Abstract
Colorimetric detection of an antigen (I) comprises (a) mixing a soln. of a tetramethylbenzidine (II) in an organic solvent with a soln. of a peroxide in a buffer just before the assay, so that a chromogenic substance (III) is formed; (b) a first antibody is adsorbed on a solid support; (c) a conjugate is formed between an immunologic reagent and an enzyme; (d) the conjugate is mixed with the sample contg. (I) and (I) binds to the first antibody and to the conjugate to form a complex in the solid phase; (e) (II) is added and the amount of (I) is determined by reaction of the complex with (III).
