A találmány tárgya eljárás fermentációban felhasználható mikroorganizmusok vagy mikroorganizmus-részek gélbe zárására.The present invention relates to a process for encapsulating microorganisms or parts of microorganisms useful in fermentation in a gel.
Rögzített mikroorganizmusok, illetve mikroorganizmus-részek alkalmazásával javítható a biokatalizátor stabilitása (élettartama), növelhető a katalizált reakció térfogati termelékenysége, és a fermentáció folyamatos üzemmódban is megvalósíthatóvá válik. It is known that fixed microorganisms or parts of microorganisms, such as spore suspensions for fermentation, cell debris, aerial mycelia, whole vegetative cell cultures or growth cultures, can be used very favorably in fermentation operations. The purpose of the fixation is to localize the microorganisms or parts of the microorganisms (which provide the biocatalysts needed for fermentation) in the reactor space and prevent them from entering the flow phase. The use of fixed microorganisms or parts of microorganisms can improve the stability (lifetime) of the biocatalyst, increase the volumetric productivity of the catalyzed reaction, and make fermentation possible in a continuous mode.
A rögzítés egyik lehetséges módszere a biokatalizátomak szálak vagy gélek üregeibe történő bezárása. Erre a célra szintetikus gélek (például poli-akrilamid) vagy természetes eredetű gélképzők alkalmazhatók. Ismert például, hogy a Penicillium chrysogeneum-konidiumok, -micéliumok és -protoplasztok gélbe zárt állapotban is felhasználhatók penicillin G bioszintéziséhez. A bezáráshoz a Biotechnoi. Bioeng. 26,318 (1984) közlemény szerint K-karragenátot, a Biotechnoi. Bioeng. 27, 261 (1979) közlemény szerint poli-akrilamidot, az Eur. J. Appl. Microbiol. Biotechnoi. 75, 211 (1982) közlemény szerint kalcium-alginát gélt használnak. Az Eur. J. Appl. Microbiol. 2, 153 (1976) közlemény poli-akrilamidba bezárt Escherichia coli sejtek használatát ismerteti 6-amino-penicillánsav előállításában. A Biotechnoi. Bioeng. 23, 2747 (1981) közlemény poli-akrilamidba bezárt Streptomyces clavuligerus sejtek felhasználását ismerteti cefalosporin C előállításában. Az Antimicrob. Agents Chemother. 75, 126 (1979) és Biotechnoi. Bioeng. J. Appl. Microbiol. Biotechnol. 75, 206 (1982) közlemény kalcium-alginátba rögzített Streptomyces sejtek felhasználását ismerteti tilozin és nikkomicin termelésében. A Biotechnoi. Lett. One possible method of attachment is to enclose the biocatalysts in the cavities of the fibers or gels. For this purpose, synthetic gels (e.g., polyacrylamide) or natural gelling agents may be used. For example, it is known that penicillium chrysogeneum conidia, mycelia and protoplasts can be used in gel-encapsulated form for penicillin G biosynthesis. To close, Biotechnoi. A fenti ismert eljárások hátrányai a következők:The disadvantages of the above known processes are as follows:
A poli-akrilamid gélbe történő bezárásnál a gélesedés során keletkező szabad gyökök súlyos sejtkárosodást okoznak, illetve egyes enzimekre gátló hatást fejtenek ki. When enclosed in a polyacrylamide gel, free radicals formed during gelation cause severe cell damage and inhibit certain enzymes. In the case of ionotropic gelling agents such as alginates and carrageenans, the metal ions required for gelling may be cytotoxic. Therefore, these methods can be successfully applied to only a limited number of microorganisms or parts of microorganisms suitable for fermentation purposes.
Célszerű megoldás lenne a fermentációra alkalmas mikroorganizmusokat, illetve mikroorganizmus-részeket agar-gélbe zárva rögzíteni. It would be convenient to fix the microorganisms or parts of microorganisms suitable for fermentation in an agar gel. The agar gel, which is the most common component of the medium used for the cultivation of microorganisms, has no adverse effect on microorganisms, provides optimal conditions for living cells and spores, and the gel network is also suitable for the in situ development of the mycelium.
Mikroorganizmusok agar-gélbe zárását ismerteti a Biotechnoi. Bioeng. Incorporation of microorganisms in agar gel is described in Biotechnol. Bioengineering. 77, 1729 (1975) and Microbiologiya 49,479 (1980). As disclosed therein, a 2-4% aqueous agar solution is homogenized by cooling with a cell suspension of Escherichia coli and Methylomonas at a temperature close to the gelling temperature, and the homogenate is added dropwise to the distilled water or phosphate buffer and left to solidify.
A megdermedt agar-géltömbök aprítása időigényes, nehézkes művelet A fermentációs célokra optimálisan felhasználható, szabályos gömb alakú, szűk mérettartományba eső átmérőjű gélrészecskék ezekkel a módszerekkel nem állíthatók elő.The disadvantage of this solution is that the gel dripped into the water will freeze in varying sizes and shapes. The use of the resulting gel particles of extremely heterogeneous shape (fiber, hemispherical or irregular shape) and size distribution for fermentation purposes is not advantageous because they do not allow a smooth flow of the medium, the particles can drift into the flow phase and are difficult to separate. Crushing of frozen agar gel blocks is a time consuming, laborious operation. The use of regular spherical, narrow-diameter gel particles which are optimally usable for fermentation purposes cannot be achieved by these methods.
Vizsgálataink során megállapítottuk, hogy a képződő gélrészecskék alakja és mérete szempontjából döntő szerepe van a gélképzés közegeként felhasznált folyadék fizikai jellemzőinek. Tapasztalataink szerint szabályos gömb alakú, szűk mérettartományba eső (rendszerint 1-5 mm méretű) gélrészecskék akkor állíthatók elő, ha a fermentációs célokra használt mikroorganizmus vagy mikroorganizmus-részek 45-50 ’C hőmérsékletű folyékony agar-oldatlal képezett keverékét olyan, a mikroorganizmust nem károsító szerves oldószerbe vagy oldószer-elegybe csepegtetjük, amelynek vízben való oldhatósága 0,03-10 g/100 g, sűrűsége 0,61,2 g/cm3, 20 ’C-on meghatározott viszkozitása 0,4510 cP, forráspontja 75-250 ’C, és amelyben a cseppek ülepedést sebessége 0,01-10 cm/sec.In our investigations, it was found that the physical properties of the fluid used as the gelling medium play a decisive role in the shape and size of the gel particles formed. In our experience, regular spherical, narrow size (usually 1 to 5 mm) gel particles can be prepared when a mixture of the microorganism or parts of the microorganism used for fermentation in a liquid agar solution at 45-50 ° C is not harmful to the microorganism. is added dropwise to an organic solvent or mixture of solvents having a water solubility of 0.03 to 10 g / 100 g, a density of 0.61.2 g / cm 3 , and a viscosity of 0.4510 cP at boiling point of 75 to 250 '. C, and wherein the droplets settle at a rate of 0.01 to 10 cm / sec.
A leírásban és az igénypontsorozatban a „fermentációra alkalmas mikroorganizmus vagy mikroorganizmusrész” megjelölésen az adott mikroorganizmusnak fermentációs célra felhasználható összes lehetséges formáját, így például a mikroorganizmus vegetatív tenyészetét, spóraszuszpenzióját, izolált sejtjeit, konidiumait, miccliumait, sejttörmelékeit, növekedésben lévő tenyészeteit, sejtkomponenseit stb. értjük.As used herein and in the claims, the term "microorganism or microorganism suitable for fermentation" refers to all possible forms of the microorganism that can be used for fermentation, such as vegetative culture, spore suspension, isolated cells, conidia, micelles, cell debris, growths, etc. We understood.
A gélképzés közegeként felhasznált szerves oldószer vagy oldószer-elegy vízben való oldhatósága célszerűen 0,1-10 g/100 g, sűrűsége pedig célszerűen 0,81,1 g/cm3 lehet További alapvető követelmény, hogy a szerves oldószeibe csepegtetett gélcseppek ne lebegjenek a szerves oldószerben; a cseppek ülepedést sebessége célszerűen 0,05-10 cm/sec.The water solubility of the organic solvent or solvent mixture used as the gelling medium is preferably 0.1 to 10 g / 100 g and the density is preferably 0.81.1 g / cm 3 Another basic requirement is that the gel droplets added to the organic solvents should not float in an organic solvent; the droplet settling rate is preferably 0.05 to 10 cm / sec.
A találmány szerinti eljárásban a gélképzés közegeként például etil-acetátot, izooktanolt, n-butil-acetátot, n-amil-alkoholt, benzil-alkoholt, acetecetésztert, a felsorolt oldószerek keverékeit, vagy 10 térfogat% metil-izobutil-ketont tartalmazó acetecetésztert használhatunk.Gel formation media used in the process of the present invention include, for example, ethyl acetate, isooctanol, n-butyl acetate, n-amyl alcohol, benzyl alcohol, acetacetate, mixtures of the solvents listed, or acetone containing 10% by volume of methyl isobutyl ketone.
A gélgyöngy-képződés gyorsítása céljából a gélképzés közegét célszerűen 0-10 ’C-ra hűtjük. In order to accelerate gel bead formation, the gelling medium is preferably cooled to 0-10 ° C. If desired, a slow injection of gas may be introduced into the gel formation medium to prevent the gel beads from adhering to one another. The gcl-2HU 202 579 Β formed beads can be isolated by filtration, washed with distilled water to remove solvent and dried if desired.
A találmány szerinti eljárást az oltalmi kör korlátozása nélkül az alábbi példákban részletesen ismertetjük.The invention is illustrated in detail by the following non-limiting Examples.

Streptomyces lavendulae ATCC 12 950 7-8 napos ferde agar tenyészetéiül steril desztillált vízzel 5x x 107 spóra/cm3 koncentrációjú spóraszuszpenziót készítünk. 10 cm3 így kapott spóraszuszpenziót 90 cm3, előzetesen 121 ’C-on 30 percig sterilizált, majd 50 ’C-ra hűtött 4%-os agar oldathoz adunk. Ezt a keveréket perisztaltikus pumpa segítségével - steril eszközök alkalmazásával - 1000 cm3 5 ’C-ra hűtött izo-oktanolba (2-etil-hexanoIba) csepegtetjük. A keletkező gélgyöngyök egymáshoz tapadását nitrogéngáz lassú átbutorékoltatásával gátoljuk. A csepegtetés befejezése után a nitrogéngáz átáramoltatását még 4-5 percig folytatjuk, majd a gélgyöngyöket kiszűrjük, 0,02 tömeg% Tween 80-at tartalmazó steril desztillált vízzel oldószermentesre mossuk. Szabályos gömb alakú, 1-5 mm átmérőjű gélgyöngyöket kapunk.Streptomyces lavendulae ATCC 12,950 is prepared by culturing a 7-8 day slant agar with sterile distilled water at a concentration of 5 x 10 7 spores / cm 3 . 10 cm 3 of the spore suspension thus obtained was added to 90 cm 3 of a 4% agar solution, previously sterilized at 121 ° C for 30 minutes and then cooled to 50 ° C. This mixture was added dropwise to 1000 cm 3 of isoctanol (2-ethylhexanol) cooled to 5 ° C using a peristaltic pump. The resulting gel beads are prevented from adhering to each other by slowly bubbling nitrogen gas through. After completion of the dropwise addition, the nitrogen gas was bubbled through for 4 to 5 minutes, and the gel beads were filtered and washed with sterile distilled water containing 0.02% Tween 80 to remove solvent. Regular spherical beads of 1-5 mm diameter are obtained.
