Abstract
Description
Találmányunk szerinti eljárás alkalmas ontíbiolikum fermentációk környezetszennyező hulladékaként képződő micéliumának állatgyógyászatban alkalmazható gyógypremixsze történő feldolgozására. Közelebbről, eljárásunk lehetővé teszi pl. oxitetraciklin, neomicin és nebramicin hatóanyagot kis mennyiségben még tartalmazó szűrt, nedves mic.éliumok gazdaságos feldolgozását és állatgyógyászati célú felhasználását.The process of the present invention is suitable for the treatment of mycelium formed as an environmental pollutant waste of ontibiotic fermentations with a veterinary drug premix. In particular, our process allows e.g. economical processing and veterinary use of filtered, wet micelles, containing small amounts of oxytetracycline, neomycin and nebramycin.
The production of fermentation antibiotics is booming in Hungary and worldwide. This is illustrated not only by the increasing amount of active compound produced each year, but also by the increasing volume of fermented volumes. Unfortunately, there are currently no active drug recovery technologies that leave inactive mycelium, so there is a general problem with the use and disposal of waste still containing the active ingredient. The use of antibiotic-containing mycelia for animal nutrition, in line with previous practice, is now in violation of health regulations.
A fentiek értelmében számos kísérlet történt arra, hogy inaktiválják a micélium hatóanyagát, azonban a béta-laktám tartalmú micéliumok kivételével kevés sikert értek el. Penicillium micéliumok inaktiválására iparilag is gazdaságosan alkalmazható eljárást ír le a T/38.232 számú magyar közzétételi irat. További kísérleteket folytattunk tetraciklin és aminoglikozid tartalmú micéliumok hatóanyagmenleBÍlésére, azonban teljes hatóanyag mentesítést nem sikerült elérni. As stated above, many attempts have been made to inactivate the mycelial agent, but little success has been achieved with the exception of beta-lactam-containing mycelia. A process which is commercially applicable to the inactivation of penicillium mycelia is described in Hungarian Patent Publication No. T / 38.232. Further attempts were made to control the mycelial content of tetracycline and aminoglycoside-containing mycelium, but complete drug release was not achieved.
This was also due to the fact that tetracycline and aminoglycoside antibiotics are much more stable and do not cleave into relatively inactive products by relatively simple chemical and / or thermal effects.
Efforts to make the active mycelium useful for veterinary purposes are well known to us. These include the process known in Hungary for treating the active ingredient formed in the fermentation of oxytetracycline in such a way that it adheres to the surface of the mycelium and, after filtration, produces a filtered, moist mycelium containing the active ingredient. The filtered wet mycelium is then sprayed and used for veterinary purposes.
A common feature of the above-described processes is that the dried active mycelium is commercially available in the form of powder or granules. It is known that drying is a very energy-intensive process {the filtered, wet mycelium takes approx. Contains 70% water) which makes the product disproportionately expensive.
Célul tűztük ki olyan eljárás kidolgozását, amellyel gazdaságosan és egyszerűen kiv i tele zh élőén tudjuk a környezetszennyező ás jelenleg hulladékanyagként jelentkező szűrt, nedves micéliumot állatgyógyászati célú felhasználásra alkalmassá tenni. Our aim is to develop a process for rendering the filtered, wet mycelium, which is a pollutant and is currently waste, economically and easily accessible for veterinary use.
Kitűzött célunk elérése érdekéhen kísérleteket. végeztünk egyrészről a micéliumok g imbás és bakteriális fertőzésének megakadályozására, másrészről stabilizálására.Attempts to achieve our goal. on the one hand to prevent g microbial and bacterial infection of the mycelium and on the other hand to stabilize it.
Λ micéliumok magas víztartalmuk és az esetenkénti magas növényi olaj tartalmuk következtében nehezen kezelhető, mechanikai behatásra pasztaszerűen össze!apadó anyagok, haLóanyagtarlalrnukat tekintve erősen inhomogének és keletkezésük állapotában csak rövid ideig, néhány napig tárolhatók. A néhány napig tartó stabilitást a viszonylag magas antibioLikuin tartalom magyarázza.Λ Mycelia, due to their high water content and occasionally high vegetable oil content, are difficult to handle, mechanically effective paste-like materials, which are highly inhomogeneous in their material content and can only be stored for a few days in their state of formation. The stability for several days is explained by the relatively high content of antibioLikuin.
A kezelt m leéli uniókat polietilén tasakokban, levegőtől elzárva szobahőmérsékleten tároltuk. To prevent bacterial and fungal infection of mycelium, only chemicals that do not hinder the downstream user, i.e., harmless to animals, can be used. In order to biologically stabilize wet mycelia, a variety of materials, especially organic acids, have been tested. It has been found that the best results are obtained by the addition of organic acids, preferably propionic acid. Λ the results of propionic acid treatments are given in Table 1. The treated m unions were stored in polyethylene sachets, sealed at room temperature.

Another important requirement for medicated premixes is the production of a constant fish content, that is, a consistently high quality formulation. It is known that the active ingredient titers of fermentations are not always the same.
Ennek továbbá az alkalmaz, ott szűrési eljárások hiányosságainak következtében a szűrt, nedves micélium hatóanyagtartnlma sem azonos az egymást kővető sarzsok esetében. In addition, due to the shortcomings of the filtration process, the content of the filtered wet mycelium is not the same for successive batches.
A hatóanyagtartalom különbségek kiküszöbölésére eljárásokat dolgoztunk ki. Egyik eljárásunk szerint a micéliumot propionsav-kénsav eleggyel kezelt takarmányliszttel keverjük össze. A keverés hatására a micélium állaga kedvezően változik, és makroszkopikusan homogén anyagot kapunk. Az, apró mirelium golyócskákat takarmányliszt veszi körül, emiatt a keverék száraz, anyagtartalma a valódi értéknél magasabbnak tűnik. Ebből a keverékből biológiai módszerrrel (F. Kavanagh (1975) Methods in Enzymology vo). 43,, p.Methods have been developed to eliminate differences in drug content. In one method, the mycelium is mixed with feed meal treated with a mixture of propionic acid and sulfuric acid. Upon mixing, mycelium changes favorably and a macroscopically homogeneous material is obtained. The tiny mirelium spheres are surrounded by fodder flour, which makes the mixture dry and its content seems to be higher than its true value. From this mixture by biological method (F. Kavanagh (1975) Methods in Enzymology vo). 43 ,, p.

A stabilitási vizsgálatok eredményeit a takarmánylisztes keverékben a 2. táblázat mutalja. The results of the stability studies in the feed meal mix are shown in Table 2.
2. táblázatTable 2
A micélium és a savakkal kezelt takarmányliszt összemérését az alábbiak szerint végeztük:The mycelium and the acid-treated feed meal were weighed as follows:
tömegrész takarmánylisztben elkevertünk 10% propionsavat és 2,0 t% kénsavat (48%-os). Az ily módon előállított savas takarmánylisztet 1 tőmegrész szűrt, nedves micéliummal kevertük össze.10% propionic acid and 2.0% sulfuric acid (48%) were mixed in a part of the feed meal. The acid fodder meal thus obtained was mixed with 1 part by weight of filtered, wet mycelium.
Mint az a táblázatból leolvasható, a kénsavas kezelés a nebramicin komplexet tartalmazó keverék esetén a pH savasra állítása révén teszi lehetővé a propionsav hatásának kifejeződését.As can be seen from the table, the sulfuric acid treatment of the mixture containing the nebramycin complex allows the effect of propionic acid to be expressed by adjusting the pH to acid.
A stabilitás vizsgálatot olyan mintákon íb elvégeztük, ahol takarmányliszt helyett propionsavat tartalmazó perlitet kevertünk a micéliumhoz. Penészedést egyik esetben sem tapasztaltunk. A perlit jó nedvszívó hatása következtében kis mennyiségben is kedvezően befolyásolja a micélium állagát, ezáltal a gyógypremix kezelhetőségét.The stability test was performed on samples in which perlite containing propionic acid was mixed with mycelium instead of feed meal. In no case did mold occur. Due to its good absorbency, perlite also has a small effect on the consistency of the mycelium and thus on the manageability of the drug premix.
Alternatively, the filtered wet mycelium is suspended in a quantity of water depending on the fermentation titer to maintain the active substance content of the mycelium produced during the re-filtration within the prescribed range. In this case, propionic acid or other organic acid used is sprayed onto the mycelium during re-filtration. The preserved mycelium thus obtained may be mixed with sulfuric acid maize flour or perlite but may be packaged directly on its own.
Ezen eljárásunk alkalmazása esetén a második szűrésnél keletkező Rzűriet sok esetben olyan mennyiségű hatóanyagot tar60 lalmaz, amelynek kinyerése a feldolgozási folyamat hatékonyságát növeli. Ilyen esetekben a reszuszpendálást az újra szűrést gyakran célszerű a főmüveletektől eltérő körülmények között végezni (pl. savazás, melegítés).In this process, the filtrate formed during the second filtration often contains an amount of the active ingredient, the recovery of which increases the efficiency of the processing process. In such cases, resuspension is often performed in conditions other than the main operation (e.g. acidification, heating).
Changes in the antibiotic content of the medicated premixes prepared by the methods developed by us were investigated during the storage period (test method as above). It has been found that in the case of drug premixes containing the active ingredient neomycin and nebramycin complex, the antibiotic activity is not reduced after two months. Λζ In the case of a drug premix containing oxyelracycline, a slight degradation of the active ingredient and a decrease in activity were observed. The results are shown in Table 3.
