Abstract

Η παρούσα εφεύρεση αναφέρεται στην ανάπτυξη αναλυτικών συσκευών βασισμένων σε ένα ή περισσότερα κύτταρα (κυτταρικών βιοαισθητήρων) των οποίων η επιφάνεια έχει τροποποιηθεί με την τεχνητή εισαγωγή μορίων κατά τέτοιο τρόπο ώστε να αντιδρούν εκλεκτικά με τις ουσίες υπό ανίχνευση ("μιμητικά κύτταρα"). Αυτά τα μόρια-υποδοχείς μπορεί να είναι πρωτεϊνες( όπως ένζυμα, αντιγόνα ή αντισώματα), νουκλεϊκάοξέα, υδατάνθρακες, λιπίδια ή να ανήκουν σε οποιαδήποτε άλλη χημική ομάδα με ικανότητα εκλεκτικής αλληλεπίδρασης με ουσίες-στόχους ("αναλύτες") οι οποίες είναι επιθυμητό να ανιχνευθούν σε ένα ή περισσότερα δείγματα. Η εισαγωγή των μορίων αυτών στην επιφάνεια των κυττάρων μπορεί να πραγματοποιηθεί μέσω ηλακτροεισαγωγής ή άλλης κατάλληλης μεθόδου. Διαφορετικές κατηγορίες μορίων μπορούν να εισαχθούνστη μεμβράνη του ίδιου κυττάρου, ιδιαίτερα αν αυτό εξυπηρετεί την εκλεκτικότητα αλληλεπίδρασης με τουςυπό προσδιορισμό παράγοντες. Η μέθοδος περιλαμβάνει τη χρήση κατάλληλου βιοαισθητήρα ο οποίος περιλαμβάνει το τροποποιημένο κυτταρικό υλικό ελεύθερο ή ακινητοποιημένο μέσα σε πλέγμα ή πάνω σε υπόστρωμα από κατάλληλο υλικό, έτσι ώστε να εξασφαλίζεταιη μέτρηση της εκλεκτικής αντίδρασης με τον υπόπροσδιορισμό παράγοντα. Η μέτρηση της αντίδρασης μπορεί να πραγματοποιηθεί με οποιαδήποτε κατάλληλη μέθοδο αναφερόμενη σε φυσικοχημική ιδιότητα του αισθητήρα, όπως τη μέτρηση της μεταβολής του δυναμικού ή διαφόρων οπτικών ιδιοτήτων (όπως φθορισμός, φωταύγεια, χημειοφωταύγεια ή ηλεκτροχημειοφωταύγεια). Επομένως είναι δυνατός ο προσδιορισμός ενός χημικού ή βιλογικού παράγοντα εφ'όσον είναι γνωστή ηκαμπύλη μεταβολής μίας ορισμένης φυσικοχημικής ιδιότητας του βιοαισθητήρα στις διάφορες συγκετρώσεις τουπαράγοντα αυτού και σε σχέσημε άλλους παρεμφερούςδομής ή δράσης παράγοντες.
