる天然レセプター
で得られる結合(ニュートラビジンなしにまたは非ビオチン化TNFを用いて測定
)と比べて、細胞へのbio-TNFの結合を少なくとも10から20倍増加させる。最良
の増加はbio-TNF cod.#B(pH6.8でビオチン化)で観察された(データは示さ
れない)。
さらに、図1(下の図)に示されているように、抗体とアビジンで前処理され
たRMA細胞上へのbio-TNFの存続時間は、同じ細胞へのTNFの存続時間(t1/2=0.
22h)よりも約30倍高い(t1/2=7h)。
この結合結果は、本発明による抗体とニュートラビジンを予め標的化させる方
法の開発により腫瘍細胞に結合するbio-TNFの最大量が10から20倍増加し約30倍
長く細胞膜上に存続できることを実証するものと結論できる。
実施例1.5 インビトロでアクチノマイシンDの存在下ビオチン化抗体とニュートラビジン
により前処理された腫瘍細胞に対するTNFとbio-TNFの細胞毒性の比較
この実施例は、ビオチン化抗体/アビジン系がビオチン-TNFを活性形で細胞表
面に届け、単純に天然リセプターを用いて得られるものと比べて、インビトロで
細胞毒性を高めることが出来る事を実証するためのものである。
この目的のために、RMAThy1.1C1.2細胞、ヒトモノクローナル抗Thy1抗体、ビ
オチン化(mAb bio-19E12)、ニュートラビジン及びbio-TNF cod.#Bが使用され
た。
実験はa)mAb bio-19E12とb)ニュートラビジン、c)bio-TNFにより細胞
を連続的にインキュベーション、洗浄を行うことにより実施された。
天然リセプターへのTNFの結合がMnスーパーオキシドディスムターゼのよう
な保護因子の合成を誘導することが知られているので、その様な因子の生産を阻
害することを目的として、さらにTNFの細胞毒性活性を高めることを目的として
、転写阻害剤(アクチノマイシンD)の存在下細胞毒性テストが実施された。方法
50μlの2%PBS/FCS中1,000,000個の細胞と1μlのmAb bio-19E12の溶液
(0.5mg/ml)が96穴丸底プレートの各穴に添加された(Costar3595)。サスペ
ンションは氷上で10分間インキュベートされた。細胞は200μl/穴の2%PBS/FC
Sの添加と1,300rpmの2分間遠心により2回洗浄された。細胞をボルテックスによ
り再サスペンドし、2%PBS/FCS(50μl/穴)と1μlの0.5mg/mlニュートラビジ
ン(2%PBS/FCS中)とを混合した。氷上で10分間インキュベートした後、細胞
を上述したように再度2%PBS/FCSで2回洗浄した。
それから50μlの2%PBS/FCS、1μlの望ましい濃度のTNFまたはbio-TNFが各
穴に添加され、氷上で15分間インキュベートされた。さらに、2%PBS/FCSで洗浄
した後、各穴の細胞を1.5mlのRPMI(5%FCS、2mMグルタミン、100 IU/mlペ
ニシリン、100μg/mlストレプトマイシン、250ng/mlのアンフォテリシンB、
50nMの2-メルカプトエタノール、500μg/mlのG418)(完全RPMI)に再サ
スペンドした。
次に細胞を96穴の平底プレート(60,000細胞数/100μl/穴)にシードし、330
ng/mlのアクチノマイシンDの溶液(完全RPMI中、10μl/穴)と混合し、5
%CO2下37℃で24時間インキュベートした。
細胞の生存率は実施例1.3に記述されているように測定された。結果
アクチノマイシンDの存在下実施された実験の結果は図2に示される。ニュー
トラビジンの非存在下50%の細胞を殺すのに必要なTNFまたはbio-TNFの量(LD
50)(吸収を50%低下することから測定できる)は100,000U/mlよりも高いけれ
ども、ニュートラビジンの存在下bio-TNFのLD50は約2000−3000U/mlである事
が分かる。さらに、TNFの細胞毒性はニュートラビジンの存在または非存在に
より影響されないけれど、bio-TNFの細胞毒性は予期されたようにニュートラビ
ジンの存在に強く依存している。この事は、a)TNF及びbio-TNFがニュートラビ
ジンの非存在下RMA細胞に弱い毒性を示すこと、b)RMA細胞膜上の人工ニ
ュートラビジンリセプターの存在はbio-TNFの細胞毒性を非常に高めること、c
)抗体-ビオチン-アビジンの橋かけにより膜に結合するbio-TNFはそれ自身の天
然のリセプターと相互作用して細胞毒性効果を起こすことができること、等を示
す。
アクチノマイシンDの非存在下実施される実験では、TNF及びbio-TNFの両方が
全然細胞毒性活性を示さなかった(データを示さない)。
実施例1.6
アクチノマイシンDの非存在化、ビオチン化抗体、ニュートラビジン及びTNFま
たはbio-TNFで前処理したRMA Thy1.1C1.2細胞のインビボ腫瘍化能(マウス)の
比較
この実験は、抗体-ビオチン-ニュートラビジン系(人工リセプター)により細
胞表面に結合するbio-TNFで前処理した細胞のインビボ腫瘍化性が、天然リセプ
ターに排他的に結合するTNFまたはbio-TNFで前処理した細胞に比べて、減少する
ことを実証するためのものである。
採用されたモデルはインビトロで前処理したRMA Thy1.1C1.2細胞の皮下投与投
与によるものである。この実験は4群のメスC57BL6マウス(各群5匹)を用いて実
施され、異なる日付で腫瘍の直径を測定して実施された。マウスは表2に示すよ
うに前処理細胞で処理された。
本研究で使用されたTNF及びbio-TNFの量(それぞれ50,OOOU/ml及び10,000U/ml
)は、実施例1.5(図2)に実証されるようにアクチノマイシンDの存在下のみに
部分的細胞毒性効果が決定されるようになっていることを思い出すのは重要であ
る。本実験では、細胞は完全にアクチノマイシンが存在しない状態、即ちTNF及
びbio-TNFの両方が非細胞毒性的である条件下で処理された。
方法
50μlの2%PBS/FCS中の180,000個の細胞が丸底プレートの4個の穴にシードさ
れ、1μlのbio-19E12 mAb(2%PBS/FCS中0.5mg/ml)と混合され、氷上で10分間
インキュベートされた。
細胞は2回200μl/穴のPBS/FCS2%と1300rpm2分間遠心で洗浄された。細胞を
ボルテックスにより再びサスペンドし、50μl/穴の2%PBS/FCSと1μlの0.5m
g/mlニュートラビジン(PBS/FCS2%中)とを混合した。氷上で10分間インキュベ
ートした後、細胞を再び上述したように2%PBS/FCSで2回洗浄した。
次に50μlの2%PBS/FCS、1μlのTNFまたはbio-TNF(それぞれの濃度は50,00
0及び10,000U/ml)が各穴に添加され、氷上で15分間インキュベートされた。
さらに2%PBS/FCSで洗浄した後、各穴の細胞が200μlのPBSに再サスペンドさ
れ、Eppendorfチューブに移され、1.2mlのPBSと混合された。インシュリン用の
シリンジを用い、30,000個の細胞/200μl/マウスがそけい部レベルで皮下投与
された。腫瘍の平均直径(横と縦)がカリブレを用いて異なる時間に測定された
。結果
結果を表3に示す。bio-19E12/ニュートラビジン/bio-TNF mAb(第4群)によ
る細胞の処理は、bio-TNF無しの同じ方法で処理したマウス(第2群)の場合と比
べて、マウスのRMA Thy1.1C1.2細胞の腫瘍化能を低下した事が分かる。bio-19E1
2/ニュートラビジン/TNF mAbによる前処理(第3群)は対照群(第2群)と比べ
て腫瘍化能の有意な低下を示さなかった。10,000の代わりに50,000U/mlのbio-TN
Fを用いてさらに実験してみると、17日後の腫瘍の体積は検出限界(15mm3)以下
であった(データは示されない)。
これらの結果は、抗体/ビオチン/ニュートラビジン"橋かけ"により細胞膜に結
合したbio-TNFはRMA Thy1.1C1.2細胞のインビボ腫瘍化能が低下するという効果
を生む事ができる事を示す。
さらに、RMA細胞の処理に使用されるbio-TNFの量はアクチノマイシンDの存在
下(実施例1.5を参照)即ちTNFが直接細胞毒性効果を起こすことができない条件
下で細胞の溶解を決める程であったけれど、やはりこの処理は投与17から19日後
腫瘍の体積を5倍以上低下した。このことは、腫瘍化の低下が腫瘍細胞に対する
宿主の間接的効果(炎症、免疫効果等)による事を示す。a)縦方向と横方向の直径の平均実施例2

された。プレートをPBSで再び3回洗浄した。
100μl/穴のサンプルを加え、37℃で1時間インキュベートした。
プレートをPBSで3回洗浄した。100μlの0.5%PBS/BSA/0.05%Tween中のストレ
プトアビジン-HRP 1:2000が各穴に添加された。37℃で1時間インキュベートし
た後、プレートを3回0.05%PBS-Tweenで洗浄した。100μlのOPD溶液を各穴に加
えた。

mAb(第3群)による細胞の処理は、bio-LPS無し(第1群)またはLPSを用いる
(第2群)以外は同じ方法で処理したマウスの場合と比べて、マウスのRMA Thy1.
1C1.2細胞の腫瘍化能を低下した事が分かる。抗体-ビオチン/ニュートラビジン
橋かけにより細胞膜に結合したbio-LPSが、RMA Thy1.1C1.2細胞の腫瘍化能がイ
ンビボで低下するような効果を起こす事をこの結果は示す。
RMA Thy1.1非処理新鮮細胞が、前処理により引き起こされた免疫応答を確かめ
るために、30日後生存した2匹のマウスの他方の脇腹に注入された。表5に示すよ
うに、2匹のマウスは2回目の注入後もまた生き残った。注入した細胞は処理され
ていなかったので、これらの結果は一回めの治療が免疫記憶を誘導したことを証
明する。a)2回目の投与:非処理新鮮RMA Thy1.1細胞が2匹の生存マウスに投与された。
