Description
A találmány tárgya eljárás az európai szőlőmoly (Lobesia botrana) hímjeire szexuális csalogató hatást kifejtő, (I) képletü 7(E),9(Z)-alkadienol-származékok előállítására. A képletben Rí 1—4 szénatomos alkilcsoportot, R2 hidrogénatomot vagy 1 -4 szénatomos acil-csoportot jelent. The present invention relates to a process for the preparation of sexually attractive sedatives of male (Lobesia botrana) European 7 (E), 9 (Z) -alkadienol derivatives of the formula (I). In the formula, R 1 is C 1 -C 4 alkyl, R 2 is hydrogen or C 1 -C 4 acyl.
Az európai szőlőmoly (Lobesia botrana) Középés Dél-Európa szőlőkultúrájának elterjedt kártevője. Szexuális csalogató anyagát, a 7(E),9(Z)-dodekadienil-acetátot (I, R^CjHs, R2=CH3CO) korábban izolálták (Buser, H. R. és munkatársai: Z. Naturforsch, C. 29, 781 (1974)], majd szintetizálták és sikeresen alkalmazzák rajzáselőrejelzésre és védekezésre [3 845 108. sz. USA szabadalmi leírás; Descoins, C. és munkatársai: C. R. Acad, Sci., Ser.The European grape moth (Lobesia botrana) is a common pest in the grape culture of southern Europe. Its sexual attractant, 7 (E), 9 (Z) -dodecadienyl acetate (I, R 1 -C 3 H 5 , R 2 = CH 3 CO), has previously been isolated (Buser, HR, et al., Z. Naturforsch, C. 29, 781). (1974)], and subsequently synthesized and successfully used for swing prediction and defense (U.S. Patent No. 3,845,108; Descoins, C., et al., CR Acad, Sci., Ser.
D. 279, 904 (1974); Biwer, G. és munkatársai: C. R. Acad, Sci., Ser. D. 280, 1469 (1975)]. A többi (I) általános képletü vegyület előállításáról a szakirodalom nem tesz említést.D. 279, 904 (1974); Biwer, G., et al., C. R. Acad, Sci., Ser. D. 280, 1469 (1975). The preparation of the other compounds of formula I is not mentioned in the literature.
A 7(E),9(Z)-dodekadienil-acetát szintetikus előállítására több eljárás is ismeretes. W. Roelofs és munkatársai (174 374. sz. magyar szabadalom) a bróm-acetaldehid dietilacetáljából és szuberinsavból kiindulva 11 lépésben állították elő, oly módon, hogy a bróm-acetaldehid acetáljából trietil-foszfáttal foszfonátot képeztek, majd savas hidrolízissel formil-metil-foszfonátot kaptak, amelyből ciklohexil-aminnal a foszfonát iminszármazékát állították elő. A szintézis másik ágán szuberinsavból metanollal metil-hidrogén-oktadionátot képeztek, majd a vegyületet higany(II)-oxiddal és brómmal kezelve metil-(7-bróm-heptanoátot) kaptak. Az utóbbiból piridin-N-o Kid dal metil-6-formil-hexanoátot állítottak elő, amit a fenti foszfonátból nátriumhidriddel képzett anionnal reagáltattak. A termékből savas hidrolízissel nyert metil[8-formil-7(E)-oktenoátot= a trifenil-propil-foszfónium-sóból nátrium-(metil-szulfinilmetilid)-del képezett foszforánnal reagáltatva metil-[7(E),9(Z)-dodekadienoátot] kaptak, amelyet lítium-alumínium-hidriddel redukálva 7(E),9(Z)-dodekadién-l-olt állítottak elő. Az utóbbit ecetsavanhidrid/piridin eleggyel acilezve kapták a célvegyületet, amelyet preparatív gázkromatografálással választottak el a mellette 20 súly% alatti mennyiségben képződő sztereoizomeijétől [7(E),9(E)-dodekadienil-acetát]. A szintézis a reakciólépések nagy száma, a drága reagensek (pl. lítium-alumínium-hidrid, piridin-N-oxid) és a költséges preparatív gázkromatográfiás eljárás miatt nem gazdaságos.Several methods are known for the synthetic preparation of 7 (E), 9 (Z) -dodecadienyl acetate. W. Roelofs et al. (Hungarian Patent No. 174,374) prepared starting materials from bromoacetaldehyde diethyl acetal and suberic acid in 11 steps by forming triethyl phosphate phosphonate from bromoacetaldehyde acetal followed by acid hydrolysis with methyl from which an imine derivative of the phosphonate was obtained with cyclohexylamine. On the other side of the synthesis, methyl hydrogen octadionate was formed from suberic acid with methanol and treated with mercury (II) oxide and bromine to give methyl (7-bromo-heptanoate). From the latter, pyridine-N-o Kid was converted to methyl 6-formylhexanoate which was reacted with the anion formed with sodium hydride from the above phosphonate. The product obtained by acid hydrolysis of methyl [8-formyl-7 (E) -octenoate = methylphosphonate from the triphenylpropylphosphonium salt with sodium (methylsulfinylmethyl) is reacted with methyl [7 (E), 9 (Z) -dodecadienoate], which is reduced to 7 (E), 9 (Z) -dodecadien-1-ol by reduction with lithium aluminum hydride. The latter was acylated with acetic anhydride / pyridine to give the target compound, which was separated from its adjacent stereoisomer (7 (E), 9 (E) -dodecadienyl acetate) by preparative gas chromatography. Synthesis is not economical due to the large number of reaction steps, expensive reagents (eg lithium aluminum hydride, pyridine N-oxide) and expensive preparative gas chromatography.
J. N. Labovitz és munkatársai (Tetrahedron L. 1975, 4209) a 7(E),9(Z)-dodekadienil-acetát előállítását 1-butinból oldották meg. Az 1-butinból metil-magnézium-bromiddal 1-butinil-magnézium-bromidot képeztek, amelyet akril-aldehiddel reagáltatva l-heptén-4-in-3-olt kaptak. A vegyületet trimetil-ortoacetáttal melegítve a metil-[4(E)-nonen-6-inoát] és a metil-[4(Z)-nonen-6-inoát] 4 :1 súlyarányú elegyét nyerték, amelyet — elválasztás nélkül - lítium-alumínium-hidriddel redukálva a 4(E)-nonen-6-in-l-ol és a 4(Z)-nonen-6-in-l-ol elegyévé alakítottak.J. N. Labovitz et al. (Tetrahedron L. 1975, 4209) resolved the preparation of 7 (E), 9 (Z) -dodecadienyl acetate from 1-butine. 1-Butine was converted to 1-butynyl magnesium bromide with methyl magnesium bromide and reacted with acrylic aldehyde to give 1-hepten-4-yn-3-ol. The compound was heated with trimethyl orthoacetate to give a 4: 1 mixture of methyl [4 (E) -none-6-inoate] and methyl [4 (Z) -none-6-inoate] which was lithium without separation. reduced with aluminum hydride to form a mixture of 4 (E) -none-6-in-1-ol and 4 (Z) -none-6-in-1-ol.
-1181328-1181328
Az utóbbiakból p-toluol-szulfonsavkloriddal a tozilátokat kapták, amelyeket l-bróm-4(E)-nonen-6-in és l-bróm-4(Z)-nonen- 6-in elegyévé alakítottak. A brómvegyületeket dilítium-tetraklór-kuprát jelenlétében 3-[(l-etoxi)-etoxi]-propil-lítiummal reagáltatták, majd savas hidrolízis után a szintézisben szükséges 7(E)-dodecen-9-in-l-olt frakcionált kristályosítással választották el a 7(Z) konfigurációjú sztereoizomerjétől. Az alkoholból trimetil-szilil-kloriddal szilil-étert képeztek, amelyet bisz(3-metil-2-butil)-boránnal reagáltatva, ecetsavas kezelés után a 7(E),9(Z)-dodecen-l-olt kapták. A vegyület ecetsavanhidríddel végzett acilezése a 7(E),9(Z)-dodekadienil acetátot adta. A szintézis a drága reaktánsok (pl. lítium-alumíniumhidrid, dialkil-borán, trimetil-szilil-klorid) és a speciális reakciókörülményeket igénylő reakciólépések miatt nem gazdaságos. The latter gave the tosylates with p-toluenesulfonic acid chloride, which were converted to a mixture of 1-bromo-4 (E) -one-6-in and 1-bromo-4 (Z) -nonene-6-in. The bromine compounds were reacted with 3 - [(1-ethoxy) -ethoxy] -propyl-lithium in the presence of dilithium tetrachloro-cuprate, followed by fractional crystallization of 7 (E) -dodecen-9-in-1-ol required for acid hydrolysis. the stereoisomer of configuration 7 (Z). The alcohol was converted to the silyl ether with trimethylsilyl chloride which was reacted with bis (3-methyl-2-butyl) borane to give 7 (E), 9 (Z) -dodecen-1-ol after treatment with acetic acid. Acylation of the compound with acetic anhydride gave 7 (E), 9 (Z) -dodecadienyl acetate. Synthesis is not economical due to expensive reactants (such as lithium aluminum hydride, dialkylborane, trimethylsilyl chloride) and reaction steps requiring special reaction conditions.
E. Negishi és A. Abramovitch (Tetrahedron L., 1977, 411) kiindulási vegyületként 2-oktin-l-olt használtak, A vegyületet az 1,3-diamino-propán káliumsójával ún. „acetilén zipper” reakciónak alávetveE. Negishi and A. Abramovitch (Tetrahedron L., 1977, 411) used 2-octin-1-ol as starting material. The compound with the potassium salt of 1,3-diaminopropane was used as a starting material. Undergoing an "acetylene zipper" reaction
7-oktin-l-olt kaptak, amelyet ecetsavanhidríddel acileztek. Az acetátot bisz-(3-metil-2-butil)-boránnal kezelve 8-acetoxi-l-oktenil-bisz(3-metil-2-butil)-boránt kaptak, amelyet az 1-butin lítium sójával reagáltatva, majd jóddal, nátrium-acetáttal, hidrogén-peroxiddal kezelve a 7(E)-dodecen-9-in-l-ol acetátját állították elő. A vegyület szén-szén hármas kötésére diziamil-boránt addicionálva, majd ecetsavval, azt követően hidrogén-peroxiddal kezelve 7(E),9(Z)-dodekadienil acetátot nyertek. A szintézis a drága kiindulási anyagok és reaktánsok (diziamilborán, alkil-lítium) alkalmazása miatt nem gazdaságos.7-Octin-1-ol was obtained, which was acylated with acetic anhydride. The acetate was treated with bis (3-methyl-2-butyl) borane to give 8-acetoxy-1-octenylbis (3-methyl-2-butyl) borane, which was reacted with the lithium salt of 1-butin and then with iodine. 7 (E) -dodecen-9-in-1-ol acetate was prepared by treatment with sodium acetate and hydrogen peroxide. Addition of disulfamylborane to the carbon-carbon triple bond followed by treatment with acetic acid followed by hydrogen peroxide gave 7 (E), 9 (Z) -dodecadienyl acetate. Synthesis is not economical due to the use of expensive starting materials and reactants (disiamylborane, alkyl lithium).
C. Descoins és munkatársai (Bul. Soc. Chim. Francé, 1977, 941) a 7(E),9(Z)-dodekadienil-acetátot l-ciklopropil-2-pentin-l-olból szintetizálták. A karbinolt dikobalt-oktakarbonillal komplexálták, majd hidrogén-bromid-cink-bromid rendszerrel l-bróm-3(E)-okten-5-inné rendezték át. A bromidot rézsók jelenlétében 4-tetrahidropiranil-oxi-butilklorid magnézium vegyületével reagáltatva a 7(E)-dodecén-9-in-l-ol tetrahidropiranil-éterét kapták, amelyet savval katalizált reakcióban 7(E)-dodecen-9-in-l-ollá hidrolizáltak. Az utóbbit katalitikus hidrogénezéssel 7(E),9(Z)-dodekadién-l-ollá redukálták, majd a redukált termék acilezése útján jutottak a célvegyülethez.Descoins et al. (Bul. Soc. Chim. Francé, 1977, 941) synthesized 7 (E), 9 (Z) -dodecadienyl acetate from 1-cyclopropyl-2-pentin-1-ol. The carbinol was complexed with dicobalt octacarbonyl and rearranged to the 1-bromo-3 (E) -octen-5-yne system by the hydrogen bromide-zinc bromide system. The bromide was reacted with magnesium compound of 4-tetrahydropyranyloxy-butyl chloride in the presence of copper salts to give the tetrahydropyranyl ether of 7 (E) -dodecen-9-yn-1-ol which was acid-catalyzed by 7 (E) -dodecen-9-yn-1. they are hydrolyzed. The latter was reduced to 7 (E), 9 (Z) -dodecadien-1-ol by catalytic hydrogenation and then acylated to the target compound.
In another process, C. Descoins et al. Used 1-hepten-4-in-3-ol as starting material. The compound was reacted with triethyl orthoacetate to give ethyl [4 (E) -one-6-in-one], which was reduced to 4 (E) -one-6-in-one by reduction with lithium aluminum hydride. The alcohol was tosylated and the tosylate was converted to l-bromo-4 (E) -one-6 with lithium bromide. The bromine compound was coupled with the magnesium compound of 1- [1- (ethoxy) ethoxy] -2-bromoethane. The resulting 7 (E) -none-6-in-1-ol was acylated with acetic anhydride and the acetate was reduced by dialkyl borane to give the target compound.
C. Descoins és munkatársainak fenti két eljárása költséges és nehezen hozzáférhető kiindulási anyagból és reaktánsokból állítja elő a kívánt vegyületet, így nem gazdaságos. The above two processes by C. Descoins and co-workers produce the desired compound from expensive and hardly available starting materials and reactants and are thus uneconomic.
More recently, a process for the preparation of 7 (E), 9 (Z) -dodecadienyl acetate has been described by G. Cassani et al., Tetrahedron L. 1980, 3497. Starting from 2 (E), 4 (Z) -heptadiene-1-acyl, acylated with 5 acetic acid chloride, the acetate was coupled with the magnesium compounds of 1-hydroxy-5-chloropentane tetrahydropyranyl ether in dilithium tetrachloro-cuprate. . The tetrahydropyranyl ether of 7 (E), 9 (Z) -dodecadien-1-ol formed in the reaction was converted to 7 (E), 9 (Z) -dodecadienyl acetate with 10 acetic acid / acetic acid. Synthesis is not advantageous due to the very expensive starting material.

C) 7(E),9(Z)-Dodekadienol előállítása (I,Ri =C2H5,R2=H)C) Preparation of 7 (E), 9 (Z) -Dodecadienol (I, R 1 = C 2 H 5 , R 2 = H)
86,4 g (0,2 mól) trifenil-propil-foszfónium jodid (VII, R!=C2H5) 900 ml száraz benzollal készült szuszpenziójához 36,6 g (0,2 mól) bisz(trimetil-szilil)-nátrium amidot adunk és az elegyet fél órán át szobahőmérsékleten keverjük, majd egy órán át refluxáltatjuk. Az oldatot szobahőmérsékletre hűtjük és hozzácsepegtetünk 15,6 g (0,11 mól) 9-hidroxi-2-nonenal 50 ml száraz benzolban készült oldatát. A reakeióelegyet 8 órán át keverjük, majd jégre öntjük. A szerves fázist elválasztjuk, a vizes fázist háromszor, összesen 600 ml pentánnal extraháljuk. A maradékot oszlopkromatografálással tisztítjuk (Kieselgel G, benzol : metanol 10 : 0,2).To a suspension of 86.4 g (0.2 mol) triphenylpropylphosphonium iodide (VII, R 1 = C 2 H 5) in 900 ml of dry benzene, 36.6 g (0.2 mol) of bis (trimethylsilyl) sodium amide was added and the mixture stirred for half an hour at room temperature and then refluxed for one hour. The solution was cooled to room temperature and a solution of 15.6 g (0.11 mol) of 9-hydroxy-2-nonenal in 50 ml of dry benzene was added dropwise. The reaction mixture was stirred for 8 hours and then poured onto ice. The organic phase is separated and the aqueous phase is extracted three times with a total of 600 ml of pentane. The combined organic phases were extracted three times with a total of 60 ml of 10% hydrochloric acid, washed with saturated sodium bicarbonate solution, water, dried over magnesium sulfate and the solvent was distilled off under reduced pressure. The residue was dissolved in pentane (400 mL), the precipitated triphenylphosphine oxide was filtered off and the mother liquor was concentrated in vacuo. The residue was purified by column chromatography (Kieselgel G, benzene: methanol 10: 0.2).
