Abstract
A microsealed pad suitable for prolonged transdermal administration of nitroglycerin comprises a biologically acceptable silicone polymer matrix affixed to a backing. The matrix is of cross-linked silicone rubber and has a plurality of 10-200 micron microsealed compartments in it which contain 10-30 vol.% of an aq. hydrophilic solvent system comprising 6-22 wt.% of a mixture contg. 10 wt.% nitroglycerin mixed with lactose and 5-15 wt.% of a hydrophobic solvent system selected from mineral oil and a glyceride of a saturated coconut oil acid and their mixtures, the hydrophilic solvent being a polyethylene glycol. The hydrophobic solvent system enhances transport and dispersion of the nitroglycerin. The hydrophilic solvent system is pref. aq. polyethylene glycol 400. The hydrophobic solvent may be e.g. mineral oil or isopropyl palmitate or their mixtures, or a mixture of migloyl oil and mineral oil.
