Stato dell’Arte State of the art
L'imaging e la spettroscopia, per esempio l'imaging e la spettroscopia di fluorescenza, sono stati sfruttati in maniera via via crescente per la caratterizzazione quantitativa e sensibile dell'attività metabolica e delle alterazioni strutturali nei tessuti biologici ex vivo ed in vivo. In particular, autofluorescence measurements are particularly interesting for clinical research, as they exploit the photo-physical properties of endogenous fluorophores (e.g. collagen, elastin, NAD (P) H, keratin) to generate tracer-free contrast and to provide information on the structural and functional properties of biological tissues, avoiding the use of potentially toxic exogenous contrast agents. The clinical potential and feasibility of autofluorescence measures for in vivo clinical diagnosis have been demonstrated in numerous studies, using both stationary and time-resolved autofluorescence measures. Stationary intensity measurement or autofluorescence emission spectroscopy have been widely used in clinical diagnostic research. However, intensity measurements alone are sensitive to intensity artifacts and therefore are difficult to compare between samples and patients. Spectrally resolved ratiometric measurements aim to provide quantitative readings but discrimination among many endogenous fluorophores is limited by their large and overlapping emission spectra. Time-resolved measurements can increase the specificity of fluorescence acquisition by resolving the fluorescence decay dynamics and thus can provide a means to distinguish endogenous fluorophores that have overlapping emission spectra but different mean fluorescence lifetimes. Fluorescence lifetime measurements are relatively insensitive to intensity artifacts affecting other techniques. Hence, when combined with fiber-optic-based instruments, fluorescence lifetime measurements of endogenous fluorophores could improve clinical diagnostics and provide guidance in surgical procedures.

In turn, for the mixture solutions of the two fluorophores, the specificity of the fluorescence increases with the spectral resolution. This is most evident when POPOP is the dominant fluorophore (see Figure 9c, third column). Specifically, when data is merged into a single channel (Figure 9c, panel 3), the corresponding fluorescence decay will be a weighted average of the fluorescence decays in the different spectral ranges. Since POPOP is predominant over FAD (see Figure 9b), the corresponding phasor will be closer to the pure POPOP phasor. If the data is binned into four spectral channels of equal amplitude (Figure 9c, panel 7), the specificity of the measurement increases and two sets of phasors are now evident in the phasor map, corresponding to the spectrally resolved fluorescence signals FAD and POPOP. A third group is visible along the line connecting the pure populations, representing the fluorescence decays within the spectral region in which both POPOP and FAD contributions are present, i.e. from ~ 450 nm to 500 nm. As the spectral resolution increases (see Figure 9c, panels 11, 15 and 19), the number of phasor groups along the line connecting the pure populations also increases and their distribution varies according to the contribution of each fluorophore in each. spectral band: fluorescence decays from bands with shorter wavelengths move towards pure POPOP (right); in turn, the fluorescence decays from longer wavelength bands are closer to pure FAD (left).
