Claims (1)
- 【特許請求の範囲】 1.反応チャンバー内で流体密閉の手段で(in a fluid-tight manner)温度 の上昇した反応(elevated temperature reactions)を実施するアッセイシステ ムであって、 (a)該反応チャンバーを包含する第一のアセンブリーと、(b)温度制御のた めの第二のアセンブリーとを含み、且つ、該第二のアセンブリーは、反応チャン バーに隣接して(adjacent to)位置決めされ得るアッセイシステム。 2.さらに、第一のアセンブリーと接触して滑らせて位置決め(slideably po sitioned)でき、そして複数の流体チャンバーを含む第三のアセンブリーを包含 し、且つ、各流体チャンバーが流体交換ポートを有し、該流体交換ポートの複数 が、第一のアセンブリーに関して第三のアセンブリーを滑らして位置決めするこ とによって、第一の流体交換チャンネルと一列に並べる(aligned with)ことが できる請求項1に記載のアッセイシステム。 3.さらに、第一の流体交換チャンネルと一列に並べることができる流体交換 ポートを有する流体チャンバーから、配列された第一の流体交換チャンネルの反 応チャンバーに流体を流させる第一の流体回転翼(fluid impeller)を包含する 請求項2に記載のアッセイシステム。 4.さらに、反応チャンバーに流体を残させる(leave)ための第二の流体回 転翼を包含し、且つ、第一のアセンブリーが、第一の流体交換チャンネルが第三 のアセンブリー中の流体交換チャンネルと一列に並んでいない場合に、流体が第 一の流体交換チャンネルを通して流れないように、流体密閉で、滑らし可能な手 段で第三のアセンブリー内に適合する(fits)請求項3に記載のアッセイシステ ム。 5.さらに、第三のアセンブリーまたは第一のアセンブリーを滑らせるための スライダーと、 (a)第三のアセンブリーまたは第一のアセンブリーの滑動、および(b)流 体を流体チャンバーから反応チャンバーに流させるための第一の流体回転翼を制 御するためのプロセッサと、 を包含する請求項2に記載のアッセイシステム。 6.請求項1に記載のアッセイシステムの第一の反応チャンバーにサンプルを 導入し、そして1つまたはそれ以上の流体チャンバーから、PCR反応を実施す るのに必要な試薬を含有する第一の反応チャンバー溶液に移動させることを包含 するPCR反応を実施する方法。 7.前記第二のアセンブリーが、約5秒以内で約25℃から約75℃の温度に 上昇させる請求項6に記載の方法。 8.前記第二のアセンブリーが、約5秒以内で約75℃から約50℃の温度に 低下させる請求項6に記載の方法。 9.反応チャンバーの温度を迅速に調節するための機構を有し、 前記アッセイシステムが(a)第一および第二のカバーを有し、および第一お よび第二のカバーの間の距離によって定義される厚みを示す反応チャンバーを包 含する第一のアセンブリーと、(b)熱源、冷却槽および第二の構成要素の表面 で開く複数の加圧流体チャンネルを包含する第二のアセンブリーと、(c)該加 圧流体チャンネル内で液圧を制御するための圧力コントローラーとを包含し、 該第二のアセンブリーが、加圧流体チャンネルが反応チャンバーのカバーと接 触しているように反応チャンバーに隣接して位置決めできるものであるアッセイ システム。 10.前記熱源が、第二のアセンブリーに付随した(attached to)少なくと も1つの電気的加熱器を包含し、且つ、前記冷却槽が、第二のアセンブリーに不 可欠な(integral)水または他の流体のための導管と、該導管を通って流体を流 れさせるための流体回転翼とを包含する請求項9に記載のシステム。 11.(a)変形できる材料から形成されたカバーを有す反応チャンバーと、 (b)該反応チャンバーの温度を迅速に調節するための機構とを包含し、 該機構が、(i)第一および第二の末端板(該第一の末端板は、該反応チャン バーカバーと接触して位置決めできる)と、 (ii)第一および第二の末端板の間に位置決めされ、且つ熱電気的(thermo electric)熱ポンプを形成するように電気的に直列に繋がった複数の対のp型お よびn型の半導体ブロックとを包含し、 前記熱ポンプが、直列に繋がった半導体ブロックを横切って(across)第一の 極性の電流が印加された際には、第一末端板から第二末端板に熱を送り(pumps heat);該直列に繋がった半導体ブロックを横切って、該第一の極性と反対の極 性の電流が印加された際には、第二末端板から第一末端板に熱を送るアッセイシ ステム。
