Abstract

901,673. Cultivating animal cells. NATIONAL RESEARCH DEVELOPMENT CORPORATION. May 19, 1959 [May 20, 1958], No. 16237/58. Class 81 (1). Animal cells, e.g. transformed or malignant cells from mammalean tissue such as lung, kidney, spleen, liver and testes, are cultured by suspending them in a liquid culture medium containing dissolved oxygen at a concentration varied to control the cell growth rate, cellcontaining medium being withdrawn and fresh medium being supplied at intervals to maintain the volume of medium substantially constant. The medium is agitated under an atmosphere containing from 5 to 25%, by volume, of oxygen and 0 to 10% carbon dioxide, the latter maintaining the pH at between 6.8 and 7.2, the preferred level, and being varied according to colorimetric readings on the medium which may contain phenol red. The process is preferably effected at 37 ‹ C. at a cell concentration of from 2 to 20 x 10<SP>5</SP> cells per millilitre of medium, a turbidimeter reactive to the cell concentration being used to control the oxygen concentration. A suitable culture medium comprises 0.5 to 2% by weight of glucose and/or fructose and/or galactose together with nitrogen sources, e.g. yeast extract, lactalbumin hydrolysate, L and DL methionine, L arginine and glutamic acid, with ammonium chloride and L-glutamine, vitamins, growth promoters, antibiotics, the medium being made isotonic with 0.9% aqueous sodium chloride solution. In the apparatus, Fig. 1, a stirred culture vessel 1 is supplied through a solenoid-operated valve 17a with culture medium from a vessel 16, cell-containing medium siphoning via a tube 2 to a cooled stirred storage vessel 5, all three vessels being supplied with manually-controlled amounts of oxygen, nitrogen and carbon dioxide from cylinders 15, 15<SP>1</SP>, 15<SP>11</SP>. Valve 17a is responsive to a time switch 17 controlled by a control box 18 connected to a recording device 18a and also to a turbidimeter comprising a light source 18b and a photo-cell 18c. An apparatus is also described, Figs. 2 and 3 (not shown), comprising several culture vessels, one above the other, in a single housing having a central common overflow tube serving each vessel.
