Abstract
The immunoassay comprises (1) collection of, or formation of, on an active support, an immune complex having an enzyme component comprising the analyte (I) bound to an antibody; (2) contact of the enzyme component with signal generating agents to form a prod. (when it is a chromophore, it must be soluble in the assay medium); and (3) interaction of the prod. with the support to result in the concn. of a detectable signal on it. The immune complex may be formed in soln. by pre-incubation of (I) with an enzyme-labelled anti-analyte antibody, and it is then collected on the support. It may be formed on the support by immunochemical capture of (I) by an anti-analyte antibody bound to the support, followed by reaction with an enzyme-labelled anti-analyte antibody. Also it may be formed by capture of enzyme-labelled anti-analyte antibody by (I) bound to the support etc. The prod. formed in step (2) may be soluble in the assay medium and capable of emitting a detectable signal. The support may be made of nylon, cellulose nitrate, ethylcellulose or modified polyamide, esp. in a disc-like shape. By the technique (I) can be determined rapidly and with sensitivity, esp. when (I) is an antigen of viral or bacterial origin. (I) may also be a hapten, cells, microorganisms and their prods. in biological samples.
