Abstract
Non-gelatinous protein hydrolysate suitable for parenteral use is lyophilized in aqueous solution containing, in addition to the protein hydrolysate, a gelatin hydrolysate having a gel point of 15 to 20 DEG C. in 26 to 35% aqueous solution and being present in a ratio of 1 to 2 parts of gelatin hydrolysate to 9 parts of the non-gelatinous protein hydrolysate; it is stated that the foaming problem normally encountered does not become apparent to any appreciable extent when the partially hydrolysed gelatin is included. The gelatin hydrolysate may be made by hydrolysing an aqueous solution of gelatin for three hours at approximately 250 DEG F. and 15 lbs. per sq. inch pressure. According to an example, to 1,470 ml. of 23% casein hydrolysate in which 69% of the total nitrogen was in the form of a -amino nitrogen, there was added 188 ml. of hydrolysed gelatin prepared as described above and containing 32% solids. Vitamins &c., dextrose and sodium ethyl mercurithiosalicylate were added and the total volume of the solution was brought up to 3,08 litres with warm pyrogenfree distilled water. After filtering, 77 ml. portions of the solution filled into 250 ml. brown-glass bottles were frozen at -60 DEG F. and then placed in a lyophiliser, the pressure being decreased to 50-150 microms. The bottles were finally removed from the lyophiliser and aseptically stoppered. The product, which contained less than 5% of moisture, reconstituted readily with pyrogen-free distilled water. Other examples are concerned with lyophilising bovine plasma and fibrin hydrolysates.
