A 3-ienil-5-fíuoro-2-[ 1 (5X9H-purin-6-ilamino)propil]kinazolin-4(37f)-on, nemzetközi szabadnevén idelalisib szerkezeti képlete az alábbi:The structural formula of 3-phenyl-5-fluoro-2- [1- (5X9H-purin-6-ylamino) propyl] quinazolin-4 (37f) -one, the international non-proprietary name, is as follows:
Az idelalisib egy foszfoinozitid-3-kináz (PI3K) enzim inhibitor. A foszfoinozitid-3-kináz enzimek számos sejtfonkeió szabályozásában vesznek részt (növekedés-szabályozás, metabolizmus, transzláció iniciálás). Az idelalisibet a leukémia különböző formáinak gyógyítására törzskönyvezték.Idealisib is an inhibitor of the phosphoinositide-3-kinase (PI3K) enzyme. Phosphoinositide-3-kinase enzymes are involved in the regulation of several cell lines (growth regulation, metabolism, translation initiation). Idealis has been registered for the treatment of various forms of leukemia.
The preparation of idelalisib was first described in WO 2005113556. The solid obtained after purification by column chromatography was dissolved in ethanol and the solution was evaporated. The composition of the pale yellow solid thus obtained is idealized by elemental analysis · EtOH 0.4 H 2 O. The analytical results for the crystallographic characterization of the solid phase are not described by the inventors.
A WO 2013134288 számú nemzetközi szabadalmi bejelentés az idelalisib hét formáját (Form I, II, III, IV, V, VI és V.I1), előállításukat és gyógyszerkészítmény előállítására történő és orvosi alkalmazásukat ismerteti. Mindegyik módosulatnak közölték a jellemző röntgen-WO 2013134288 discloses seven forms of idelalisib (Form I, II, III, IV, V, VI and V.I1), their preparation and their use in the manufacture of a medicament and in medicine. For each modification, the characteristic X-ray
SZTNH-1004 40243 ponti ffinkciós felvételét (a Form I, IL III, IV, V formák esetében az elemi cella paramétereket is megadták), az egyes kristályíörmákat termoanalitikai módszerekkel (TG, DSC) is vizsgálták. A Fönn I és a Form II oldoszennent.es kristályfonna, a további kristályfonnák mindegyike az idelalisib bázis valamilyen szolvátja. A Form I polimorfot metanol-vizes elegyböl kristályosítják, az elegy hőmérsékletének pontosan meghatározott szabályozásával. A Form II polimorfot a Form I polimorf őrlésével, adott esetben ehhez kapcsolódó oldószeres szuszpendálásával vagy a Form I polimorf kompressziójával nyerik. Tehát a Form I polimorf egyes készítményfejlesztési műveletek (pl. tablettázás) során az általánosan alkalmazott körülmények között részben Form 11 módosulattá alakulhat. A két forma együttes jelenléte a gyógyszerkészítményben - az elvégzett gyógyszertechnológiai műveletek sajátosságait tekintve várhatóan változó arányban - mindenképpen technológiai és minőségellenőrzési nehézséget jelent.SZTNH-1004 40243 point functional recording (in the case of Form I, IL III, IV, V the elementary cell parameters were also given), the individual crystal forms were also examined by thermoanalytical methods (TG, DSC). Fönn I and Form II are soluble crystalline fibers, each of the other crystal fibers being a solvate of the idelalisib base. The Form I polymorph is crystallized from a methanol-water mixture by precisely controlling the temperature of the mixture. The Form II polymorph is obtained by grinding the Form I polymorph, optionally with associated solvent suspension, or by compressing the Form I polymorph. Thus, a Form I polymorph may be partially converted to a Form 11 modification under commonly used conditions during certain formulation operations (e.g., tableting). The combined presence of the two forms in the medicinal product - in a proportion that is expected to vary in terms of the characteristics of the pharmaceutical technology operations performed - is in any case a technological and quality control difficulty.
A WO 2015014315 számú nemzetközi szabadalmi bejelentés az idelalisib tíz formáját ismerteti, melyek közül négy oldószermentes kristályforma, a többi valamilyen szolvát. Az oldószermentes kristályformák közül kettő megegyezik a WO 2013134288 számú szabadalmi bejelentésben ismertetett Form I és Form II polimorffal. A két további forma (Form IV és Form Vili) egyik legfontosabb előnyeként csekély higroszkopieitásukat említik: Form IV esetében 0,3 %, míg a Fönn Vili módosulatnál 0,84 % a környezet nedvességtartalmából megkötött víz mennyisége a 20-80 % relatív páratartalom tartományban.WO 2015014315 discloses ten forms of idelalisib, four of which are solvent-free crystalline forms and the others are solvates. Two of the solvent-free crystal forms are identical to the Form I and Form II polymorphs described in WO 2013134288. One of the main advantages of the other two forms (Form IV and Form Vili) is their low hygroscopicity: 0.3% for Form IV and 0.84% for Fönn Vili, the amount of water bound from the ambient moisture content in the range of 20-80% relative humidity.
Given the many crystalline forms, it is clear that the production of the amorphous form is only possible under special, tightly controlled conditions. A further disadvantage of the amorphous form in such cases is that it can be converted to one of the more thermodynamically stable crystalline modifications during storage. According to the examples described in the patent application, the amorphous form was obtained exclusively by spray-drying technology or by evaporation of an ideal solution in the presence of a pharmaceutical excipient (polyvinylpyrrolidone, hydroxypropylmethylcellulose).

A találmány részletes ismertetéseDETAILED DESCRIPTION OF THE INVENTION
More particularly, the present invention relates to a crystalline Form E modification of idelalib base having characteristic X-ray powder diffraction peaks as follows: 20 (-0.2.2 ° 20): 11.56; 12.80; 14.96; 20.85. A találmányunk szerinti új idelalisib bázis módosulat (Form E) kristályszerkezetének mechanikai behatásokkal szembeni ellenálló-képességét préseléses vizsgálatokkal tanulmányoztok. A beállított nyomás értéke (kb. 50 MPa) összemérhető a tablettázáskor szokásosan alkalmazott nyomásértékekkel. Az idelalisib bázis Form E módosulatának nagynyomású préselésével előállított pasztilla mintáról felvett röntgen-pordiffraktogramot a 2. ábrán mutatjuk be. A difiraktogram láthatóan megegyezik a kiindulási (préselés előtti) mintáéval, tehát a préselés nem eredményezett szilárdfázisú átalakulást a mintában. The resistance of the crystal structure of the novel idealizib base modification (Form E) according to the invention to mechanical influences was studied by compression tests. The value of the set pressure (approx. 50 MPa) is comparable to the pressure values normally used for tableting. An X-ray powder diffractogram of a lozenge sample prepared by high pressure compression of the Form E modification of the idelalisib base is shown in Figure 2. The diffractogram is apparently the same as the initial (pre-compression) sample, so compression did not result in solid phase transformation in the sample.
Hasonló kísérletet végeztünk el a WO 2013134288 számú szabadalmi bejelentés szerinti idelalisib Form I módosulattal, az eredmények a 3. ábrán láthatók. Ebben az esetben a kapott pasztillához tartozó diffraktogramon - a hivatkozott bejelentésben foglaltaknak megfelelően megjelentek az idelalisib bázis Form II módosulatára jellemző diffrakciós csúcsok (nyilakkal jelölve a 3. ábrán), tehát a minta részben átalakult, nem egységes krístályszerkezetü. Az idelalisib bázis Fönn I módosulata ezek alapján kevésbé ellenálló a mechanikai behatásokkal szemben, a minta nagynyomású préselés hatására szilárdfázisú átalakulást szenvedett. A Form I módosulatnál tapasztalt jelenség hátrányos, mivel készímiény-formulálás során (pl. tablettázáskor) a hatóanyagot ebbe a nagyságrendbe eső préserő érheti, melynek hatására a készítményben lévő hatóanyag nem lesz egységes krístályszerkezetü.A similar experiment was performed with the idelalisib Form I modification according to WO 2013134288, the results of which are shown in Figure 3. In this case, the diffractogram of the obtained lozenge showed diffraction peaks characteristic of the Idelalisib base Form II modification (indicated by arrows in Figure 3), so that the sample had a partially transformed, non-uniform crystal structure. Based on these, the Fönn I modification of the ideal ideal base is less resistant to mechanical influences, the sample underwent solid-phase transformation under high-pressure compression. The phenomenon experienced with the Form I modification is disadvantageous because during formulation (e.g. tableting) the active ingredient may be subjected to a compression force of this order, as a result of which the active ingredient in the formulation will not have a uniform crystal structure.
Az idelalisib bázis Form E módosulatának 25 °C-on felveti vizszorpciós izotermáját a 4. ábrán mutatjuk be. The water absorption isotherm of the Form E modification of the idelalisib base at 25 ° C is shown in Figure 4. It can be clearly seen that the material does not show hygroscopic properties: in the range of 0-85% relative humidity the maximum water uptake is below 0.15%. The measurement results show that the hygroscopicity of the Idelalisib base Form E variant in the study range is lower than that of the Fönn I and Fönn II variants according to WO 2013134288 and from the two new solvent-free forms disclosed in WO 2015014315 ( Form IV and Form Vili) are also preferably omitted. The lower hygroscopicity observed with Form E is also beneficial in terms of stability and formulation.

A narancssárga oldatot 45-50 °C-ra hütjük, majd vákuumban bepároljuk (50 °C, 30-35 mbar). A bepárlási maradékot 200 ml diklórmetánban (DKM) oldjuk, 2x50 ml vízzel mossuk. A szerves fázist 10 g magnézium-szulfáttal szárítjuk (1 órán keresztül kevertetvé), 0,6 g aktív szénnel derítjük. A maradék gyantaszerű anyaghoz szobahőmérsékleten 100 ml acetonitrilt (ACN) adunk. Kevertetésre homogén oldatot kapunk, amelyet tovább kevertetve, néhány perc után megindul az idelalisib Form E-S acetonitrii szóival kristályosodása. Két óra kevertetés után a kivált anyagot szűrjük, apránként, mindig kevés ACN-rel fedve addig mossuk, míg a szőriét színtelenné nem válik (kb. 6’5 ml). Az így kapott anyag az idelalisib acetonitrii szolvátja (Form E-S).16.4 g (49.13 mmol) of (S) -2- (1-aminopropyl) -5-fluoro-3-phenylquinazolin-4 (3H) -one hydrochloride (II), 9.9 A mixture of 6-chloropurine (63.9 g, 63.9 mmol), isopropyl alcohol (IPA) (330 mL) and triethylamine (TEA) (20.5 mL, 14.9 g, 147.4 mmol) was added at 82-84 ° C. stirred at room temperature for 63 hours. The orange solution was cooled to 45-50 ° C and then concentrated in vacuo (50 ° C, 30-35 mbar). The residue was dissolved in dichloromethane (DCM) (200 ml) and washed with water (2 x 50 ml). The organic layer was dried over magnesium sulfate (10 g) (stirred for 1 hour) and clarified with charcoal (0.6 g). The desiccant and clarifier were filtered off, washed with DCM (2 x 20 ml) and the solvent removed in vacuo. To the remaining resin was added acetonitrile (ACN) (100 mL) at room temperature. Upon stirring, a homogeneous solution is obtained, which, after further stirring, starts to crystallize idelalisib Form E-S with acetonitrile after a few minutes. After stirring for two hours, the precipitate is filtered off and washed little by little, always with a little ACN, until the hair is colorless (about 6'5 ml). The material thus obtained is the acetonitrile solvate of idelalisib (Form E-S).
