[Also in Group V] Substantially dry spherical pharmaceutical beads are made by directing preferably under pressure and through an orifice, a fine continuous stream of a composition comprising a sol of a congealable material e.g. gelatin, zein or carbowax and a pharmaceutical material, the temperature of said composition being above the congealing temperature of the sol, into a body of an immiscible liquid for said composition at a temperature below the congealing temperature of the sol, the body of the immiscible liquid moving transversely relative to the direction of movement of the stream of the composition and the depth and spread of the body of immiscible liquid being such that successive spherical beads are formed of substantially uniform diameter without interference of one bead with the other. An apparatus for making said beads comprises a reservoir 10 of congealable material containing said pharmaceutical material connected to a chamber 12. A plurality of orifices 14 are provided in chamber 12. Air under pressure in chamber 12 forces the sol through the orifices 14 into the immiscible liquid provided by a pump 16 from a supply 17 said liquid being directed over a trough 18 in a layer below the orifices, said layer being of a sufficient depth to prevent the beads formed from striking the trough. Heating coil 19 maintains the sol above its congealing temperature in reservoir 12. Cooling means 20 maintains the immiscible liquid below the congealing temperature of the sol. The beads formed range in size from 10-80 mesh U.S. standard and after formation are washed in a solvent e.g. naphtha or carbon tetrachloride and dried. The sol may also contain plasticizing agents e.g. sorbitol and substances reducing solubility e.g. for a gelatin sol, aldehydes such as formaldehyde, vanellin benzaldehyde or phenol. The beads may also be tanned. Specified pharmaceutical materials are D-amphetamine sulphate, Vitamin B12, Riboflavin, Thiamine hydrochloride, sulphaguanadine, D - amphetamine. Other constituents of the sol include isopropyl alcohol and flavouring agents. The spherical beads formed may be mixed with excipients e.g. lactose and magnesium stearate and compressed into tablets. The beads may also be coated with carbowax, shellac starch paste, glucose or acacia dried and compressed or may be enteric coated.
