Abstract

51-Guanylic acid is prepared by adding to a solution containing guanosine and a phosphate donor comprising one or more aromatic phosphates and/or one or more nucleotides other than 51-guanylic acid an enzyme source comprising or obtained from cells of a species of microorganism selected from the genera Flavobacterium, Serratia, Staphylococcus and Pseudomonas, allowing biochemical phosphorylation of the guanosine to take place while maintaining the pH from 3,5 to 4,0 and thereafter recovering the accumulated 51-guanylic acid. Suitable aromatic phosphates include nitrophenyl phosphates, phenyl phosphate, benzyl phosphate and their salts. Copper and/or zinc ions are preferably present. The product may be recovered by separating the cells, extracting liberated phenol with an organic solvent, absorbing the guanylic acid on an ion-exchange resin, eluting with a mixture of HCl and NaCl, concentrating the eluate and adding ethanol to precipitate sodium 51-guanylate. The guanosine starting material may be derived from a fermentation process and may contain other nucleotides which are converted to their nucleotides such as 51-inosinic and 51-adenylic acids as byproducts.
