R rappresenta un atomo di idrogeno, un residuo alchilico ramificato, un gruppo cicloalchilico avente da 5 a 10 atomi di carbonio, un gruppo fenilico, eventualmente sostituito da uno o diversi atomi di cloro, gruppi metilici 0 metossilici oppure gruppi alchenilici a catena diritta o ramificata aventi da 3 a 11 atomi di carbonio, ed a loro sali farmaceuticamente accettabili.
Nel brevetto europeo EP 284 3S9 sono descritte piperazine 1,4-sostituite con un gruppo 3,4,5-(trimetossi)benzoilico ma soltanto uno dei sostituenti è un gruppo 3,4,5-(trimetossi)benzoilico, l'altro essendo un gruppo carbonilico policiclico condensato.
La presente invenzione si riferisce a composti di piperazina 1,2,4-sostituiti che presentano una 1,4-bis-sostituzione mediante un gruppo 3,4,5-(trimetossi)benzoilico. Questa serie di nuovi derivati piperazinici viene preparata in pochi stadi e fornisce attività farmaceutiche maggiori delle -piperazine 1,4 -sosti tu ite della tecnica anteriore.
Questa invenzione riguarda anche un procedimento di preparazione di composti di formula generale I, procedimento che comprende il portare a reagire un composto di formula generale in cui R ed n sono come sopra definiti ed X è un atomo di bromo o cloro con pirazilmetil sodio. Il pirazilmetil sodio può essere preparato da metilpirazina e sodioammide in ammoniaca liquida. La reazione viene convenientemente effettuata in una miscela di ammoniaca tiquida/dietiletere a -30 C (raffreddamento con ghiaccio secco).
che in seguito ad acilazione per trattamento con 3,4,5-trimetossibenzoilcloruro in benzene, in presenza di trietilammina, a temperatura ambiente, fornisce il prodotto finale di formula generale I.
L'invenzione si riferisce, infine, a composizioni terapeutiche contenenti uno dei composti I come ingrediente attivo.
ESEMPIO 1
N.N'-bis-(3’.4’.5 -trimetossibenzoin-2-n-ottil piperazina
R = H, n = 8
Stadio a
Preparazione di n-ottil pirazina
(Composto II in cui R = H ed n = 8)
A 15,6 g (0,4 moli) di sodio ammide in 400 mi di ammoniaca liquida anidra raffreddata a -50°C sono stati aggiunti 37,6 g (0,4 moli) di metilpirazina. La miscela è stata agitata per mezz'ora alla stessa temperatura.
Quindi è stato aggiunto n-eptil bromuro (35,8 g 0,2 moli), diluito con un egual volume di dietil etere anidro, in un periodo di 20 minuti e la miscela è stata agitata per un'altra ora. La reazione è stata raffreddata rapidamente per aggiunta di 25 g di cloruro di ammonio solido e l’ammoniaca è stata sostituita con dietil etere. La miscela è stata riscaldata fino a che il dietil etere ha appena iniziato a ricadere ed è stata quindi versata in ghiaccio, resa fortemente acida per aggiunta di HC1 concentrato ed estratta con dietil etere. Lo strato acquoso è stato quindi reso basico per aggiunta di NaOH ed estratto con CHClj.
Dopo essicamento su MgSO4 e filtrazione, l'evaporazione della fase organica ha portato ad un residuo che è stato cromatografato su una colonna di gel di silice usando dietil etere/etere di petrolio (15:85, v.v) come eluente.
Il composto II del titolo (33,4 g, resa 87%) è stato ricuperato sotto forma di liquido giallo pallido.
IR (film) : 3050 (C-H aromatico), 2940, 2860 (C-H), 1580, 1525 (anello aromatico) cm-1.
1HNMR (60 MHz), CDCI3, HMDS) 6ppm : 8,5 (ampio s, 3H, aromatico H), 2,75 (t, 2H, CH2-C = N), 1,7 (m, 2H, CH2-C-C = N), 1,25 (ampio s, 10H, (CH2)5), 0,8 (t,3H, CH3).
stadio b
Preparazione di 2-n-ottil piperazina
(Composto III in cui R = H ed n = 8).
Una soluzione di 3,4 g (20 millimoli) del composto Π, preparato nello stadio (a) precedente e 50 mg dij pd (10%)/carbone in 100 mi di etanolo è stata trattata con H2 sotto una pressione di 2,8 bars sotto agitazione per 3 ore a temperatura ambiente. Dopo filtrazione ed evaporazione dell’etanolo, il composto del titolo è stato ricuperato con resa del 100% come solido molto igroscopico.
IR (in olio paraffinico) : 3240 (N-H), 2920, 2850 (C-H) cm<-1>.
Stadio c
Preparazione di N,N’-bis-(3’,4’,5’-trimetossibenzoil)-2-n-ottil piperazina (Composto I in cui R = H ed n = 8)
Una soluzione di 3 g (15 millimoli) del composto III preparato nello stadio (b) precedente in 30 mi di benzene secco ed 1,5 mi di (C2H5)3N è stata aggiunta a gocce ad una soluzione di 7,6 g (33 millimoli) di 3,4,5-trimetossibenzoilcloruro in 30 mi di benzene secco. La miscela è stata agitata per una notte a temperatura ambiente. Quindi l’eccesso di acilcloruro è stato decomposto per aggiunta di 2 mi di etanolo mantenendo sotto agitazione per un’ora a temperatura ambiente. E’ stata aggiunta acqua e lo strato organico è stato lavato con una soluzione al 5% di NaHCO3, quindi con acqua.
Procedendo come descritto nel esempio 1 stadio (a) ma a partire da neopentil bromuro invece che da n-eptil bromuro, il composto del titolo è stata ricuperato sotto forma di liquido.

Procedendo come descritto nell’esempio 2, stadi (a), (b) e (c),ma a partire da p-(metil)fenetil bromuro anziché da n-eptilbromuro è stato ricuperato il composto del titolo come cristalli bianchi; p.f. 83-85%.

Una prova dell'interesse farmaceutico dei composti dell'invenzione è stata stabilita dalla seguente sperimentazione farmaceutica:
1. - Inibizione dell'aggregazione piastrinica_su conigli New Zealand
I derivati della presente invenzione hanno rivelato un’interessante attività PAF-antagonista rispetto all’aggregazione piastrinica indotta da 2,5 nm di PAF su plasma arricchito in piastrine (PRP) secondo il metodo di Bom.
Questa attività è stata posta in evidenza da studi di legame su omogenato di piastrine di coniglio. In questa preparazione in effetti questi derivati inibiscono il legame specifico mentre essi non hanno effetto significativo sulla formazione di legame dei leganti appropriati su omogenati di tessuto ricchi in recettori benzodiazepinici (centrali e periferici), istaminergici (Hj e H2) ed adrenergici (aj, a2, β2, β3) rispetto ad angiotensina II oppure ad ANF. La loro affinità è pertanto caratteristica dei recettori PAF.
La sperimentazione è stata condotta su piastrine con plasma di conigli New Zealand.
Campioni di sangue sono stati prelevati dal'arterìa auricolare e collocati in un tampone citrato (3,8%; pH 7,4); il sangue è stato ulteriormente centrifugato per 15 minuti a 1200 giri al minuto.
Il campione sottoposto a prove è stato preparato in DMSO, quindi versato su plasma arricchito in piastrine per un minuto, quindi è stata aggiunta una dose di 2,5 nm di PAF.
La determinazione viene fatta su un apparecchio Cronolog Coultronics che determina la percentuale di trasmissione corrispondente alla massima altezza del picco prima della disaggregazione.
Viene -calcolata la percentuale di variazione dell’inibizione rispetto alla percentuale di trasmissione (controllo:DMSO puro).
Questo metodo è stato descritto in dettaglio in LABORATORY INVESTIGATIONS, Voi. 41, No. 3, p. 275, 1979, JEAN-PIERRE CAZENAVE, Dr. MED., JACQUES BENVENISTE, DR.MED., AND J. FRASER MUSTARD, M.D., "Aggregation of rabbits platelets by platelet-activating factor is independent of thè release reaction and thè arachidonate pathway and inhibited by membrane-active drugs".

2. - Anafilassi cutanea-passiva (ratti
I derivati secondo l'invenzione, quando somministrati a 6,5 e 12,5 mg/kg, per via intraperitoneale, in ratti hanno antagonizzato significativamente con effetto dipendente dalla dose la reazione di anafilassi cutanea passiva (sensibilizzazione locale indotta per iniezione di immunosiero omologo ricco in IgE, quindi immediata reazione di ipersensibilizzazione mediante iniezione sistemica dell’antigene). Questa attività protettiva è stata ricuperata dopo somministrazione orale di 25 mg/kg di ognuno dei composti dell’invenzione (protezione variante tra il 39% e l'83% a seconda del composto).
3. Broncosoasmo immune su cavie
I composti dell’invenzione sono stati anche studiati sul broncospasmo immune indotto da iniezione di antigene su una cavia condizionata passivamente per iniezione di un immunosiero eterologo contenente IgE. La dose di 25 mg/kg per os di ciascuno dei composti riduce questo, broncospasmo al 15-49% e la dose da 5 mg/kg per via i.v. al 25-53% a seconda del composto.

- RIVENDICAZIONIrappresenta un gruppo alchilico a catena diritta o ramificata avente da n=l a n=20 atomi di carbonio, e R rappresenta un atomo di idrogeno, un residuo alchilico ramificato, un gruppo cicloalchilico avente da 5 a 10 atomi di carbonio, un gruppo fenilico, eventualmente sostituito da uno o diversi atomi di cloro, gruppi metilici o metossilici oppure gruppi alchenilici a catena diritta o ramificata aventi da 3 a ll atomi di carbonio, e loro sali terapeuticamente accettabili.
- 2. Procedimento di preparazione dei composti della rivendicazione 1 comprendente gli stadi di (a) far reagire un composto di formula generale in cui R ed n sono come sopra definiti ed X è un atomo di bromo o cloro con pirazilmetil sodio;c) acilare il composto IIΙ mediante 3,4,5-trimetossibenzoil cloruro, in benzene, in presenza di trietilammina, a temperatura ambiente.
- 3. Composizioni terapeutiche contenenti una quantità efficace di almeno un composto secondo la rivendicazione 1, associato con gli usuali veicoli per la via di somministrazione scelta.
